천마의 흑질 내 도파민성 신경세포 보호 효과에 대한 단백체학적 분석 (Proteomic Analysis for Neuroprotective Effect of Gastrodia elata Blume in the Substantia Nigra of Mice)

  • Objectives : Parkinson's disease (PD) is a neurodegenerative disorder threatening the quality of life and highly occurred in over 65 years old. Gastrodia elata Blume (GEB), a traditional medicine used for the treatment of headache and convulsion, has been reported to have neuroprotective effect. This study was designed to investigate the neuroprotective effect of GEB and the proteomic changes in the substantia nigra (SN) of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated mice. Methods : Male eleven-week-old C57BL/6 mice were intraperitoneally injected with 30 mg/kg of MPTP at 24-h intervals for 5 days. Two hours after the daily MPTP injection, the mice were orally administered 800 mg/kg of GEB extract, which continued for 7 days beyond the MPTP injections, for a total of 12 consecutive days. Two hours after the final GEB administration, the brain samples were collected, and dopaminergic neuronal death and proteomic changes in the SN were evaluated. Results : GEB prevented the MPTP-induced dopaminergic neuronal death and regulated the expression of 11 proteins including thimet oligopeptidase, T-complex protein 1, glycine tRNA ligase, and pyruvate kinase isozymes M1. Conclusions : GEB prevents MPTP-induced dopaminergic neuronal death by regulating the proteins in the SN.

황련 열수추출물을 처치한 인간 대장암 세포 SNU-81에서의 단백질 발현 변화 (Monitoring the Change of Protein Expression in Human Colon Cancer Cell SNU-81 treated with the Water-Extract of Coptis japonica)

  • Background : Anticancer effects of herbal medicine have been reported in various types of cancer, but the systematic approaches to explain molecular mechanism(s) are not established yet. Objective : To find the anticancer-effect and mechanism(s) of Water Extract of Coptis japonica (WECJ) colon cancer cell (SNU-81). Methods : We first selected 11 herbals, and anti-cancer effects of water-extracts from those herbals have been tested in human colon cancer cell line, SNU-81. Among the tested herbals, the WECJ significantly reduced proliferation of SNU-81. To establish a basis of understanding for anti-cancer mechanism, whole proteins have been obtained from SNU-81 harvested at 48 and 96 hrs after the treatment of WECJ, protein expression has been profiled by 2DE-based proteomic approach. Results : Various changes of the protein expression have been monitored, and most frequent dysregulation was found in the molecular chaperons including heat shock protein 90-alpha (Hsp90-alpha), 14-3-3 protein epsilon, T-complex protein 1 subunit alpha, protein disulfide-isomerase A3, and calreticulin. Interestingly, proliferation-associated protein 2G4 has been up-regulated, and it suggests the possible effect of Coptis japonica on ErbB3-regulated signal transduction pathway and growth control of human colon cancer cells. Conclusion : Based upon the present findings, the further study will focus on monitoring various cancer survival factors after artificial regulation of the proteins identified, and it would be the basis for the understanding of the Coptis japonica anti-cancer effect(s) at the molecular level.

한국 일부지역 농촌인의 질병개념에 대한 탐색적 연구 (The Concepts of illness of Rural Korean Peoples)

  • The problem addressed by this study was to reveal what people of Korean rural villages think about the cause, treatment and prevention of illness. The purpose was to contribute to the building of a concept of health toward the development of Korean Nursing Theory. Subjects were residents of five districts among four counties in a farming area of Chonbuk province recommended by health workers as appropriate informants. They were interviewed in their homes, using ethnoscientific methods developed in anthropology. The research tool consisted of open questions developed through the literature and preliminary exploratory interviews. Data were analyzed by classifying each concepts of cause, treatment and prevention of illness or illness symptoms collated by frequency and percentage. The causes of illness are conceived as primarily concrete physical and natural, for examples, overeating, lack of energy, changes in the season and extreme temperatures. Compared to others studies, few supernatural causes related to traditional view of illness were identified. Concepts of the treatment of illness included formal treatments used by modern western or oriental physicians and traditional therapists. But folk medicine used by traditional healers or by the family in the home was most prevalent. The concept of illness prevention originated in the concept of the cause of illness, thus primarily physical and natural, for examples, nutritious food, limiting the amount of food, avoiding becoming cold. When the concept of illness of rural Korean is researched from a sociocultural aspect, the traditional views of an evil cause of ill health and treatment by supernatural methods is not found to be prevalent but folk medicine still occupies a large place in treatment which si often a complex mixture from many mysterious sources. The significance of this study lies in the fact that ethnonursing research can contribute basic data toward the development of Korean nursing theories. Modern western medical concepts have not been accepted unconditionally: traditional concepts are alive and dynamic in Korea and must be recognized in Korean nursing.

