THE EFFECT OF PDGF-BB AND IGF-I COMBINATION ON THE HEALING OF ARTIFICIAL PERIAPICAL LESIONS IN BEAGLE DOGS (PDGF-BB와 IGF-I 혼합 투여가 비글견 인공 치근단 병소의 치유에 미치는 영향에 관한 연구)

  • It is difficult to treat the endodontic apical perforation successfully. In this study, we hypothesized that the application of PDGF-BB and IGF-I into periapical perforation site may accelerate periapical healing and lead to bone deposition. And the specificity of osteonectin in periapical healing was investigated. The experiments were performed on the upper and lower 51 premolar teeth of 4 beagle dogs. The pulp chamber of each tooth was opened and the dental plaque was inserted into the canal for developing the periapical lesion for 5 weeks. Then, the roots were artificially perforated at the apex with the number 4 profile of .06 taper. In each step, standard periapical radiographs were taken to compare the size of lesion each other. The radiographs were scanned and analyzed by image analysis system. The mean and standard deviation of periradicular radiolucency ratios were calculated in each group. ANOVA was used for comparison. 51 premolars were grouped into 3 groups; control group, calcium hydroxide-treated group and calcium hydroxide plus growth factors-treated group. In the control group, the apical perforations were not sealed and obturated with gutta-percha and ZOE sealer by lateral condensation technique. In the experimental groups, the apical perforation were sealed with calcium hydroxide and with/without $4{\mu}g$ of PDGF-BB & IGF-I in cellulose gel and obturated by lateral condensation technique. Fluorescent bone markers were used to measure new bone formation. Following 2, 4, 12 weeks after experiment the dogs were sacrificed and histologic sections were prepared. Each tooth block including periapical lesion was sectioned mesiodistally. One half of the sections were decalcified with 6% nitric acid and processed by standard paraffin embedding technique. The sections were stained by hematoxylin and eosin, and immunostained for osteonectin. Histomorphometrical measurement of neoformed bone was performed using a light microscope. And the other half of the sections were prepared by undecalcified preparation, and confocal laser scanning microscopic investigations were done.

Effects of NaOCl on Neuronal Excitability and Intracellular Calcium Concentration in Rat Spinal Substantia Gelatinosa Neurons

  • Recent studies indicate that reactive oxygen species (ROS) can act as modulators of neuronal activity, and are critically involved in persistent pain primarily through spinal mechanisms. In this study, we investigated the effects of NaOCl, a ROS donor, on neuronal excitability and the intracellular calcium concentration ($[Ca^{2+}]_i$) in spinal substantia gelatinosa (SG) neurons. In current clamp conditions, the application of NaOCl caused a membrane depolarization, which was inhibited by pretreatment with phenyl-N-tert-buthylnitrone (PBN), a ROS scavenger. The NaOCl-induced depolarization was not blocked however by pretreatment with dithiothreitol, a sulfhydryl-reducing agent. Confocal scanning laser microscopy was used to confirm whether NaOCl increases the intracellular ROS level. ROS-induced fluorescence intensity was found to be increased during perfusion of NaOCl after the loading of 2',7'-dichlorofluorescin diacetate ($H_2DCF$-DA). NaOCl-induced depolarization was not blocked by pretreatment with external $Ca^{2+}$ free solution or by the addition of nifedifine. However, when slices were pretreated with the $Ca^{2+}$ ATPase inhibitor thapsigargin, NaOCl failed to induce membrane depolarization. In a calcium imaging technique using the $Ca^{2+}$-sensitive fluorescence dye fura-2, the $[Ca^{2+}]_i$ was found to be increased by NaOCl. These results indicate that NaOCl activates the excitability of SG neurons via the modulation of the intracellular calcium concentration, and suggest that ROS induces nociception through a central sensitization.

