Detection and Phylogenetic Analysis of Viruses Linked with Fig Mosaic Disease in Seventeen Fig Cultivars in Palestine

  • Fig mosaic is a viral disease (FMD) that spreads in Palestinian common fig (Ficus carica L.) orchards. Recognizing the economic value of fig plants and the harmful nature of FMD, the disease poses a significant threat to the economy of the fig production in Palestine. We applied the reverse transcription and amplification (RT-PCR) and PCR technique to leaf samples of 77 trees and 14 seedlings of 17 fig cultivars. The samples were collected from orchards in the main fig-growing provinces of the Palestinian West Bank, to assess the prevalence of viruses associated with FMD, and confirm a possible link of symptoms with viruses detected. Four viruses were detected: Fig mosaic virus (FMV), Fig badnavirus-1 (FBV-1), Fig leaf mottle-associated virus 2 (FLMaV-2), and Fig fleck-associated virus (FFkaV). FMV and FBV-1 were found in all tested fig plants (100%), while FLMaV-2 and FFkaV were detected in 61.5% and 33% of the fig samples, respectively. The high incidence of FBV-1 in the newly propagated symptomatic and symptomless seedlings from different cultivars may be an indication that FBV-1 is integrated into the genome of the fig in a cultivar nondiscriminatory manner. Very weak or no association was detected between FMD symptoms severity in the 17 Palestinian fig cultivars with the various viruses' combinations observed (i.e., number of the viruses infecting the plant). These results support the notion that FMD symptom severity expression is likely to be controlled by a combination of FMV infection, cultivars, and environmental factors, rather than the number of viruses infecting the plant.

Occurrence of Gray Mold Caused by Botrytis cinerea on Common Fig in Korea (Botrytis cinerea에 의한 무화과 잿빛곰팡이병)

  • In 2010 and 2011, gray mold was found on common fig (Ficus carica) fruit grown at the research field of Jeollabuk-do Agricultural Research and Extension Services, Korea. Gray mold symptoms on common fig fruit mainly occurred after harvest season until December. The typical symptom included brown water-soaked rot and fruit decay. The diseased fruit was covered by gray to brown colored conidiophore and conidia. The conidiophores were tree shape and measured $15-33{\times}2{\mu}m$. Conidia on conidiophore were ellipsoidal or lemon shape, colorless, single cell, and measured $7.3-14.6{\times}6.8-11.1{\mu}m$. The nucleotide sequences of the rDNA ITS region obtained from the pure culture of the gray mold on common fig were 100% similar to the sequences of the GenBank accession number HQ171052, EU519210, HQ171053, FN812726, HM849615, and EU563120 of B. cinerea isolates. In phylogenetic tree, the representative isolate was placed within same clade of B. cinerea. Based on the morphological characteristics and analysis of rDNA ITS sequence data, the fungus was identified as B. cinerea.

Identification of Pseudocercospora bolleana Associated with Angular Leaf Spot on Common Fig in Korea (무화과나무의 모무늬잎마름 증상에 관여하는 Pseudocercospora bolleana 동정)

  • A cercosporoid fungus associated with angular leaf spots on the leaves of common fig (Ficus carica) in Korea is known to be morphologically similar to Passalora, but phylogenetically similar to Pseudocercospora. To clarify the ambiguity, six fig samples with angular leaf spots were collected and examined using a microscope, and two representative isolates were sequenced for multiple genes. The morphological characteristics were consistent with previous descriptions of Passalora bolleana. Molecular phylogenetic analysis based on the internal transcribed spacer and large subunit ribosomal DNA (rDNA) regions showed that the Korean isolates, as well as previously published Korean and Romanian isolates, formed a well-supported group in the clade of Pseudocercospora species. Consequently, the current Korean isolates should be correctly described as Pseudocercospora bolleana. Additionally, Pseudocercospora fici-caricae, a cercosporoid fungus previously described as a leaf pathogen on common fig in Taiwan and Korea, was also compared and discussed.

  • In this study, it aims to research for unified system of "the surveying and geospatial information management" and new National geoSpatial Information Infrastructure(NSDI) as new paradigm against the strategy of "global geospatial information management". The country's existing NGIS projects and the policies of spatial information were examined in this paper, then it was defined newly by modification of NSDI's data coverage with bottom-up method. The new NSDI strategy is based on large scale digital map which was influenced by the local and global trend such as open data, e-Government, Earth observation, etc. (refer to Fig. 1). It was also suggested with new concept of NSDI model that the public-private sharing data can be added to digital map on equal term with spatial core data. (refer to Fig. 2) It is proposed the institutional model of MOLIT(Ministry of Land, Infrastructure and Transport) as new concept of NSDI which was applied(refer to Fig. 4). The new model is improving localization and reinforcing cooperation system with not only the other departments within the MOLIT but also the other ministries(forestry, environment, agriculture, heritage, etc.) from independent operation system as a part informatization of land, infrastructure and transport. At the new SDI institutional model of the MOLIT, the spatial information is reorganized as common data infrastructure for all applications, Goverment 3.0 can be feasible according to common data related to government agencies and local government's data vertically or horizontally. And then, it can be practical strategy model to integrate and link all the map and the register which are managed by the laws and institutions if this unified system as a common data can include all spatial core data(digital map), such as base map data of NGA(national gespatial agency), land data and facility data of local government.

