Development and Characterization of Monoclonal Antibodies against Nucleoprotein for Diagnosis of Influenza A Virus

  • Influenza, which is a highly contagious disease caused by the influenza A virus, continues to be a major health concern worldwide. Although the accurate and early diagnosis of influenza virus infection is important for controlling the spread of this disease and rapidly initiating antiviral therapy, the current influenza diagnostic kits are limited by their low sensitivity. In this study, we developed several new influenza nucleoprotein (NP)-specific monoclonal antibodies (mAbs) and compared their sensitivity and specificity of those with commercially available anti-NP mAbs. Three mAbs, designated M24.11, M34.3, and M34.33, exhibited higher reactivities to recombinant NPs and A/Puerto Rico/8/1934 (H1N1) viral lysates compared with the commercial mAbs, as assessed using enzyme-linked immunosorbent assays. M34.3 and M34.33 showed higher reactivities with A/California/04/09 (pandemic H1N1) and A/Philippines/2/82 (H3N2) viral lysates than the commercial mAbs. In contrast, M24.11 had marked reactivity with H3N2 but not with pandemic H1N1. Immunofluorescent confocal microscopy showed that the three mAbs effectively detected the presence of influenza virus in lung tissues of mice infected with A/Puerto Rico/8/1934. These results indicate that the newly developed M34.3 and M34.33 mAbs could be useful for the development of influenza diagnostics.

실험 배합사료, 상품사료 및 생사료 공급이 육성기 조피볼락(Sebastes schlegeli)의 성장, 육질 및 조직 성상에 미치는 영향 (Effects of Different Pellets on the Growth, Flesh Quality and Histopathological Changes of growing Korean Rockfish Sebastes schlegeli)

  • This study compared the effects of diets consisting of experimental extruded pellets (EP), commercial soft extruded pellets (SEP), and raw-fish-based moist pellets (MP) on the growth, flesh quality, and histology of the Korean rockfish Sebastes schlegeli. Three replicated groups of 60 fish each (initial mean weight 152 g) were fed one of two experimental EPs (EP1 and EP2), two commercial SEPs (SEP1 and SEP2), or MP for 11 weeks. The survival, weight gain, and final mean weight of the fish did not differ significantly among the groups. The highest feed efficiency was in fish fed MP (P<0.05), and the protein efficiency ratio of fish fed MP was significantly higher than that of fish fed EP1, EP2, or SEP1, but not significantly different from that of fish fed EP2. The daily feed intake of the fish fed the EPs and SEPs was significantly higher than that of the fish fed MP (P<0.05). The condition factor, Hepatosomatic Index, and Viscerasomatic Index of the fish did not differ significantly among the groups. No notable differences in the textural properties of the dorsal muscle, plasma biochemical parameters, or histological features of the fish were observed in fish fed any diet. This suggests that extruded pellets can be used to feed growing Korean rockfish without compromising growth, flesh quality, or histopathological change in comparison to raw fish-based moist pellets.

GIS를 활용한 부산시 도심재생에 관한 연구 (City center Regeneration in Busan Metropolitan Using GIS)

  • 도심공동화를 단순히 상주인구나 유동인구의 감소, 상업기능의 쇠퇴 등만으로 보고 단편일률적인 방안을 제시하는 것은 또 다른 문제를 가져오게 된다. 그래서 본 연구에서는 부산시의 도심을 조사 분석하여, 그에 적합한 도심재생방안을 부산시의 특성을 고려하여 제시하는데 그 목적이 있다. 도심의 기능별 지역을 알기위해서 부산시의 도심에 속하는 15개 동을 대상으로 1996년, 2000년, 2004년 세 개 연도를 분석하여 도심의 공간적 특성과 도심공동화현상을 파악하고자 하였다. 이들지역에 20개의 변수를 선택하여 요인분석과 군집분석을 사용하였다. 각 연도별 주요요인은 각각 3개씩 나타났으며, 누적 설명력은 84.2%, 87.1%, 85.5%로 높게 나타났다. 그리고 이들 요인은 주거적 요인, 상업적 요인, 관리적 요인을 설명하는 성분으로 나타났다. 본 연구의 중요한 결과들은 다음과 같다. 첫째, 전체적인 기능별 도심공동화현상이 나타나고 있으므로 종합적인 도심재생방안의 모색이 시급하다고 할 수 있겠다. 둘째, 도심의 지역별 특성이 다르게 나타나고 있으므로 지역별 특성에 부합되는 도심재생방안이 수립되어야 하겠다.

