CRISPR/Cas9-mediated knockout of Rag-2 causes systemic lymphopenia with hypoplastic lymphoid organs in FVB mice

  • Recombination activating gene-2 (RAG-2) plays a crucial role in the development of lymphocytes by mediating recombination of T cell receptors and immunoglobulins, and loss of RAG-2 causes severe combined immunodeficiency (SCID) in humans. Rag-2 knockout mice created using homologous recombination in ES cells have served as a valuable immunodeficient platform, but concerns have persisted on the specificity of Rag-2-related phenotypes in these animals due to the limitations associated with the genome engineering method used. To precisely investigate the function of Rag-2, we recently established a new Rag-2 knockout FVB mouse line ($Rag-2^{-/-}$) manifesting lymphopenia by employing a CRISPR/Cas9 system at Center for Mouse Models of Human Disease. In this study, we further characterized their phenotypes focusing on histopathological analysis of lymphoid organs. $Rag-2^{-/-}$ mice showed no abnormality in development compared to their WT littermates for 26 weeks. At necropsy, gross examination revealed significantly smaller spleens and thymuses in $Rag-2^{-/-}$ mice, while histopathological investigation revealed hypoplastic white pulps with intact red pulps in the spleen, severe atrophy of the thymic cortex and disappearance of follicles in lymph nodes. However, no perceivable change was observed in the bone marrow. Moreover, our analyses showed a specific reduction of lymphocytes with a complete loss of mature T cells and B cells in the lymphoid organs, while natural killer cells and splenic megakaryocytes were increased in $Rag-2^{-/-}$ mice. These findings indicate that our $Rag-2^{-/-}$ mice show systemic lymphopenia with the relevant histopathological changes in the lymphoid organs, suggesting them as an improved Rag-2-related immunodeficient model.

Translation and Adaptation of the Children's Home Inventory for Listening Difficulties (CHILD) into Korean (가정환경 아동듣기평가(CHILD) 부모용 설문지의 한국어 번역 및 적용 연구)

  • The Children's Home Inventory for Listening Difficulties (CHILD) questionnaire has been applied for assessing listening and communication difficulties in various home situations for children with hearing loss. The purpose of the study was to translate the CHILD questionnaire for parents into Korean and verify reliability and validity of Korean version of CHILD (CHILD-K). CHILD-K was completed by 55 parents of children (from ages 3~12 years) using cochlear implants (CI). Among the 55 children, 27 were in preschool and 28 in elementary. Internal consistency reliability of CHILD-K was verified by Chronbach's alpha. The mixed factorial ANOVA was conducted to compare the effects of the age group and situation factors (Quiet, Noise, Distance, Social, and Media factors) on the score of CHILD. The results indicated that CHILD-K showed excellent internal consistency reliability (${\alpha}=.96$). The CHILD scores among age groups were significantly different as the older age group resulted in higher scores in all situations except Distance. For both groups the mean scores for the Quiet situation were significantly higher than other situations, and the mean scores for the Social situation were significantly lower than other situations. Moreover, analysis showed that children with CI had difficulties in the Social situation combined with other situation factors. The results indicate that the Korean version of CHILD questionnaire is a reliable tool for the assessment of communication abilities in home situation in Korean-speaking children using CI.

Asymmetric Timeliness of Market Information According to Corporate Losses and Earnings (기업의 손실과 이익에 따른 시장정보의 비대칭적 적시성)

  • This study investigates the sensitivity reflected in the accounting earnings differs according to the difference in the characteristics of accounting information such as profit and loss for the same market information. For this, market information and accounting data were analyzed for 11,462 non-financial listed companies listed on the KOSPI and KOSDAQ markets from 2012 to 2020 by using Basu's measurement of conditional conservatism and Ball and Shivakumar's measurement of conservatism. Accounting earnings sensitivity was analyzed according to the combination of information. As a result of the study, it was confirmed that both earnings and losses corporates recognize losses with delay, while losses are recognized quickly by loss corporates and delayed recognition by earnings companies. It was confirmed that more strict conservatism was applied to the losses corporates compared to the earnings corporates by delaying the recognition of earnings while the early recognition of the losses. It provides empirical data on the causality between the asymmetric timeliness and the combined effect of market information and accounting information by verifying that the losses corporates responds sensitively to market information while the earnings corporates does not react sensitively to the market information.