  • Purpose : Facial palsy goes together not only physical difficulties but also social life's of relationship to other people. Therefore we was devised correction splint and rehabilitation set for facial palsy proofreading. Method : This article was used by questionnaire survey that intended for 140 patients who had got facial palsied such as universities hospitals and oriental hospitals over the country in Korea. The subject matter that faced consciousness of a patient as opposed the awkward rehabilitating tool that a general matter and patient. In the object that the rehabilitation tool which now patient used through a wraps a face in only as a treatment object. Results : 1. The most chief complaints among the facial palsy patients were eating activity (41%), relationship to other people (29%), communication (20% )(Fig. 3). 2. The most needs of the facial palsy patients was aids for early treat (53%), prevented face deformity (16%) etc, (Fig. 4). 3. So we are going to make a correction splint and rehabilitation set for facial palsy, that makes common use broadly in based of medical utility (CAD. 1~7). Conclusion : We invented a correction splint and rehabilitation set for facial palsied patients in based of questionnaire survey.

  • 1. The inflorescence of the oak has been evolved through a serial reduction of the panicle as is presented in the figure 1. 2. Perianth lobes though they range from one to twelve in a flower six loves occur more than 50% of tested flowers as is tabulated in the table 1. They have been increased partly by a fusion of flowers and partly by a reversion of the stamen into the perianth lobe. They also have been decreased by a fusion of perianth lobes each other. 3. Stamen numbers in a flower range from one to twenty four, but eight stamens are more common than the others as is shown in the table 1. They have been increased by a fusion of flowers and decreased by a fusion of stamens in a flower as are presented in the figure 4. 4. Floral position in the inflorescence, environmental factors, hybridity, and abnormal nutrition supply seem to accelerate fusion and reduction of the stamen in a flower. 5. Flowers which consist of eight stamens and six perianth lobes occur more common than the others, however, the number of the stamen (Y) and the perianth lobes (X) appear to be correlated as follows (see also fig.5.). Y=1.09X+3.78, r=0.4 for Q. dentate Thumb. (7004 flowers investigated) Y=0.71X+4.18, r=0.48 for Q. mongolica Fischer (7409 flowers investigated) Y=1.03X+2.91, r=0.4 for Q. allena B1. (8662 flowers investigated) Y=0.69X+2.00, r=0.4 for Q. serrata Thumb. (9048 flowers investigated) 6. Morphological similarity of the bracteoles and perianth lobes as illustrated in the fig. 3, phenomena of a fusion of bracteoles and separation of perianth lobes into three groups in a flower, and increasing and decreasing phenomena of the stamen number and perianth lobes in a flower as presented in the figures 3 and 4 support the proposal on the oak flower evolution made by H. Hjelmquist in 1948.

  • The present study deals with dynamical relationships between variations of common mackerel and horse mackerel catches in the waters adjacent to Korea for the years 1926 to 1969. The materials used here were obtained from Fishery Statistics published by the Bureau of Fisheries and the Fisheries Research and Development Agency of Korea, Secular variations of common mackerel and horse mackerel catches were classified intc five stages from A to E as shown in Fig. 2. As to variations of catch, the two species have alternating cycle trends occurring every ten years. With regard to the long period trend of catch, there is no correlation between annual variations of the species caught. Dynamical relationships between the two species showed a somewhat different pattern of catch as they transferred from one stage to another of the secular variations.

Identification of Tumor Suppressor Loci on the Long Arm of Chromosome 5 in Primary Small Cell Lung Cancers (원발성 소세포폐암에서 염색체 5번의 장완에 위치한 종양억제유전자좌의 확인)

  • Backgrounds : Recent cytogenetic studies indicated that long of the long arm of chromosome 5 is a frequent event in small cell lung canær (SCLC), suggesting the presence of a tumor suppressor gene in its place. To map the precise tumor-suppressor loci on the chromosome arm for further positional cloning efforts, we tested 15 primary SCLCs. Methods : The DNAs extracted from paraffin-embedded tissue blocks with primary tumor and corresponding control tissue were investigated. Nineteen polymorphic microsatellite markers located in the long arm of chromosome 5 were used in the microsatellite analysis. Results : We found that ten (66.7%) of 15 tumors exhibited LOH in at least one of tested microsatellite markers. Two (13%) of 10 tumors exhibiting LOH lost a larger area in chromosome 5q. LOH was observed in five common deleted regions at 5q. Among those areas, LOH between 5q34-qter and 5q35.2-35.3 was most frequent (75%). LOH was also observed in more than 50% of the tumors at four other regions, between 5q14-15 and 5q23-31, 5q31.1, 5q31.3-33.3, and 5q34-35. Three of 15 tumors exhibited shifted bands in at least one of the tested microsatellite markers. Shifted bands occurred in 2.5% (7 of 285) of the loci tested. Conclusion : Our data demonstrated that at least five tumor-suppressor loci exist in the long arm of chromosome 5 and that they may play an important role in small cell lung cancer tumorigenesis.

  • Background : Loss of the short arm of chromosome 16 is a frequent event in various cancers, which suggests the presence of tumor suppressor gene(s) there. To map precise tumor suppressor loci on the chromosome arm for further positional cloning efforts, we tested 23 primary small cell lung cancers. Method : The DNAs extracted from paraffin embedded tissue blocks with primary tumor and corresponding control tissue were investigated. Twenty polymorphic microsatellite markers located in the short arm of chromosome 16 were used in the microsatellite analysis. Results : We found that six (26.1%) of 23 tumors exhibited LOH in at least one of tested microsatellite markers. Two (8.7%) of 6 tumors exhibiting LOH lost a larger area in chromosome 16p. LOH was observed in five common deleted regions at 16p. Among those areas, LOH between D16S668 and D16S749 was most frequent (21.1%). LOH was also observed at four other regions, between D16S3024 and D16S748, D16S405, D16S420, and D16S753. Six of 23 tumors exhibited shifted bands in at least one of the tested microsatellite markers. Shifted bands occurred in 3.3% (15 of 460) of the loci tested. Conclusion : Our data demonstrated that at least five tumor suppressor loci might exist in the short arm of chromosome 16 and that they may play an important role in small cell lung cancer tumorigenesis.
