Effects of 17β-estradiol, Interleukin-1β, and Human Chorionic Gonadotropin on Activity and mRNA Expression of Plasminogen Activators in Porcine Endometrial Cells

  • This study aimed to investigate changes in the activity and mRNA expression of plasminogen activators (PAs) induced by $17{\beta}$-estradiol ($E_2$), human chorionic gonadotropin (hCG), and interleukin-$1{\beta}$ ($IL-1{\beta}$) in porcine endometrial cells. Endometrial cells were isolated from the epithelium and cultured to 80% confluence. They were then treated for 24 h with $E_2$ (0.2, 2, 20, and 200 ng/mL), $IL-1{\beta}$ (0.1, 1, 10, and 100 ng/mL), and hCG (0.5, 1, 1.5 and 2 IU/mL). mRNA expressions of urokinase-type (uPA) and tissue-type (tPA) PAs were analyzed using reverse transcription PCR, and activities were measured using a PA activity assay. mRNA expressions of uPA and tPA increased with $E_2$ treatment; however, this was not significant. Similarly, treatment with hCG did not influence the mRNA expressions of PAs. Interestingly, treatment with 0.1 ng/mL $IL-1{\beta}$ significantly reduced the mRNA expression of uPA, but did not affect that of tPA. Treatment with 2, 20, and 200 ng/mL $E_2$ increased PA activity compared with the control group; treatment with 0.1 and 1 ng/mL $IL-1{\beta}$ significantly increased PA activity compared with the other $IL-1{\beta}$ treatment groups, whereas treatment with 10 and 100 ng/mL $IL-1{\beta}$ decreased. Treatment with 2 IU/mL hCG increased PA activity compared with the other treatment groups, although there were no significant differences between the hCG and control groups. In conclusion, the activity and mRNA expression of PAs were differently regulated by the hormone/cytokine and its concentration in porcine endometrial cells. Therefore, understanding PA regulatory mechanisms may help to improve the reproductive potential of domestic animals.

A Study on the Prediction of Turbidity near the Confluence of Banbyeoncheon by Using the KU-RLMS Model (KU-RLMS 모형을 이용한 반변천 합류부 탁도 예측에 관한 연구)

  • 댐 하류로 탁수를 선택적으로 배제하기 위해서는 방류 탁수가 하류에 미치는 영향을 정확히 예측할 수 있는 하천 탁도 예측 및 관리시스템 구축이 필요하다. 낙동강과 반변천의 합류부에서의 이차원적인 혼합에 관한 수치해석 결과는 완전혼합을 가정하는 일차원 수질모델링의 초기 입력자료에 사용됨으로써 낙동강 본류 전체구간의 탁도 모의결과의 정확성을 높이는 데 사용될 수 있다. 본 연구는 낙동강의 중상류에 위치한 반변천 합류부에 평면 이차원 비정상 수치모형인 KU-RLMS 모형을 적용하여 탁도 변화 특성을 규명할 목적으로 수행하였다. KU-RLMS 모형은 하천 및 저수지의 국부적인 수리, 수질, 유사이동 해석을 위해 개발된 평면 이차원 비정상 수치모형이다. 직사각형 격자를 사용하는 유한차분법의 단점을 보완하기 위해, 수심적분된 2차원 연속방정식, 운동량방정식, 이송확산방정식을 불규칙한 경계를 현실적으로 모사할 수 있는 직교곡선 좌표계로 변환한 방정식을 사용한다. 이 모형은 흐름, 농도, 지형변화를 조합하여 계산할 수 있는 모형으로서 점착성 및 비점착성 유사의 이동을 모의할 수 있다. 수치모형 적용을 위한 현황분석으로 안동 및 임하 조정지댐의 방류량, 안동 수위관측소의 수위, 법흥교 및 포진교 지점의 탁도 자료를 분석하였다. 이송확산모형의 보정을 위해, 안동대교 지점의 탁도 횡분포 측정 자료를 사용하여 확산계수에 대한 매개변수 추정 및 검증을 수행하였다. 또한, 안동조정지댐과 임하조정지댐의 방류량 및 방류탁도을 고려하여 수치모의조건을 결정하였으며, 각 조건에 대한 탁도 변화 특성을 분석하였다.된 주변국이 될 수밖에 없을 것이다. 21세기 문화산업에서 우리가 판단하게 될 디자인의 가치는 계몽의 원리에 대한 '역사성'과 '현재성'의 변증법에 달려있는 것이며, 새로운 철학, 새로운 문명, 새로운 세계를 열어가는 것이다.r$ (地理志)에는 추현리와 이미 외리를 언급하면서 상주의 자기제작의 위상을 짐작하는 기록이 언급되면서 전국의 상품의 절반을 담당하고 있었음을 알 수 있었다. $\ulcorner$경상도지리지$\lrcorner$(慶尙道地理志)에는 상주가 8곳으로 1/3의 자기 생산을 담당하고 있었다. $\ulcorner$경상도지리지$\lrcorner$(慶尙道地理志)에는 $\ulcorner$세종실록$\lrcorner$(世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와 동년대에 동일한 목적으로 찬술되었음을 알 수 있다. $\ulcorner$경상도실록지리지$\lrcorner$(慶尙道實錄地理志)에는 $\ulcorner$세종실록$\lrcorner$(世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와의 비교를 해보면 상 중 하품의 통합 9개소가 삭제되어 있고, $\ulcorner$동국여지승람$\lrcorner$(東國與地勝覽) 에서는 자기소와 도기

