Dyeing Properties on Polylactic Acid (PLA) Fabrics by Disperse Dyes (분산염료에 의한 PLA 직물의 염색성)

  • This study optimizes a suitable dyeing method for polylactic acid (PLA) fabrics using disperse dyes. For this, disperse red 60 (DR 60), disperse blue 56 (DB 56), and disperse yellow 54 (DY 54) were used and dyed on PLA fabrics dependent of dyeing temperature and time. The fastness of PLA fabrics dyed with three disperse dyes were evaluated; in addition, dye exhaustion, color strength (K/S value), and colorimetric properties of PLA fabrics were compared with PET fabrics. The experiments indicated optimum dyeability of PLA fabrics with disperse dyes. The dyeing temperature was $90^{\circ}C$ for every dye and the dyeing time were 20 min, 60 min, and 40 min for DR 60, DB 56, DY 54, respectively. PLA fabrics had good color fastness to washing, dry cleaning fastness, hot pressing fastness, rub fastness, and perspiration fastness by DR 60, DB 56, and DY 54. The dye exhaustion of PLA fabrics were lower than PET fabrics; however, K/S values were higher than PET fabrics.

  • Rhodamine chromophore/fluorophore have been attracted to many researchers due to its excellent photophysical properties. In this study, we have designed and synthesized a strong emissive fluorescent dye chemosensor for toxic elements. A rhodamine-based sensor was prepared by incorporation the rhodamine fluorophore and several functional host groups with high affinity to hazardous metal and anion. This sensor shows a high selectivity and an excellent sensitivity and is a dual-responsive colorimetric and fluorescent metal/anion-specific sensor. In addition, the 1:1 binding mode was proposed based on Job's plot method. Finally, computational calculation was simulated and calculated to approach for HOMO/LUMO of this dye chemosensor.

Effect of Cadmium on $C_6$ Glioma Cells in Culture

  • It is demonstrated that cadmium has cytotoxic effect on glial cells, oxygen radicals are involved in cadmium-induced cytotoxicity. However, the toxic mechanism of cadmium is left unknown so far. The purpose of this study was to examine the cytotoxicity of $CdCl_2$ on $C_6$ glioma cells. The cytotoxicy was measured by cell viability via XTT assay in $C_6$ glioma cells. Colorimetric assay such as XTT assay is regarded as a very sensitive screening method for the determination of the cell viability on a lots of chemicals. In this study, $CdCl_2$ decreased cell viability according to the dose- and time dependent manners after $C_6$ glioma cells were treated with various concentrations of $CdCl_2$ for 48 hours. $IC_{90}\;and\;IC_{50}$ values for XTT assay was determined at $5{\mu}M\;and\;55{\mu}M$ of $CdCl_2$, respectively. These results suggest that $CdCl_2$ has highly cytotoxic effect on $C_6$glioma cells by the decrease of cell viability.

Determination of Goitrogenic Metabolites in the Serum of Male Wistar Rat Fed Structurally Different Glucosinolates

  • Glucosinolates (GLSs) are abundant in cruciferous vegetables and reported to have anti thyroidal effects. Four GLSs (sinigrin, progoitrin, glucoerucin, and glucotropaeolin) were administered orally to rats, and the breakdown products of GLSs (GLS-BPs: thiocyanate ions, cyanide ions, organic isothiocyanates, organic nitriles, and organic thiocyanates) were measured in serum. Thiocyanate ions were measured by colorimetric method, and cyanide ions were measured with CI-GC-MS. Organic isothiocyanates and their metabolites were measured with the cyclocondensation assay. Organic nitriles and organic thiocyanates were measured with EI-GC-MS. In all treatment groups except for progoitrin, thiocyanate ions were the highest among the five GLS-BPs. In the progoitrin treated group, a high concentration of organic isothiocyanates (goitrin) was detected. In the glucoerucin treated group, a relatively low amount of goitrogenic substances was observed. The metabolism to thiocyanate ions happened within five hours of the administration, and the distribution of GLSs varied with the side chain. GLSs with side chains that can form stable carbocation seemed to facilitate the degradation reaction and produce a large amount of goitrogenic thiocyanate ions. Because goitrogenic metabolites can be formed without myrosinase, the inactivation of myrosinase during cooking would have no effect on the anti-nutritional effect of GLSs in cruciferous vegetables.

Real-Time Detection of Residual Free Chlorine and pH in Water Using a Microchannel Device

  • A microfluidic device for real-time monitoring of residual free chlorine and pH in water based on optical absorption is proposed. The device consists of a serpentine micromixer for mixing samples with a reagent, and a photodiode and light emitting diode(LED) for the detection of light absorbance at specific wavelengths, determined for specific reagent combinations. Spectral analyses of the samples mixed with N, N'-diethyl-p-phenylenediamine(DPD) reagent for chlorine determination and bromothymol blue(BTB) for pH measurement are performed, and the wavelengths providing the most useful linear changes in absorbance with chlorine concentration and pH are determined and used to select the combination of LED and photodiode wavelengths for each analyte. In tests using standard solutions, the device is shown to give highly reproducible results, demonstrating the feasibility of the device for the inexpensive and continuous monitoring of water quality parameters with very low reagent consumption.

