Development of a Functional Mixed-Starter Culture for Kefir Fermentation (Kefir 배양용 기능성 복합 Starter 개발)

  • Kefir, which originates in the Caucasian mountains, is a cultured milk beverage produced by a combination of acidic and alcoholic fermentation. Kefir products are commonly used as food vehicles to deliver health-promoting materials including kefran and lactic acid bacteria to consumers. The aim of this study was to develop a freeze-dried starter culture without yeast and assess the suitability of kefir-like dairy products for the growth of lactic acid bacteria and the acidification of milk. Pasteurized whole milk (SNF 8.5%) stored at $25^{\circ}C$ was aseptically inoculated with starter cultures (0.002% w/v); it was kept at $25^{\circ}C$ until the pH attained a value of 4.6. Ten grams of the kefir-like product sample was diluted with 90 mL of 0.15% peptone water diluent in a milk dilution bottle, followed by uniform mixing for 1 min. Viable cells of Lactobacillus species were enumerated on modified-MRS agar (pH 5.2), with incubation at $37^{\circ}C$ for 48 h. Viable cells of Lactococcus species were enumerated on M17-lactose agar, with incubation at $32^{\circ}C$ for 48 h. The pH attained a value of 4.6 after fermentation for 9 h 30 min (Starter 1), 9 h 45 min (Starter 2), and 12 h (Starter 3). The viable cell count of Lactobacillus sp. and Lactococcus sp. was initially $10^5{\sim}10^6CFU/g$; it increased significantly to $10^9CFU/g$ after 12 h of incubation. During the storage of the kefir-like products at $4^{\circ}C$ for 1 4 days, the total viable cell numbers were unchanged, but the pH decreased slightly. The consistency of the kefir products increased gradually during the storage. The organoleptic properties of the kefir products fermented using the new starter culture are more desirable than those of commercial kefir. These results suggest that the newly developed starter culture without yeast could be suitable for kefir fermentation.

Evaluation of Cell-Wall Microstructure and Anti-Swelling Effectiveness of Heat-Treated Larch Wood (낙엽송 열처리재의 세포벽 미세구조 및 항팽윤효율 평가)

  • In this study, the cell-wall microstructure and anti-swelling effectiveness (ASE) of heat-treated larch wood were evaluated and the correlation between them was analyzed. For this purpose, some larch lumbers were heat-treated for 12, 18, and 24 hours at temperatures of 190℃ and 220℃. By observing the scanning electron microscopy cross-sectional image of the heat-treated larch, it was confirmed that the shape of heat-treated wood cell changed, the cut-section of the wood cell wall was rough, and the intercellular space has become wide as the intercellular bonds had broken because of heat-treatment. In addition, the evaluation of the swelling for each treatment condition revealed that, as the heat-treatment temperature and duration increased, the amount of absorbed water and swelling decreased and the ASE increased. The decrease in the amount of absorbed water is thought to be affected by the chemical change in the cell wall by heat-treatment. On the contrary, the decrease in the swelling and the increase in the ASE are thought to be due to a combination of chemical changes and physical changes such as structural changes in the cell wall.

Effect of Packaging Method on the Lipid Oxidation, Protein Oxidation, and Color in Aged Top Round from Hanwoo (Korean Native Cattle) during Refrigerated Storage

  • The objective of this study was to investigate the effects of the packaging method on the lipid and protein oxidation, and color in aged top round from Hanwoo (Korean native cattle) for 14 d at $4^{\circ}C$. Catalase activity was the highest (p<0.05) in vacuum packaging (VP) treatment during storage, and was higher (p<0.05) in 50% Ox-MAP and 50% Ox-MAP+vacuum skin packaging (VSP) treatments than in other treatments at d 14. Superoxide dismutase activity was higher (p<0.05) in VP, 50% Ox-MAP, and 50% Ox-MAP+VSP treatments than in other treatments at d 14. During storage, total antioxidant activity was the highest (p<0.05) in VP treatment and was higher (p<0.05) in 50% Ox-MAP+VSP treatment than in 80% Ox-MAP treatment. TBARS value was the lowest (p<0.05) in VP treatment during storage and was lower (p<0.05) in 50% Ox-MAP and Ox-MAP+VSP treatments than in 80% Ox-MAP and Ox-MAP treatments, respectively. Carbonyl content was the lowest (p<0.05) in VP treatment from 10 d. From 7 d, the $a^*$ value was the highest (p<0.05) in VP treatment and was higher (p<0.05) in 50% Ox-MAP and 50% Ox-MAP+VSP treatments than in other treatments. The $b^*$ value was the highest (p<0.05) in VP treatment from 3 d, and was higher (p<0.05) in 80% Ox-MAP+VSP, 50% Ox-MAP, and 50% Ox-MAP+ VSP treatments than in 80% Ox-MAP treatment at d 14. Therefore, VP improved the oxidation and red color stabilities in stored-aged top round compared with Ox-MAP. In addition, 50% Ox-MAP improved the lipid oxidation and red color stabilities compared with 80% Ox-MAP, and its inhibitory effect on lipid oxidation was enhanced by combination with VSP.