Rapid Induction of mRNA for Prostaglandin H Synthase in Ovine Meningeal Fibroblasts

  • We examined effects of interleukin $1{\alpha}$ ($IL1{\alpha}$) and phorbol 12, 13 dibutyrate (PDB), an activator of protein kinase C, on mRNA for Prostaglandin H synthase (PGHS) and prostanoid production in cultured ovine meningeal fibroblasts. Immuno- and morphologically-identified fibroblasts were derived from cerebral cortex and white matter from fetal lambs (approximately 120 days gestation) and grown to confluence on glass coverslips in 12 well plates. Levels of prostaglandin $F_{2{\alpha}}$ and the stable hydrolysis product of prostacyclin (i.e., $6-keto-PGF_{1{\alpha}}$) were determined using enzyme immunoassay. Relative amounts of mRNA were determined by in situ hybridization using ovine cDNA for PGHS1. $IL1{\alpha}$ (10 ng/ml) increased mRNA levels over baseline by $62{\pm}19%$ (p<0.05) at 60 min., $37{\pm}12%$ (NS) at 120 min., and $36{\pm}18%$ (NS) at 240 min (n=12). Levels of $6-keto-PGF_{1{\alpha}}$ were $148{\pm}18%$ pg/ml during baseline, $246{\pm}41%$ pg/ml at 60 min., $248{\pm}40%$ pg/ml at 120 min., and $259{\pm}62%$ pg/ml at 240 min (all p<0.05) (n=12). $PGF_{2{\alpha}}$ was increased although it wasn't statistically significant. However, $IL1{\alpha}$ decreased $PGE_2$ level significantly (all p<0.05). PDB $(10^{-6}M)$ increased mRNA levels over baseline by $25{\pm}6%$ after 30 min., $40{\pm}6%$ after 60 min., and $20{\pm}8%$ after 90 min. (n=9) (all p<0.05). Levels of $6-keto-PGF_{1{\alpha}}$ were $200{\pm}43%$ pg/ml during baseline, $202{\pm}43%$ pg/ml after 30 min. (NS), $268{\pm}58%$ pg/ml after 60 min. (p<0.05), and $296{\pm}60%$ pg/ml after 90 min. (p<0.05) (n=9). Levels of $PGF_{2{\alpha}}$ were $178{\pm}26%$ pg/ml during baseline, $300{\pm}30%$ pg/ml after 30 min., $299{\pm}35%$ pg/ml after 60 min., and $355{\pm}32%$ pg/ml after 90 min (all p<0.05) (n=6). Actinomycin-D (1 mg/ml) prevented increases in mRNA, $6-keto-PGF_{1{\alpha}}$, and $PGF_{2{\alpha}}$ at 60 min. for both $IL1{\alpha}$ and PDB. We conclude that cerebral fibroblasts are avid producers of prostanoids, and that enhanced production of PGHS is responsible for augmented $PGF_{2{\alpha}}$ and prostacyclin production in the presence of an activator of protein kinase C and for decreased $PGE_2$ and increased prostacyclin production in the presence of $IL1{\alpha}$.

