Similarities and differences between alpha-tocopherol and gamma-tocopherol in amelioration of inflammation, oxidative stress and pre-fibrosis in hyperglycemia induced acute kidney inflammation

  • BACKGROUND/OBJECTIVES: Diabetes mellitus (DM) is a major chronic disease which increases global health problems. Diabetes-induced renal damage is associated with inflammation and fibrosis. Alpha (AT) and gamma-tocopherols (GT) have shown antioxidant and anti-inflammatory effects in inflammation-mediated injuries. The primary aim of this study was to investigate effects of AT and GT supplementations on hyperglycemia induced acute kidney inflammation in alloxan induced diabetic mice with different levels of fasting blood glucose (FBG). MATERIALS/METHODS: Diabetes was induced by injection of alloxan monohydrate (150 mg/kg, i.p) in ICR mice (5.5-week-old, male) and mice were subdivided according to their FBG levels and treated with different diets for 2 weeks; CON: non-diabetic mice, m-DMC: diabetic control mice with mild FBG levels (250 mg/dl ${\leq}$ FBG ${\leq}$ 450 mg/dl), m-AT: m-DM mice fed AT supplementation (35 mg/kg diet), m-GT: m-DM mice with GT supplementation (35 mg/kg diet), s-DMC: diabetic control mice with severe FBG levels (450 mg/dl < FBG), s-AT: s-DM mice with AT supplementation, s-GT: s-DM mice with GT supplementation. RESULTS: Both AT and GT supplementations showed similar beneficial effects on $NF{\kappa}B$ associated inflammatory response (phosphorylated inhibitory kappa B-${\alpha}$, interleukin-$1{\beta}$, C-reactive protein, monocyte chemotactic protein-1) and pre-fibrosis (tumor growth factor ${\beta}$-1 and protein kinase C-II) as well as an antioxidant emzyme, heme oxygenase-1 (HO-1) in diabetic mice. On the other hands, AT and GT showed different beneficial effects on kidney weight, FBG, and oxidative stress associated makers (malondialdehyde, glutathione peroxidase, and catalase) except HO-1. In particular, GT significantly preserved kidney weight in m-DM and improved FBG levels in s-DM and malondialdehyde and catalase in m- and s-DM, while AT significantly attenuated FBG levels in m-DM and improved glutathione peroxidase in m- and s-DM. CONCLUSIONS: the results suggest that AT and GT with similarities and differences would be considered as beneficial nutrients to modulate hyperglycemia induced acute renal inflammation. Further research with careful approach is needed to confirm beneficial effects of tocopherols in diabetes with different FBG levels for clinical applications.

Effects of Bio-Ion Water on Growth Performance, Blood Characteristics and Meat Quality in Growing and Finishing Pigs (Bio 이온수 급여가 비육돈의 성장, 혈액성상 및 육질 특성에 미치는 영향)

  • This study was conducted to investigate the effects of bio-ion water on growth performance, blood characteristics and meat quality in pigs. Ninety nine crossbreed pigs $(Landrace{\times}Yorkshire{\times}Duroc)$ were randomly allotted to three treatments; CON (basal diet), T1 (basal diet with bio ion water from growing period), T2 (basal diet with bio ion water from finishing period). There were no significant differences in growth performance and carcass characteristics of pigs among treatments. The red blood cell and white blood cell were significantly higher (P<0.05) in diet added with bio ion water than the control. Proximate analysis (%), meat color, pH, drip loss (%), cooking loss (%) and shear force $(kg/cm^{2})$ were not significantly different (P>0.05) among treatments. The treatment 1 had lower saturated fatty acid (SFA) to unsaturated fatty acid ratio, but higher UFA concentration than those of control. The aroma of cooked meat in T1 was higher than other treatments. Thereby, overall acceptability sensory score of cooked meat in T1 tended to be higher than other treatments.

