대장암 세포주 SNU C2A에서 etoposide 처리에 의한 apoptosis 유도 (Induction of apoptosis by etoposide treatment in colon cancer cell line SNU C2A)

  • Background: Inactivation of tumor suppressor genes is believed to be important in the development of many human malignancies. Recently, several lines of evidence have indicated that the wild type p53 gene located at 17p13.3, may function as a tumor suppressor gene and that a mutant p53 gene could promote transformation by inactivating normal p53 function in a dominant negative fashion. These broad spectrum of p53 mutation in human cancers provide that mutant p53 and their protein may be potential targets of tumor diagnostic and therapeutic interventions. Method: Colony formation was performed to investigate growth suppressional ability. p53 expression pattern was examined by western blot and p53-mediated transactivation ability was assessed by CAT activity. SNU C2A cells were observed in apoptotic aspects induced by etoposide and $H_2O_2$ treatment, detecting sensitivity on agent, DNA fragmentation through agarose gel, chromatin condensation by fluorescence microscope, and cell cycle distribution by FACS. Result: 1) p53 mutant his179arg ($histidine{\rightarrow}arginine$) detected in SNU C2A cells lost transcriptional activity and growth suppression ability, showing dominant negative effect on its wild type p53. 2) Etoposide-treated SNU C2A cells induced apoptosis, exhibiting dramatic reduction of cell growth, DNA fragmentation, nuclear condensation formation of apoptotic body and increment of sub-G1 cell fraction. 3) Etoposide and $H_2O_2$-treated SNU C2A cells have no high increase of p53 expression and overexpressed p53 protein changed localization, from cytoplasm to nucleus. Also, p53-mediated transcriptional activity was increased by agents-treatment. Conclusion: SNU C2A cells coexpress wild-type and mutant p53 protein induced apoptosis in the condition on DNA damage, through localizational shift from cytoplasm to nucleus of p53 protein rather than the induction of p53 protein. SNU C2A cells derived mutant p53 his179arg abrogated both the growth supression ability and transactivational activity, showing inhibition effect on transcriptional activity of wild type p53, but did not repress the activity of wild type p53 in SNU C2A cells owing to dominant activity of wild type. These cell condition may provide new gene therapeutic implications leading effective antiproliferation of cell when mutant and wild-type p53 protein were co-expressed in cell.

  • Introduction: Bone resorption is a unique function of osteoclasts. Osteoclasts are a specialized macrophage polykaryon whose differentiation is regulated principally by macrophage colony-stimulating factors, receptor activator of nuclear factor ${\kappa}B$ ligand (RANK) ligand, osteoprotegerin (OPG), and interleukins (IL). Reflecting the integrin-mediated signals, osteoclasts develop a specialized cytoskeleton that allows it to establish an isolated micro-environment between itself and the bone, wherein matrix degradation occurs by a process involving proton transport. The levels of IL-1, IL-6, OPG, and prostaglandin $E_2$ ($PGE_2$) expression were evaluated to study the correlations between dental implant teeth and the adjacent teeth. Materials and Methods: The exudate of the gingival crevice acquired from dental implants, adjacent teeth, opposite teeth and contralateral teeth of 24 patients. Results: 1. The levels of IL-1, IL-6, OPG and $PGE_2$ expression in dental implant teeth were higher than those of the contralateral teeth. 2. IL-1 revealed a higher expression level in the adjacent teeth than in dental implant teeth. 3. The dental implant teeth and adjacent teeth did not show a remarkable difference in the level of IL-1 expression. 4. All the other cytokines were strongly expressed in the dental implant compared to the adjacent teeth. Conclusion: These results suggest that there might be close correlation between dental implant teeth and adjacent teeth in terms of the expressions of cytokines that affect the development and regulation of osteoclasts.

급식소 식품접촉표면 위생 모니터링 도구로서의 ATP Luminometer 적합성 확인 (Verification of the Suitability of the ATP Luminometer as the Monitoring Tool for Surface Hygiene in Foodservices)

  • 창원시 소재 단체급식소 4개소에서 사용하는 칼($12\;cm^2$), 도마($100\;cm^2$), 고무장갑(손바닥과 손가락 사이)의 표면을 대상으로 ATP Luminometer와 일반세균수용 건조필름을 이용한 미생물 배양법을 함께 적용하여 ATP(RLU)와 APC (CFU)을 측정하였다. 통계분석 결과, 3가지 조리도구 모두에서 ATP(RLU)와 APC(CFU) 간에 유의적인 양의 상관성이 존재하였다. 각 도구별 상관계수는 칼 0.84(p<0.001), 도마 0.79(p<0.001), 고무장갑 0.78(p<0.001)로 나타났다. 본 실은 단체급식 현장에 적합한 ATP Luminometer 가이드라인을 설정하기에 앞서 실시간적인 표면 위생 모니터링 도구로서의 사용 적합성을 살펴 본 예비실험이며, 도출된 회귀식을 이용하여 도구별 ATP(RLU) 가이드라인을 설정하고 검증할 예정이다.

