팽이버섯 자실체 색택의 유전연구 (Studies on the Inheritance of fruitbody color in Flammulina velutipes)

  • 팽이버섯 야생종과 재배종들의 esterase isozyme 밴드패턴을 비교하고 그 중 특정 야생종 ASI 4019균을 단포자 분리후 교배형을 결정하였다. 단포자의 PDA plate상에 색소 집적과자실체 형성관계를 비교한 결과, 갈색 색소를 집적시킨 단포자와 색소를 집적시키지 않은 단포자간 교배시 갈색 자실체를 형성하였으며 색소를 집적시키지 않은 백색 단포자간 교배한 결과 백색 자실체를 형성하였다. 백색자실체는 재배종의 백색과 색택이 다르게 나타났다. 백색 균사체를 나타내는 단포자와 재배종 단포자를 교배한 결과 연갈색 자실체가 나타났으며 이 Fl의 단포자를 재배종 단포자와 여교배하여 BC1F1을 비교한 결과 1계통은 연갈색 자실체만 형성하고 또 다른 한계통은 연갈색 자실체 8개, 연갈색과 백색 자실체가 혼합된 것이 2개 있었다.

메밀, 감자, 들깨를 이용한 항고혈압 기능성 식이가 정상혈압쥐 및 본태성고혈압쥐에서 혈압 및 혈장지질에 미치는 영향 (Effects of Antihypertensive Diets Mainly Consisting of Buckwheat, Potato, and Perilla Seed on Blood Pressures and Plasma Lipids in Normotensive and Spontaneously Hypertensive Rats)

  • The study was carried out to investigate a new type of functional foods with hypotensive effect which is critical in the prevention and treatment of hypertension and related circulatory diseases. The experimental diets(A, B, C) were prepared from plant based ingredients such as buckwheat, potato, perilla seed with different ratios formulated as an edible form appropriate for human consumption according to AIN-77 standard. Control group(D) was fed commercial rat chow. Twety-four 15-week-old SDR(Sprague Dawley rats) and twenty-four 20-week-old SHR(spontaneously hypertensive rats), weighing 200g respectively, were assigned to 4 treatments of 6 rats each in a completely randomized design. Blood pressure was measured at 7 day interval by tail-cuff sphygmomanometer using an IITC cuff pump and amplifier. The growth rates of both SHR and SDR were not statistically different in comparions with the control except those of diet B and C in SDR and SHR, respectively(p<0.05). When exprimental diets were fed, systolic blood pressure of SDR and SHR at day 28 had been lowered by 17-20 mmHg(ave. of 19.6mmHg for both strains) compared with the reference pressure at day 0. The levels of HDL-cholesterol were increased, while the levels of LDL-cholesterol consistently decreased in both strains when experimental diets were fed(p<.05). Plasma total cholesterol levels were not different among treatments. Plasma triglyceride levels were higher in control diet(commercial rat chow) due to two times higher fat content of control diet itself(p<.05). Atherogenic indices were lower compared to those of control when the experimental diets were fed in both strains (p,.05). The results suggest that the antihypertensive diets mainly consisting of plant-based ingredients may possibly effective in relieving hypertension as well as circulatory diseases.

  • To estimate the effect of selective breeding on the improvement of growth in olive flounder Paralichthys olivaceus, we compared the growth of a strain selected for rapid growth to that of an unselected strain from a commercial hatchery. The fish strains were fed with either moist pellets (MP) (dry matter, 59.8% crude protein; 14.1% lipid) or extruded pellets (EP) (dry matter, 50.4% crude protein; 13.8% lipid) for 190 days and were reared under similar conditions. The mortality rates were less than 2% and were not significantly different among the experimental groups. The growth rate of the selected fish was significantly greater than that of the unselected fish regardless of the diet type, and both strains fed MP grew significantly faster than those fed EP. The selected fish consumed more feed than the unselected fish. However, there was no significant difference in the feed efficiency between the selected and unselected fish. These results demonstrate that the selected fish exhibited superior growth rates, and that this was associated with a greater intake of food. Thus, selective breeding may be useful for improving the growth of commercial olive flounder.

Incorporation of Selected Strains of Pediococcus spp. on Quality Characteristics of Dry Fermented Sausage during Fermentation and Ripening

  • This research investigated the physio-chemical and microbial quality characteristics of dry fermented sausage from selected Pediococcus strains: P. acidiliactici (PE1) and P. pentosaceus (PE2) as compared to commercial starter culture (COS) during fermentation and ripening. Treatments showed no substantial variation (p<0.05) in water activity (aw) values across the study period. PE1 and PE2 treatments exhibited similar (p>0.05) pH values and presented remarkable (p<0.05) lower volatile basic nitrogen (VBN) and thiobarbituric acid reactive (TBARS) content than COS treatment throughout the ripening period. However, the pH values in COS batch were considerably lower than others. PE1 samples presented a significant highest (p<0.05) counts both in lactic acid bacteria (LAB) and total plate count (TPC) than COS and PE2 treatments at 7 days fermentation, and it resulted in a similar and higher TPC count as COS after the ripening period. After the ripening process, treatments are ordered based on LAB counts as follows: COS>PE1>PE2. All batches presented similar redness and yellowness attributes since the 7 days of fermentation and in lightness across the study period. Treatments were similar (p>0.05) in springiness and chewiness traits across the study period and in hardness characteristics in the ripened products. Cohesiveness was higher in PE1 and COS batches. No variation (p>0.05) in aroma and sourness sensory attributes of treatments. The color attribute was highest (p<0.05) in PE1 and PE2 treatments and PE1 had the highest overall acceptability. The overall outstanding merit exhibited by PE1 can be utilized in the commercial production of high-quality dry fermented sausage.

