Simultaneous Quantitative Analysis of Major Constituent of Ethanol Extract from Leaves of Helianthus annuus L. (향일규 잎의 에탄올 추출물에 대한 주요 성분 동시 정량분석)

  • Helianthus annuus L. has been reported with various pharmacological activities such as antibacterial, antidiabetic, and antioxidant effects. According to recent studies, H. annuus L. is known to contain components such as phenolic compounds, flavonoids, alkaloids, sesquiterpenoids, and lignans. The seeds of H. annuus L. have been reported to contain chlorogenic acid and di-O-caffeoylquinic acid as major components. However, studies on the main components and content of leaves of H. annuus L. are still incomplete. Therefore, in this study, the contents of four major components of H. annuus L. were evaluated by simultaneous quantitative analysis with high performance liquid chromatography (HPLC)-diode array detector (DAD). The isolated four compounds Caffeoylquinic acid(CQA), 3,4-dicaffeoylquinic acid(3,4-DCQA), 3,5-dicaffeoylquinic acid(3,5-DCQA) and 4,5-dicaffeoylquinic acid(4,5-DCQA) were shown in a large linearity with a correlation coefficient (R2) of 0.99. In addition, as a result of intra-inter day analysis of four major compounds by the analysis method of this study, the accuracy of 88.46% or more and less than 112.85% and excellent precision of less than 3% were shown, the content analysis showed CQA (0.383±0.018 mg/g), 3,4-DCQA (0.282±0.017 mg/g), 3,5-DCQA (1.109±0.068 mg/g), and 4,5-DCQA (0.673±0.020 mg/g).

Comparison of the metabolic profile of the mycelia and fruiting bodies of artificially cultured Cordyceps militaris

  • Cordyceps militaris, a well-known traditional Chinese medicine, has multiple health-promoting effects. It is used as a herbal remedy and health food in Asian countries. Cultured mycelia are often used as a substitute for natural C. militaris. In the present study, the mycelia and fruiting bodies of artificially cultured C. militaris were analyzed using a metabolomics approach. The protein and crude fat contents of the mycelia were substantially higher than those of the fruiting bodies. The top three abundant amino acids in the mycelia were proline (3.9 g/100 g), aspartic acid (2.9 g/100 g), and glutamic acid (2.7 g/100 g). The carbohydrate content was similar in the fruiting bodies and mycelia. Analysis revealed that both the fruiting bodies and mycelia are rich in phenolic compounds and exhibit antioxidant activity. Further, six metabolites were significantly different between the mycelia and fruiting bodies. The levels of Ca, glucose, Mg, and Se were higher in the mycelia than in the fruiting bodies. In contrast, mannitol and Zn were more abundant in the fruiting bodies. The current study provides a comprehensive metabolic profile of the mycelia and fruiting bodies of artificially cultured C. militaris. Such an exercise is potentially important for understanding the metabolism of C. militaris and facilitating the use of cultured mycelia as a supplement to C. militaris fruiting bodies in traditional Chinese medicine.

Analysis of the current status of quantitative literature evidence for the prescription of 56 herbal medicines covered by health insurance (건강보험 급여 한약제제 56종 처방의 계량적 문헌 근거 현황 분석)

  • Objectives: The purpose of this study is to analyze the current state of quantitative literature evidence for the prescription of 56 herbal medicines covered by health insurance that have been studied in Korea for the past 30 years, to evaluate the reliability of the evidence, and to find out the research direction of herbal medicine prescription in the future. Methods: 56 kinds of herbal medicine prescriptions were searched in domestic literature search databases OASIS, DBpia, and overseas PubMed, classified into chemistry, toxicity, cells, animals, clinical cases, and clinical trial studies, and built into an EBM pyramid structure. Results: When classified according to research contents, there were 61 cases (7.5%) of physicochemical analysis to identify constituent substances, 80 cases (9.8%) of toxicity evaluation, and 672 cases (82.7%) of efficacy evaluation. The efficacy evidence was classified according to the evidence-based medical pyramid structure: 196 cell trials (29.1%), 372 animal trials (55.4%), 89 case and case reporting series (13.3%), 7 comparative case studies (1.1%), and 8 randomized control clinical trials (1.2%). In the pyramid composition, the basis for the validity of 56 kinds of herbal medicines prescribed was 568 cases (84.5%) in cell and animal units, which could not be said to be highly reliable. There was no relationship between the ranking of quantitative literature evidence for herbal medicine prescriptions and the ranking of salary administration. Conclusions: In an era that continues to require scientific evidence for herbal medicine, traditional herbal medicine should secure the basis for safety validity even for the 10th most frequent prescription among 56 herbal medicine prescriptions for consumers. In particular, traditional herbal medicine should increase the quantitative and qualitative level of case reports on related herbal medicine prescriptions, focusing on each clinical society, and move toward comparative case studies and randomized clinical trial so that traditional herbal medicine is positioned as Evidence-based medicine.

