Recent trends in tissue culture and genetic transformation of Phalaenopsis (팔레놉시스 조직배양 및 형질전환 최근 연구동향)

  • This report describes recent advances in tissue culture and genetic transformation of commercial Phalaenopsis. Recently, an importance of Phalaenopsis has been increased due to its popularity with beautiful flowers and is widely used for pot plants as well as cut-flower. Its use is rapidly enlarging in worldwide. Thus, demands for the release of new elite cultivars in Phalaenopsis have been increased. During the last several decades, some critical progresses have been made in tissue culture and genetic transformation in Phalaenopsis species. Cooperation with these biotechnological methods are supposed to promote the release of commercial Phalaenopsis cultivars in the near future. Until now, no technical review on tissue culture and genetic transformation in Phalaenopsis has been reported in Korea. Therefore, we inquired the brief history and techniques of tissue culture system in Korea.

Isolations of the Orchid mycorrhizal Fungi from the Roots of the Korean native Orchids and Inoculations of the Isolates to Four different Orchids (한국 자생란에 난 균근균의 분리와 유묘난에 접종)

  • Ten isolates of the orchid mycorrhizal fungi were isolated from the roots of Korean native orchid plants (Cymbidium goeringii) which inhabitate mainly in southern and western areas of Korea. The growth rates and color of the isolates in potato dextrose agar (PDA) were various. Microscopic observations of the hyphae isolated were identified as Rhizoctonia repens and R. endophytica var endophytica or their related species. R. repens was isolated from the roots of the Korean native orchids, but R. endophytica var endophyica was only isolated from the roots of the commercial orchids introduced from foreign countries. Also, the polymorephic patterns of genomic DNA extracted from selected isolates were compared with those of DNA extracted from the orchid mycorrhizal fungi isolated previously and similar band patterns were observed among those isolates. Five isolates of R. repens were selected and cultured at the oatmeal agar for investigating their symbiosis with orchid plants. The symbiotic specificity between orchid plants and isolated orchid mycorrhizal fungi was observe by growing orchids about six months in the greenhouse. The symbiotic responses of the commercial orchid plants with selected isolates were quite different form different isolates due to the genetic variations.

  • Formalin, 37% formaldehyde, has been employed as a chemical for controlling ectoparasites and aquatic fungi responsible for infectious fish of diseases in hatcheries and culture facilities (Roberts, 1978; Schnick, 1991; Rach et al., 1997). Regarding the commercial usage of formaldehyde in the aquaculture industry, the U.S. Food and Drug Administration (FDA) approved three commercial products as parasiticides in a species-specific manner: Paracide-F and Formalin-F for bluegill, catfish, largemouth bass, salmon, and trout and Parasite-S for all finfish (FDA, 1998). Withdrawal time for these products was legally zero when used as permitted under the regulations. With the increased production of cultured fish in Korea, such as olive Hounder Paralichthys olivaceus and black rockfish Sebastes schlegeli, application of formalin to diseased fish has become more frequent. Moreover, there is still some concern about environmental exposures caused by effluents from fish culture facilities. The purpose of the present study was to evaluate residues in fish resulting from therapeutic usage of formalin in the aquaculture industry and to document the rate of disappearance of formaldehyde in seawater treated with formalin. Isolation of Fungal Pathogens to an Edible Mushroom, Pleurotus eryngii, and Development of Specific ITS Primers

  • Fungal pathogens have caused severe damage to the commercial production of Pleurotus eryngii, the king oyster mushroom, by reducing production yield, causing deterioration of commercial value, and shortening shelf-life. Four strains of pathogenic fungi, including Trichoderma koningiopsis DC3, Phomopsis sp. MP4, Mucor circinelloides MP5, and Cladosporium bruhnei MP6, were isolated from the bottle culture of diseased P. eryngii. A species-specific primer set was designed for each fungus from the ITS1-5.8S rDNA-ITS2 sequences. PCR using the ITS primer set yielded a unique DNA band for each fungus without any cross-reaction, proving the validity of our method in detection of mushroom fungal pathogens.

