Beauty food activities of wild-cultivated Ginseng (Panax ginseng C.A. Meyer) ground part (산양삼(Panax ginseng C.A. Meyer) 지상부위의 미용 식품 활성)

  • This study aimed to investigate the beauty food activities of wild-cultivated ginseng (Panax ginseng C.A. Meyer). wild-cultivated ginseng extracts were analyzed for antioxidant, skin whitening, anti-wrinkle effect was measured in water and 70% ethanol extract. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical decolorization activities of water and 70% ethanol extracts were 16.69 and 2.18% as well as 4.04 and 3.25% at a solid content of $200{\mu}g/mL$, respectively. The antioxidant protection factors (PF) of water and 70% ethanol extracts at a solid content of $200{\mu}g/mL$ were 1.06 PF and 1.09 PF, respectively. Thiobarbituric acid reactive substance (TBARs) were both 96% at a solid content of $200{\mu}g/mL$. As PF and TBARs showed higher activity than DPPH and ABTS, we could know that antioxidant activity in the lipophilic component of wood-cultivated ginseng were superior to water-soluble component of wood-cultivated ginseng. Tyrosinase inhibitory activity was 10.97 and 52.39% in water and 70% ethanol extracts at a solid content of $200{\mu}g/mL$. The collagenase and elastase inhibitory activities as anti-wrinkle effect were 15.71 and 20.43% in water extracts as well as 32.26 and 86.74% in 70% ethanol extract at a solid content of $200{\mu}g/mL$. The results show that anti-wrinkle effect was the best among the other experiments. This extracts from wood-cultivated ginseng, therefore, seems to be a potent beauty food resource against wrinkles.

  • This study was designed to compare the effects on antioxidant, whitening, and anti-wrinkle improvement of 60% ethanol extracts from Trigonotis radicans var. sericea (TR) and Lactobacillus brevis-fermented T. radicans var. sericea (FTR). The total phenolics contents of TR was 18.6 mg/g, and FTR was 18.2 mg/g. The HPLC analysis confirmed more phenolic compounds were produced in FTR by bioconversion than in TR. The FTR showed higher 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity than TR. The results of the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation decolorization assay, FTR and TR showed higher activity than BHT (positive control). The FTR (1.21-1.29 PF) showed higher antioxidant protection factor (PF) activity than the TR (1.15-1.22 PF) and showed higher tyrosinase, collagenase and elastase inhibitory activities. This results showed L. brevis -fermentation of Trigonotis radicans var. sericea changed the composition profile and has effects on antioxidant, whitening and anti-wrinkle improvement.

Anti-wrinkle Effect of Cambodian Phellinus linteus Extracts (캄보디안 상황버섯 추출물의 주름개선 효과 연구)

  • The skin of human is constantly being exposed to environmental irritants such as ultraviolet, smoke and chemicals. These irritants cause free radicals and reactive oxygen species which leave serious damages on the cells of skin. The water and ethanol extracts of Cambodian Phellinus linteus were investigated for the activities of anti-lipid peroxidation and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. As the result of evaluation of liquid oxidation rate by add $Fe^{2+}$ and $Cu^{2+}$ to Cambodian Phellinus linteus extracts, Cambodian Phellinus linteus ethanol extracts were higher than Cambodian Phellinus linteus water extracts in the chealting ability of $Fe^{2+}$ and $Cu^{2+}$. The Cambodian Phellinus linteus ethanol extracts exhibited that anti-lipid peroxidation higher than butylated hydroxytoluene (BHT) at the concentration of 0.1 mg/ml, 0.5 mg/ml and 1 mg/ml. Cambodian Phellinus linteus water and ethanol extracts showed a higher inhibitory effect on $Fe^{2+}$-induced lipid peroxidation compared to $Cu^{2+}$-induced lipid peroxidation. In the case of anti-wrinkle effect, the elastase inhibition activity of Cambodian Phellinus linteus ethanol extracts was 50.7%, and it is higher than urosolic acid at the concentration of 0.01 mg/ml. Also, in collagenase inhibition activity, Cambodian Phellinus linteus water extract showed low effect, but Cambodian Phellinus linteus ethanol extract was about 50% at a 0.1 mg/ml. concentration. These results proved that Cambodian Phellinus linteus had anti-lipid peroxidation and anti-wrinkle effect. Therefore, Cambodian Phellinus linteus could be useful as an anti-wrinkle cosmetic ingredient.