An ELISA-on-a-Chip Biosensor System for Early Screening of Listeria monocytogenes in Contaminated Food Products

  • An enzyme-linked immunosorbent assay (ELISA)-on-a-chip (EOC) biosensor combined with cell concentration technology based on immuno-magnetic separation (IMS) was investigated for use as a potential tool for early screening of Listeria monocytogenes (L. monocytogenes) in food products. The target analyte is a well-known pathogenic foodborne microorganism and outbreaks of the food poisoning typically occur due to contamination of normal food products. Thus, the aim of this study was to develop a rapid and reliable sensor that could be utilized on a daily basis to test food products for the presence of this pathogenic microorganism. The sensor was optimized to provide a high detection capability (e.g., 5.9 ${\times}\;10^3$ cells/mL) and, to eventually minimize cultivation time. The cell density was condensed using IMS prior to analysis. Since the concentration rate of IMS was greater than 100-fold, this combination resulted in a detection limit of 54 cells/mL. The EOC-IMS coupled analytical system was then applied to a real sample test of fish intestines. The system was able to detect L. monocytogenes at a concentration of 2.4 CFU/g after pre-enrichment for 6 h from the onset of cell cultivation. This may allow us to monitor the target analyte at a concentration less than 1 CFU/g within a 9 h-cultivation provided a doubling time of 40 min is typically maintained. Based on this estimation, the EOC-IMS system can screen and detect the presence of this microorganism in food products almost within working hours.

An Analysis of Ginseng Advertisements in 1920-1930s Newspapers During Japanese Colonial Period (일제강점기 중 1920-1930년대 신문에 실린 인삼 광고 분석)

  • The influx of modern culture in the early 20th century in Korea led to numerous changes in the country's ginseng industry. With the development of ginseng cultivation technology and commerce, the production and consumption of ginseng increased, and various ginseng products were developed using modern manufacturing technology. Consequently, competition for the sales of these products became fierce. At that time, newspaper advertisements showed detailed trends in the development and sales competition of ginseng products. Before 1920, however, there were few advertisements of ginseng in newspapers. This is thought to be because newspapers had not yet been generalized, and the ginseng industry had not developed that much. Ginseng advertisements started to revitalize in the early 1920s after the launch of the Korean daily newspapers Dong-A Ilbo and Chosun Ilbo. Such advertisements in this period focused on emphasizing the traditional efficacy of Oriental medicine and the mysterious effects of ginseng. There were many advertisements for products that prescribed the combination of ginseng and deer antler, indicating the great popularity of this prescription at the time. Furthermore, advertisements showed many personal experience stories about taking such products. Mail order and telemarketing sales were already widely used in the 1920s . In 1925, there were advertisements that ginseng products were delivered every day. The advertisements revealed that ginseng roots were classified more elaborately than they are now according to size and quality. Ginseng products in the 1920s did not deviate significantly from the scope of traditional Oriental medicine formulations such as liquid medicine, pill, and concentrated extract. In the 1930s, ginseng advertisements became more active. At this time, experts such as university professors and doctors in medicine or in pharmacy appeared in the advertisements. They recommended ginseng products or explained the ingredients and medicinal effects of the products. Even their experimental notes based on scientific research results appeared in the advertisements to enhance the reliability of the ginseng products. In 1931, modern tablet advertisements appeared. Ginseng products supplemented with vitamins and other specific ingredients as well as ginseng thin rice gruel for the sick appeared at this time. In 1938, ginseng advertisements became more popular, and advertisements using talents as models, such as dancer Choi Seunghee or famous movie stars, models appeared. Ginseng advertisements in the 1920s and 1930s clearly show a side of our rapidly changing society at the time.