Resonance May Elucidate New Bone Formation Induced by Low amplitude and High frequency Mechanical Stimuli (고주파 미세자극에 의한 뼈의 생성에 관한 모델링)

  • Bone is a self-assembly material. It is known that the low amplitude and high frequency mechanical stimulus, which is much less amplitude but higher frequency than those induced by the normal activity, can induce new bone formation. The vibrating resonance is employed to elucidate why new bone is formed by this kind of mechanical stimulus. For example, as 30 Hz and $5{\mu}{\epsilon}$ mechanical stimulus is applied at the wall of canaliculus (the tiny tube type pathway of bone fluid flow and the diameter of canaliculus is less than 200nm), the osteocytic cell membrane experiences $1,000{\mu}{\epsilon}$ strain due to the vibrating resonance. Two experiments will follow after this pilot study; (1) observing the MAPK pathway of osteocytes by using in-vitro cell culture and (2) visualizing the actin filament network in the osteocytes by using the imaging technique, such as confocal laser scanning microscope.

SURFACE ROUGHNESS OF EXPERIMENTAL COMPOSITE RESINS USING CONFOCAL LASER SCANNING MICROSCOPE (공초점 레이저 주사 현미경을 이용한 실험적 레진의 표면 조도에 대한 연구)

  • The purpose of this study was to evaluate the effect of a new resin monomer, filler size and polishing technique on the surface roughness of composite resin restorations using confocal laser scanning microscopy. By adding new methoxylated Bis-GMA (Bis-M-GMA, 2,2-bis[4-(2-methoxy-3-methacryloyloxy propoxy) phenyl] propane) having low viscosity, the content of TEGDMA might be decreased. Three experimental composite resins were made: EX1 (Bis-M-GMA/TEGDMA = 95/5 wt%, 40 nm nanofillers); EX2 (Bis-M-GMA/TEGDMA = 95/5 wt%, 20 nm nanofillers); EX3 (Bis-GMA/TEGDMA = 70/30 wt%, 40 nm nanofillers). Filtek Z250 was used as a reference. Nine specimens (6 mm in diameter and 2 mm in thickness) for each experimental composite resin and Filtek Z250 were fabricated in a teflon mold and assigned to three groups. In Mylar strip group, specimens were left undisturbed. In Sof-lex group, specimens were ground with #1000 SiC paper and polished with Sof-lex discs. In DiaPolisher group, specimens were ground with #1000 SiC paper and polished with DiaPolisher polishing points. The Ra (Average roughness), Rq (Root mean square roughness), Rv (Valley roughness), Rp (Peak roughness), Rc (2D roughness) and Sc (3D roughness) values were determined using confocal laser scanning microscopy. The data were statistically analyzed by Two-way ANOVA and Tukey multiple comparisons test (p = 0.05). The type of composite resin and polishing technique significantly affected the surface roughness of the composite resin restorations (p < 0.001). EX3 showed the smoothest surface compared to the other composite resins (p < 0.05). Mylar strip resulted in smoother surface than other polishing techniques (p < 0.05). Bis-M-GMA. a new resin monomer having low viscosity, might reduce the amount of diluent, but showed adverse effect on the surface roughness of composite resin restorations.

A Novel Method for 3D Surface and Solid Construction Analysis of Fabric Microstructure (직물 미세구조의 3차원 표면 및 솔리드 형성 방법)

  • In-depth knowledge of fabric microstructure is essential for understanding clothing comfort since it plays a significant role in heat and mass transfer between the human body and clothing. In this study, a novel method was employed for investigating 3D surfaces and solid construction characteristics of specific fabrics by using a reverse engineering technique. The surface construction data were obtained by a confocal laser scanning microscope and then manipulated by a 3D analysis program. Triangle mesh was used for connecting each 3D point, with clouds and fabric surface characteristics created by rendering techniques. For generating a 3D solid model, determinants of radius of curvature was used. According to the proposed method, actual surface expression of the real fabric was achieved successfully. The results from this methodology can be applied to the detailed analysis of clothing comfort that is highly influenced by the microstructure of the fabric.

