  • Douglas-fir (Pseudotsuga menziesd Mirb. Franco) is highly regarded as a commercial timber species throughout the world in part due to its fast growth relative to many other species. In this study, basal area per hectare equation for Douglas-fir plantations in Southland of New Zealand has been developed based on medium measurement cycles of permanent sample plots data set. The function was developed using the algebraic difference equation method, and various sigmoid-shaped projection equations were used. Parameter estimation was obtained by non-linear routine of the SAS. As a result, of the functions tested a variant of the Schumacher polymorphic function including site index and thinning term as predictor variables showed the higher precision of the fitting. The results indicate that site index is positively correlated with basal area growth. And the thinning term was found to be useful to increase precision of the model.

Comparative Assessment of Diagnostic Performances of Two Commercial Rapid Diagnostic Test Kits for Detection of Plasmodium spp. in Ugandan Patients with Malaria

  • Prompt diagnosis of malaria cases with rapid diagnostic tests (RDTs) has been widely adopted as an effective malaria diagnostic tool in many malaria endemic countries, primarily due to their easy operation, fast result output, and straightforward interpretation. However, there has been controversy about the diagnostic accuracy of RDTs. This study was conducted to evaluate the diagnostic performances of the 2 commercially available malaria RDT kits, RapiGEN Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) for their abilities to detect Plasmodium species in blood samples collected from Ugandan patients with malaria. To evaluate the diagnostic performances of these 2 RDT kits, 229 blood samples were tested for malaria infection by microscopic examination and a species-specific nested polymerase chain reaction. The detection sensitivities for P. falciparum of Malaria Ag Pf/Pv (pLDH/pLDH) and Asan $EasyTest^{TM}$ Malaria Ag Pf/Pv (HRP-2/pLDH) were 87.83% and 89.57%, respectively. The specificities of the 2 RDTs were 100% for P. falciparum and mixed P. falciparum/P. vivax infections. These results suggest that the 2 RDT kits showed reasonable levels of diagnostic performances for detection of the malaria parasites from Ugandan patients. However, neither kit could effectively detect P. falciparum infections with low parasitaemia (<$500parasites/{\mu}l$).

Purification and Identification of Paenibacillus sp., Isolated from Diseased Larvae of Allomyrina dichotoma (Linnaeus, 1771) (Coleoptera: Scarabaeidae) in Insect Farms

  • In reared populations of Allomyrina dichotoma, commercial insects, the skin of last instar larvae was changed softer with opaque white, and infested grubs eventually died. To clarify the cause of the symptom, we collected the larvae of A. dichotoma from five farms and examined their intestinal bacterial florae using pyrosequencing technique. From those results, a member of Paenibacillus was found only in the larvae showing the symptom of disease. Through PCR analysis using a Paenibacillus specific primer set, we obtained the partial 16S rRNA gene sequence and confirmed the microbe as Paenibacillus sp. For clear identification, a whole guts was extracted from each larva showing the sign of the disease and incubated at $70^{\circ}C$ for 15 min to isolate spore forming bacteria. After then, each content of guts was cultured on $MYPGP_{NAL}$ agar medium($12.5{\mu}g/ml$ of nalidixic acid) at $30^{\circ}C$. The 16S rRNA gene sequence analysis for the isolated bacteria showed that they were closely related to P. rigui(97.9% similarity), to P. chinjuensis(96.1% similarity), and to P. soli(95.3% similarity). Additional tests including API test and cellular fatty acid composition analysis were performed, but the strain couldn't be identified at species level, suggesting it may represent novel species of the genus Paenibacillus.

A Study on the Use of Wood Waste for Slope Revegetation Techniques (임목폐기물 파쇄칩을 활용한 녹화기술의 비탈면 적용성에 관한 연구)

  • The main objectives of this comparative study were 1) to quantify the potential contribution of wood chips to revegetation on the cut slope in a constructed area, 2) to compare the floristic similarity of species composition whether existing of wood chips in the soil media or not. Wood chips were combined with soil media in the ratio of 30%. The thin layer (1 cm, 3 cm) soil media revegetation methods and the thick layer (10 cm) soil media revegetation method were constructed on the cut slopes by machineries respectively. Also, comparative experiments the existence of wood chips in the soil media were conducted to evaluate the effects of wood chips on revegetation works. The total size of quadrat was $300m^2$ ($100m^2{\times}3$). The results of monitoring for 3 years on cut slopes were as follows: 1) All the quadrat existed in the proper range for vegetation. 2) Species richness of quadrat including wood chips was 10% more than those of commercial soil medias. 3) The coverage rates of quadrat including wood chips were similar or superior to those of conventional methods. It can be concluded that the use of wood chips would be effective to promote establishment of diverse landscape and vegetation. However, it behooves to continue monitoring on succession of vegetation for ecological restoration. Finally, adequate wood chips deposit and gathering methods should be studied properly.

