Non-Aflatoxigenicity of Commercial Aspergillus oryzae Strains Due to Genetic Defects Compared to Aflatoxigenic Aspergillus flavus

  • Aspergillus oryzae is generally recognized as safe, but it is closely related to A. flavus in morphology and genetic characteristics. In this study, we tested the aflatoxigenicity and genetic analysis of nine commercial A. oryzae strains that were used in Korean soybean fermented products. Cultural and HPLC analyses showed that none of the commercial strains produced detectable amount of aflatoxins. According to the molecular analysis of 17 genes in the aflatoxin (AF) biosynthetic pathway, the commercial strains could be classified into three groups. The group I strains contained all the 17 AF biosynthetic genes tested in this study; the group II strains deleted nine AF biosynthetic genes and possessed eight genes, including aflG, aflI, aflK, aflL, aflM, aflO, aflP, and aflQ; the group III strains only had six AF biosynthetic genes, including aflG, aflI, aflK, aflO, aflP, and aflQ. With the reverse transcription polymerase chain reaction, the group I A. oryzae strains showed no expression of aflG, aflQ and/or aflM genes, which resulted in the lack of AF-producing ability. Group II and group III strains could not produce AF owing to the deletion of more than half of the AF biosynthetic genes. In addition, the sequence data of polyketide synthase A (pksA) of group I strains of A. oryzae showed that there were three point mutations (two silent mutations and one missense mutation) compared with aflatoxigenic A. flavus used as the positive control in this study.

Comparative Study on the Resistance of Three Commercial Strains and Balady (Local) Breed of Chickens to Infection With Salmonella gallinarum

  • Three commercial strains bred in the US (Hubbard), Holland (Lohman), and Canada (Shaver) and Balady breed of chickens were orally infected with $10^6$ cfu of Salmonella gallinarum per chick. Chicks were compared for weight gain, feed intake, feed conversion ratio, mortality rate and contamination of the liver, spleen and intestine with Salmonella gallinarum on the day of slaughter. The Balady chicks had significantly the lowest mortality rate (p<0.01) and weight gain (p<0.001) and poorest feed conversion ratio (p<0.001). Salmonella gallinarum could be shed from the liver, spleen and intestine with alternative rates in all strains. Lohman was the most resistance over the other two commercial strains, while Hubbard had the highest susceptibility. The Balady chicks were the most resistance. However, they may serve as a possible reservoir of Salmonella gallinarum and it may play a role of spreading the infection to the commercial farms in Jordan.

Characteristics of Peach Wine with Different Commercial Yeast Strains (시판 와인 효모에 대한 복숭아주의 발효 특성)

  • This study investigated the effects of different commercial yeast strains on the characteristics of peach wine. Peach fruit was inoculated with Fermivin, K1-V1116, EC-1118, D-47 and AR2. Peach wines fermented with the D-47, K1-V1116, and AR2 strains showed faster fermentation than wines fermented with the other strains. Wine fermented with EC-1118 had the lowest titratable acidity and highest pH. The ethanol content of wines fermented with different commercial yeast strains was in the range of 13~14%. In sensory evaluation, the wine made with EC-1118 showed the highest overall scores in color and acceptability.

Toxin Genes and Antimicrobial Resistance of Clostridium perfringens Strains Isolated from Commercial Jeotgals (시판 젓갈에서 분리한 Clostridium perfringens의 독소 유전자 및 항균제 내성 분석)

  • Clostridium perfringens causes diarrhea and other diseases in humans and animals. We investigated the prevalence, toxin gene profiles, and antimicrobial resistance of C. perfringens isolated from commercial jeotgal sample. C. perfringens was isolated from 11 of 22 commercial jeotgals. All C. perfringens strains were positive for the alpha toxin gene, but not for the beta, epsilon, iota, CPE or NetB toxin genes; therefore, all strains were identified as type A C. perfringens. However, the beta2 toxin gene was identified in 54.5% of isolates. Disk diffusion susceptibility tests showed that most isolates were resistant to kanamycin (90.9%), nalidixic acid (72.7%), oxacillin (54.5%), erythromycin (27.3%), ciprofloxacin (9.1%) and clindamycin (9.1%). However, all strains were susceptible to 14 other antimicrobial including amoxicillin, ampicillin, and chloramphenicol. The average minimum inhibitory concentrations against C. perfringens of clindamycin, kanamycin, and nalidixic acid were 128.0, 128.0, and 54.0 ㎍/mL, respectively. These results provide new insight into the necessity for sanitation of commercial jeotgal, and provide evidence to help reduce the risk of contamination with antimicrobial-resistant bacteria.

