回春凉膈散과 龍石散이 抗炎作用에 미치는 影響 (Experimental study on the Anti-inflammatory and wound healing effect of Hoichunyanggyuksan and Yongseksa.)

  • Hoichunyonggyuksan(HCYGS) and Yongseksan(YSS) are important prescriptions that have been used in oriental medicine for stomatitis and wound healing. The study was done to evaluate the inhibitory effects of cytotoxicity, formation of superoxide on the macrophage and neutrophil, prostaglandins($PGE_2$), interleukins($IL-1{\Beta}$), collagenase activity and synthesis of collagen and DNA. The results were obtained as fo1lows: 1. HCYGS and YSS were not showed the proliferation difference of human fibroblast and monocyte in all concentrations to be experimented and in result, it was concluded that they have no cytotoxicity. 2. YSS inhibited the formation of superoxide to $48\% at the concentration of $0.001\%$ in the mouse monocyte. 3. HCYGS inhibited the formation of superoxide to $13\%$ at the concentration of $0.001\%$ as compared with control in the human monocyte. 4. HCYGS and YSS inhibited the formation of superoxide to $25\%\;and\;35\%$ respectively at the concentration of $0.0001\%\;and\;HCYGS\;showed\;38\%$ inhihitory rate on the formation of superoxide at concentration of $0.001\%$ in the human neutrophil. 5. The concentration of inhibiting the production of prostaglandins($PGE_2$) to $11\%$ in the human monocyte stimulated with E. coli were $0.01\%$ of HCYGS. 6. The concentration of inhibiting the production of interleukins($IL-l{\Beta}$) to $43\%\;and\;39\%$ in the human monocyte stimulated with E. coli were $0.01\%$ of HCYGS and YSS. 7. HCYGS and YSS didn't influence on collagen synthesis and total protein in fibroblasts. 8. HCYGS and YSS inhibited the collagenase activity to $22\%\;and\;19\%\;at\;0.1\%,\;45\%\;and\;56\%\;at\;0.2\%,\;57\%\;and\;52\%$ at $0.5\%$ respectively, but YSS exerted the inhibitory effect on collagenase activity to $27\%$ at the lower concentration of $0.01\%$. 9. HCYGS and YSS didn't show the faster recovary than control group hut exerted the consistant effect on the anti-inflammations and the formation of granulation tissue.

노회(蘆薈)(알로에), 자화지정(紫花地丁)의 항산화, 항염증, 주름, 미백에 미치는 영향 (Effects of Aloe and Violae herba Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening)

  • Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.

Effects of Polygonati Rhizoma Extracts on the Collagenase Activity and Procollagen Synthesis in Hs68 Human Fibroblasts and Tyrosinase Activity

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.

우절 에탄올추출물의 미백활성능과 주름저해 효능평가 (Evaluation of Whitening Activity and Wrinkle Inhibitory Effect of Ethanol Extracts of Nelumbinis Rhizomatis Nodus)

  • 본 연구는 화장품 소재로서 우절 에탄올추출물의 가능성을 확인하기 위한 것이다. 이를 위해 우리는 연근의 마디부분을 70%에탄올로 추출하여 사용하였으며 미백효능 및 주름개선 효과에 대한 생물학적 활성 평가를 수행하였다. 시료의 미백활성을 알아보기 위해 멜라닌세포(B16F10 cells)의 MTT assay를 이용한 샘플의 독성평가와 멜라닌 생성에 영향을 주는 관련 단백질의 발현량을 확인하였다. 시료의 주름활성억제 평가를 위해 collagenase inhibition assay를 수행하였다. 또한 UVB로 유도된 섬유아세포(CCD-986sk cells)의 MTT assay를 이용한 샘플의 독성평가와 matrix metalloprotease (MMP-1) inhibition 및 procollagen synthesis를 평가하였다. 미백활성 평가를 알아보기 위한 western bolt 결과, 멜라닌 생성과 관련된 두 단백질 Tyrosinase related protein-1 (TRP-1, Tyrosinase related protein-2 TRP-2)의 합성이 농도의존적으로 감소됨을 나타내었다. 게다가 우절 에탄올추출물의 collagenase inhibiion 실험의 결과, $500{\mu}g/ml$의 농도에서 대조군인 EGCG와 효능이 비슷한 80% 이상의 효과를 나타내었다. Procollagen synthesis 결과 UVB 자극군에 의해 콜라겐 합성은 68.8%로 감소되었으며 시료의 $25{\mu}g/ml$의 농도에서 80.2%의 콜라겐 합성에 회복능을 보였다. MMP-1 inhibition 실험결과는 $25{\mu}g/ml$ 농도에서 20.2%의 저해능을 나타내었다. 실험 결과는 NRN의 미백 및 주름 억제 효과를 검증하고, 향후 안전한 천연 화장품 재료로 사용될 수 있음을 확인했다.

