In vitro Organogenesis and Propagation of Heloniopsis orientalis Thunb

  • Heloniopsis orientalis (Liliaceae) is an important horticultural crop native to Korea. Under natural conditions, germination is poor and plant growth is delayed. Therefore, we have developed a vegetative propagation method to produce plants with vigorous growth characteristics via tissue culture. The regenerated shoots were then initiated directly from leaf explants on an MS medium containing either 0.5 to 2.0 mg/L 2,4-D or 1.0 to 3.0 mg/L BA. Healthy plantlets with adventitious roots were formed on the medium supplemented with 1.0 mg/L BA (81%). BA triggered callus initiation without caulogenesis or rhizogenesis, and callus formation was better on the half-strength MS medium than on the full-strength medium. This in vitro propagation protocol will be useful for conservation, as well as for mass propagation.

  • The production of spring wheat, the major crop in Mongolia, is accounting for 98% of the cultivated area. Collection, conservation and utilization of wheat germplasm resources play an important role in wheat breeding and production in Mongolia. Understanding genetic variability in the existing genebank accessions is important for collection and conservation of wheat germplasms. To determine the genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with 3 to 5 alleles per locus and a mean genetic diversity value of 5.66. The average genetic diversity index was 0.68, with values ranging from 0.37 to 0.80. The 200 Mongolian wheat accessions were divided into two subgroups based on STRUCTURE, un-rooted NJ cluster and principal coordinate analyses. The results from this study will provide important information for future wheat germplasm conservation and improvement programs with Mongolian genebank.

  • Kelp grouper ($Epinephelus$ $bruneus$ Bloch 1793) is a commercially important fish in Korea. In recent years, the catch of kelp grouper in the coastal waters of Korea has significantly declined. Despite its importance, little is known about its genetic diversity and conservation efforts are hampered. In this study, we isolated and characterized 12 microsatellite loci using an enrichment method based on magnetic/biotin capture of microsatellite sequences from a size-selected genomic library. All loci were readily amplified and contained TG/CA denucleotide repeats. To characterize each locus, 30 individuals from a natural E. bruneus population in the coastal waters of Jeju Island, Korea, were genotyped. All loci except three, KEm118, KEm154, and KEm219, were polymorphic, with an average of 8.1 alleles per locus (range 2-18). The mean observed and expected heterozygosities were 0.47 (range 0.19-1.00) and 0.61 (range 0.29-0.92), respectively. A significant deviation from Hardy-Weinberg equilibrium was observed at three loci (KEm134, KEm184, and KEm283). These findings will be useful for effective monitoring and management of genetic variation of kelp grouper as well as for the implementation of a fisheries conservation program.

  • Visual conservation center is a facility for the preservation of visual materials such as film and digital image data, and its construction project is currently being considered. This study evaluates the economic benefit of the project using CVM (Contingent Valuation Method), the main method of nonmarket evaluation in the both domestic and foreign literature. Survey data used in the analysis was collected in seven major metropolitan cities using person-to-person interviews and a logit model is used for the econometric estimation. The economic benefit measured by a household's average willing to pay (WTP) per year is shown to amount to 8,958 won.

Metallurgical Characteristics and Manufacturing Techniques of Ring-Pommel Swords Excavated from Ancient Tombs in Hadae, Ulsan (울산 하대고분 출토 민고리자루칼의 재질 특성과 환두부 제작 방법)

  • This study analyzed the microstructure of ring-pommel swords, excavated from Ancient Tombs, Hadae, Ulsan and examined their production technique, using non-destructive testing and a metallurgical method. The results confirmed that the five ring-pommel swords, unearthed in Ancient Tombs, Hadae, Ulsan, as identified by radiographic non-destructive testing, had been solely manufactured using iron, through forging based on the single-piece technique. Furthermore, these results were compared with previous studies, and the manufacturing techniques of single-piece ring-pommel swords were categorized into three types: pure iron - changing the shape, pure iron - changing the shape - carburization, and steel - changing the shape - quenching. The ring-pommels of four swords had around 0.7% of carbon content, which is as much as for eutectoid steel and higher than for other parts of these swords, such as the backs of their blades and handles. The weapon function of a small ring-pommel sword, under 60cm in length, was maximized by quenching focusing on its blade. Conversely, the martensite quenching structure was not observed in four ring-pommel swords shorter than 75cm. In other words, the same types of single-piece ring-pommel swords(late in 2C~early in 4C) were unearthed from Ancient Tombs, Hadae, and the group who has manufactured these swords is presumed to have limited their effectiveness, functionally depending on purposes, through an iron-making process and heat-treatment techniques.

