재래식 및 개량식 고추장 효모의 분포 및 생리특성 (Distribution and Physiological Characteristics of Yeasts in Traditional and Commercial Kochujang)

  • 개량식 고추장으로부터 동정한 효모들은 Candida glabrata C. guilliermondii. C. humicola. C. rugosa, C. zeylanoides, Cryptococcus uniguttulatus, Pichia farinosa, Rhodotorula glutinis, Saccharomyces cerevisiae Zygosaccharomyces rouxii로 분류되었다. 재래식 고추장에서 동정한 효모는 C. glabrata, P. farinosa, S. cerevisiae, Z. rouxii로 분류되었으며 분포율이 높아 발효숙성에 깊이 관여할 것으로 생각되는 효모는 재래식과 개량식고추장에서 Candida, Pichia, Saccharomyces, Zygosaccharomyces등 4속이었다. 개량식 고추장의 발효숙성 중기와 후기에 나타난 Z. rouxii와 S. cerevisiae는 고추장용 메주에서 분리되어 그 기원이 되는 것으로 생각되며, 특히 Z. rouxii는 고추장용 메주에서 분포된 효모중 46%를 차지하였다. 발효숙성중 분포는 Z. rouxii가 15일 후에 33%, S. cerevisiae가 21일 후에 13%펄 각각 가장 높은 분포율을 보였다. Candida속의 경우 개량식 메주, 소맥코지, 콩코지 모두에서 분리되었으나 고추장의 발효숙성이 진행됨 에 따라 40%에서 10%로 점차 감소하는 경향을 보였다. 재래식 고추장에서는 발효음성 3개월 후에 Z. rouxii가 53%, 7개월 후에 S. cerevisiae가 26%로 가장 높은 분포율을 보였다. NaCl 과 glucose를 혼용한 배지에서 S. cerevisiae와 Z. rouxii 등 2종의 효모가 다른 효모에 비해 생육이 현저히 나타난 것은 이들이 고추장의 발효숙성 과정에서 분리된 빈도수가 높았다는 것과 무관하지 않을 것으로 사료된다. 특히 S. cerevisiae와 Z. rouxii 두 균주는 무염 또는 7% NaCl이 첨가된 배지에서 가장 먼저 가스를 생성하였으며 7% NaCl이 첨가된 조건하에서의 가스생산량은 Z. rouxii 및 S. cerevisiae 각각에서 0.024 ml/ml/hr, 0.013ml/ml/hr로 나타났다.

시판 메밀차 열수 추출물의 항산화 및 신경세포 보호효과 (Neuronal Cell Protection and Antioxidant Activities of Hot Water Extract from Commercial Buckwheat Tea)

  • 본 연구에서는 시판 메밀차 열수 추출물의 항산화 효과 및 신경세포 보호효과를 조사하였다. 시판 메밀차 열수 추출물의 ABTS 라디칼 소거 활성, FRAP 및 MDA 생성 저해 실험결과 농도 의존적인 경향이 나타났으며 또한 높은 항산화 활성을 보여주었다. 과산화수소로 유발된 산화적 손상에 의한 ROS 축적량을 조사한 결과 $H_2O_2$ 단독 처리구보다 메밀차 열수 추출물 처리구에서 낮은 ROS 축적량을 나타내었다. MTT 및 LDH 분석을 통한 PC12 세포 중의 신경세포 보호효과를 측정한 결과 MTT 분석에서는 시판 메밀차 열수 추출물의 모든 농도에서 높은 세포 생존율을 나타냈고, LDH 분석에서는 추출물에 의한 농도 의존적인 세포질 효소 (LDH) 방출량 감소가 관찰되었다. 총 페놀성 화합물, rutin 및 quercitrin의 함량은 각각 9,608.10 mg/g, 13.42 및 0.90 mg/100 g이었다. 본 연구결과를 종합해 볼 때 rutin 및 quercitrin과 같은 다양한 페놀성 화합물을 함유한 시판 메밀차 추출물은 항산화 활성과 산화적 스트레스로 유발된 신경세포 보호효과를 나타내어 퇴행성 신경질환 등을 예방 할 수 있는 기능성 식품 소재로서의 활용 가치가 높을 것으로 판단된다.