Characteristics of Compressive Strength Development of High Strength Cement Composites Depending on Its Mix Design (고강도 시멘트 복합체의 배합조건에 따른 압축강도 발현 특성)

  • This study investigates the compressive strength of high-strength cement composites with 64 mixture designs and 2 curing conditions. The cement composites were designed with varying water-to-binder ratios, silica fume content to cement, and binder content per unit volume of cement composite to explore compressive strength development depending on its mix design. An increase in the water-to-binder ratio decreased the compressive strength of the composites, having consistency with the trend in normal concrete. The compressive strength increased with ages at an ambient curing temperature, but it was not identified at high-temperature curing. The compressive strength development was negligible in case that silica fume content to OPC is 15%~25%, but a decrease in the con ten t below 15% reduced compressive stren gth. It was more obvious in the specimen of low water-to-binder ratio. The specimen with 840kg/m3 of binder content per unit volume had the highest compressive strength in this study, and the decrease in binder content reduced the compressive strength of high strength cement composites in low silica fume content.

Anti-skin-aging Effect of Mori Folium through decreased Advanced glycation end product (AGEs) (최종당화산물 억제를 통한 상엽(桑葉)의 항피부노화 효과)

  • Objectives : Recently, numerous studies reported that excessive generation of advanced glycation end products (AGEs) stimulated expression of skin wrinkle related proteins. This study aimed to evaluate inhibits skin wrinkle formation effect of Mori Folium (MF) through decreased AGEs. Methods : To evaluate the skin wrinkle inhibition effect of MF, SD-rats were divided into three groups; normal rats (Nor), AGEs-induced rats (Con), AGEs-induce rats treated with MF at dose of 100mg/kg body weight (MF). To induced AGEs, streptozotocin (50mg/kg) was injected intraperitoneally, and after 3 days, 100mM methyl glyoxal was administered orally for 3 weeks. After the experiment, the animal's dorsal skin tissues and serum were separated and tested. Results : The oral administration of MF suppressed the AGEs level in serum. Also, the AGEs in skin tissues was significantly reduced through treatment of MF compared with control group. Moreover, the expressions of AGEs related proteins such as polyclonal anti-$N^e$-(carboxymethyl) lysine (CML), anti-$N^e$-(carboxyethyl)lysine (CEL), AGE receptors (RAGE) were reduced in MF group compared with the control group in kidney and skin tissues. The matrix metallo proteinase-1 (MMP-1) reduced by MF treatment with the result that collagen type 1 alpha 2 (COL1A2) was improved that reduced by accumulation of AGEs. Conclusion : The evidence of this study indicate that oral administration of MF reduces the levels of AGEs in serum, skin, and kidney tissues. In conclusion, MF inhibit skin wrinkle formation, suggesting the potential of anti-wrinkle material.

Skin Wrinkle Improvement Effect of Paeoniae radix and processed Paeoniae radix Through inhibition of Advanced glycation end products (AGEs) (작약 및 포제작약의 최종당화산물 억제를 통한 피부 주름 개선 효과)

  • Objectives : Collagen decrease of Skin appears through various path ways. One of causes may be the Advanced glycation endproducts (AGEs) that combine formation of glucose and protein. The aim of this study was to explore the prevent wrinkle formation of Paeoniae radix (PR) and heated Paeoniae radix (HPR) via AGEs path way. Methods : AGEs formation inhibitory activities of PR and HPR measured using bovine serum albumin, glucose, and fructose. To evaluate the protective effects of PR and HPR in diabetic rats induced with streptozotocin (STZ) and methyl glyoxal (MGO), SD rat were distributed into four groups. Normal rats (Nor), AGEs-induced rats (Con), AGEs-induced rats treated with 100 mg/kg PR(PR), AGEs-induced rats treated with 100 mg/kg HPR (HPR). To induce AGEs, streptozotocin (50 mg/kg) was administered intraperitoneally and after 3 days administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of HPR inhibited AGEs in skin tissues compared with PR. The increased reactive oxygen species (ROS) levels in the serum were diminished by HPR treatment. The analyses of kidney and skin tissues proteins indicated that HPR treatment effectively reduced AGEs related protein levels as compared to that by PR treatment. Also, HPR decreased anti-oxidant related protein levels in skin tissues such as catalase, glutathione peroxidase. Moreover, it inhibited the reduction of COL1A2 by decreasing MMP-1. Conclusion : Based on these results, it was suggested that PR and HPR could have Improving effects on wrinkle formation. These evidences provide useful information for the development wrinkle formation treated agent.