부루세라백신(RB51)의 안전성에 관한 연구 I. Brucella abortus RB51 백신균주의 생화학적 및 유전학적 성상비교 (Studies on the safety of Brucella abortus RB51 vaccine I. Comparison of the biochemical and genetic characteristics of Brucella abortus RB51 vaccine strains)

  • Biochemical and genetic analysis were carried out to investigate the potential recovery of pathogenecity or related mutations of Brucella abortus RB51 vaccine strains. RB51 strains were recovered from commercial vaccines, including related seed stocks from private companies in Republic of Korea, strain from USA, a reference strain from C university and a field isolate (Daehungjin) from aborted dairy cow after RB51 vaccination were compared with two identified virulent wild strains (S2308 and a field strain isolated from dairy cow in Korea) at the same conditions. All the strains examined, except identified pathogenic strains, revealed the identical characteristics to the original RB51 in biochemical properties, antigen and bacteriophage typing. Outer membrane protein (OMP) profiles from strains of RB51 showed the same patterns with standard RB51 in SDS-PAGE. In addition, Western blotting with the brucella specific monoclonal antibody also indicated that all the vaccine strains were completely deficient in their LPS compared to the pathogenic Br abortus strains. The differences in DNA structures among strains were also possible to detect after PCR. All vaccine strains, except S19, S1119-3, S1075, S544 and Br suis, were amplified a 178bp DNA fragment of eri-gene, and 364bp of IS711 elements. In contrast, 498bp DNA product was only found with Br abortus. Overall evidences in the present study confirmed that the RB51 strains for vaccine production in Korea did not originated from the phenomena of possible recovery of pathogenicity or related to any potential mutation event at all.

미각센서를 이용한 산양발효유의 저온저장 중 풍미분석 (Taste Analysis of Fermented Goat Milk during Cold Storage using an Electronic Tongue)

  • 본 연구는 상업유산균인 단일 균주와 복합 균주를 이용한 산양유의 발효형태와 저장 중 풍미변화를 조사한 결과가 있다. 복합 균주는 단일 균주보다 단시간에 있어서 낮은 pH와 높은 산도를 나타내었다. 또한 복합 균주에서는 유산균 수의 증식 속도도 빠르게 나타내었다. 관능검사는 패널의 숙련도에 따라 느끼는 맛이 다르다. 따라서 수많은 시료를 분석할 경우, 재현성이 있는 객관적인 데이터를 얻기 어려운 단점이 있다. 더욱이 우리는 미각센서를 이용하여 저장 중 풍미변화를 측정하였다. 미각센서는 단일 균주와 복합 균주를 이용한 산양발효유에 대하여 사용 균주별 다양한 풍미의 변화가 측정 가능하였다. 따라서 본 연구는 국내에서 처음으로 발효유에 있어서 미각센서를 이용하여 저온 저장기간 동안 맛 성분의 변화를 측정한 자료로 있다. 이러한 결과는 차후 유제품의 소비기한 유지에 대한 자료로서 이용가치가 크게 기대되고 있다.

Natural Lactic Acid Bacteria Population and Silage Fermentation of Whole-crop Wheat

  • Winter wheat is a suitable crop to be ensiled for animal feed and China has the largest planting area of this crop in the world. During the ensiling process, lactic acid bacteria (LAB) play the most important role in the fermentation. We investigated the natural population of LAB in whole-crop wheat (WCW) and examined the quality of whole-crop wheat silage (WCWS) with and without LAB inoculants. Two Lactobacillus plantarum subsp. plantarum strains, Zhengzhou University 1 (ZZU 1) selected from corn and forage and grass 1 (FG 1) from a commercial inoculant, were used as additives. The silages inoculated with LAB strains (ZZU 1 and FG 1) were better preserved than the control, with lower pH values (3.5 and 3.6, respectively) (p<0.05) and higher contents of lactic acid (37.5 and 34.0 g/kg of fresh matter (FM), respectively) (p<0.05) than the control. Sixty LAB strains were isolated from fresh material and WCWS without any LAB inoculation. These LAB strains were divided into the following four genera and six species based on their phenotypic, biochemical and phylogenetic characteristics: Leuconostoc pseudomesenteroides, Leuconostoc citreum, Weissella cibaria, Lactococcus lactis subsp. lactis, Lactobacillus buchneri, and Lactobacillus plantarum subsp. plantarum. However, the prevalent LAB, which was predominantly heterofermentative (66.7%), consisted of Leuconostoc pseudomesenteroides, Leuconostoc citreum, Weissella cibaria, and Lactobacillus buchneri. This study revealed that most of isolated LAB strains from control WCWS were heterofermentative and could not grow well at low pH condition; the selective inoculants of Lactobacillus strains, especially ZZU 1, could improve WCWS quality significantly.