Seedborne Fungi and Fungicide Seed Treatment of Ginseng

  • The incidence of fungi and their possible contribution to low vigour were examined in a collection of ginseng (Panax quiquefolius) seed from Ontario. When examined after one winter of stratification in the field and storage at 4f for five months in the laboratory, the collection exhibited low vigour (plant stand 16.7% of seeding rate six weeks after seeding) and high incidence (94%) of discolored or soft seed. Fungi isolated (and incidence) from 1,304 endosperm halves recovered from surface-sterilized seed were, in order of abundance, Fusarium rostrum (22.2%), Chaetomium crispuum (14.3%), Funriud oxysporum (9.0%), Fusarium sdani (9.0%), iwmor sp. (8.4%), Alternaria sp. (8.1%), Zowieua lucotricha (7.8%), Cylindruarpn sp. (0.9%), Fusarium avenacmm (0.9%), and Vdudla iliata (0.4%). Most of these fungi, including known and potential pathogens of ginseng (species of Alerraria, Cylindrocarpon, Fusarium, and Trichodirma), were associated with both healthy and diseased seed. Application of Benlate (benomyl), Thiram (thiram), or UBI-2584 (tebuconazole) to seed caused slight to pronounced reduction in emergence and did not significantly affect plant stand six weeks after seeding. The study demonstrated the high level of infection by fungi, including known and potential pathogens of the cry, in an arbitrary collection of ginseng seed from commercial sources, and the phytotoxicity of the fungicides tested when applied to moist stratified seed. The lack of efficacy of the fungicides precluded determination of the contribution of seedborne fungi to low vigour of the seed.

Biochemical Performance and Quantitative Assessment of F1 Hybrid of Two Ecoraces of Tropical Tasar Silkworm Antheraea Mylitta Drury (Lepidoptera: Saturniidae)

  • Antheraea mylitta Drury is basically a crossbreeding species, as such it seems to be potentially a good material for the exploitation of heterosis. In the present study F1 hybrid of wild ecorace Laria (L) and semi-domestic Daba (D) was raised and evaluated for various quantitative traits and biochemical parameters during larval stage. Improved fecundity ($+18{\pm}1.8%$ and higher egg hatching rate ($+10.96{\pm}1.3%$) was recorded in the F1hybrid ($L{\times}D$). Biochemical parameters studied in the hemolymph, midgut and fatbody of the larva showed significantly higher (P<0.05) total proteins and carbohydrate concentration besides digestive enzyme activity. Correspondingly SDS-PAGE revealed more number of protein bands in the hemolymph sample of F1s, ranging between 29 kDa to 66 kDa compared to parental lines. The present study demonstrates the positive heterosis effect in the F1 hybrid of Laria ${\times}$ Daba. Biochemical analysis indicates that, there is possibilities of exploitation of hybrids with specific parents targeted for desirable commercial traits (silk yield and fecundity). Moreover, most of these biochemical parameters can be used as markers to analyze the genetic improvement in the tasar silkworms.

Current Status of Plasmodiophora brassicae Researches in Korea

  • Clubroot disease is caused by the soil-born obligate plant pathogen Plasmodiophora brassicae. This pathogen can infect all cruciferous vegetables and oil crops, including Brassica rapa, B. oleracea, B. napus, and other Brassica species. Clubroot disease is now considered to be a major problem in Chinese cabbage production in China, Korea, and Japan. We collected several hundreds of P. brassicae infected galls from Korea, and isolated the single spore from the collection. For establishment of novel isolation, and mass-propagation methods for singe spore isolates of P. brassicae pathogen, we developed new filtration method using both cellulose nitrate filter and syringe filter. Accurate detection of P. brassicae pathogen in the field was done by using real-time PCR in the potential infested soil. When we tested the different pathogenicity on commercial Chinese cabbage varieties, P. brassicae from collected galls showed various morphological patterns about clubroot symptom on roots. To date, 8 CR loci have been identified in the B. rapa genome using the quantitative trait loci (QTL) mapping approach, with different resistant sources and isolates. We are trying to develop the molecular marker systems for detect all 8 CR resistant genes. Especially for the study on the interaction between pathogens and CR loci which are not well understood until now, genome wide association studies are doing using the sequenced inbred lines of Chinese cabbage to detect the novel CR genes.

  • Predicting coal combustion by computational fluid dynamics (CFD) requires a combination of complicated flow and reaction models for turbulence, radiation, particle flows, heterogeneous combustion, and gaseous reactions. There are various levels of models available for each of the phenomena, but the use of advanced models are significantly restricted in a large-scale boiler due to the computational costs and the balance of accuracy between adopted models. In this study, the influence of coal devolatilization model and turbulent mixing rate was assessed in CFD for a commercial boiler at 500 MWe capacity. For coal devolatilization, two models were compared: i) a simple model assuming single volatile compound based on proximate analysis and ii) advanced model of FLASHCHAIN with multiple volatile species. It was found out that the influence of the model was observed near the flames but the overall gas temperature and heat transfer rate to the boiler were very similar. The devolatilization rate was found not significant since the difference in near-flame temperature became noticeable when it was multiplied by 10 or 0.1. In contrast, the influence of turbulent mixing rate (constant A in the Magnussen model) was found very large. Considering the heat transfer rate and flame temperature, a value of 1.0 was recommended for the rate constant.