Water-Environment-Economic nexus analysis of household food waste impacts: A case study of Korean households

  • Food waste has increasingly become a global issue of concern among the researchers and policymakers due to its significant environmental and economic impacts, and other associated unsustainable use of resources, including water resources. While food wastage occurs at each stage of the supply chain with food loss at the upstream and food waste at the downstream, the impacts of food waste occurring at the consumption side are enormous due to the accumulated added values. In this study, the embedded water resources, greenhouse gas emissions, and economic loss of household food waste were investigated. The primary granular data of household food waste was collected through direct sampling from 218 selected households of the Buk-gu community in Daegu, South Korea from July 2019 to May 2020. The water footprint, which was based on the water footprint concept, i.e., indirect water use, and GHG emission potential factor for each of the food items were adopted from the literature, while the retail prices and disposal cost were used to assess the economic cost of wasted food items. The water footprint, GHG emission associated with environmental impacts, and the economic cost of 42 major identified wasted food items were conducted. The findings showed that an average of 0.73 ± 0.06 kg/household/day edible food waste was generated among the sampled households, with leafy vegetable, watermelon, and rice responsible for 10, 9, and 4%, respectively, of the total weight of the 42 food wasted items. The water footprint and environmental impact of the household food waste resulted in 0.46 ± 0.04 m3 and 0.71±0.05 kg CO2eq, respectively. Beef, pork, poultry, and rice accounted for 52, 9, 5, and 4% of the total water footprint, while beef, pork, rice, tofu/cheese had 52, 8, 6, and 6% of the total emissions, respectively, embedded in the food wasted. Furthermore, the average estimated economic cost associated with wasted food items was 3855.93±527.27 Korean won, with beef, fish, and leafy vegetable responsible for 21, 13, and 10%, respectively, of the total economic cost. A combined assessment using water-environmental-economic nexus indicated that animal-based food had the highest footprint impacts, with beef, pork, and poultry indicating high indices of 0.3, 0.08, and 0.06 respectively, on a scale of 0 to 1, compared to corn and lettuce with lowest impacts of 0.02. Other food items had moderate impact values ranging from 0.03 to 0.05. This study, therefore, provides insight into the enormity of environmental and economic implications of household food waste among Korean households.

  • Objectives : This study retrospectively analyzed the medical records of patients admitted to Korean medicine ophthalmology, otolaryngology and dermatology department of Daejeon Korean medicine hospital from March 2018 to February 2023 to analyze the characteristics of patients receiving inpatient treatment. Methods : We retrospectively analyzed inpatients who admitted to ophthalmology & otolaryngology & dermatology clinic of Daejeon Korean medicine hospital from March 1st, 2018 to February 28th, 2023, according to gender, age, year, season, detailed subdivision. The statistical analysis performed using IBM SPSS 29.0 for Windows. Results : 1. Examining the gender distribution of the patient group, there were 367 female patients, accounting for 71.7% of the total patients, and 145 male patients, accounting for 28.3% of the total patients. 2. When analyzing inpatients by subdivision, otology accounted for more than half of the total number of inpatients, and the combined number of otology and dermatology accounted for more than 80% of the total. 3. As a result of analyzing inpatients by frequent disease, Sudden hearing loss was a significantly higher number of patients, accounting for 22.7% of all inpatients. Conclusions : It was found that the proportion of patients with otologic diseases was very prominent. It is thought that further research is needed to see if the trend of increasing demand for otologic diseases continues.