Effects of organic amendments on lettuce (Lactuca sativa L.) growth and soil chemical properties in acidic and non-acidic soils

  • Soil acidification challenges global food security by adversely influences soil fertility and agricultural productivity. Carbonized agricultural residues present a sustainable and ecofriendly way to recycle agricultural waste and mitigate soil acidification. We evaluated the effects of organic amendments on lettuce growth and soil chemical properties in two soils with different pH levels. Carbonized rice husk was produced at 600℃ for 30 min and rice husk was treated at 1% (w·w-1). Carbonized rice husk increased soil pH, electrical conductivity, total carbon content, and nitrogen content compared with untreated and rice husk treatments. Furthermore, this study found that lettuce growth positively correlated with soil pH, with increasing soil pH up to pH 6.34 resulting in improved lettuce growth parameters. Statistical correlation analysis also supported the relationship between soil pH and lettuce growth parameters. The study findings showed that the use of carbonized rice husk increased the constituent elements of lettuce, such as carbon, nitrogen, and phosphate content. The potassium content of lettuce followed a similar trend; however, was higher in acidic soil than that in non-acidic soil. Therefore, improving the pH of acidic soil is essential to enhance agricultural productivity. It is considered advantageous to use agricultural residues following pyrolysis to improve soil pH and agricultural productivity.

Nondestructive Measurement of Chemical Compositions in Polished Rice and Brown Rice using NIR Spectra of Hulled Rice acquired in Transmittance and Reflectance Modes (정조 상태에서 투과법과 반사법을 이용한 백미 및 현미 성분의 비파괴 측정)

  • The purpose of this study is to measure fundamental data required for the prediction of rice quality and to develop regression models to predict protein, amylose, moisture and fatty acid contents, and Toyo taste meter value (TTMV) of brown and polished rice from hulled rice NIR spectra. NIR spectra of hulled rice measured in transmittance mode (850-1050 nm) and in reflectance mode (400-2500 nm) were used to predicted chemical compositions of brown rice and polished rice. For most chemicals, the transmittance spectra could provide better calibration results than the reflectance ones. Beside the Toyo taste meter value (TTMV), the hulled rice spectra could predict chemical contents with the determination coefficients higher than 0.8. Spectra of hulled rice measured in transmittance mode could be used for the prediction of chemical compositions in brown rice and polished rice precisely. However, taste value of polished rice was a constituent that was hardly to be predicted.

Variation of Glucosinolate Contents among Domestic Broccoli (Brassica oleracea L. var. italica) Accessions (국내 브로콜리(Brassica oleracea L. var. italica) 유전자원 내 Glucosinolate 함량 변이)

  • A total of 95 broccoli (Brassica oleracea L. var. italica) accessions were evaluated for the identification of desulfo-glucosinolates and their content variation in the flower head using ultra performance liquid chromatography, to select the potentially functional broccoli breeding lines. The six individual desulfo-glucosinolates, including progoitrin, glucoraphanin, sinigrin, gluconapin, glucobrassicanapin, and glucobrassicin, were commonly identified, based on the chromatogram peak comparison with those of the nine individual glucosinolate standards. The total glucosinolate contents varied from 4.2 to $29.0{\mu}mol{\cdot}g^{-1}$ DW and the glucoraphanin (1.6 to $13.9{\mu}mol{\cdot}g^{-1}$ DW) was confirmed as a major constituent in the total glucosinolate profile among the six identified individual glucosinolate species, whereas the progoitrin, which was only detected in 13 accessions, showed accession-specific variation and negative correlation with glucoraphanin content. It was also revealed that the four major glucosinolates, such as glucobrassicanapin, glucoraphanin, glucobrassicin, and gluconapin, affected major content variation and showed higher positive inter-correlation. These results might be used for the selection of potential breeding materials as functional broccoli germplasm through the further evaluation on the stability and reproducibility of glucosinolate profile depending on environmental factors or cultural managements using the selected accessions.

A Survey on Middle School Teachers' Perception of Character Education (인성교육에 대한 중학교 교사들의 인식 조사)

  • The purpose of this study is to analyze the perception of middle school teachers of character education and to suggest implications for middle school character education. A total of 161 middle school teachers in Korea were surveyed and their responses were analyzed by frequency analysis and descriptive statistical analysis on the approach to character education, problem of character education, general view on character education, constituent of character education program, and teaching and learning method, First, character education was intended to be carried out through life instruction and subject education. Second, the problem of character education is that it is not carried out properly because senior school is given priority. Third, the overall view of character education is becoming more and more problematic for the personality development of students. Fourth, character components to be taught in character education program are consideration, manners, self-control, and responsibility. Fifth, the teaching and learning methods for character education were ranked in order of reflection, mutual learning, experiential activities, and student centering. Sixth, the proper period of the character education program should be continuous throughout school life with consent for continuous and repetitive education. Based on this, it is suggested that it is more important to consider how to organize character education for guidance throughout life and for curriculum than to develop it as one time program.