영수보사(迎隨補瀉)와 원방보사(圓方補瀉) 수기법(手技法)의 정량적(定量的) 연구(硏究) (Analysis of YoungSu & Wonbang Acupuncture Method by the Measurement of Physiological Signals on Acupoints)

  • Previously two papers dealing with YoungSu(against the meridian course and following the course of the meridian) Acupuncture were published by our group. Here we are reporting the further analysis of YoungSu and WonBang(by twisting and rotating the needle) acupuncture methods. It is very important to understand objectively the Qi variation induced by the reinforcing-reducing manipulation method in the acupuncture therapy. We decided the medical treatment by utilizing the PyongChi Method (a kind of method to figure out the way of treatment by observing the unbalanced state of five phases). The Qi variation in the meridian treated by YoungSu and WonBang, the recovery of five phases deviation were measured by choosing single acupoint instead of complex acupoints. By using Youngsu and WonBang, we increased or decreased the Qi of the phase which caused the unbalanced state. We observed whether the Qi of the treated meridian can be increased and if the state of unbalance can be recovered. To achieve the effect of reinforcing-reducing, we needed a correct choice of treating method and a selection of a proper meridian in advance. This study was carried out by adding another way of acupuncture from the previous paper. We discovered that the effects of reinforcing-reducing by each manipulation method could be superposed each other when two counteracting Youngsu and WonBang methods were treated at the left and the right side of human body which was correspondent with our previous paper. We found that the Qi variation of the treated meridian, which was induced by Youngsu and WonBang, was linearly proportional to the reduction of five phase deviations. The slope of Qi variation was almost similar (y = -0.413x - 0.138) as that of previous paper (y = -0.266x - 0.038, Y = -0.446x - 0.079). It is assumed that the addition of other basic methods on the top of reinforcing-reducing manipulation method would magnify the effect of acupuncture.

항암제 내성 유방암 MCF7/adR 세포주에 대한 보정방암탕과 홍삼산성다당체의 세포고사 유도효과 (Apoptotic Effect of Ethanol Extracts of Bojungbangamtang and Acidic Polysaccharide of Korea Red Ginseng in a MCF7/adR Multidrug-resistance Breast Cancer Cells)

  • This study was undertaken to determine whether the 9 herbal complex induces apoptosis in human breast cancer MCF-7 cells and adriamycin-resistant MCF7/adR cells. Ethanol extracts of Bojungbangamtang (BBTE) and acidic polysaccharide of Red Ginseng (GIN) induced cell death in both MCF-7 and MCF7/adR cells. Ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng also induced $G_2/M$ cell cycle arrest and increased TUNEL positive cells in MCF7/adR cells. In addition, flow cytometric analysis revealed the decreased expression of P-glycoprotein (P-gp) in ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng treated MCF7/adR cells. Similarly, decreased protein levels of P-glycoprotein and multidrug resistance associated proteins-1 were also determined by immunocytometry in ethanol extracts of Bojungbangamtang treated MCF7/adR cells. Taken together these data indicate that ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng inhibit the function of ABC transporters such as multi drug resistance associated proteins (MRPs) and P-glycoprotein as well as induce apoptosis in MCF7/adR cells. Thus, these data suggest that ethanol extracts of Bojungbangamtang and polysaccharide of Red Ginseng can be candidates for the treatment of multidrug-resistant MCF7/adR cells.

녹각 추출물과 CoenzymeQ10 복합제가 운동능력에 미치는 영향 (Ergogenic Effect of Cervi Cornu and CoenzymeQ10 Complex)

  • Objectives: We aimed to evaluate the effect of Cervi Cornu and coenzymeQ10 on exercise and endurance capacity in rats and mice. Methods: The extract of Cervi Cornu was manufactured by the pharmacy department of Kyung Hee Oriental Medical Hospital, and CoQ10 soft cap (Ildong Pharmaceutical) was used. In total, 24 rats and 30 mice were divided into 3 groups: Control (rat=8, mouse=10), CoQ10 alone (rat=8, mouse=10), Cervi Cornu extract, and CoQ10 (rat=8, mouse=10). Ergogenic effect was evaluated by administering the Cervi Cornu extract and coenzymeQ10 to rats and measuring the time to exhaustion during treadmill running; endurance capacity was assessed by measuring cold water swimming time, serum lactate level, and serum corticosterone level in each group. At 1 week from the end of treatment, we recalculated time to exhaustion during treadmill running in rats to investigate the long-term effect of the Cervi Cornu extract and coenzymeQ10. Results: Cervi Cornu extract has long-term benefits in that it preserves the ergogenic effect caused by exercise. Cervi Cornu and coenzymeQ10 have no effect on increasing cold water swimming time in ICR mice. CoenzymeQ10 decreases the serum corticosterone level in ICR mice performing cold water swimming test. Conclusions: Cervi Cornu seems to preserve the ergogenic effect caused by exercise, but a larger study is needed to investigate effect of Cervi Cornu and coenzymeQ10 on improving endurance capacity. CoenzymeQ10 decreases serum corticosterone level and it is related with the anti-psychological fatigue effect.