Analysis of Nutrient Cycling Structure of a Korean Beef Cattle Farm Combined with Cropping as Affected by Bedding Material Types (깔개물질의 종류에 따른 한우-경종 결합 농가의 양분순환 구조 분석)

  • In this study, we analyzed nutrient cycling structure of a small farm (cattle of 100 heads and arable lands of 2.5 ha) in Jeonnam province to investigate the effects of nutrients input by the addition of bedding materials (sawdust and rice hull) and nutrients loss before the application to the soils (the period during manure storage in the feedlot and composting process) on nutrient cycling structure. Sawdust and rice hull added as bedding materials increased N by 1.6% and 14.2% and $P_2O_5$ by 3.1% and 27.4%, respectively, relative to the amount of nutrients produced by excretion. This result suggests that the addition of nutrients via bedding materials should be considered for better estimation of nutrient balance. The most significant characteristics of the nutrient cycling structure was loss of mass and nutrients during the storage (21 days) and composting period (90 days). During this period, 78.4% of N and 9.5% of $P_2O_5$ was lost from sawdust compost; meanwhile, the percentages of loss for rice hull compost were 81.6% and 10.3%, respectively. A lower percentage of nutrients loss in sawdust compost than that in rice hull compost was attributed to the relatively slow decomposition rate of organic materials in the sawdust compost which has higher C/N ratio and lignin contents. Therefore, it was concluded that estimation of nutrient balance should be conducted based on nutrient contents in the final compost being applied to the lands rather than the amount of nutrients contained in the livestock excretion. In addition, the effects of bedding materials on nutrient losses should be also taken into account.

Clinical Long-term Assessment of Bioactive Glass Graft (Bioactive glass의 장기 임상적 평가)

  • The ultimate goal of periodontal therapy is the regeneration of periodontal tissue which has been lost due to destructive periodontal disease. To achieve periodontal regeneration, various kinds of methods have been investigated and developed, including guided tissue regeneration and bone graft. Bone graft can be catagorized into autografts, allografts, xenografts, bone substitutes. And materials of all types have different biological activity and the capacity for periodontal regeneration, but ideal graft material has not been developed that fits all the requirement of ideal bone graft material. Recently, bioactive glass that has been utilized in plastic surgery is being investigated for application in dental practice. But, there has not been any long-term assessment of bioactive glass when used in periodontal intrabony defects. The present study evaluates the long-term effects of bioactive glass on the periodontal regeneration in intrabony defects of human and the effect of plaqu control on long term treatment results after dividing patients into those who underwent 3-month regular check-up and those who didn't under go regular check-up The clinical effect on 74sites from 17 infrabony pockets of 11 patients were analyzed 36months after treatment. 51 sites which underwent regular check up were classified as the Follow-up group(F/U group), and 23 sites which did not undergo regular check up were classified as Non Follow-up group(Non F/U group). After comparing the probing depth, attachment loss, bone probing depth before and 36months after treatment, the following results could be concluded. 1. The changes of probing pocket depth showed a statistically significant decrease between after baseline and 36 months after treatment in F/U group(1.79${\pm}$0.68mm) and did no show astatistically significant decrease between after baseline and 36months after treatment in Non F/U group(0.61${\pm}$0.54mm) (P<0.05). 2. The changes of loss of attachment showed a statistically significant decrease between after baseline and 36 months after treatment in F/U group(1.44${\pm}$0.74mm) and did no show astatistically significant decrease between after baseline and 36months after treatment in Non F/U group(1.18${\pm}$1.54) (P<0.05). 3. The changes of bone probing depth showed a statistically significant decrease between after baseline and 36 months after treatment in both F/U(1.35${\pm}$0.28) and Non F/U group(0.78${\pm}$0.55mm) (P<0.05). The results suggest that treatment of infrabony defects with bioactive glass resulted in significan reduction of attachment loss and bone probing depth 36months after the treatment. The use of bioactive glass in infrabony defects, combined with regular check-up and proper plaque control generally shows favorable clinical results. This measn that bioactive glass could be a useful bone substitute.