십전대보탕(十全大補湯)이 암전이(癌轉移) 억제(抑制)에 미치는 영향(影響) (Anti-metastatic Effects of Shiquandabutang on Human Fibrosarcoma cells, HT1080)

  • To examine the effect of Shiquandabutang on the metastasis of cancer, the following experiments were made. Before the main experiments, the cytotoxicity was measured by putting Shiquandabutang sample in HT1080. Then zymography was made to examine the change of gelatinolytic activity. And western blotting was carried out to examine the changes of Fos, Jun, Ets, the transcription factors of MMP-2, MMP-9, and Erk, JNK on signal transduction pathway to AP-1. Third, in vitro invasion assay with transwells coated by collagen and matrigel was carried out. From the results of the above the following conclusions were obtained. 1. The experimental result about cytotoxicity of Shiquandabutang against HT1080 was as below. The stained cell count after being treated by Shiquandabutang sample $400{\mu}g/ml$ for 24 hours was 0.9% of total cells, and the stained cell count by Shiquandabutang sample $100{\mu}g/ml$ was 1.5% of total cells. Both were near the level of control group which showed 0.6% stained. 2. The result of collagenase assay was as below. In Shiquandabutang sample $400{\mu}g/ml$, MMP-2 was reduced as compared with TPA control group, and the band of MMP-9 induced by TPA disappeared. In Shiquandabutang sample $800{\mu}g/ml$, both bands of MMP-2 and MMP-9 disappeared. 3. The results of western blots for Jun, Fos, Ets, Erk, JNK were as below. In Shiquandabutang sample $200{\mu}g/ml$, Ets was reduced, and Fos were increased. 4. The result of invasion assay was as below. The number of cells which migrated across transwell membrane in Shiquandabutang-treated group was less than that of +TPA control group. From the above results, it was concluded that Shiquandabutang might control the appearing and acting of collagenase not by the MMP-2, -9 promoter but by other way.

한약재 복합 추출물이 항산화 및 항노화에 미치는 영향 (Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

현지초(Geranium thunbergii) 추출물의 생리활성 및 Raw 264.7 cells에서의 항염활성 검증 (Verification of the Physiological Activity of Geranium thunbergii Extract and Anti-inflammatory Activity in Raw 264.7 Cells)

  • 본 연구에서는 현재까지 화장품 소재로써 다양한 연구가 진행되지 않은 현지초 추출물의 효능평가 및 항염 관련 활성 연구를 진행하였다. 현지초 추출물의 항산화능을 확인하기 위해 전자공여능 및 ABTS+ 라디칼 소거능을 측정한 결과, 각각 50 ㎍/ml의 농도에서 91%, 94%를 나타내어 항산화능이 우수함을 확인할 수 있었다. 미백활성 측정을 위해 tyrosinase 저해활성 측정을 실시하였으며 최고 농도인 1,000 ㎍/ml에서 24.8%의 저해능이 나타났다. 현지초 추출물의 주름개선 활성을 알아보기 위해 elastase 및 collagenase 저해활성 측정을 실시하였으며 그 결과, 각각 최고 농도인 1,000 ㎍/ml에서 30.6%, 90%의 저해능이 나타났고 collagenase 저해활성에서 우수한 저해능을 확인할 수 있었다. 세포 실험 진행을 위해 현지초 추출물 처리에 따른 대식세포 Raw 264.7의 생존율을 MTT assay에 의거하여 진행하였으며 100 ㎍/ml의 농도에서 83.6% 이상의 세포 생존율을 나타내어 이하의 세포 관련 실험 진행은 100 ㎍/ml 이하의 농도의 현지초 추출물을 가하여 실험을 실시하였다. NO assay에 의하여 현지초 추출물 처리에 따른 NO 생성 저해 활성을 측정한 결과, 100 ㎍/ml의 농도에서 74.9%의 저해율을 확인하였다. 단백질 발현 억제능을 알아보기 위해 western blot을 진행하였으며 현지초 추출물은 COX-2 및 iNOS 두 인자 모두 농도의존적으로 단백질 발현량이 저해됨을 확인할 수 있었다. 이러한 결과들에 의해 현지초 추출물은 항염 관련 기능성 화장품 소재로써 활용 가능성이 있다고 사료된다.