개 생균제 사용에 적합한 유산균주의 선발 및 면역활성 평가 (Selection and immunomodulatory evaluation of lactic acid bacteria suitable for use as canine probiotics)

  • This study was conducted to isolate lactic acid bacteria (LAB) from dog intestine and identify potential probiotic strains for canine use. One hundred and one LAB were isolated from feces of 20 healthy dogs. Acid, bile, and heat resistance along with adherence to Caco-2 cells and antimicrobial activity against pathogens were examined. To analyze immunomodulative effects, the production of nitric oxide (NO), TNF-${\alpha}$, and IL-$1{\beta}$ was measured using RAW 264.7 macrophages. Additionally, RAW BLUE cells were used to evaluate nuclear factor-${\kappa}B$ (NF-${\kappa}B$) generation. Ultimately, three strains were selected as canine probiotics and identified as Lactobacillus reuteri L10, Enterococcus faecium S33, and Bifidobacterium longum B3 by 16S rRNA sequence analysis. The L10 and S33 strains showed tolerance to pH 2.5 for 2 h, 1.0% Oxgall for 2 h, and $60^{\circ}C$ for 5 min. These strains also had strong antimicrobial activity against Escherichia coli KCTC 1682, Salmonella Enteritidis KCCM 12021, Staphylococcus aureus KCTC 1621, and Listeria monocytogenes KCTC 3569. All three strains exerted better immunomodulatory effects than Lactobacillus rhamnosus GG (LGG), a well-known commercial immunomodulatory strain, based on NO, NF-${\kappa}B$, IL-$1{\beta}$, and TNF-${\alpha}$ production. These results suggested that the three selected strains could serve as canine probiotics.

Evaluation of Yeast Diversity During Wine Fermentations with Direct Inoculation and pied de cuve Method at an Industrial Scale

  • The diversity and composition of yeast populations may greatly impact wine quality. This study investigated the yeast microbiota in two different types of wine fermentations: direct inoculation of a commercial starter versus pied de cuve method at an industrial scale. The pied de cuve fermentation entailed growth of the commercial inoculum used in the direct inoculation fermentation for further inoculation of additional fermentations. Yeast isolates were collected from different stages of wine fermentation and identified to the species level using Wallersterin Laboratory nutrient (WLN) agar followed by analysis of the 26S rDNA D1/D2 domain. Genetic characteristics of the Saccharomyces cerevisiae strains were assessed by a rapid PCR-based method, relying on the amplification of interdelta sequences. A total of 412 yeast colonies were obtained from all fermentations and eight different WL morphotypes were observed. Non-Saccharomyces yeast mainly appeared in the grape must and at the early stages of wine fermentation. S. cerevisiae was the dominant yeast species using both fermentation techniques. Seven distinguishing interdelta sequence patterns were found among S. cerevisiae strains, and the inoculated commercial starter, AWRI 796, dominated all stages in both direct inoculation and pied de cuve fermentations. This study revealed that S. cerevisiae was the dominant species and an inoculated starter could dominate fermentations with the pied de cuve method under controlled conditions.

재조합 lysozyme-HJP34 단백질의 다양한 병원성 세균에 대한 항균 효능 및 자돈 사료첨가제의 가능성 평가 (Antibacterial evaluation of recombinant lysozyme-HJP24 proteins against various bacterial pathogens, and of its possibility test as a feed additive in piglets)

  • The recombinant lysozyme-HJL34 proteins were expressed and purified using commercial Escherichia (E.) coli expression system. Stx2e+ F18+ E. coli, Actinobacillus pleuropneumoniae (APP), Streptococcus (S.) suis, and Clostridium (C.) perfringens strains were isolated from pigs. The minimum inhibitory concentrations (MICs) of the recombinant lysozyme-HJP34 proteins were examined by means of the microtiter plate method, according to the NCCLS recommendations. The possibility of its as the alternatives to antibiotics was tested in piglets. The MICs were determined as 75 ㎍/mL, 300 ㎍/mL, 75 ㎍/mL, 35.5 ㎍/m against Stx2e+ F18+ E. coli, APP, S. suis, C. perfringens, respectively. A total of 25 piglets were divied 5 groups. The piglets in group A~C were fed with commercial feed and those in groups D, E were fed with commercial feedstuff. All piglets in groups B~E were challenged with virulent Stx2e+ F18+ E. coli, APP, S. suis strains. Groups C and D were treated with antimicrobial from 24 h after challenge. All piglets in group B died within 3 days after challenge. Among 5 piglets in groups C and D piglets, 80% survived after challenge. Among group E piglets, 60% were alive until the end of this study. Therefore, this study indicates that recombinant lysozyme-HJP34 proteins is a suitable possibility as a feed additive for reduction of diseases by bacterial pathogens in piglet feed.