Effect of 35% Hydrogen Peroxide with Dicalcium Phosphate Dihydrate on the Tooth Whitening and Microhardness (35% 과산화수소에 제2인산칼슘를 함유한 치아미백제가 치아의 색과 경도에 미치는 영향)

  • The purpose of this study was to evaluate tooth whitening and microhardness after treatments with tooth bleaching agents containing dicalcium phosphate dihydrate (DCPD) and 35% hydrogen peroxide (HP) which were used in-office bleaching. Thirty enamel specimens were obtained from human premolars and randomly divided into 3 groups(n=10). Tooth bleaching agents were prepared with DCPD (0 g for controls, 0.1 g and 1 g for experimental groups) and HP solution (35% HP). All groups were applied to enamel surfaces for 60 min for 1 day. The pH of each tooth bleaching agent was measured. Tooth color, microhardness of enamel surfaces were also measured. The tooth bleaching agents containing DCPD showed a significant increase in pH compared to the ones without DCPD(p<0.05). Paired t-tests showed significant difference in color values of enamel before and after bleaching in all the groups(p<0.05). As a result, changes in color, containing DCPD group does not contain a statistically significant difference between groups was observed.(p>0.05). In all groups, tooth hardness after bleaching showed a significant decrease in microhardness (p<0.05). However, the DCPD concentration increased in the bleaching, microhardness values slightly decreased. Based on the above results, tooth bleaching agents containing DCPD and 35%HP were equally effective. Due to increases in pH and effective reduction of tooth surface decalcification, the surface characteristics are exposed to a reduced degree of negative effects, resulting in fewer constituent enamel alterations. Thus, commercial availability of the constituents of tooth whitening materials can be achieved.

Comparison of Food Components in Various Parts of White Muscle from Cooked Skipjack Tuna Katsuwonus pelamis as a Source of Diet Foods (다이어트 식품 소재로서 자숙 가다랑어(Katsuwonus pelamis) 백색육의 부위별 식품성분 특성)

  • This study evaluated the possible use of white muscle from cooked skipjack tuna as a constituent of diet foods. White muscles from the belly and dorsal area of cooked skipjack tuna were identified as anterior, median, and posterior. The skipjack tuna white muscle contained more moisture and ash (except for part I in both the belly and dorsal muscles) than chicken muscle, while it had less crude protein and crude lipid (except for part II in belly muscle). The yield was the highest in part I of both the dorsal and belly parts among the various parts of white muscles. The skipjack tuna white muscle contained 14-18% fewer calories than chicken breast muscle. Part I from both the belly and dorsal muscles had higher total amino acid contents than the other parts, but lower contents than chicken breast muscle. White muscle of skipjack tuna was rich in minerals, such as phosphorus, iron, and zinc. The total free amino acid content of part I in the belly and dorsal muscles was 1,152.1 and 1,215.7 mg/100 g, respectively, and was 1.7-1.8 times higher than in chicken breast muscle. The major amino acids in the white muscles from skipjack tuna were taurine, histidine, anserine, and carnosine. Based on these results, if it is possible to mask the fish odor, all parts of the white muscle from skipjack tuna could be used as constituents of diet foods.

Lipid Contents and Fatty Acid Composition of Three Lipid Classes in Korean Pinenut (잣지질 성분의 분획정량 및 각 획분의 지방산 조성)

  • Total lipids (TL) from Korean pinenut (Pinuskoraiensis S & Z) were extracted, purified and fractionated into three lipid classes (neutral lipid: NL, glycolipid; GL, phospholipid; PL). Lipid contents(constituent components) and fatty acid composition of three lipid classes were determined by thin layer chromatography and gas chromatography. TL ranged from 69.0% to 69.8% in fresh pinenut and consisted of 95.9% to 96.7% NL, 3.2% to2.5% GL and 0.9% to 0.8% PL. In the NL, triglycerides were predominant (80.8%) with the smaller amounts of sterol, diglycerides, free fatty acids, sterol esters and hydrocarbons. Monogalactosyl diglycerides and esterified steryl glycosides (23.5%) were the major components of GL, but cerebrosides, steryl glycosides and digalactosyl diglycerides were also found as minor components. Of the PL, phosphatidyl choline (40.2%) and phosphatidyl ethanolamine (19.4%) were the major components, comprising over 60% of this class. Phosphatidyl inositol, lysophosphatidyl choline were also present in the PL. The major fatty acids in the NL were linoleic acid (48.6%), oleic acid (28.8%) and arachidic acid(14.4%), The fatty acid composition in the GL was similar to the pattern in the NL, but PL contained a higher percentage of palmitic acid (17.7%) and stearic acid (6.0%) than other lipid classes.