Characterization of Chitinase in Oak Tissues and Changes in Its Activity Related to Water Stress and Inoculation with Hypoxylon atropunctatum

  • Chitinase activities from Shumard oak tissues were determined to study changes in chitinase activities related to water stress. The enzyme extracted in sodium acetate buffer (0.1M, pH 4.5) was assayed by a colorimetric method. In addition, the fungal hyphae of Hypoxylon atropunctatum in xylem tissues of oak were observed through scanning electron microscopy. The enzyme in oak tissues was mainly endochitinase, and optimum pH for enzyme activity was 5. Specific chitinase activities from both of stems held under high relative humidity (ranges of 0.63-1.11 pKatal/$\mu\textrm{g}$ of protein) and stems held under low relative humidity (ranges of 0.41-0.99 pKatal/$\mu\textrm{g}$ of protein) were significantly increased following fungal inoculation with H. atropunctatum. However, there was no significant difference in chitinase activities between tissues held under high and low humidities, which might be due to fungal chitinase. Scanning electron microscopy showed holes in fungal hyphae in the xylem tissues of stems held under high humidity but not in the stems held under ow humidity, suggesting that hyphae might be hydrolyzed by plant hydolases such as chitinase.

  • Purpose: The purpose of this study was to compare color parameters of different fabrication method of Lava ceram powder, Lava DVS blocks, IPS e.max ceram and IPS e.max zirpress. Methods: The three available shades(A1, A2, A3.5) of Lava & Kavo zirconia blocks were examined. For each of three colors, three different thickness were tested, 5 times of measurement frequency. The measurements were made using a spectrophotometer equipped with an integrating sphere using the CIE $L^*$, $a^*$, $b^*$ colorimetric system. Results: Lava build-up group and Lava DVS group showed clinically perceived color difference, on the other hand IPS e.max build-up group and IPS e.max zirpress group did not show clinically perceived color difference to shade allowed on core. When the thickness of porcelain veneer increased from Lava and IPS e.max groups, color differences(${\Delta}E$) were decreased. Conclusion: All-ceramic system has color characteristics that clinicians have to consider when selecting materials. Also, manufacturers of different porcelain systems must make every effort to achieve color reproducibility.

Antioxidant and Cholinesterase Inhibitory Activities of the By-products of Three Pandalid Shrimps

  • The antioxidant and cholinesterase inhibitory activities of the acetone and dichloromethane ($CH_2Cl_2$) extracts of the by-products (heads, shells, and tails) of Pandalus borealis, Pandalus hypsinotus, and Pandalopsis japonica belonging to the family Pandalidae were investigated and their bioactivities were compared. The antioxidant and cholinesterase inhibitory activities of the organic solvent extracts of three shrimp by-products were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-Azino-bis[3-ethylbenzothiazoline-6-sulfonic acid] ($ABTS^+$) radical scavenging activities, reducing power and xanthine oxidase (XO) inhibitory activity assays and Ellman's colorimetric method. The extracts of P. hypsinotus exhibited the highest antioxidant and cholinesterase inhibitory activities. The acetone extracts showed more potent activities toward antioxidant and cholinesterase inhibition compared with the $CH_2Cl_2$ extracts. Furthermore, the total carotenoid contents of the acetone extracts were higher than those of the $CH_2Cl_2$ extracts. Thus, the carotenoid contents may affect antioxidant and cholinesterase inhibition. Our results suggest that the shrimp by-products could act as a nutraceutical agent to prevent oxidative stress and Alzheimer's disease.

Antibacterial Effect of Huanggeumjakyak-tang against Methicillin-resistant Staphylococcus aureus (황금작약탕(黃芩芍藥湯)의 MRSA에 대한 항균활성에 관한 연구)

  • Objectives The aim of this study was to investigate antimicrobial activity of Huanggeumjakyak- tang water extract against MRSA. Methods The antibacterial activities of Huanggeumjakyak-tang were evaluated against 3 strains of Methicillin-resistant Staphylococcus aureus (MRSA) and 1 standard Methicillinsusceptible S. aureus (MSSA) strain by using the disc diffusion method, minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test and time-kill assay was performed under dark. Results The MIC (minimum inhibitory concentration) of Huanggeumjakyak-tang water extract against S. aureus strains ranged from 1,000 to $2,000{\mu}g/ml$. So we confirmed that it has a strong antibacterial effect. Also the combinations of Huanggeumjakyak-tang water extract and conventional antibiotics exhibited improved inhibition of MRSA with synergy effect. Conclusions The results obtained in this study suggest that Huanggeumjakyak-tang water extract showed antibacterial effect against MRSA, and it also showed reducing effect on the side-effect problems that are the major weak points of traditional antibiotics.

Development of Anticancer Agents from Korean Medicinal Plants (Part 4). Antitumor Activity of the Butanol Soluble Fraction of Perilla frutescens (한국산 생약으로부터 항암물질의 개발(제4보) 소엽 부탄올 가용분획의 항암활성)

  • This study was carried out to develop antitumor effect of the n-butanol soluble fraction of Perilla frutescens on (KB cells) human oral epitheloid carcinoma cells. The cytotoxictty of methanollc extract of Perilla frutescens on KB cells was evaluated by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) assay. The antitumor activity of various fractions obtained from n-butanol soluble fraction of Perilla frutescens was evaluated in human oral epithelold carcinoma cells. The antitumor acavity of the n-butanol soluble fraction on human oral epitheloid carcinoma cells was evaluated by MTT assay of colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured human oral epitheloid carcinoma cells. These results were obtained as follows; 1. The fractions 1,2 and 3 of the n-butanol soluble fraction of Perilla frutescens were shown significant antitumor activities. 2. The number of human oral epitheloid carcinoma cells were decreased and tend to form cell cluster by treatment with fractions 1,2,3 and 4 of the n-butanol soluble fraction of Perilla frutescens. 3. The fraction 1 of the n-butanol soluble fraction of Perllla frutescens showed the highest antitumor activity on Perilla frutescens. It has been selected as a lead fraction for further examinations.