Skin elasticity improvement effect of Young persimmon and Heated young persimmon by decreased Advanced glycation end products(AGEs) (떫은감과 포제 떫은감의 최종당화산물 생성 억제를 통한 피부 탄력 개선 효과)

  • Objectives : Advanced glycation end products (AGEs) is bind formation of glucose and protein. Acceleration of AGE formation during hyperglycemia is associated with the pathogenesis of diabetic complications and causes kidney and skin damage. The aim of this study was investigated the AGEs inhibitory activity and antioxidant activity of water extracts from young persimmon (YP) and heated young persimmon (HYP). Methods : Paeoniae Radix Alba (YP) is prepared by heating with 30% ethanol. AGEs formation inhibitory activities of YP and HYP measured using bovine serum albumin. To evaluate the protective effects of YP and HYP in diabetic rats induced with streptozotocin (STZ) and methyl glyoxal (MGO), SD rats were distributed into four groups; normal mice (Nor), AGEs-induced rats (Con), AGEs-induced rats treated with 100 mg/kg YP (YP), AGEs-induced rats treated with 100 mg/kg heated YP (HYP) for 3weeks. Heated young persimmon respectively decrease AGEs construction. Results : YP and HYP administration inhibited the biomarkers of AGEs in serum, kidney and skin tissues. AGE-induced rats revealed that the significant decreased collagen however, heat processing methods of young persimmon up regulated inhibits AGEs-induced collagen decrease. The expressions of AGEs were decreased in YP and HYP treated group compared with the control group in tissues. It specifies that HYP has potential to serve as a positive regulator of via AGEs path way. Conclusion : It has proposed that may have an improvement effect on diabetic complications, heated young persimmon has AGEs inhibitory excellent activities and antioxidant effect.

Effects of Aerobic Exercise upon Cytosolic GAPHD and Mitochondrial MnSOD Activity of Pancreatic Cells in the Type 1 Diabetic Rats (유산소운동이 제1형 당뇨쥐의 췌장 세포질 GAPHD 및 미토콘드리아 MnSOD 활성에 미치는 영향)

  • Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and manganese superoxide dismutase (MnSOD) has been hypothesized as a mediator in the activation of multiple pathways implicated in the pathogenesis of diabetic disease. The objective of this study was to understand the mechanism that aerobic exercise activate GAPDH and MnSOD in pancreatic cells. To achieve the purpose of this study, thirty male Sprague-Dawley rats were assigned to control group, diabetic group and diabetic exercise group. 10 rats were forced to exercise according to exercise protocol for 8weeks and 20 rats were untrained for control and diabetic group. Pancreatic tissue were extracted from the each. Expressions of GAPDH and MnSOD in diabetic pancreatic tissues were significantly decreased compare to control group. However, swimming (trained diabetic group) significantly increased expressions of GAPDH and MnSOD compare to diabetic group, respectively. In hyperglycemia, GAPDH and MnSOD in pancreatic cells is activated by aerobic exercise, and this inactivates multiple pathways implicated in the pathogenesis of diabetic disease. In conclusion, these findings suggest that increased activity of GAPDH and MnSOD by exercise have beneficial effects on mitochondrial dysfunction and arresting the progression of diabetic disease.

A Generalized Model for the Prediction of Thermally-Induced CANDU Fuel Element Bowing (CANDU 핵연료봉의 열적 휨 모형 및 예측)

  • The CANDU element bowing is attributed to actions of both the thermally induced bending moments and the bending moment due to hydraulic drag and mechanical loads, where the bowing is defined as the lateral deflection of an element from the axial centerline. This paper consider only the thermally-induced bending moments which are generated both within the sheath and the fuel and sheath by an asymmetric temperature distribution with respect to the axis of an element The generalized and explicit analytical formula for the thermally-induced bending is presented in con-sideration of 1) bending of an empty tube treated by neglecting the fuel/sheath mechanical interaction and 2) fuel/sheath interaction due to the pellet and sheath temperature variations, where in each case the temperature asymmetries in sheath are modelled to be caused by the combined effects of (i) non-uniform coolant temperature due to imperfect coolant mixing, (ii) variable sheath/coolant heat transfer coefficient, (iii) asymmetric heat generation due to neutron flux gradients across an element and so as to inclusively cover the uniform temperature distributions within the fuel and sheath with respect to the axial centerline. As the results of the sensitivity calculations of the element bowing with the variations of the parameters in the formula, it is found that the element bowing is greatly affected relatively with the variations or changes of element length, sheath inside diameter, average coolant temperature and its variation factor, pellet/sheath mechanical interaction factor, neutron flux depression factor, pellet thermal expansion coefficient, pellet/sheath heat transfer coefficient in comparison with those of other parameters such as sheath thickness, film heat transfer coefficient, sheath thermal expansion coefficient and sheath and pellet thermal conductivities.