Effects of irradiation on TGF-${\beta}_1$ mRNA expression and calcific nodule formation in MC3T3-E1 osteoblastic cell line (방사선조사가 MC3T3-E1 골모세포주의 TGF-${\beta}_1$ mRNA 발현과 석회화결절 형성에 미치는 영향)

  • Purpose : To investigate the effects of irradiation on transforming growth factor ${\beta}_1$ (TGF-${\beta}_1$) mRNA expression and calcific nodule formation in MC3T3-E1 osteoblastic cell line. Materials and Methods : Cells were cultured in alpha-minimum essential medium ($\alpha$-MEM) supplemented with 10% fetal bovine serum and antibiotics. When the cells reached the level of 70-80% confluence, culture media were changed with $\alpha$-MEM supplemented with 10% FBS, 5 mM $\beta$-glycerol phosphate, and $50\;{\mu}g/mL$ ascorbic acid. Thereafter the cells were irradiated with a single dose of 2, 4, 6, 8 Gy at a dose rate of 1.5 Gy/min. The expression pattern of TGF-${\beta}_1$ mRNA, calcium content and calcific nodule formation were examined on day 3, 7, 14, 21, 28, respectively, after the irradiation. Results : The amount of TGF-${\beta}_1$ mRNA expression decreased significantly on day 7 after irradiation of 4, 6, 8 Gy. It also decreased on day 14 after irradiation of 6, 8 Gy. and decreased on day 21 after irradiation of 8 Gy. The amount of calcium deposition decreased significantly on day 7 after irradiation of 4, 8 Gy (P < 0.01) and showed a decreased tendency on day 14, 21 after irradiation of 4, 6, 8 Gy. The number of calcific nodules was decreased on day 7 after irradiation of 4, 8 Gy. Conclusion: Irradiation with a single dose of 4, 6, 8 Gy influences negatively the bone formation at the molecular level by affecting the TGF-${\beta}_1$ mRNA expression that was associated with proliferation and the production of extracellular matrix in MC3T3-E1 osteoblastic cell line.

Soil Erosion and river-bed change of the Keum river basin using by GIS and RS (GIS와 RS를 이용한 금강유역 토양침식과 하상변화 연구)

  • Flooding hazard caused by natural and artificial environmental changes is closely associated with change in river bed configuration. This study is aimed at explaining a river-bed change related to soil erosion in the Keum river basin using GIS and RS. The USLE was used to compute soil erosion rate on the basis of GIS. River-bed profiles stretching from Kongju to Ippo were measured to construct a 3D-geomorphological map. The river-bed change was also detected by remote sensing images using Landsat TM during the period of 1982 to 2000 for the Keum river. The result shows that USLE indicates a mean soil erosion rate of $1.8\;kg/m^2/year$, and a net increase of a river-bed change at a rate of $+5\;cm/m^2$/year in the Kangkyeong area. The change in river-bed is interpreted to have been caused by soil erosion in the downstream of the Keum river basin. In addition river-bed change mainly occurred on the downstream of the confluence where tributaries and the main channel meet. Other possible river-bed change is caused by a removal of fluvial sand aggregates, which might have resulted in a net decrease of exposed area of sediment distribution between 1991 and 1995, while a construction of underwater structures, including a bridge, a reclamation of sand bars for rice fields and dikes, resulted in an increase of the exposed area of river-bed due to sediment accumulation.