The Growth Promoting Effect of Enterobacteria Bifidobacterium infantis KCTC 3270 by Combination of Natural Products Bearing hntioxidative Capacity (장내 유용세균 Bifidobacterium infantis KCTC 3270의 성장 촉진을 위한 항산화 천연산물의 조합구성)

  • The growth of enterobacteria, Bifidobacterium infantis KCTC 3270, was promoted by natural products bearing antioxidative capacity and mixed combinations of two, three and four kinds of them. B. infantis was expressed a good growth by Aurantii nobilis pericarpium majus L., Astragali radix, Epimedii herba, alone, and two mixed combinations of Epimedii herba and Theae folium, Epimedii herba and Crataegi fructus, Epimedii herba and Eucommiae cortex, and three mixed combinations of Theae folium, Epimedii herba and Chelidonium majus L., Theae folium, Epimedii herba and Aurantii nobilis pericarpium, Theae folium, Chelidonium majus L. and Auantii nobilis pericar pium, Theae folium, Chelidonium majus L. and Astragali radix, Theae folium, Aurantii nobilis pericarpium and puerariae radix, Epimedii herba, Chelidonium majus L. and Aurantii nobilis pericarpium, and four mixed com-binations of Theae folium, Chelidonium majus L., Astragali radix and Epimedii herba, and Theae folium, Chelidonium majus L., Astragali radix and Puerariae readix, and Theae folium, Epimedii herba, Aurantii nobilis pericarpium and Puerariae radix, and Theae folium, Chelidonium majus L., Aurantii nobilis pericarpium and Puerariae radix. The best four mixed combination ofr the growth of B. infantis was Theae folium, Chelidonium majus L. Astragali radix and Epimedii herba, which promoted 2.1 times than that of control, and its antioxidative capacity was also 5 times higher, and the ratio of elimination of hydroxyl radical was more than 80% in each dilution rate. As these combinations of natural products might activate some parts of body, they could be applied to pharmaceutical applications, functional foods, antiaging tea, also expected to promote useful bacterial growth for fermentative beverage bearing multifunction.

Combination of Natural Products Removing ROS for Growth Promoting Effects of the Useful Enterobacteria Lacobacillus sp. (ROS 제거기능을 갖는 천연산물로부터 장내 유용세균 Lacobacillus속의 성장을 촉진시키는 조합의 구성)

  • The growth of Enterobacteria, Lactobacillus acidophilus and L. plantarum was promoted by natural products themselves bearing antioxidative capacity and combined two, three and four kinds of them.1. acidophilus was promoted by Paeonia japonica, Acori graminei rhizoma, Aurantii nobilis pericarpium, alone, and two mixed combinations were composed of Acori graminei rhizoma and Cinnamomi cortex, Aurantii nobilis pericarpium and Puerariae radix, and three mixed combinations were formed with Cimicifugae rhi- zoma, Cinnamomi cortex and Glycyrrhizae radix, and four mixed combinations were made up Glycyrrhizae radix, Theae folium, Scutellaria baicalensis george and Cinnamomi cortex. There four combination promoted growth of 1. acidophilus with 1.3 times than that of control, and their antioxidative capacity also higher than that off times, the ratio of elimination of hydroxyl radical was more than 70% in dilution rate of 100 times. In the case of 1. plantarum was premoted by Corm fructus, Nelumbo nuclfera gaertner Sophorae flos, alone, and two mixed combinations were organized with Schizandrae Iructus and Theae folium, Paeonia japonica and Epimedii herba, and three mixed combinations were combined with Corni fructus and Theae folium and Paeonia japonica, Corni fructus and Coptidis rhizoma and Schieandrae fructus, and four mixed combinations were composed of Corm fructus, Theae folium, Coptidis rhizoma, Scutellaria baicalensis george, and Corm fructus, Schizandrae fructus, Paeonia japonica, Theae folium. These four combinations assisted growth of 1. plantarum with 1.5 times than that of control, and their antioxidative capacity also higher than that of 4-5 times, the ratio of elimination of hydroxyl radical was more than 75% in dilution rate of 100 times. As these combinations of natural products could activate some parts of body, they might be applied pharmaceuitcal sources, functional foods, and expected to fermentative beverages.