Morphological Characteristics of Medium-Leaf Type Zoysiagrasses (Zoysia spp.) and Their Classification Using RAPDs (중엽형 한국잔디(Zoysia spp.)류의 형태적 특성과 RAPDs를 이용한 분류)

  • Zoysiagrass, especially Jungji implicating medium-leaf type zoysiagrass has been widely used in South Korea recently. This study was carried out to classify 36 selected medium-leaf type zoysiagrass accessions compared to 5 basic zoysiagrass species using RAPDs. Morphological characteristics such as leaf width, leaf angle, leaf sheath length, existance of trichomes and stolon color were measured as useful characteristics for identification of species in Zoysia genus. Nineteen RAPD markers were identified using 8 selected random primers. The dissimilarity coefficient of variants ranged from 0 to 0.736. Three zoysiagrass groups were clustered by dissimilarity coefficient analysis. Group 1 consisted of Z. japonica and some US varieties including 'Zenith' and 'Meyer'. Group 2 consisted of Z. sinica, Z. macrostachya and Korean commercial varieties such as 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type grasses. Group 3 was genetically distinct from Group 1 and Group 2, and included Z. matrella and Z. tenuifolia. 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type zoysiagrasses showed very close genetic relationship with Z. sinica and Z. macrostachya.

Body Weight Changes of Laboratory Animals during Transportation

  • The majority of laboratory animals were transported from commercial breeders to a research facility by ground transportation. During the transportation, many biological functions and systems can be affected by stress. In this experiment, the change of body weight during the transportation was measured and the recovery periods from the transportation stress established based on the body weight changes. Total 676 laboratory animals which were aged between 3 to 9 wk old were studied. The transportation time taken from container packing to unpacking the container was approximately 24 h. The temperature of animal container was constantly maintained by air-conditioning and heating equipment. Rats were found to be more sensitive than mice. The body weight of rats was significantly decreased 3.71% (p<0.05) compared to the body weight of mice which decreased 0.9% There was no significant difference between the strains in the same species. When the changes of body weights were compared between delivery days, C57BL/6 mice showed the most variable changes compared to other species and strains. Consequently, C57BL/6 was more sensitive to stress than the other strains and the transportation process needs to be standardized to reduce between day variability. To establish the recovery periods from transportation stress, the body weight changes were measured during the acclimation period. Although the body weight of animals decreased during transportation, animals recovered their weight loss after the next day.

A Comparative Study on Fisheries Resource Management System between Korea and China (한·중 어업자원관리제도에 관한 비교연구)

  • Korea and China are two opposite countries located aside Yellow Sea and co-utilize the East China Sea. The two countries are close together from geological point of view, however, the competitive development of resources was more emphasized than the cooperative development of resources between the two countries because the special policy relationship. Additionally, after the communist government of China was founded in 1949, the political conception between the two countries was quite different. Therefore the establishment of appropriate international fisheries co-operation was impossible, and the international management problems of fisheries resources in Yellow Sea and East China Sea were let alone. UN convention on the Law of the Sea came to force in 1994, Korea and China adopted the exclusive economic zone system in 1996. On the other hand, Fisheries Law in Korea was enacted in 1953 in order to management of fisheries resources, and also China was enacted fisheries law in 1986. The two countries control the fisheries effort through fisheries license system, meanwhile through prohibition fishing area, prohibition fishing period, limitation of net size, and limitation of body length to conserve and manage the fisheries resource. The serious management methods of resource management in the two countries are similar such as the creation of promptly decreased species and those species that have commercial value, discharge of fish seedling stock, settlement of artificial reef and clean of fishing ground. Therefore, the two countries should consider not only the improvement of formal law system, but also how to recover the fisheries resources in circumference water zone and how to improve the efficiency of fisheries resource management. Specially the settlement and management of artificial reef should be chosen in the area that have the highest benefit to two countries, and should establish the common management system of discharge of fish seedling stock. And the two countries should adopt the same criteria through technical management and limitation of net size, limitation of body length, and prohibition area of special fisheries to ensure the highest fisheries benefit of fisherman in the two countries and the highest efficiency of fisheries resource management.

