Systemic Amyloidosis in an African Penguin (Spheniscus Demersus) with Bumblefoot

  • A female, 14-year-old, African penguin (Spheniscus demersus) weighing 2.5 kg with the anorexia for one day and 22 months history of bumblefoot was found dead in aquarium. Grossly, severe bumblefoot in the central surface of the right foot, severe enlarged liver and kidneys with orange color were observed. Histopathologically, pinkish amorphous materials were accumulated in liver, spleen, kidney, and stomach. The pinkish materials in the liver, kidney, spleen, and stomach were confirmed as amyloid with typical orange color reactions using Congo red stain under the light microscope and with bright red color expressions using Congo red stain under the fluorescence microscope. Based on the typical gross and histopathologic findings and special staining, this case was confirmed as systemic amyloidosis. A long period time of moderate bumblefoot might be closely associated with the occurrence of systemic amyloidosis in the captive penguin.

Staining Response of KMnO4-Treated Animal Amyloid Proteins to Congo red (과망간산카리움 처리(處理)한 동물(動物) 아밀로이드 단백(蛋白)의 Congo red에 대한 염색반응(染色反應))

  • The characteristics of the amyloid proteins were histochemically investigated with the amyloidladen organs from bovine(15 cases), canine(1 case), feline(1 case), and artificially induced rabbit amyloidosis(1 case). The amyloid-laden organs from bovine, canine and experimental rabbit amyloidosis showed potassium-permanganate-senstive reaction, revealing complete disappearance for Congo red affinity, loss of brick-red colored fluorescence and green birefringence. From these findings, the amyloids from bovine, canine and experimentally induced rabbit amyloidosis were thought to be equivalent to amyloid protein A (AA protein). In the present feline case with amyloidosis, however, the amyloid proteins revealed potassium permanganate-resistant reaction, showing unchanged affinity for Congo red, and immunoglobulin was also deposited in the glomeruli of the kidney. From these findings, the amyloid proteins from feline case with amyloidosis were considered to be equivalent to amyloid light chain-related proteins (AL protein).

Biofilm Forming Ability and Production of Curli and Cellulose in Clinical Isolates of Enterobacteriaceae (생물막형성 장내세균의 Curli 및 Cellulose 세포외 바탕질 분석)

  • In this study, 22 clinical isolates of Enterobacteriaceae including Citrobacterfreundii (6 strains), Enterobacter cloacae (5 strains), Enterobacter aerogenes (3 strains), Serratia marcescens (7 strains) and Pantoea spp. (1 strain) were investigated for the biofilm forming ability and biosynthesis of curli and cellulose. Biofilm forming ability was the highest among the isolates of E. cloacae and the lowest among the isolates of E. aerogenes. The expression of the biofilm-forming extracellular matrix components, cellulose and curli fimbriae, was examined by Congo-red (CR) staining and calcofluor staining methods. PCR screening for the presence of curli gene (csgA) revealed that 4 strains of E. cloacae and 1 strain of C. freundii carried the csgA, showing a good correlation between the phenotypic detection of curli fimbriae by CR staining method and the genotypic detection of curli gene by PCR in E. cloacae.

  • Amyloidosis is a rare disease which is characterized by the deposition of a histochemically specific substance called amyloid in many tissue bodies, and causes various symptoms according to the organs involved. Amyloid is usually recognized by its staining reaction with Congo red stain. Primary pulmonary amyloidosis is very rare. Nodular pulmonary amyloidosis is an uncommon entity that usually manifests itself as an asymptomatic incidental finding on the chest roentgenogram and is misdiagnosed as lung cancer or pulmonary tuberculosis.