Mixing Pyroligneous Acids with Herbicides to Control Barnyardgrass (Echinochloa crus-galli)

  • Alternatives to commercial chemical herbicide are currently being searched and tested due to the numerous adverse effects of commercially available herbicides to the environment. Barnyardgrass (Echinochloa crusgalli) is an important weed species around the world, especially in paddy rice fields. This study focuses on the favorable effects of mixing pyroligneous acids with commercial liquid herbicides. Seedlings were transplanted and grown under greenhouse conditions. The effect of treatment time or leaf-stage on herbicide-pyroligneous acid efficacies was checked, coupled with isolation and quantification of biochemical compounds. Results revealed that herbicide treatment at early post emergence (2~3 leaf stage) of Echnochloa crus-galli leads to effective control. Both liquid herbicides affected fatty acid, protein, and amino acid syntheses as reflected on their contents. The influence of wood vinegar (WV) or rice vinegar (RV) on these compounds was not thoroughly verified due to lack of information on the pyroligneous products. We observed that mixing WV or RV with BCB (bentazone + cyhalof-butyl) gives more favorable results than BUC (butachlor + clomazone), mixed with WV or RV. The result would indicate the potential of mixing pyroligneous acid in reducing herbicide application rate.

First Report of Botrytis Mold Caused by Botrytis cinerea on Peonies (Paeonia lactiflora Pall.)

  • In 2019, symptoms of Botrytis mold on the peony (Paeonia lactiflora Pall.) 'Sarah Bernhardt' were observed during a survey of the commercial greenhouses of Gangjin County, South Korea. The initial symptoms, small brown spots, were observed mainly at the leaf margins. The lesions extended to the interior of leaves forming irregular spots in which abundant conidia developed. Fungal colonies were obtained from surface-sterilized tissue excised from growing edges of the lesions that were transferred to potato dextrose agar. Melanized irregular sclerotia were formed in these colonies after 40 days at 8℃. Molecular phylogeny based on sequences of genes for glyceraldehyde-3-phosphate dehydrogenase, heat-shock protein 60, and RNA polymerase subunit II were highest for the PBC-2 isolate to the type strains of Botrytis cinerea, rather than other Botrytis species associated with peony diseases. Following Koch's postulates, healthy Sarah Bernhardt plants were inoculated with a foliar application of conidial suspensions of the isolate PBC-2. Following incubation under humidity with a 12 hr photoperiod for 7 days, symptoms developed on the leaf margins that were identical to those observed in the greenhouses. This study is the first report of Botrytis blight caused by B. cinerea on peonies grown in commercial greenhouses in South Korea.

Comparison of Seven Commercial TaqMan Master Mixes and Two Real-Time PCR Platforms Regarding the Rapid Detection of Porcine DNA

  • A pig-specific real-time PCR assay based on the mitochondrial ND5 gene was developed to detect porcine material in food and other products. To optimize the performance of assay, seven commercial TaqMan master mixes and two real-time PCR platforms (Applied Biosystems StepOnePlus and Bio-rad CFX Connect) were used to evaluate the limit of detection (LOD) as well as the PCR efficiency and specificity. The LODs and PCR efficiencies for the seven master mixes on two platforms were 0.5-5 pg/reaction and 84.96%-108.80%, respectively. Additionally, non-specific amplifications of DNA from other animal samples (human, dog, cow, and chicken) were observed for four master mixes. These results imply that the sensitivity and specificity of a real-time PCR assay may vary depending on master mix and platform used. The best combination of master mix and real-time PCR platform can accurately detect 0.5 pg porcine DNA, with a PCR efficiency of 100.49%.