Isolation and Characterization of Microbial Strains with Hydrolytic Enzyme Profile from Clay Minerals

  • A total of 262 bacterial strains were isolated from clay minerals, bentonite and zeolite, in Gyeongsangbukdo, Republic of Korea, and their hydrolytic enzyme activities were analyzed. Most of the isolated strains belonged to Micrococcales and Bacillales order. Of strains, 96 strains produced α-amylase activity, 42 strains showed cellulase activity, 111 strains had pectinase activity, and 70 strains showed protease activity. Among them, 177 isolates exhibited one or more of the hydrolytic enzyme activities and in particular Bacillus cereus MBLB1321, B. albus MBLB1326 and KIGAM017, B. mobilis MBLB1328, MBLB1329 and MBLB1330 showed all of the enzyme activities. These results demonstrate the diversity of functional Bacillus species in clay minerals as vital sources for the discovery of industrially valuable hydrolytic enzymes, which have a great commercial prospect in various bio-industrial applications.

Penetrating behavior of target prawns (Sicyonia penicillata) contacting netting panels in an experimental water tunnel

  • Capture efficiencies of commercial shrimp trawls may improve if their designs took into better account behavioral responses of wild shrimp to approaching cod-end of the trawls. Here we report results of water tunnel-based experimental studies of responses of wild California target prawns to several different near-realistic netting configurations over a range of water velocities (0.3-0.7 m/s). Netting panels were oriented at parallel to water flows (FP) on the bottom of test section, vertical (VT) or diagonal sloping backward (DG), bottom to top. Behavioral responses were recorded by video camera and analyzed frame by frame. Measured responses included rates of penetrating through netting by behavioral features and tail-flip frequencies. Frequencies of prawn passing through the nets increased with flow speed for both orientations and were higher at given speeds for sloped nets. Other behavioral features (e.g., passage head-or tail-first) also varied significantly with water velocities and netting orientation. Interactions of penetrating rates between netting orientations and flow speeds also were significantly dependent, except for prawn size. Additional studies are needed of other shrimp species and at higher water velocities more similar to actual field operations using trawls to improve size selectivity.

황금·황련 추출물의 항균활성이 막걸리 저장성에 미치는 영향 (Antimicrobal Activity of Sutellaria baicalensis·Coptidis rhizoma Extract on the Preservation of Makgeolli)

  • This research was carried out to find herbal preservatives for Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. When Makgeolli was kept at $25^{\circ}C$ to find the changes in acidity, total microbial cell number, yeast cell number, and bacterial species variety, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, and Makgeolli was changed to complete off-flavor. Acetobacter pasteurianus is the main acidifier in Makgeolli and shows a synergy effect in acid formation when cultured in combination with Lactobacillus casei. Among 12 herbs, the ethanol extract of Sutellaria baicalensis showed antimicrobial activity against A. pasteurianus, whereas the ethanol extract of Coptidis rhizoma showed antimicrobial activity against L. casei. Makgeolli added with Sutellaria baicalensis extracts demonstrated a lower acidity than that with Coptidis rhizoma extracts, which indicates that the inhibition of an acetic acid former is more important than that of a lactic acid former in Makgeolli preservation. Sutellaria baicalensis extracts prolonged the shelf life of Makgeolli by 1~2 weeks at a minimal inhibitory concentration ($0.63mg/m{\ell}$) during storage at $10^{\circ}C$.

단백질 분해효소억제제를 이용한 담배의 품질평가 (Evaluation of Cigarette Quality by Use of ??1-Protease Inhibitor)

  • Current studies indicated that emphysema in smokers might be due, in part, to the local suppression of G, -protease inhibitor(u, -Pl) in lung by reactive oxygen species in cigarette smoke or smoke-activated lung neutrophiles. In the present works, we examined the possibility that a measure which inactivated $\alpha$l-Pl by cigarette smoke could be an alternative method to evaluate the cigarette quality, In order to determine the inactivation of $\alpha$1, -Pl, trypsin inhibitory capacity(TIC) was assayed. A rapid loss of $\alpha$1, -Pl activity occurred when $\alpha$1-Pl solutions was exposed the gas phase or total particulate matter(TPM) obtained from various brands. The inactivation of $\alpha$1-Pl by gas phase was dependent upon the number of puffs and the age of the smoke. However, that by TPM was rather decreased since 2 puffs and also showed no more change over 24hrs after exposing. Inactivation of $\alpha$1-Pl determined by our suggested method(5 puffs, 24hours of aging after exposing) using various commercial cigarettes exhibited that high tar brands has inactivated it more strongly than low tar cigarettes. But the ability of some brands to inactivate $\alpha$1-Pl does not correlate with the content of tar or nicotine. These results so여esc that the degree of $\alpha$1-Pl inactivation by cigarette smoke may be a useful index for the evaluation of cigarette quality and that it should be also contribute to the manufacture of less hazardous cigarettes.