Whitening and anti-wrinkle effect of Pinus koraiensis leaves extracts according to the drying technique (건조방법에 따른 잣나무(Pinus koraiensis) 잎 추출물의 주름개선 및 미백 효과)

  • This study provide activity for beauty food of water and 80 % ethanol extracts from Pinus koraiensis leaves. Total phenolic content of extracts from Pinus koraiensis leaves were each 12.22 mg/g (Drying under hot air) and 17.93 mg/g (Drying under shade), 14.36 mg/g (Lyophilization) in water extracts (WE) and 11.9 mg/g and 20.63 mg/g, 17.96 mg/g in 80 % ethanol extracts (EE). The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of extracts from Pinus koraiensis leaves was 96.20 % in EE from drying under shade at extracts concentration. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical decolorization activity of extracts from drying under shade was 99.85 % in WE and 99.80 % in EE at extracts concentration. The antioxidant protection factor (PF) extracts from drying under shade type was 9.63 PF in WE and 10.48 PF in EE at extracts concentration. The thiobarbituric acid reactive substance from Pinus koraiensis leaf was 89.39 % in EE from drying under shade at extracts concentration. The elastase inhibition activity of EE for anti-wrinkle effect showed an excellent wrinkle improvement effect, showing 71.46 % in EE from lyophilization. Collagenase inhibition activity of EE from drying under shade was 97.48 % in extracts. Tyrosinase inhibition activity which was related to anti-melanogensis was observed. The tyrosinase inhibitory effect of extracts from lyophilization was confirmed to be 60.4 % in EE more than another drying methods at extracts concentration. Through out all results, it can be expected Pinus koraiensis leaves extracts to use as a functional material for anti-oxidant and functional beauty food.

Antioxidation and Anti-photoaging Effects of White Taraxacum Coreanum Extract by Lactobacillus plantarum (Lactobacillus plantarum에 의한 흰 민들레 발효물의 항산화 및 광노화 억제 효과)

  • Lactobacillus plantarum SM4, a strain producing β-glucosidase, was isolated from kimchi and fermented with white Taraxacum coreanum to enhance the production of bioactive compounds. The total polyphenol content(TPC), total flavonoid content(TFC), radical scavenging activity(RSA), tyrosinase inhibitory activities(TIA), and collagenase inhibitory activities(CIA) were measured to evaluate the skin whitening and anti-wrinkle effects of the fermented product. The TPC of fermented white T. coreanum was 41.8±0.26 mg GAE/g DW, which was approximately two times higher than the hot-water extraction of 21.4±0.67 mg GAE/g DW. RSA, an indicator of antioxidant activity, was 65.6±4.7% in fermentation, which is four times higher than that of the hot-water extract. TAI and CAI, which are indicators of the whitening and anti-wrinkle effects, were 87.9±4.73% and 66.7±3.48%, respectively, which were 2.4 and 1.5 times higher than those of hot-water extraction. When comparing the UVA(320 nm) protection effects of fermented and hot-water extraction, the fermented white T. coreanum showed higher protection with an absorption rate of 64.7% and 15.2%, respectively. The white T. coreanum fermented product showed higher bioactive properties and improved skin whitening, anti-wrinkle, and UV protection effects through the production of β-glucosidase from L. plantarum SM4.

Beauty food activities of Polygala japonica Houtt. (영신초(Polygala japonica Houtt.)의 미용 식품 활성)

  • The objective of this study was to identify beauty food activity of Polygala japonica Houtt., which were widely used as a folk medicinal plant to treat inflammatory diseases. Both hot water and 50% ethanol extract of P. japonica Houtt. were evaluated for their anti-oxidant, whitening, anti-wrinkle and anti-inflammatory effect based on their phenolic concentration. To measure anti-oxidant activity, four experiments were carried out. 1,1-diphenyl-3-picrylhydrazyl, 2,2-azinobis (3-ethylbenzothia-zoline-6-sulfonic acid) and thiobarbituric acid reactive substances all showed more than 80% efficacy in both hot water and 50% ethanol extract of P. japonica Houtt. and likewise antioxidant protection factor value more than 2. To investigate the whitening effect of P. japonica Houtt., the inhibition effect of tyrosinase on hot water and 50% ethanol extract was inhibited by 12.46 and 22.10%, respectively. To examine the anti-wrinkle effect, the inhibition effect of elastase and collagenase on hot water and 50% ethanol extract were inhibited by 28.28, 58.36% and 59.73, 78.90% respectively. To investigate the anti-inflammatory effect and moisturizing effect by hyaluronidase inhibitory effect, hot water and 50% ethanol extract were 35.65 and 86.80%, respectively. The result of all the above experiments show values at a concentration of $200{\mu}g/mL$, and based on these experimental results, P. japonica Houtt. can be expected as a functional material in beauty foods.