  • This is the result of the study on the temporary fortifier using sublimation type Consolidate is Cyclododecane to prepare plan for reinforcement of the surface part that can be damaged during the dismantling of stone cultural heritages. To supplement the disadvantages of the existing reinforcement methods using intumescent urethane foam, Cyclododecane was diluted in solvent to reinforce the surface and inside desquamation, and after dismantling the framework, it sublimated by imposing heat of about $60^{\circ}C$. Such method can guarantee the strength needed for reinforcement of the damaged surface with outstanding reversibility of Cyclododecane being entirely sublimated. But, it shows big difference of effect according to the solvent, so it shall be diluted in petroleum ether or heated in a double boiler. Therefore, considering the working conditions at the site, it seems the most appropriate to use petroleum ether double boiler heating method for injection and filling of the desquamation part and temporary reinforcement processing with Cyclododecane diluted in petroleum ether for surface spraying.

  • To verifying the stability of Natural pigments for Dancheong which is available on the market was carried out as light resistance and gas corrosion test. In this particular case, we will confirm the characteristics of the only pigments except of Dancheong technique and influence of substance. Medium using for coloring are animal glue and synthetic resin that widely used for Dancheong in currently. Selected to artificial light source is a xenon arc lamp, contaminated gas is nitrogen dioxide gas in experiment. Degree of degradation of pigment were compared by measuring the degree of color change. The result of light resistance test, mineral pigments were good; color is green and blue type, soil pigments also good; color is yellow ocher, white clay, red clay etc. Gamboge, unghwang, cinnabar, red lead were confirmed not good. The result of gas corrosion test, most of the natural pigments were good condition except the red lead. Generally, animal glue samples had better than a synthetic resin samples.

  • Stupa of State Preceptor Jigwang at Beopcheonsa temple site, Wonju, was conserved and restored in 1957. It was necessary to assess the degree of damage to the used mortar and to provide objective data on the necessity of its removal. Therefore, the HCP(half-cell potential) measurement used in concrete fields was applied. Multi-regression analysis of HCP data, following the rapid corrosion test of the roofstone mortar used by the rebar, resulted in a high correlation of HCP values(0.86), depending on the amount of corrosion and cover depth used for the steel bar. As a result, high correlation values(0.86) for the coefficients of determination were derived. The showed that the measurement of the wet conditions -431 to -663 mV on the roofstone indicated a corrosion damage rate of 90% or more after removal and restoration.

  • The purpose of this study was to examine the effects of environmental education through the modern art appreciation on the children's environmental conservation knowledge, environmental conservation attitude, and art appreciation ability of 30, 5 year old children (15 experimental groups and 15 comparative groups) of D Daycare Center in Pyeongtaek, Gyeonggi-do, were selected for the study. In conslusion, environmental education through modern art appreciation had a positive effect on environmental conservation knowledge of children, environmental conservation attitude and art appreciation ability. Through this, this study would be to present teaching methods and basic materials that could improve children's environmental preservation knowledge, environmental preservation attitude and art appreciation ability.

  • During the conservation and maintenance of the birthplace center yard of President Asan Yoon Bosun, four porcelain enamel dishware were excavated from the central yard well. The glaze layer of excavated enamel was severely damaged; hence, the conservation process was done rapidly. In addition, scientific investigation and analysis were conducted to confirm the material properties of the glaze layer. It was confirmed that the outer surface was inverted and dried, while the inner surface was upright and fired during the glazing and drying process by measuring the film thickness. By examining the breakup phenomenon, the breaking up of the white enamel on the colored enamel was confirmed. This indicates that the colored glaze rose to the surface depending on the density of the colored glaze and white glaze. The investigation of the cross-section of the film confirmed that the lower layer formed according to the bonding properties with metal during the glazing process. Analysis of the constituents of the identified lower layer confirmed that there are differences between the specific components of the metal oxide of the lower layer and the surface color development of the upper layer.