죽순식초의 화학적 특성 및 향기분석 (Chemical Characteristics and Flavors of Bamboo-shoot Vinegar)

  • 담양군 죽순으로 제조한 죽순식초와 시판 과실식초의 이화학적 특성인 산도, 무기질, 아미노산, 플라보놀을 비교 분석하고 SBSE-GC/MS방법을 사용하여 향기성분을 분석하였다. 죽순식초의 산도는 4.49%로 제일 낮았으며 무기질 중 Na의 경우 8.36 mg/100 g으로 다른 과실식초에 비해 그 함량이 낮았다. 아미노산은 다른 시판 식초에 비해 tyrosine과 lysine의 함량이 높았으며 flavonol aglycone 중 quercetin이 3.29 mg/L 들어있었다. SBSE방법으로 향기성분을 분석한 결과 aldehyde류의 함량이 시판식초에 비해 높았으며 특이적으로 수면 유도물질로 알려진 oleamide가 처음으로 검출되었다.

Nanoparticle Formation from a Commercial Air Freshener at Real-exposure Concentrations of Ozone

  • Occupational nanomaterial exposure is an important issue in the manufacture of such products. People are also exposed to various nanoparticles in their living environments. In this study, we investigated nanoparticle formation during the reaction of ozone and volatile organic compounds (VOCs) emitted from a commercial air freshener, one of many widely used consumer products, in a $1-m^3$ reaction chamber. The air freshener contained various VOCs, particularly terpenes. A petri dish containing 0.5 mL of the air freshener specimen was placed in the bottom of the chamber, and ozone was continuously injected into the center of the chamber at a flow rate of 4 L/min with an ozone concentration of either 50, 100 or 200 ppb. Each test was conducted over a period of about 4 h. The higher ozone concentrations produced larger secondary nanoparticles at a faster rate. The amount of ozone reacted was highly correlated with the amount of aerosol formation. Ratios of reacted ozone concentration and of formed particle mass concentration for the three injected ozone concentrations of 50, 100 and 200 ppb were similar to one other; 4.6 : 1.9 : 1.0 and 4.7 : 2.2 : 1.0 for ozone and aerosol mass, respectively.

해상가두리 양식장에서 배합사료 및 생사료 공급에 따른 조피볼락(Sebastes schlegeli)의 성장 및 육질 비교 (Evaluation of Commercial Extruded Pellets and Raw Fish-Based Moist Pellets for the Growth and Quality of Korean Rockfish Sebastes schlegeli Cultured in Net-Cages)

  • This study was conducted to compare extruded pellets (EP) and soft extruded pellets (SEP) with a raw fish-based moist pellet (MP) diet on the growth and flesh quality of Korean rockfish Sebastes schlegeli. Three groups of 20,000 fish (initial mean weight 133 g) per net-cage ($6{\times}12{\times}7m$) were fed commercial EP, SEP or MP for 16 months. The survival of fish fed SEP was higher than those of fish fed EP or MP. The highest growth performances were observed in the mean weight gain, total weight gain, and feed efficiency of fish fed MP, followed by those fed EP and SEP. Among the fish fed on extruded pellets, the total weight gain of fish fed SEP was higher than that of those fed EP, while fish fed EP grew faster than those fed SEP. No notable differences in body composition, sensory scores or textural properties of the dorsal muscle were observed in fish fed on EP, SEP or MP. Thus, it is suggested that extruded pellets, rather than raw fish-based moist pellets, could be fed to Korean rockfish without compromising flesh quality.