Antibiotic Resistance of Pectobacterium Korean Strains Susceptible to the Bacteriophage phiPccP-1

  • Commercial products with antibiotics like streptomycin as active ingredients have been used to control soft rot disease caused by Pectobacterium species for a long time. In this study, antibiotic resistance of twenty-seven Korean strains of Pectobacterium species including P. carotovorum, P. odoriferum, P. brasiliense, and P. parmenteri, which were previously shown to be susceptible to the bacteriophage phiPccP-1 was surveyed using a disk diffusion assay. While all strains were highly susceptible to ampicillin, kanamycin, chloramphenicol, tetracycline, and rifampicin, some strains showed weak susceptibility to 300 ㎍/ml of streptomycin. Furthermore, some of them are partially or completely resistant to commercial pesticides-Buramycinand streptomycin at the concentration of 250 ㎍/ml that is recommended by the manufacturer for streptomycin-based pesticides. These results indicate the presence of streptomycin-resistant Pectobacterium strains in South Korea, and the development of antibiotic alternatives to control soft rot is needed.

Assessment of Characteristics and Functional Properties of Lactobacillus Species Isolated from Kimchi for Dairy Use

  • The objective of this study was to identify lactic acid bacteria (LAB) isolated from kimchi and to evaluate its characteristics and functional properties for application in fermented dairy products as a probiotic or commercial starter culture. Eight stains isolated from kimchi were selected through an investigation of phenotypic characteristics. Two strains (DK211 and DK303) were identified as Lactobacillus plantarum, another two (DK207 and DK215) as Lactobacillus paracasei, and one (DK301) as Lactobacillus sakei. The remaining three strains were identified as species of Weissella. All selected Lactobacillus strains had acid and bile tolerance, even though there was wide variation in the ability of each strain. DK303 showed a remarkably higher proteolytic activity. There were no significant differences in β-galactosidase activity among the tested strains, except that DK301 showed no activity. Auto-aggregation varied between 82.1 and 90.0%, and hydrophobicity values ranged from 0.5 to 51.6%.The strongest auto-aggregation and hydrophobicity were observed in DK211. All selected strains showed better 1,1-diphenyl-2-picrylhydrzyl (DPPH) scavenging activity than commercial strains. DK211, DK215, DK301, and DK303 had effective inhibitory activity against all pathogens tested except E. coli. When selected strains were used for yogurt preparation as a single starter culture, the time required to reach target titratable acidity (0.9) was 11-12 h. The yogurt fermented with DK211 had favorable panelists ratings for most sensory attributes, which were comparable with yogurt fermented with a commercial strain. The results suggest that strains isolated from kimchi could be potential probiotic and starter cultures for use in yogurt manufacturing.

Trends of commercial strain development and spawn industry in mushrooms (버섯의 품종 육성과 종균 산업의 동향)

  • According to UPOV (International Union for Protection of New Varieties of Plants), mushroom spawn market have to be opened by the year 2009. Number of commercial strains distributed by the year 2005 were 179 of 24 species of edible and medicinal mushrooms. Only nine strains of oyster mushrooms were registered as protected variety, which is not compatible with those recorded in other advanced countries. Meaning of spawn in broad sense contains commercial strains. Development of commercial strains faces two main problems in Korea : One is the complicated genetic patterns and sexuality of mushroom species, and the other is expensive experimental equipments and fruiting body growing houses. Resolution of these problems leads to development of mushroom strains. This could be achieved as follows; genetic resources collection and assessment, molecular characterization of useful genetic characters, development of new commercial strains by hybridization using typical genetic resources, strengthening of breeding research using "Mushroom Breeding Group", management of spawn research company by consortium, foundation of mushroom general industry, promotion of consumption, and upgrade of competition ability for other countries. Antibiotic Resistant Characteristics of Bifidobacterium from Korean Intestine Origin and Commercial Yoghurts (한국인 장관과 유산균 식품 유래 Bifidobacterium의 항생제 내성 특성)