Molecular Cloning and Expression of Cellulase of Gene of Pseudomonas sp. in Escherichia coli (Pseudomonas sp.의 Cellulase 유전자의 대장균에의 클로닝 및 발현)

  • The genes for cellulases of Pseudomonas sp. LBC505 and CYC10, potent cellulase complex-producing strains, were cloned in Escherichia coli with pUC19. Recombinant plasmids pLCl and pLC2 were isolated from transformants producing cellulase by Congo red staining, and their genes cloned were 0.7 kb and 4.6 kb HindIII fragments, respectively. The inserts of pLCl and pLC2 were hybridized to chromosomal DNAs digested with HindIII from Pseudomona~ sp. LBC505 and CYC10, respectively. Immunodiffusion assays revealed that pLC1-and pLC2-encoded cellulase showed similarity with that of host strains. About 24% of cellulase activity was observed in the extracellular fraction of E. coli carrying pLC1, and its activity was higher about 1.4 times than that of LBC505. The enzymatic properties of pLC1 and pLC2 encoded cellulase were the same as those of cellulase from host strains. HPLC analysis and substrate specificity showed that cellulases were the same as those of cellulase from host strains. HPLC analysis and substrate specificity showed that cellulases cloned were endocellulase.

Characterization of ${\beta}$-1,4-Glucanase Activity of Bacillus licheniformis B1 in Chungkookjang (청국장 발효균주 Bacillus licheniformis B1의 ${\beta}$-1,4-glucanase 특성)

  • Fermented soybeans contain microorganisms, diverse enzymes, and bioactive compounds. Few studies on cellulase in Chungkookjang exist. Oligosaccharides play diverse roles of bioactivity. Through Congo red test and activity staining, it was confirmed that the enzyme solution contained cellulase. Optimal pH and temperature of the cellulase produced by Bacillus licheniformis B1 were 10 and $40^{\circ}C$, respectively. Through TLC analysis, it was demonstrated that the enzyme solution degraded carboxymethyl cellulose (CMC), whose main products contained dimer and trimer oligosaccharides. Cellulase activity of barley-Chungkookjnag fermented by the strain increased, compared with that of Chungkookjang. The cellulase was found to be a ${\beta}$-1,4-glucanase through the analysis of the cloned gene, showing polymorphism at 32 amino acid sites in the coding range of amino acid 10 and 460.

Secondary renal amyloidosis in a 13-year-old girl with bronchiectasis

  • A 13-year-old girl was diagnosed with non-cystic fibrosis (CF)-related multifocal bronchiectasis accompanied by nephrotic-range proteinuria of unknown cause. On renal biopsy, there were many segmental homogeneous deposits of amyloid tissue with positive Congo red staining in the glomeruli and interstitium. On electron microscopy, relatively straight, non-branching, randomly arranged amyloid fibrils were showed in the mesangium of the glomeruli. These fibrils were approximately 10 nm in diameter, compatible with secondary amyloidosis. Her level of serum amyloid A was remarkably elevated. To our knowledge, this girl is the first case of secondary renal amyloidosis induced by bronchiectasis in Korean children.

The Signal Sequence of Sporulation-Specific Glucoamylase Directs the Secretion of Bacterial Endo-1,4-β-D-Glucanase in Yeast (효모에서 포자형성 특이 글루코아밀라제의 분비서열에 의한 세균 endo-1,4-β-D-glucanase의 분비)

  • The sporulation-specific glucoamylase (SGA) of Saccharomyces diastaticus is known to be produced in the cytoplasm during sporulation. For the purpose of proving that SGA has secretory potential, we constructed a hybrid plasmid, pYESC25, containing the promoter and the putative signal sequence of the SGA fused in frame to the endo-1,4-${\beta}$-D-glucanase (CMCase) gene of Bacillus subtilis without its own signal sequence. The recipient yeast strain of S. diastaticus YIY345 was transformed with the hybrid plasmid. CMCase secretion from S. diastaticus harboring pYESC25 into culture medium was confirmed by the formation of yellowish halos around transformants after staining with Congo red on a CMC agar plate. The transformant culture was fractionated to the extracellular, periplasmic, and intracellular fraction, followed by the measurement of CMCase activity. About 63% and 13% enzyme activity were detected in the culture supernatant (extracellular fraction) and periplasmic fraction, respectively. Furthermore, ConA-Sepharose chromatography, native gel electrophoresis, and activity staining revealed that CMCase produced in yeast was glycosylated and its molecular weight was larger than that of the unglycosylated form from B. subtilis. Taking these findings together, SGA has the potential of secretion to culture medium, and the putative signal sequence of SGA can efficiently direct bacterial CMCase to the yeast secretion pathway.