SIMS Protein imaging with nanoparticle tagged antibody for simultaneous omic imaging

  • One of the major problems of biological ToF-SIMS imaging is the lack of protein and peptide imaging. Most of biological story telling is mianly based on proteins. The biological implication of lipid ToF-SIMS imaging would be much higher if protein imaging is provided together. Utilizing high secondary ion yields of metals, proteins can be ToF-SIMS imaged with nanoparticle tagged proteins. Nanoparticles such as Fe3O4, SiO2, PbS were used for imaing NeuN, MCH, Orexin A, ${\alpha}$ synucline, TH(Tryosine Hydroxylase) in mouse tissues with a spatial resolution of ${\sim}2{\mu}m$ using a TOF-SIMS. Lipids and neurotransmitters images obtained simultaneously with protein images were overlayed for more deeper understanding of neurobiology, which is not allowed by any other bioimaging technqiues. The protein images from TOF-SIMS were compared with confocal fluorescence microscopy and NanoSIMS images. A new sample preparation method for imaging single cell membranes in a tissue using the vibrotome technique to prepare a tissue slice without any fixation and freeze drying will be also presented briefly for Hippocampus and Hypothalamus tissues.

Conjugated Oligomers Combining Fluorene and Thiophene Units : Towards Supramolecular Electronics

  • Conjugated oligomers, used as models for fluorene-thiophene copolymers, are compared in terms of the microscopic morphology of thin deposits and the optical properties. The AFM images and the solid-state absorption and emission spectra are interpreted in line with the structural data, in terms of the assembly of the conjugated molecules. The compound with a terthiophene central unit and fluorene end-groups shows well-defined monolayer-by-monolayer assembly into micrometer-long strip-like structures, with a crystalline herringbone-type organization within the monolayers. Polarized confocal microscopy indicates a strong orientation of the crystalline domains within the stripes. In contrast, the compound with a terfluorene central unit and thiophene end groups forms no textured aggregates. The difference in behavior between the two compounds most probably originates from their different capability of forming densely-packed assemblies of ${\pi-pi}$ interacting molecules. These assemblies are used as active elements in organic field effect transistors designed by using soft lithography technique.

  • With the advent of CLSM in the end of 1980s, it has been applied to the field of pulp and paper science in various ways. This study showed the potentials of CLSM In analyzing a change of pulp fiber and paper properties before and after mechanical treatment. In particular, a quantification of internal fibrillation has been done using cross-sectional images of fibers and image analysis technique, then evaluated the effects of fiber wall delamination on fiber and paper properties. It showed that the delaminated fibers were closely associated to development of the interfiber bonding in a fiber network. The CLSM made it possible to investigate a density profile along the sheet thickness, which was created by some papermaking processes like pressing, drying and calendering. Through the attempt to observe the forming procedure of a fiber network during handsheet making, the CLSM images showed that the pressing stage was considered greatly to contribute to generation of interfiber bonding with removing a free water and partly a bound water between fibers. In addition, the CLSM could be used to illustrate not only a surface profile of paper showing the extent of smoothness or roughness, but also a density profile in a B-direction of the network. Finally it became evident that the CLSM could be used as an excellent tool to predict development in fiber and paper properties before and after mechanical treatment during papermaking processes.

  • A tunable pulsed dye laser was operated in a single longitudinal mode by injection locking with a cw ring dye laser. A doubled Nd:YAG laser was used as the pumping source. Phase sensitive detection technique was applied to maintain the required match between the master laser frequency and the slave resonator cavity length. The fluctuation of the center frequency of the pulsed laser was < 10 MHz, and the pulse duration (FWHM) was 6 ns. The linewidth measured by scanning confocal interferometer was 130 MHz. When pumped by 50 mJ of the doubled Nd:YAG laser, the output energy of the pulsed dye laser was 2 mJ and the peak power was 330 kW. 30 kW.

  • In precision laser microfabrication, focusing is essential to acquire good machining precision and uniform machining quality. If it does not perform, laser machining cannot be realized. So, confocal scanning method with high depth resolution is used for focus detection technique. This paper is concerned with a procedure for design, analysis and performance test of an autofocus fine actuating stage, which is composed of flexure-hinge type lever mechanism and piezoelectric actuator. Through series of analytical design, the stage is simplified as a rigid bodies(lever and main body) and springs(flexure hinges). The simplified model was applied to determine the dimension of flexure hinges and lever. After structural analysis confirmed design requirement, an actual stage was made and verified through an experiment on the static and dynamic characteristics(maximum stroke and 1st natural frequency). The fabricated stage was satisfied with the design requirement.