고려 의서 『향약구급방』의 임상 의학 특징 분석 (An Analysis of Clinical Characteristics on the Medical book of Koryŏ Dynasty, Hyang'yak Kugŭppang (鄕藥救急方, Medical Recipes of Local Botanicals for Emergency Use))

  • Objectives : In this paper, We pointed out medico-historical meaning of Hyang'yak Kugŭppang (鄕藥救急方, Medical Recipes of Local Botanicals for Emergency Use), a medical book of Koryŏ Dynasty, breaking from the existing analysis view based on Hyang'yak (鄕藥), local herbs produced in Korean Peninsula. Methods : For that purpose, we analyzed the clinical characteristics of the diseases and symptoms, strategies of treatment, therapeutic tools, drug formulations, composition of herbal formulas in Hyang'yak Kugŭppang. Results : The following characteristics were confirmed in Hyang'yak Kugŭppang. First, it contained a way to cope with the diseases and symptoms that do not have to check the exterior, interior, cold, and heat. Also, it introduced basic strategies such as reducing, facilitating flow, draining downward, warming, or tonifying, etc. It mainly included herbal medicine as the therapeutic tools, and introduced formulas composed by single herb than complex recipes. It suggested external applications more than internal use, and proposed formulations in the form of decoctions or powders, etc. The herbs of formulas were used by experience without explanations of nature & taste, efficacy of each herbs. Conclusions : Hyang'yak Kugŭppang is a book of medical recipes for emergency use that has been distributed to help gentries, even if they do not have professional medical knowledge, to instantly cope with diseases and symptoms with medicines that are easy to obtain. Previous researches has analyzed the value of Hyang'yak Kugŭppang as a collection of information of hyang'yak, and used it as a historical basis of the uniqueness of Korean medicine. Since Hyang'yak Kugŭppang is not a book specialized for herbs, but rather a clinical medical book that introduces recipes for specific symptoms of diseases, if we pay attention to the clinical characteristics of Hyang'yak Kugŭppang, it will provide an opportunity to describe Koryŏ or Korean medicine in a new way.

Non-Ionic Surfactants Antagonize Toxicity of Potential Phenolic Endocrine-Disrupting Chemicals, Including Triclosan in Caenorhabditis elegans

  • Triclosan (TCS) is a phenolic antimicrobial chemical used in consumer products and medical devices. Evidence from in vitro and in vivo animal studies has linked TCS to numerous health problems, including allergic, cardiovascular, and neurodegenerative disease. Using Caenorhabditis elegans as a model system, we here show that short-term TCS treatment ($LC_{50}$: ~0.2 mM) significantly induced mortality in a dose-dependent manner. Notably, TCS-induced mortality was dramatically suppressed by co-treatment with non-ionic surfactants (NISs: e.g., Tween 20, Tween 80, NP-40, and Triton X-100), but not with anionic surfactants (e.g., sodium dodecyl sulfate). To identify the range of compounds susceptible to NIS inhibition, other structurally related chemical compounds were also examined. Of the compounds tested, only the toxicity of phenolic compounds (bisphenol A and benzyl 4-hydroxybenzoic acid) was significantly abrogated by NISs. Mechanistic analyses using TCS revealed that NISs appear to interfere with TCS-mediated mortality by micellar solubilization. Once internalized, the TCS-micelle complex is inefficiently exported in worms lacking PMP-3 (encoding an ATP-binding cassette (ABC) transporter) transmembrane protein, resulting in overt toxicity. Since many EDCs and surfactants are extensively used in commercial products, findings from this study provide valuable insights to devise safer pharmaceutical and nutritional preparations.

뇌해마의 장기양 조직배양을 이용한 한약물의 뇌신경세포손상 보호효능 연구 (Neuroprotective Effects of Medicinal Herbs in Organotypic Hippocampal Slice Cultures)

  • Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. 한방소재 화장품의 아토피 피부염 개선 효과 (The Enhancing Effect of Atopic Dermatitis by cosmetics containing estern medical herbs)

  • 한방소재를 이용한 화장품이 아토피피부염을 개선하는지를 알아보고자, 중등증의 아토피 피부염환자 16명을 대상으로, 진경한방컴플렉스(감초, 백지, 작약, 지실, 천궁, 갈근, 현호색), 편백수, 녹차, 황기, 홍삼, 당밀발효액, 사과 발효여과물이 함유된 화장수와 크림을 하루에 3회 피부염병변에 6주간 도포하도록 하였다. 그 결과 SCORAD 지수는 $26.9{\pm}11.8$에서 $14.9{\pm}9.7$로 감소(p<0.05), Total IgE(IU/mL)는 $641.9{\pm}1294.6$에서 $565.8{\pm}1076.8$로 감소, Eosinophil의 수(ug/dL)는 $246.7{\pm}203.5$에서 $203.3{\pm}130.7$로 감소하였다(p<0.05). 피부수분함유량은 $36.6{\pm}10.0$에서 $44.0{\pm}10.3$로 증가(p<0.01), 경피수분손실량(g/h/$m^2$)은 $16.7{\pm}5.1$에서 $15.4{\pm}8.7$로 감소(p<0.05)하였으며, 피부 pH는 약산성을 유지하였다. 또한 의사 및 환자의 총괄유효성 평가도 증가되었다.