  • The study was to introduce the revegetation measures for the improvement of the landscape in October, 2012 on the slope; two and four years after the introduction, the slope was examined to see the status of the revegetation. The result is as follows. The result from examining the soil on the slope, the soil was Loamy sand; which infers that the soil is in a poor condition for improving the scenery. Examining the characteristics of the vegetation-base materials, the soil acidity was pH 6.4 to 6.7; and the soil hardness was 21 to 24mm (hardness quotient of soil), this can be considered a favourable condition, which has no effect on the growth and development of plants. When it comes to the discovery of the species in the area, Lotus corniculatus var., Silene armeria L., Papaver rhoeas, Trifolium pratense L. developed and grew in spring after two years. And in the summer, Lotus corniculatus var., Dianthus chinensis L., Cosmos bipinnatus, Coreopsis drummondii L., Trifolium pratense were founded. And there were Lotus corniculatus var., Dianthus chinensis L., Aster yomena, Coreopsis tinctoria, Trifolium pratense L. in the autumn. According to the examination conducting after four years, Lotus corniculatus var., Dianthus chinensis L., Silene armeria, Cosmos bipinnatus, Coreopsis drummondii L., Papaver rhoeas, Trifolium pratense L. were founded in the spring, and Lotus corniculatus var., Dianthus chinensis L., Cosmos bipinnatus, Coreopsis drummondii L., Trifolium pratense grew in the summer. Lotus corniculatus var., Dianthus chinensis L., Astragalus sinicus, Aster yomena, Coreopsis tinctoria, Trifolium pratense L. were discovered in the autumn. Among the exotic species founded in the area, were Festuca arundinacea, Lolium perenne, Festuca rubra. When it comes to the protection of vegetation and dominance, Lotus corniculatus var., Metaplexis japonica, Coreopsis drummondii L. are the dominant species among the spring plants; and Lotus corniculatus var., Trifolium repens L., Erigeron annuus (L.) Pers. were the dominant plants in the summer. In the autumn, Lotus corniculatus var., Coreopsis drummondii L., Poa pratensis appear to be the dominant plants. The colonies of Pinus densiflora and Castanea crenata var. were formed around the South Sejong IC. Part of the area is occupied by the colony of Alnus hirsuta Turcz. ex Rupr. In terms of shrub, there were Indigofera pseudo-tinctoria, Robinia pseudoacacia, and Rosa polyantha var. genuina NAKA. As ground cover plants, Sasa borealis, Rubus crataegifolius Bunge were founded.

Parthenogenetic Activation of Porcine Oocytes and Isolation of Embryonic Stem Cells-like Derived from Parthenogenetic Blastocysts

  • These experiments were carried out to optimize the parameters of electrical activation, methods of parthenogenetic activation and embryo culture in vitro and meanwhile to isolate embryonic stem cells-like (ESCs) derived from porcine parthenogenetic blastocysts (pPBs). These results showed that, as the electric field strength increased from 1.0 to 2.7 kV/cm, the cleavage rate of parthenogenetic embryos increased gradually but the rate of oocyte lysis was significantly increased when using 2.7 kV/cm field strength. The rate of cleavage in 2.2 and 2.7 kV/cm groups was significantly increased in comparison with that of the 1.0 kV/cm group. A voltage field strength of 2.2 kV/cm DC was used to investigate blastocyst development following activation with a single pulse of 30 or $60-{\mu}sec$ pulse duration. The optimum pulse duration was 30-${\mu}sec$, with a blastocyst rate of 20.7%. Multiple pulses were inferior to a single pulse for blastocyst yield (8.0% vs. 29.9) (p<0.05). For porcine oocyte parthenogenetic activation methods, the rates of cleavage (79.0% vs. 59.8%) and blastocysts (19.4% vs. 3.4%) were significantly increased in electrical activation in contrast to chemical activation with ionomycin/6-DMAP (p<0.05). Rates of cleavage and blastocyst formation in NCSU-23 and PZM-3 embryo media were higher than those of G1.3/G2.3 serial culture media, but there was no significant difference among the three groups. The total cell number of blastocysts in PZM-3 embryo culture media containing $5{\mu}g/ml$ insulin was significantly higher than that of the control (no insulin) ($44.3{\pm}9.1$ vs. $33.9{\pm}11.7$). For isolation of PESCs-like, the rates of porcine blastocysts attached to feeder layers and ICM colony formation in Method B (nude embryo culture) were better than those in Method A (intact embryo culture).