Graphite-Ni계 분말의 제조 및 용사 코팅 특성에 미치는 공정변수의 영향 (Fabrication of Graphite-Ni Composite Powders and Effect of Thermal Spray Coating Parameters on Mechanical and Microstructural Properties)

  • Graphite-Ni composite powders were synthesized by mechanical alloying(MA) and spray drying(SD). Fabricated powders as well as commercial graphite-Ni powders were thermally sprayed on mild steel substrates using high velocity oxygen fuel (HVOF) thermal spray process and flame thermal spray process. The effects of several process parameters on related properties in thermally sprayed coatings have been investigated and correlated with microstructures in this study. The results indicated that the desired properties can be obtained when commercial powders were applied using HVOF process, while coating properties in case of MA powder application were inferior to those in HVOF process in so far. However, it is suggested that property enhancement can be obtained if the fraction of hexagonal graphite phase can be increased in mechanically alloyed powders.

증강현실을 활용한 원도심 상권 활성화방안 (A Method of Commerce Activation of the Old City-Center of Jeju City Using Augmented Reality Technology)

  • 제주시는 원도심의 거주인구와 유동인구 모두가 줄어드는 원도심쇠퇴 현상이 나타나는 도시 중 하나이다. 제주시 원도심 쇠퇴는 1990년대 이후 본격화되고 있으며, 2000년대 들어 점점 심화되고 있는 실정이다. 이에 따라 제주시는 원도심의 역사문화자원을 활용한 도심관광 활성화 방법과 산업경제 영역 정비를 하여 원도심 상권 활성화 방안을 추진하고 있다. 본 논문에서는 제주시 원도심 공영관광지인 제주목관아에 증강현실기술을 이용하여 원도심 상권 활성화에 도움이 되는 방문객을 증가시키는 방법을 제안한다. 제주목관아를 소재로 한 증강현실 콘텐츠를 스마트폰기반으로 구현하고 시범운영을 하면서 효과를 분석하기 위한 설문조사를 수행한다. 설문조사는 온라인/오프라인을 통한 홍보를 통하여 제주목관아를 찾은 관람객 574명을 대상으로 자기기입 방식으로 수행한다. 설문조사 결과를 바탕으로 제안한 방법이 유동인구를 증가시킴으로써 원도심 상권 활성화에 효과적으로 사용될 수 있음을 보인다.

육성기 넙치 배합사료의 성장 및 수질 평가 (Effects of a Commercial Extruded Pellet on Growth Performance and Water Quality in Growing Olive Flounder Paralichthys olivaceus)

  • This study was conducted to evaluate the effect of commercial extruded pellet (EP) diet on the growth and water quality compared with a raw fish moist pellet (MP) diet for flounder on field feeding experiments. Three replicate groups of 130 fish per each tank (initial weight of 380 g) were fed one of the EP and Moist pellet (MP) for 7 months. In field feeding experiment, no significant difference was observed in final mean weight of fish fed the experimental EP (1,233 g) and MP (1,237 g), and any noticeable problem caused by feeding the experimental EP was not found during the whole period of feeding trial. Feed efficiency of fish fed the experimental EP was higher than that of MP, and there was no significant difference in survival between the fish fed experimental EP and MP. The concentrations of SS, COD, TN and TP of seawater in tanks of MP groups were 1-5 times higher than those of EP groups. Therefore, these results strongly suggest that diet EP could be developed to replace MP diet for the marketable size of production for flounder without adverse effects on growth performance and water quality.

Analysis and Comparison of Volatile Flavor Components in Rice Wine Fermented with Phellinus linteus Mycelium and Regular Commercial Rice Wine

  • This study identified and compared the volatile flavor components of two commercial rice wines: one fermented using the mycelium of Phellinus linteus and a regular commercial rice wine. The volatile flavor components were isolated from the infusions by Porapak Q (50-80 mesh) column adsorption. The concentrated aroma extracts were then analyzed and identified by GC and GC-MS. Thirty-four kinds of flavor components were identified in the mycelium-fermented rice wine, including 11 alcohols, 8 esters, 3 ketones, 6 acids, 3 hydrocarbones, and 4 others. In the regular commercial rice wine, 36 kindss of flavor compounds were identified, including 9 alcohols, 6 esters, 4 ketones, 6 acids, 9 hydrocarbones, and 2 others. Therefore, the data indicate that the primary flavor components in the rice wines were alcohols and esters.