Strength Property Improvement of OCC-based Paper by Various Mechanical and Chemical Treatments of its Fiber (골판지 고지의 물리화학적 처리에 의한 강도향상)

  • To increase the strength properties of recycled fiber, especially OCC (Old Corrugated Container) in this study, we used the mechanical pretreatment on the fibers before refining. The mechanical action in the Hobart mixer induced high shear and compression on the fibers, which resulted in the breakdowns of fiber internal structure, and microcompressions on the surface of the fibers. We evaluated the degree of mechanical treatment by fiber curl index. Four different refining techniques were applied to the pretreated fibers (Valley beater, Kady mill, PFI mill, and Impact refining) to find the best combination of the pretreatment and the refining methods. Conclusions were summarized as followed. 1. In keeping the fiber length from shortening, Kady mill and PFI mill refining were effective. Kady mill and Valley beater application tended to straighten out the fiber shapes. 2. Valley beating increased the breaking length of the handsheets better than other methods, while lowering the tear strength most. The mechanical pretreatment increased breaking length about 10% in average irrespective of four different refining methods. 3. Tear strength was increased by the mechanical pretreatment and by the PFI mill refining. 4. Burst strength was increased by the mechanical pretreatment and by valley beating method. 5. In increasing the breaking length and burst strength while keeping tear strength, combination of mechanical pretreatment and Valley beating were most effective.

Development of Organic liquid Fertilizer for leaf Vegetable under Greenhouse (하우스 엽채류를 위한 관비재배용 유기액비 개발)

  • This experiment was conducted to develop liquid fertilizer for leaf vegetable using the agricultural by-products such as dry chicken dropping, bone meal, rice bran, soybean oil cake and fish meal. Combination of 50% dry chicken dropping, 30% bone meal and 20% rice bran among several combinations of by-products was selected as materials for liquid fertilizer of head lettuce and cabbage. 50kg of materials with combination selected got mixed to 200ι of water, which kept under room temperature in greenhouse. EC and pH of fertilizer was stabilized after 35 days. On the decrease of bad smell during fermentation, addition of materials such as bioceramics, woody vinegar and active charcoal was not effected. And on rapid fermentation, addition of microorganisms and sugar had a little effect, but decreased the content of inorganic nutrients. Fertigation of liquid fertilizer developed on cabbage by dropping made head weight increased by 0.9kg per plant and nitrate content decreased by 276ppm in comparison with chemical fertilizer which was 3.5kg, and 2,426ppm, respectively. By use of organic liquid fertilizer developed in this experiment, yield of cabbage could be more obtained by 26% than in use of chemical fertilizer and income by 24%.

Synergistic Effects of Cinnamomum camphora Leaves Extract against Clinical Isolated Methicillin-resistant Staphylococcus aureus (임상에서 분리한 메치실린 내성 황색포도상구균에 대한 녹나무잎 추출물의 항균활성)

  • Methicillin-resistant Staphylococcus aureus (MRSA) has been emerging worldwide as one of the most important hospital and community pathogens. At the same time, because of the difficulty in developing chemical synthetic drugs and because of their side-effects, scientists are making more efforts to search for new drugs from plant resources to combat clinical multidrug-resistant microbial infections. Cinnamomum camphora (C. camphora) is a plant of family Lauraceae, and grown Jeju island in South Korea that are used as a drug to treat neurasthenia, epilepsy, cystitis, pyelonephritis, digitalis, cancer, and diabetes mellitus in folk remedies. In this study, antibacterial activites of 80% ethanol extract of C. camphora leaves (CCE) were investigated in combination with antibiotics against clinical isolates of MRSA. The results showed that CCE was determined with MIC and MBC values ranging from 156 to 313 and 313 to $625{\mu}g/ml$, oxacillin from 128 to 256 and 128 to $512{\mu}g/ml$, ampicillin from 4 to 64 and 8 to $128{\mu}g/ml$. The combination of CCE with oxacillin or/and ampicillin were synergistic effect against MRSA 1, 6, 7, 8, 10, 11, 12, 13, and 15/ MRSA 1, 2, 6, and 7.