Analysis of Inundation Characteristics for EAP of Highway in Urban Stream - Dongbu Highway in Jungrang Stream - (도시하천도로의 EAP수립을 위한 침수특성분석 - 중랑천 동부간선도로를 중심으로 -)

  • An hydraulic and hydrologic analysis procedure was proposed to reduce the inundation damage of highway in urban stream, that could contribute the EAP and Traffic control planning of Dongbu highway in the Jungrang stream basin which is one of the representative urban area in Korea. We performed the HEC-HMS runoff analysis, and the UNET unsteady flow modeling to decide the inundation reaches and their characteristics. The high inundation risk areas were of Emoon railway bridge and the Wollueng bridge, which are inundated in the case of 10 year and 20 year frequency flood respectively. We also analyze the inundation characteristics under the various conditions of the accumulation rainfall and the duration. Flood elevation at the Wolgye-1 bridge exceed over Risk Flood Water Level(EL.17.84 m) when the accumulation rainfall is over 250 mm and shorter duration than 7 hr. When neglecting backwater effect from the Han river, inundation risk are highly at the reach C2(Wolgye-1 br. ${\sim}$Jungrang br., left bank), C1(Wolgye-1 br. ${\sim}$Jungrang br., right bank), D(Jungrang br. ${\sim}$Gunja br.) in order, but when consider the effect, the inundation risk are higher than the others at the reach D2(Jungrang br. ${\sim}$Gunja br., left bank) and E(Gunja br. ${\sim}$Yongbi br.), which are located downstream near confluence.

Effects of 2-deoxy-D-glucose and quercetin on the gene expression of bone sialoprotein and osteocalcin during the differentiation in irradiated MC3T3-E1 osteoblastic cells (2-deoxy-D-glucose와 quercetin이 방사선조사 MC3T3-E1 골모세포주의 분화시 bone sialoprotein과 osteocalcin 유전자의 발현에 미치는 영향)

  • Purpose : To investigate the effects of 2-deoxy-D-glucose (2-DG) and quercetin (QCT) on gene expression of bone sialoprotein (BSP) and osteocalcin (OC) during the differentiation in irradiated MC3T3-E1 osteoblastic cells. Materials and Methods : When MC3T3-E1 osteoblastic cells had reached 70-80% confluence, cultures were transferred to a differentiating medium supplemented with 5 mM 2-DG or $10{\mu}M$ QCT, and then irradiated with 2, 4, 6, and 8 Gy. At various times after irradiation, the cells were analyzed for the synthesis of type I collagen, and expression of BSP and OC. Results : The synthesis of type I collagen in cells exposed to 2 Gy of radiation in the presence of 2-DG or QCT showed no significant difference compared with the control group within 15 days post-irradiation. When the cells were irradiated with 8 Gy, 2-DG facilitated the irradiation mediated decrease of type I collagen synthesis, whereas such decrease was inhibited by treating with QCT. During MC3T3-E1 osteoblastic cell differentiation, the mRNA expression of BSP and OC showed the peak value at 14 days and 21 days, respectively. 2-DG or QCT treatment alone decreased the level of BSP mRNA, but increased the OC mRNA level only at early time of differentiation (day 7). In the cells irradiated with 2, 4, 8 Gy, the mRNA expression of BSP and OC decreased at 7 days after the irradiation. The cells were treated with various dose of radiation in the presence of 2-DG or QCT, the mRNA level of both BSP and OC increased although this increase was observed at low dose of radiation (2 Gy) and at the early stage of differentiation. However, when the cells were exposed to 4, 6, or 8 Gy, the increase of BSP and OC mRNAs was detected only in cells co-incubated with QCT. Conclusion : This study demonstrates that 2-DG and QCT affect differently the expression of bone formation related factors, type I collagen, BSP, and OC in the irradiated MC3T3-E1 osteoblasic cells, according to the dose of radiation and the times of differentiation. Overall, the present findings suggest that 2-DG and QCT could have the regulatory roles as radiation-sensitizer and -protector, respectively.