  • 본(本) 시험(試驗)은 양송이 복토중(覆土中) 미생물(微生物)의 Mycogone perniciosa Magn.에 대한 항균작용(抗菌作用)및 M. perniciosa 와 Agaricus bisporus (Lange) Sing. 사이의 상호관계(相互關係)를 구명(究明)하기 위하여 실시(實施)되었으며 시험(試驗)한 결과(結果), 1. In vitro test에서 M. perniciosa는 복토(覆土)에서 분리(分離)된 미동정(未同定)의 미생물(微生物)들에 의해서 생장(生長)이 억제(抑制)되었으며 재배시험(栽培試驗)에서 M. perniciosa의 포자현탁액(胞子懸濁液)을 복토표면(覆土表面)에 국소접종(局所接種)하였을 때 라병(羅病)버섯은 접종(接種)된 부분(部分)에서만 발생(發生)되었고 접종부분(接種部分) 주변(周邊)에서는 발생(發生)되지 않았다. 2. M. perniciosa와 A. bisporus사이의 길항적(拮抗的) 관계(關係)는 in vitro test에서 배양기(培養基)의 종류(種類)에 따라 다소(多少) 차이(差異)가 있었으나 A. bisporus의 균사(菌絲)는 M. perniciosa 균총(菌總) 위를 덮어서 자랐다. 재배시험(栽培試驗)에서 M. perniciosa의 포자현탁액(胞子懸濁液)을 A. bisporus의 곡립종균(穀粒種菌) 표면(表面) 혹은 복토층(覆土層)의 중간부위(中間部位)에 분무접종(噴霧接種)하였을 때에는 라병(罹病)버섯이 발생(發生)되지 않았으나, 복토표면(覆土表面)에 접종(接種)하였을 때에만 리병(罹病)버섯이 발생(發生)되었다.

효소분리 및 용매정제법으로 제조한 고농도 Sialic Acid(23%)가 함유된 GMP 가수분해분말의 미생물복귀돌연변이시험 연구 (Bacterial Reverse Mutation Test Evaluation of Hydrolyzed GMP Powder Containing Highly Concentrated Sialic Acid (23%) produced by Enzyme Separation and Solvent Enrichment Method)

  • 본 시험은 안전식품인 우유단백질에서 분리되어 영유아 식품 등에 세계적으로 사용되고 있는 GMP 내 기본적으로 7%로 결합되어 있는 지표성분인 sialic acid의 함유량이 높게 제조한 23%-GNANA를 기능성 식품 원료 개발함에 최종 연구 목표를 두었다. 시험물질은 23% sialic acid와 GMP 단백질로 구성(제품명: HELICOBACTROL-23)되어 있으며, (주)한일바이오메드사(한국)에서 공여 받아 미생물복귀돌연변이시험을 실시하였다. 미생물에 대한 돌연변이 유발성 유무를 검색하기 위해 히스티딘 요구성 균주인 Salmonella typhimurium TA98, TA100, TA1535 및 TA1537과 트립토판 요구성 균주인 Escherichia coli WP2uvrA를 이용하였다. 미생물복귀돌연변이시험은 시험물질을 5단계의 농도군(0, 61.7, 185, 556, 1,670, $5,000{\mu}g/plate$)으로 하여 평가하였다. 평가결과로서, 대사활성계 존재 유무와 관계없이 5균주 모두에서 시험물질의 각 농도에 의한 복귀돌연변이 유발원 양성기준인 콜로니 생성수치가 재현성있는 증가를 나타내지 않았으며, 용량의존성도 확인되지 않았다. 결론적으로, 시험물질인 23%-GNANA의 식품첨가물로서 등록을 위하여 수행한 미생물돌연변이시험에서 안전성이 확인되었다.