Inhibitory Effects of Trichosanthis Radix in the Activity and Proliferation of Th2 T Cells and Eosinophils in vitro : Implications on its Regulatory Roles for Asthma (과루근(瓜蔞根)이 Th2 T 세포와 호산구에 대한 활성 및 증식 억제에 미치는 영향)

  • Objectives : The present study was carried out to investigate the effect of Trichosanthis Radix extract (TRE) on the proliferation and activation of eosinophils which were prepared from lung cells of asthma-induced mice by ovalbumin (OVA) treatment. Methods : C57BL/6 mouse was exposed to OVA three times a week for 6 weeks. The mouse lung tissues were dissected out, chopped and dossiciated with collagenase (1 $\mu$g/ml). Eosinophils were activated by rmIL-3/rmIL-5 co-treatments. The lung cells were treated with TRE, incubated for 48 hr at 37$^{\circ}C$, and analyzed by flow cytometer, ELISA and RT-PCR methods Results : To measure cytotoxicity, mouse lung fibroblast cells (mLFCs) were pretreated with various concentrations of TRE. TRE at 100 $\mu$g/ml, the highest concentration, examined did not have any cytotoxic effects on mLFCs. In FACS analysis, number of granulocyte/lymphocyte, CD3e-/CCR3+, CD3e+/CD69+, CD4+/CD8+ T cells in asthma-induced lung cells were significantly decreased by TRE treatment compared to the control group. But CD4+/CD25+ T cells were not examined significant change in lung cells treated with TRE. In ELISA analysis, production levels of IL-3, IL-5, IL-13 and histamine in asthma-induced lung cells, which were induced by rIL-3 plus rmIL-5 co-treatment, were significantly decreased by TRE treatment. Conclusions : The present data suggested that Trichosanthis Radix on the inhibition of parameters associated with asthma responses in eosinpophils, and thus implicate the possibility for the clinical application of Trichosanthis Radix.

Various Physiological and Anti-inflammatory Effects of Sanguisorba officinalis L. Roots as a Functional Cosmetic Material (기능성 화장품 소재로써 오이풀 뿌리(Sanguisorba officinalis L. roots)의 다양한 생리 활성 및 항염증 효과)

  • In this study, the various physiological and anti-inflammatory activities of Sanguisorba officinalis L. roots (SR) were assessed for potential use as functional cosmetic materials. As a result of measuring electron-donating abilities to determine the antioxidant ability of SR extract, activity increased as the concentration increased, showing an excellent antioxidant capacity of 93.8% at a 1,000 ㎍/ml concentration. Further, the antioxidant power of SR extract, which was determined using an ABTS+ assay measurement, was more than 99% at concentrations of 50 ㎍/ml or more, while the tyrosinase inhibition rate was 37.7% at the highest concentration of 1,000 ㎍/ml. Consequently, the elastase and collagenase inhibition of SR extract measured 84.9% and 90.3%, respectively, at a 1,000-㎍/ml concentration. As a result of confirming the survival rate of Raw 264.7 cells, the cell survival rate was determined to be 80% or more below a 100 ㎍/ml concentration, and subsequent cell-related experiments were conducted at concentrations below 100 ㎍/ml. Furthermore, after applying a NO assay to identify anti-inflammatory activity, it was confirmed that SR extract had an inhibitory rate of 50.8% at a concentration of 500 ㎍/ml, and it was excellent at suppressing the inflammatory expression. As a result of verifying protein expression by treating SR extract in Raw 264.7 cells, it was confirmed that expression was inhibited concentrated in all factors. Therefore, it is judged that SR can be used as a functional cosmetic material with antioxidant, whitening, and wrinkle-improving physiological effects and anti-inflammatory activities.

Effect of Cervi Pantotrichum Cornu Herbal acupuncture on protease activities, antioxidant in Rheumatoid arthritis rats (류마티스 관절염 실험용쥐의 활액에서 단백분해효소의 활성 및 항산화에 대한 녹용약침의 효과)