A Study on the Dynamic Lateral Displacements of Caisson Quay Walls in Moderate Earthquake Regions (중진지역에서 케이슨 안벽의 동적수평변위 특성에 관한 연구)

  • In this study, 28 earthquake records with magnitudes from 5.3 to 7.9 are selected for dynamic analysis in order to assess applicability of the earthquakes for domestic seismic design. The assessment is performed using the seismic spectrum analysis of energy and acceleration. Based on results of the analysis, four acceleration time histories, which satisfy the Korean design standard response spectrum, are proposed. From the dynamic analysis using earthquake magnitudes from 6.4 to 7.9, it is found that horizontal displacements corresponding to earthquake magnitudes greater than 7 are two times larger than those with magnitude 6.5. Therefore, it can be stated that use of strong earthquakes, such as Miyagiken-ken-oki earthquake (Ofunato, $M_{JMA}=7.4$) and Tokachi-oki earthquake (Hachinohe, $M_{JMA}=7.9$), for the seismic design in Korea is not applicable, and may prove to be excessively conservative due to overestimated seismic force. From the dynamic analyses using the proposed acceleration time histories, effects of caisson quay wall dimension and the subsoil condition are investigated as well. The simplified design charts to evaluate horizontal displacements of caisson quay wall are also proposed based on earthquake magnitude 6.5 that is appropriate in Korea.

  • Al centro dell'interesse di Strehler ci sono sempre l'uomo e le sue azioni, e l'uomo e la posizione del teatro. Inoltre, nei suoi teatri vengono alla luce i rapporti che interessano l'uomo e la societa', l'uomo e se stesso, l'uomo e la storia, l'uomo e la politica. Strehler interpreta sempre i testi prendendo in considerazione la contemporaneita' e completa la rappresentazione strutturandola in un sistema implacabilmente coerente di segni scrulolosi. Poiche' le sue opere, anche se le stesse, sono rappresentate ogni volta con significati diversi a secondo del tempo, non si tratta soltanto di rivivere le opere, ma di metterle in scena in nuove versioni che hanno significati che interpretano i cambiamenti avvenuti. Questo e' anche il caso de I giganti della montagna. Nella prima regia di quest'opera, Strehler cerca di mettere in evidenza una concreta e generale storia umana attraverso il mito, e un messaggio che è al tempo stesso di disperazione e di fede nell'uomo. Un messaggio di perdono e di pietà. Pietà per tutti. Nella seconda regia, egli incarna, attraverso il vuoto dell'immobilita' e il silenzio, la vita della poesia. Cerca di rappresentare l'estremo degli effetti scenici attraverso la minima mobilita' in una scena immobile, e attraverso il massimo grido di dolore in una scena di silenzio. E anche i personaggi sono divisi in due diverse categorie. 'Gli scalognati', che creano 'teatro puro' non per un pubblico ma per puro giuoco, e 'la compagnia dei comici', che crea 'teatro rappresentato' che implica la possibilita' dell'interprete condizionato dai rischi, dagli errori di valutazione e d'interpretazione e che resta legato ad una realta' concreta. Attraverso questo contrasto egli vuole esprimere "il timore che nonostante tanta "rivoluzione" giovanile intorno, il cammino della societa' degli uomini verso l'abbandono di alti ideali e valori, fosse accelerato e quasi ineluttabile". Nella terza regia, i due mondi si incontrano in uno spazio dove realta' e sogno si confondono. Non sono piu' contrapposti. E' come "una tragica contestazione che i Giganti hanno vinto e che ci hanno, consapevoli o inconsapevoli, travolti, che non siamo piu gli stessi, che stiamo precipitando". Ma egli non dispera, benche' rappresenti la condizione tragica, dice al pubblico di "comunicare in silenzio la reciprocita' delle sconfitte. al di la' delle parole che non esistono".