하동 지역에 서식하는 바지락의 미생물총 분포에 관한 정량 및 정성적 분석 (Quantitative and Qualitative Studies of Commensal Bacterial Flora of Clam, Ruditapes philippinarum in Hadong Area)

  • 바지락 조직과 주변 환경에 있는 세균의 특성과 분포를 여러 배지와 온도 그리고 생화학적 동정 kit를 사용하여 비교 분석하였다. 장내, 갯벌, 아가미 그리고 외부체액에 있는 미생물의 집락수는 사용한 BHIA, STA 그리고 SNA 배지에 영향을 받지 않는 것으로 나타났다. 그리고 $15^{\circ}C$ 배양조건에서의 증식속도는 $25^{\circ}C$$35^{\circ}C$의 조건에 비해 느린 것으로 나타났으나 총 세균 집락수는 $15^{\circ}C$에서의 것이 가장 높게 나타났다. 영양배지에서 자란 세균집락을 무작위적으로 선택배지에 옮기면 조직 시료를 직접 도말 한 경우에 비해 높은 비율로 집락형성을 하는 것을 보여 주었다. API 20E와 API 20NE를 사용하여 각 장기 또는 외부에 있는 세균의 종류를 동정한 결과 바지락 조직간에는 서로 유사한 종의 분포를 보이나 주위 환경이 되는 갯벌이나 외부체액에 있는 미생물군의 종류와는 다르게 나타났다. 바지락 주위의 갯벌이나 외부체액의 미생물군은 Pseudomonas 가 주종을 이루고 있는 조직내의 미생물군에 비해서 훨씬 높은 다양성을 보여 주었다. 이러한 결과는 바지락 조직이 주위 환경중의 세균에 대한 선택적 친화력이 있음을 의미한다고 할 수 있다.

The Effect of Bacterial Inoculants and a Chemical Preservative on the Fermentation and Aerobic Stability of Whole-crop Cereal Silages

  • Three microorganisms and one chemical preservative were tested for their effects on the fermentation and aerobic stability of whole-crop wheat, sorghum and maize silages. Wheat at the early dough stage, sorghum at the late milk stage and maize at the one-third milk line stage were harvested and ensiled in 1.5-l anaerobic jars untreated or after the following treatments: control (no additives); Lactobacillus plantarum (LP) at $1.0{\times}10^6$ colony-forming units (CFU)/g of fresh forage; L. buchneri (LB) at $1.0{\times}10^6$ CFU/g; Propionibacterium acidipropionici (PA) at $1.0{\times}10^6$ CFU/g; and a formic acid-based preservative (FAP) at 3 ml/kg of fresh forage weight. Three jars per treatment were sampled on d 90 after ensiling, for chemical and microbiological analysis. At the end of the ensiling period, 90 d, the silages were subjected to an aerobic stability test lasting 5 d. In this test, $CO_2$ produced during aerobic exposure was measured along with chemical and microbiological parameters which serve as spoilage indicators. The silages inoculated with LP had higher concentration of lactic acid compared with the controls and the other treated silages (p<0.05). The controls and LP-inoculated silages spoiled upon aerobic exposure faster than LB, PA and FAP-treated silages. The controls and LP-inoculated silages spoiled upon aerobic exposure faster than LB, PA and FAP-treated silages due to more $CO_2$ production (p<0.05) in these two groups and development of yeasts unlike the other groups. In the experiment, the silages treated with LB, PA and FAP were stable under aerobic conditions. However, the numbers of yeasts was higher in the LP-inoculated wheat, sorghum and maize silages compared with the LB, PA and FAP-treated silages. The LB, PA and FAP improved the aerobic stability of the silages by causing more extensive heterolactic fermentation that resulted in the silages with high levels of acetic and propionic acid. The use of LB, PA and FAP as silage additives can improve the aerobic stability of whole-crop wheat, sorghum and maize silages by inhibition of yeast activity.

Development of High-specificity Antibodies against Renal Urate Transporters Using Genetic Immunization

  • Recently three proteins, playing central roles in the bidirectional transport of urate in renal proximal tubules, were identified: two members of the organic anion transporter (OAT) family, OAT1 and OAT3, and a protein that designated renal urate-anion exchanger (URAT1). Antibodies against these transporters are very important for investigating their expressions and functions. With the cytokine gene as a molecular adjuvant, genetic immunization-based antibody production offers several advantages including high specificity and high recognition to the native protein compared with current methods. We fused high antigenicity fragments of the three transporters to the plasmids pBQAP-TT containing T-cell epitopes and flanking regions from tetanus toxin, respectively. Gene gun immunization with these recombinant plasmids and two other adjuvant plasmids, which express granulocyte/macrophage colony-stimulating factor and FMS-like tyrosine kinase 3 ligand, induced high level immunoglobulin G antibodies, respectively. The native corresponding proteins of URAT1, OAT1 and OAT3, in human kidney can be recognized by their specific antibodies, respectively, with Western blot analysis and immunohistochemistry. Besides, URAT1 expression in Xenopus oocytes can also be recognized by its corresponding antibody with immuno-fluorescence. The successful production of the antibodies has provided an important tool for the study of UA transporters.