  • We have compared(using the same series of experimental tissue samples) the levels of proteolytic enzyme activities and free radical-induced protein damage in synovial fluid from RA and CPH cases. Many protease types showed significantly increased (typically by a factor of approximately 2-3-fold) activity in RA, compared to normal rats. However, CPH significantly reduced the cytoplasmic enzyme activities of arginyl aminopeptidase, leucyl aminopeptidase, pyroglutamyl aminopeptidase, tripeptidyl aminopeptidase, and proline endopeptidase to almost about 1/10 each. For the Iysosomal proteases, synovial fluid samples from RA rats, CPH significantly reduced the enzyme activities of cathepsin B, dipeptidyl aminopeptidase I and dipeptidyl aminopeptidase II. In extracellular matrix degrading(collagenase, tissue elastase) and leukocyte as sociated proteases (leukocyte elastase, cathepsin G), CPH decreased these enzyme activities of collagenase, tissue elastase and leukocyte associated elastase in RA. In cytoplasmic and lysosomal protease activities in plasma from RA. CPH and normal plasma samples were not significantly different, suggesting that altered activity of plasma proteases (particularly those enzymes putatively involved in the immune response) is not a contributory factor in the pathogenesis of RA. In addition, the level of free radical induced damage to synovial fluid proteins was approximately twice that in RA, compared with CPH. CPH significantly decreased the level of ROS induced oxidative damage to synovial fluid proteins (quantified as protein carbonyl derivative). Therefore we conclude that both proteolytic enzymes and free radicals are likely to be of equal potential importance as damaging agents in the pathogenesis of inflammatory joint disease, and that the design of novel therapeutic strategies for patients with the latter disorder should include both protease inhibitory and free radical scavenging elements. In addition, the protease inhibitory element should be designed to inhibit the action of a broad range of protease mechanistic types (i.e. cysteine-, metallo- and serine- proteinases and peptidases). However, increased protein damage induced by ROS could not be rationalised in terms of compromised antioxidant total capacity, since the latter was not significantly altered in RA synovial fluid or plasma compared with CPH.

Protection of UV-derived Skin Cell Damage and Anti-irritation Effect of Juniperus chinensis Xylem Extract (향나무추출물의 광손상으로부터 피부세포 보호와 자극완화 효과에 대한 연구)

  • The human skin is constantly exposed to environmental irritants such as ultraviolet, smoke, chemicals. Free radicals and reactive oxygen species (ROS) caused by these environmental facts play critical roles in cellular damage. These irritants are in themselves damaging to the skin structure but they also participate the immensely complex inflammatory reaction. The purpose of this study was to investigate the skin cell protective effect of Juniperus chinensis xylem extract on the UV and SLS-induced skin cell damages. We tested free radical and superoxide scavenging effect in vitro. We found that Juniperus chinensis xylem extracts had potent radical scavenging effect by 98% at 100 $\mu\textrm{g}$/mL. Fluorometric assays of the proteolytic activities of matrix metalloproteinase-l(MMP-1, collagenase) were performed using fluorescent collagen substrates. UV A induced MMP-1 synthesis and activity were analyzed by enzyme-linked immunosorbent assay (ELISA) and gelatin-based zymography in skin fibroblasts. The extract of Juniperus chinensis showed strong inhibitory effect on MMP-1 activities by 97% at 100 $\mu\textrm{g}$/mL and suppressed the UVA induced expression of MMP-1 by 79% at 25 $\mu\textrm{g}$/mL. This extract also showed strong inhibition on MMP-2 activity in UVA irradiated fibroblast by zymography. We also examined anti-inflammatory effects by the determination test of proinflammatory cytokine, interleukin 6 in HaCaT keratinocytes. In this test Juniperus chinensis decreased expression of interleukin 6 about 30%. Expression of prostaglandin E$_2$, (PGE$_2$) after UVB irradiation was measured by competitive enzyme immunoassay (EIA) using PGE$_2$ monoclonal antibody. At the concentrations of 5-50 $\mu\textrm{g}$/mL of the extracts, the production of PGE$_2$ by HaCaT keratinocytes (24 hours after 10 mJ/$\textrm{cm}^2$ UVB irradiation) was significantly inhibited in culture supernatants (p〈0.05). The viability of cultured HaCaT keratinocytes was significantly reduced at the doses of above 10 mJ/$\textrm{cm}^2$ of UVB irradiation, but the presence of these extracts improved cell viability comparing to control after UVB irradiation. We also investigated the protective effect of this extract in sodium lauryl sulfate (SLS)-induced irritant skin reactions from 24 hour exposure. Twice a day application of the extract for reducing local inflammation in human skin was done. Irritant reactions were assessed by various aspects of skin condition, that is, erythema (skin color reflectance) and transepidermal water loss (TEWL). After 5 days the extract was found to reduce SLS-induced skin erythema and improve barrier regeneration when compared to untreated symmetrical test site. In conclusion, our results suggest that Juniperus chinensis can be effectively used for the prevention of UV and SLS-induced adverse skin reactions such as radical production, inflammation and skin cell damage.
