Efficient Transformation of Klebsiella oxytoca by Electroporation

  • A protocol for the transformation of Klebsiella oxytoca by electroporation was developed. Preparation of competent cells at early exponential phase was most critical to obtain high transformation efficiency. The highest efficiency of 1.6$\times$106 transformants per $\mu\textrm{g}$ DNA(pBR322) could be obtained by electroporation of K. oxytoca cells prepared at the OD600 of 0.2 with 1.25$\mu\textrm{g}$ DNA at the filed strength of 2.5kV, the parallel resistance of 200$\Omega$ and capacitance of 25$\mu$F.

  • Herpes zoster is a cutaneous infection that is characterized by an acute vesicobullous rash with ipsilateral one or two dermatomal distribution and painful allodynia, while predominantly being found in the elderly. Extensive cutaneous dissemination has been reported in immune-compromised patients, such as those who suffer from HIV infections, cancer, chemotherapy, and corticosteroid therapy patients. However, we report a case of disseminated herpes zoster infection in an immuno-competent elderly individual.

  • Implantation is a highly organized process that involves an interaction between a receptive uterus and a competent blastocyst. In humans, natural fecundity suggests that the chance of conception per cycle is relatively low (~30%) and two-third of lost pregnancies occur because of implantation failure. Defective implantation leads to adverse pregnancy outcomes including infertility, spontaneous miscarriage, intrauterine fetal growth restriction and preeclampsia. With use of advanced scientific technologies, gene expression analysis and genetically-engineered animal models have revealed critical cellular networks and molecular pathways. But, because of ethical restrictions and the lack of a mechanistic experiment, comprehensive steps in human implantation have still not been completely understood. This review primarily focuses on the recent advances in mechanisms of implantation. Because infertility is an emerging issue these days, gaining an understanding the molecular and hormonal signaling pathway will improve the outcome of natural pregnancy and assisted reproductive technology.

Transformation of Bacillus Subtilis by Streptomyces bobili R-Plasmid DNA (Streptomyces bobili의 R-Plasmid. DNA에 의한 Bacillus subtilis의 Transformation)

  • The penicillin resistant plasmid DNA was prepared from Streptomyces bobili YS-40, producing penicillinase, by the phenol extraction method and introduced into Bocillus subtilis IAM 12118 by the transformation procedure of Mahler method. The optimal pH and temperature on the transformation was 7.0, 3$0^{\circ}C$ respectively. Above 20 minutes contact of plasmid DNA and recipient cell was shown the high transformation frequency. The transformant of penicillin resistance was proportionally increased as increase of the DNA concentration. The addition of lysine in transformation system increased the transformation frequency about 6-fold and the addition of the chloramphenicol did not affect the transformation frequency.

  • Assisted reproductive technologies (ART) merely depend on improving the oocyte maturation and their developmental competence to produce good quality embryos. Oocyte maturation passes through long and complex molecular steps starts from the early embryonic life and ends with sperm fertilization. Oocyte developmental competence can be attained by improving the nuclear and cytoplasmic mechanisms together with some epigenetic maturation. In this review, we highlight the cornerstones of oocyte maturation on both nuclear and cytoplasmic levels. Interfering or supporting these molecular mechanisms would help in the development of novel regulating agents for reproductive performance of humans and livestock species.

  • In this paper, we have designed a platform with MSC8101 processor for networked converter monitoring and diagnosis. MSC8101 is a dual processor type SOC(System On a Chip), which is consist of 16bit DSP and 32bit RISK CPM. As it have DSP and CPM, MSC8101 is competent for networking and data processing application. This MSC8101 platform is designed for networked monitoring and diagnosis, so it is important processing ability and networking capability.

  • This is a genuine case report of the Ross operation without the use of homografts or heterografts in reconstruction of the right ventricular outflow tract. A 8-year-old boy with congenital aortic stenosis underwent aortic valve replacement with a pulmonary autograft and right ventricular outflow tract reconstruction with a pericardial conduit bearing autologous aortic monocusp. The postoperative echocardiography and cardiac angiography revealed good ventricular function and competent neoaortic valve. He has been followed up for 19 months.

  • N crassa mutant strain nap showed reduced growth rate, decreased electric membrane potential, and elevated intracellular ATP content in comparison to the wild type. Blue light induced a hyperpolarization of the membrane potential in both strains. The analysis of oxidative and phosphorylation activities of mitochondria isolated from the two strains has revealed that nap utilized more efficient oxidative pathways. The higher intracellular ATP content in the nap was presumably due to impaired transport systems of the plasma membrane, and to a lesser extent to the functioning of the fully competent respiratory chain. The excess ATP possibly accounts for carotenoid accumulation in the mutant.

  • The MTF1 gene of Saccharomyces cerevisiae encodes a 43 kDa MITOCHONDRIAL RNA polymerase specificity factor which recognizes mitochondrial promoters to initiate correct transcription. To better understand structure-function of the MTF1 gene as well as the transcription mechanism of mitochondrial RNA polymerase, two cold-sensitive alleles of the MTF1 mutation were isolated by plasmid shuffling method after PCR-based random mutagenesis of the MTF1 gene. The mutation sites were analyzed by nucleotide sequencing. These cs phenotype mtf1 mutants were respiration competent on the nonfermentible glycerol medium at the permissive temperature, but incompetent at 13.deg.C. The cs phenotype allele of the MTF1, yJH147, encoded an L146P replacement. The other cs allele, yJH148, contained K179E and K214M double replacements. Mutations in both alleles were in a region of Mtflp which is located between domains with amino acid sequence similarities to conserved regions 2 and 3 of bacterial s factors.

  • Numerical studies were carried out to investigate the mechanical properties of competent hysteric isolators for seismic design of bridge. For dynamic analysis, bridges with isolator were simplified to a model with single degree of freedom. The initial stiffness and the yielding forces of hysteric isolators were varied. Seismic responses obtained by time history analysis show that about 4% of the weight acting as the inertia force is appropriate for the yielding force of isolator. And also better results could be achieved with the values about two times the weight per unit displacement for the initial stiffness of isolator.