Effect of Inclusion of Hard Versus Soft Wheat Bran with Different Particle Size on Diet Digestibility, Growth Performance and Carcass Traits of Fattening Rabbits

  • Effect of inclusion of hard vs. soft wheat bran with different particle size on diet digestibility, growth performance and some slaughter traits was evaluated in fattening rabbits. Four isonitrogenous and isocaloric diets were used according to the origin of wheat bran (hard (HWB) - Triticum durum - and soft (SWB) - Triticum aestivum) combined with wheat bran particle size sieved by 2 mm (fine: 2) or by 8 mm (coarse: 8) in a bifactorial (2${\times}$2) study. A growth trial was conducted to measure the effect of treatments on performance in one hundred and twenty New Zealand White${\times}$Californian rabbits fed experimental diets from 50 to 87 days of age. Faecal apparent digestibility was determined within the last week in twenty animals per diet. Digestibility of nutrients was higher (p<0.05) in the diet containing HWB2, except for crude protein, ether extract and ash, than fine and coarse soft wheat bran diets. Final live weight, feed intake and feed consumption of rabbits on the diet with fine hard wheat bran were higher and resulted in greater daily weight gains (p<0.01) than for animals on the other diets. The slaughter yield and percentage value of organs were not significantly (p>0.05) affected by the diets fed; however, the diet containing fine hard wheat bran led to lower (p<0.05) percentages of skin, abdominal fat and carcass drip loss than the other dietary treatments. It is concluded that fine hard wheat bran can be better included in the diet than soft wheat bran to maximize growth performance without affecting carcass traits of fattening rabbits.

Induction of Apoptosis in Arsenic Trioxide-treated Lung Cancer A549 Cells by Buthionine Sulfoximine

  • Arsenic trioxide (ATO) affects many biological processes such as cell proliferation, apoptosis, differentiation and angiogenesis. L-buthionine sulfoximine (BSO) is an inhibitor of GSH synthesis. We tested whether ATO reduced the viability of lung cancer A549 cells in vitro, and investigated the in vitro effect of the combination of ATO and BSO on cell viability in relation to apoptosis and the cell cycle. ATO caused a dose-dependant decrease of viability of A549 cells with an $IC_{50}$ of more than $50{\mu}m$. Low doses of ATO or BSO ($1{\sim}10{\mu}m$) alone did not induce cell death. However, combined treatment depleted GSH content and induced apoptosis, loss of mitochondrial transmembrane potential (${\Delta}{\Psi}_m$) and cell cycle arrest in G2. Reactive oxygen species (ROS) increased or decreased depending on the concentration of ATO. In addition, BSO generally increased ROS in ATO-treated A549 cells. ROS levels were at least in part related to apoptosis in cells treated with ATO and/or BSO. In conclusion, we have demonstrated that A549 lung cells are very resistant to ATO, and that BSO synergizes with clinically achievable concentration of ATO. Our results suggest that combination treatment with ATO and BSO may be useful for treating lung cancer.

Effects of Winter Cover Crop of Ryegrass (Lolium multiflorum) and Soil Conservation Practices on Soil Erosion and Quality in the Sloping Uplands

  • Most of the uplands in alpine regions during off-season are left as bare soil and thus vulnerable to severe erosion due to the inherent topographical conditions. Appropriate management strategy to cope with this problem is urgently needed, yet few researches have been reported on the effects of winter cover crop and management on soil erosion. We assessed effects of ryegrass (Lolium multiflorum) as cover crop, green manure or mulching residue on soil erosion and quality through field and segment plot lysimeter experiments in alpine uplands. Ryegrass successfully adopted to winter in alpine region based on biomass, nutrient contents, and vigors of top and root systems. Incorporation of ryegrass into soil maintained soil fertility, nutrient uptake, and yield of cabbage exerting potential use as green manure. Cultivation of ryegrass suppressed occurrence of Chinese cabbage pests. Surface coverage by ryegrass as cover crop and mulching residue significantly reduced soil loss up to 96%, when combined with soil conservation management practices. Results revealed maintaining cover crop over winter was beneficial in reducing soil erosion, and sustaining soil quality and Chinese cabbage productivity. This study suggested winter cover crop, followed by green manure and mulching, and conservation tillage system could be one of the best management practices in alpine sloping uplands cultivating Chinese cabbage.