Effect of a Low Temperature-Induced Quiescence on Short Term Storage of an Egg Parasitoid, Trichogramma sp. Nabis 101 (알기생봉(Trichogramma sp. Nabis101)의 저온 발육 휴지와 단기 저장 기술)

  • A commercial egg parasitoid, Trichogramma sp. Nabis101, was released into agricultural cultivating areas in Korea due to its wide host spectrum against insect pests. Moreover, an application technique has been recently developed to enhance its control efficacy by mixture treatment with a microbial control agent. Despite its expansion of commercial availability, any genetic identification on this commercial strain was not determined. Also, to meet inconsistent demands from consumers, the live parasitoids need to be stored without significant loss of their survival and parasitic activity. This study determined nucleotide sequence of internal transcribed spacer (ITS) of the wasp species. The identified ITS sequences indicate that this wasp species is most similar to T brasiliensis. Optimal storage condition of this wasp required young parasitized stage at $10^{\circ}C$. Under these conditions, survival, sex ratio, longevity, and parasitic behavior were not much impaired for 5 weeks.

A Study on the Changes of the Molluscan and Crustacean Fauna after the Construction of the Naktong Estuary Barrage (낙동강 하구언 건설에 의한 연체동물상 및 갑각류상의 변화에 관한 연구)

  • The materials of this study consist of both specimens and survey records of the mollusks and crustaceans which were obtained from 30 collection sites in the Naktong Estuary during Aug.-Nov., 1991. The mollusks and crustaceans collected and identified in this study are 31 and 50 species, respectively. Of these two species of mollusks and six species of crustaceans were newly recored in this area. Including the previous records, the occurrence and distribution of 63 molluscan species and 138 crustacean species were compared in eight zones, before and after the construction of the Naktong Estuary Barrage. During this investigation, only a few species of mollusks occurred in the northern freshwaters off the Barrage. The habitats for the intertidal species were greatly destructed along the eastern part of the UIsuk Is. and the riverside of Tadaep'o where the bank was recently constructed. The commercial bivalves including Corbicula fluminea and Ruditapes Philkippinarum were greatly decreased in number in all the eatuarine waters. A freshwater crab, Eriocheir japonicus, seems to be extinguished in the Naktong River and its neighbouring streams.

Studies on Discoloration of 16 Commercial wood Grown in Korea (한국산(韓國産) 유용목재(有用木材)의 변색(變色)에 관한 연구)

  • Change of color of woods is a trouble frequently found in decorative wood products. In this paper, studies were carried out on discoloation sensitivities of wood specimens to iron (0.1%, $FeCl_3{\cdot}6H_2O$), alkali (pH12.0, NaOH), acid (pH 1.0 $C_2H_2O_4$) and exposing to sunlight (40 hrs.). Four soft-woods and eleven hard-woods grown in Korea were used in this test. The results obtained were as follows: 1. In iron stain, strong discolored wood species were Larix leptolepis, Quercus mongolica. Zelkova serrata and Prunus yedoensis, and light discolored were Paulownia lomentosa and Pinus rigida. 2. In alkali stain. light discolored wood species were Abies holophylla, Kalopanax pictum, Populus ${\times}$ albaglandulosa, Acer mono, Pinus rigida, Pinus densiflora, Quercus mongolica, Betula platyphylla var. japonica, Prunus yedoensis, Zelkova serrata, Populus euramericana, Pinus koraiensis and Paulownia tomentosa, but there was no wood species of strong discolored compared with the color difference values of other reports. 3. In acid stain, light discolored wood species were Pinus rigida, Paulownia tomentosa. Pinus koraiensis, Quercus mongolica, Kalopanax piclum and Acer mone, but there was no strong discolored species compared with the color difference values of other reports. 4. In exposing to sunlight, strong discolored species were Larix leptolepis, Acer mono, Paulownia tomentosa, Populus euramericana, Pinus densijlora, Robinia pseudoacacia, Abies holophylla, Populus ${\times}$ albaglandulosa, Zelkova serrata, Prunus yedoensis and Pinus rigida, and light discolored was Alnus japonica. In general, it was shown that Korean wood species were susceptible to change of color by exposing to sunlight and iron stain compared with by alkali stain and acid stain.