Influence of forestry host plants and rearing seasons on silk gland weight of tropical tasar silkworm, Antheraea mylitta (Lepidoptera: Saturniidae) under Doon valley conditions of Uttarakhand in India

  • Tropical tasar silkworm, Antheraea mylitta is a commercial forest silkworm in India that produces tasar silk, but never experimented in Uttarakhand, a Himalayan state of India. A. mylitta express divergent phenotypic characters under different ecological conditions; so, we studied the effect of seven forest tree species in two rearing seasons on variability in silk gland weight of Daba (bivoltine) ecorace of A. mylitta at Forest Research Institute in Dehra Dun, Uttarakhand during 2012 and 2013. We used two-way completely randomized block factorial design and Post HOC Tukey's HSD test to analyse the collected data and there after carried out multiple regression analysis. Results indicated that silk gland weight differed significantly between rearing seasons (DF=1, F=2333.98, p <0.05), host plants (DF 6, F= 1516.25, p <0.05) and their interactions (DF=6, F=7.10, p <0.05). Higher silk gland weight was found in second rearing season than the first on all the host tree species. Terminalia alata fed A. mylitta larvae showed the highest silk gland weight of 8.03 and 9.47 g in first and second rearing seasons, followed by T. tomentosa (7.19 & 9.01g), T. arjuna (6.8 & 8.08 g) and L. speciosa (6.57 & 7.83 g) fed larvae, respectively. Post HOC Tukey's HSD test indicated that silk gland weight of L. speciosa and T. arjuna fed larvae in both the rearing seasons did not differ significantly. E.I. analysis also confirmed that T. alata, T. tomentosa, T. arjuna and L. speciosa are better in their order of merit than T. bellirica , T. chebula and L. tomentosa. Multiple regression analysis indicates that larval weight gain is a strong predictor (β=1.002, t=346.777, p = <0.05) for the silk gland weight of A. mylitta ; however, larval duration had significant negative regression weight (β=-0.270, t=-8.436, p = <0.05) on mean weight of silk gland.

Application of Electronic Nose in Discrimination of the Habitat for Black Rice (전자코를 이용한 검정현미의 산지판별)

  • The discrimination of the agricultural origin, especially locally produced of imported products such as black rices was investigated by using electronic nose. Volatile components from these products were discriminated by six metal oxide sensors without pretreatment. Pattern recognition was carried out. Principal component analysis showed the differences between imported and locally produced ones. The number of 57 from 69 species of black rices were recognized as locally produced one (83.33%) and 11 from 13 species one (imported black rices) was correctly discriminated. Unknown habitat of black rice could be identified by artificial neural network system whether the imported or not. Also commercial electronic nose (E-nose 5000) that was combined with metal oxide sensor and conducting polymer sensor showed 92.75% (locally produced black rices) and 92.31% (imported one) of discrimination.

Fishing investigation of trawl fisheries in sea mount of central northern Pacific (북태평양 중부 해산어장에 있어서 저층 트롤의 어획실태)

  • This study was conducted in order to the experiment of bottom trawl fisheries in the sea mount of central northern pacific($30^{\circ}-40^{\circ}N$, $170^{\circ}-175^{\circ}E$), during the period of July 1 to August 25, 2004 by commercial trawl ship. Fishing experiment was carried out by bottom trawl, with the codend of 110mm mesh size. The number of 101 test fishing was done in the sea mount of central northern pacific during 56 days and the total catches were 198,593kg as 18 fish species, and CPUE/day and catches/day were 8.3 hours towing time and 3,546kg. Catch ratio according to each fishing ground was higher with the order of B, C, and E, and catches for B was 7,171.0kg during the towing time of 13.2 hours. Main target species were Pseudopentaceros richardsoni and Beryx splendens, there was 14.9% of damage ratio for fishing gears on catching the target fishes inhabiting on the rough sea bottom. Catches by water depth was the order of 300m below, 301-350m, 451-500m and 351-400m. It is thought that there was no problem on the mesh size because most of fishes caught from the experimental fishing was 30cm over.