그린볼 사과(Green ball apple; Malus pumila Mill.) 적과의 항산화 및 elastase, collagenase, hyaluronidase 저해 효과 (Inhibition effect against elastase, collagenase, hyaluronidase and anti-oxidant activity of thinning Green ball apple)

  • 그린볼 사과 적과의 phenolic compound를 50-200 ㎍/mL 농도로 처리하여 DPPH radical 소거능을 측정한 결과 200 ㎍/mL 농도의 물과 ethanol 추출물은 94.69과 92.24%의 DPPH radical 소거능을 나타내었다. ABTS radical 소거능은 200 ㎍/mL 농도의 물과 ethanol 추출물에서 각각 100.30과 99.16%의 ABTS radical 소거능을 나타내었다. PF는 200 ㎍/mL 농도의 물과 ethanol 추출물에서 각각 1.76, 1.76 PF를 나타내었다. TBARs는 phenolic compound 농도가 200 ㎍/mL에서 물과 ethanol 추출물 각각 101.46, 99.64%의 항산화능을 나타내었다. 염증에 관련이 있는 HAase의 저해 효과를 측정한 결과 150 ㎍/mL 농도의 물과 ethanol 추출물은 각각 33.28, 32.14%의 저해효과를 나타내었다. 미용식품활성을 나타내는 elastase 저해활성을 측정해 본 결과 phenolic compound 농도가 200 ㎍/mL에서 물과 ethanol 추출물 각각 47.33, 40.92%의 저해 효과를 나타내었다. 피부의 주름과 탄력에 관련이 있는 collagenase 저해 효과를 측정한 결과 200 ㎍/mL 농도의 물과 ethanol 추출물은 각각 46.19, 65.58%를 나타내었다. 따라서 그린볼 사과 적과는 항산화, 항염증, 주름 개선 등 다양한 생리활성을 나타내는 기능성 소재로 개발이 가능할 것으로 판단되었다.

흰쥐의 외과적 창상에 대한 야관문 에탄올 추출물의 피부재생 효과 (Regeneration Effects of Lespedeza cuneata Ethanol Extract on Experimental Open Wound in Rat)

  • 야관문 추출물의 피부재생능력을 알아보고, 창상을 유발한 실험동물 모델에서 창상 치유 효과를 검증하고자 6개 군(NO, CO, PC, LCL, LCM, LCH)으로 나누어 5주간 실험하였다. Elastase 저해 활성을 평가한 결과 LCM군에서 항산화제인 BHA보다 2.7%($52.39{\pm}4.52$ vs $53.88{\pm}1.85$) 낮은 수치를 나타내었으며, collagenase 저해 활성을 평가한 결과 LCM군에서 BHA보다 7%($37.68{\pm}2.91$ vs $35.19{\pm}7.80$) 더 높은 활성을 나타내었다. 창상유발 21일에 LCL군, LCM군 및 LCH군의 왼쪽 창상 면적은 CO군과 비교하여 각각 54.2%, 53.5% 및 48.7% 현저한 창상면적 감소를 보였다. 이로서 야관문 추출물은 피부상피조직의 재생과 교원질의 합성을 도와 외과적 창상에 치료 효과가 있는 것으로 판단된다.

어피의 효소적 가수분해물을 이용한 천연조미료의 개발 (The Development of a Natural Seasoning Using the Enzymatic Hydrolysate of Fish Skin)

  • 수산물 가공시 부산물로 얻어지는 어피(魚皮)를 보다 효율적으로 이용하기 위하여 효소로 대구피를 가수분해할 때 가수분해조건을 구명하였으며 그 가수분해물을 이용한 천연조미료(모조간장 및 복합조미료)의 개발을 시도하였다. pH-drop법으로 대구피분해에 있어서의 collagenase, trypsin 및 pronase의 활성을 비교해 본 결과 pronase가 가장 활성이 높았으며 이 pronase의 $K_m$$V_{max}$값은 각각 1.82 mgN/ml, 0.06 mgN/mL/min였다. Pronase에 의한 대구피 가수분해조건은 반응온도 $50^{\circ}C$, 반응시간 3시간, pH6, 효소농도 0.03%였으며, 이 조건하에서의 가수분해도는 78.8%였다. 그러나 대구피를 collagenase로 1시간 분해시킨 후 pronase로 처리할 경우 가수분해도는 90.83%였다. 이때 가수분해물의 분자량은 8,000 dalton이었다. 아미노산 조성은 glycine(27.95%), glutamic acid(10.94%), proline(10.48%), aspartic acid(7.47%), serine(7.39%)이 전체 아미노산의 64.23%였으나 쓴맛을 내는 valine, methionine, isoleucine, leucine, phenylalanine, histidine 등은 13.05%에 불과하였다. 가수분해물과 기타 부원료로 제조한 모조간장원액과 시판 100% 양조간장을 8:2(v/v)로 혼합한 모조간장은 시판대두간장에 비해 최소유의차 검정 결과 맛, 색 및 종합평가면에서 5% 유의수준내에서 유의차가 없었다. 가수분해물 31.7% 함유한 복합조미료도 시판 복합조미료와 관능적으로 비교해 본 결과 복합조미료로서의 손색이 없는 제품이었다.