Effect of protein transduction domain fused-bone morphogenetic protein-2 on bone regeneration in rat calvarial defects (단백질 전달 영역 융합-Bone Morphogenetic Protein-2가 백서 두개골 결손부에서 골 조직 재생에 미치는 효과)

  • Purpose: Recombining bone morphogenetic protein (BMP) is usually acquiredfrom high level animals. Though this method is effective, its high cost limits its use. The purpose of this study was to evaluate the effect of bone morphogenetic protein-2 with protein transduction domain (BMP-2/PTD;TATBMP-2) on bone regeneration. Rat calvarial defect model and osteoblastic differentiation model using MC3T3 cell were used for the purpose of the study. Materials and Methods: MC3T3 cells were cultured until they reached a confluence stage. The cells were treated with 0, 0.1, 1, 10, 100, 500 ng/ml of BMP-2/PTD for 21 days and at the end of the treatment, osteoblastic differentiation was evaluated usingvon Kossa staining. An 8mm, calvarial, critical-size osteotomy defect was created in each of 48 male Spraque-Dawley rats (weight $250{\sim}300\;g$). Three groups of 16 animals each received either BMP-2/PTD (0.05mg/ml) in a collagen carrier, collagen only, or negative surgical control. And each group was divided into 2 and 8 weeks healing intervals. The groups were evaluated by histologic analysis(8 animals/group/healing intervals) Result: In osteoblastic differentiation evaluation test, a stimulatory effect of BMP-2/PTD was observed in 10ng/ml of BMP-2/PTD with no observation of dose-dependent manner. The BMP-2/PTD group showed enhanced local bone formation in the rat calvarial defect at 2 weeks. New bone was observed at the defect margin and central area of the defect. However, new bone formation was observed only in 50% of animals used for 2weeks. In addition, there was no new bone formation observed at 8 weeks. Conclusion: The results of the present study indicated that BMP-2/PTD(TATBMP-2) have an positive effect on the bone formation in vitro and in vivo. However, further study should be conducted for the reproducibility of the outcomes.

Experimental Study on Mechanism Analysis of Headcut Erosion in the Noncohesive Sediment Bed (비점착성 하상에서의 두부침식 메커니즘 분석에 관한 실험 연구)

  • The headcut erosion at the confluence section of a mainstream and tributary can migrate up the tributary streams, and rapid degradation can threaten the stability of hydraulic structures installed in the channel. Therefore, quantitative analysis for the development and mechanism of headcut erosion is needed to prevent damage due to the headcut. In this study, hydraulic experiments for headcut erosion in the channel with noncohesive materials were performed and the knickpoint movement and final bed slope change were analyzed based on the different hydraulic conditions. As a result, the knickpoint movement was 1.5 times faster when the difference in velocity between the upstream and downstream sections was 2.5 times greater and the central part of the cross-section was eroded and collapsed earlier than the left and right sides. The movement length of headcut erosion was longer and the final bed slope was milder as the velocity difference between the upstream and downstream sections was increased. This study showed that a correlation between the knickpoint movement and bed slope change by headcut erosion and the water level difference of upstream and downstream sections was not constant compared to the velocity difference.

Cause Analysis for Reduced Effect of Sewer Pipe Improvement Project Based On Investigation of Interceptor Sewers (차집관로의 조사 및 분석을 통한 하수관로정비 사업의 효과 감소 원인 분석)

  • Interceptor sewer is installed underground near to the river side mostly ofstate-owned land and the management efficiency of public sewage disposal facilities is decreasing as too much infiltration/inflow(I/I) and river flow to interceptor sewer are caused by broken or deteriorated sewer. This also affects the sewer pipeline project and decreases its efficiency. Therefore, the aim of this study is to investigate interceptor sewer which has influence on the reduction of the project effect. The investigation were performed for three study areas. The study includes the investigation of current condition of interceptor sewer(sewer extension, pipe diameter, pipe type, installed year, installed locations, etc), investigation of inside of sewer by CCTV accompanied by pumping and dredging works where required, investigation of inside of manholes by eyes, calculation of pollutant load using the results of investigation of flow quantity and quality. Multipoint investigations were simultaneously performed for flow quantity at confluence area and other investigations were also performed for flow quantity and BOD for interceptor sewer and comparison of pollutant load, investigation of infiltration/inflow(I/I) caused by deterioration of interceptor sewer. As the result of the study, a main reason for reduced effect of sewer pipe improvement project was analyzed as the low-density sewage and I/I in public seweage treatment Facility due to deteriorated and unmanaged interceptor sewers.