  • To obtain antibiotic resistant profiles of Bifidobacterium, minimum inhibitory concentrations (MIC) of 14 antibiotics for 93 Bifidobacterium isolates from Korean intestine origin were determined. All strains tested were sensitive to chloramphenicol, rifampicin, and amoxicillin, whereas resistant to aminoglycoside family, nalidixic acid, and vancomycin. Among vancomycin-resistant strains, 34% were resistant at more than $100\;{\mu}g/mL$, and showed variant resistances toward tetracycline, erythromycin, and penicillin. Their resistances against penicillin, cephalothin, and tetracycline were higher than ten years ago. MIC of ten isolates from commercial yoghurt products were very similar to those of strains from Korean intestine origin, and 20% strains showed resistance at higher than $100\;{\mu}g/mL$ vancomycin. These results indicated patterns of antibiotic resistance against Bifidobacterium from Korean intestine origin and commercial yoghurts were very similar,and prevalence of vancomycin resistance for Bifidobacterium was 20%. To develop new probiotic, antibiotic resistance of vancomycin and risks involved should be evaluated.

Profiles of Toxin Genes and Antimicrobial Resistance of Bacillus cereus Strains Isolated from Commercial Jeotgal (시판 젓갈에서 분리한 Bacillus cereus의 독소 유전자 및 항균제 내성 분석)

  • Twenty-three Bacillus cereus strain isolated from commercial jeotgal were investigated for 11 toxin genes and susceptibility to 25 different antimicrobials. The hemolytic enterotoxins hblA, hblC, and hblD were detected in 13.0%, and non-hemolytic enterotoxins nheA, nheB, and nheC were detected in 26.1%, 100%, and 100% of the isolates, respectively. The positive rates of cytK, entFM, becT, hlyII, and ces were 73.9%, 60.9%, 26.1%, 8.7%, and 0.0%, respectively. According to the disk diffusion susceptibility test, all of the strains studied were resistant to cefuroxime, followed by cefoxitin (78.3%), oxacillin (78.3%), ampicillin (69.6%), penicillin G (69.6%), and amoxicillin (65.2%). However, all the strains were susceptible to 11 other antimicrobials, including amikacin, chloramphenicol, and ciprofloxacin. The average minimum inhibitory concentrations of amoxicillin, ampicillin, and cefuroxime against B. cereus were 462.9, 235.0, and 135.0 ㎍/mL, respectively. These results highlight the need for sanitizing commercial jeotgal, and provide evidence to help reduce the risk of jeotgal contamination by antimicrobial-resistant bacteria.

Differentiation Among Commercial Strains of Pleurotus spp. Based on DNA Fingerprinting Using Universal Rice Primer (URP) (Universal Rice Primer(URP)에 의한 DNA 핵산지문법을 이용한 느타리의 유통 품종간 구분)

  • To distinct the commercial strains in Pleurotus spp., 81 strains in eight Pleurotus species were used. DNA fingerprinting using URP-PCR was conducted to determine the phylogenetic relationships among Pleurotus strains. DNA profiles of Pleurotus species obtained by twelve URP primers were analyzed for genetic similarity by NTSYS program. We could divide strains into ten clusters, in which three of them belong to P. ostreatus and the others to the different species, respectively. At the 76% similarity level, 70 P. ostreatus strains were distinguished into three clusters. Cluster I contained 35 strains and some of them showed almost 100% similarity, one strain closely related to Weonhyeong and six strains closely related to Wangheukpyeong. In cluster II, twenty-one out of 23 strains showed 100% to Suhan. Cluster III contained twelve strains, including six strains closely related to Chunchu-2. The results suggested that there are many same strains with different names in mushroom spawn market.
