Screening of Potato Cultivars for Infestation by Selatosomus puncticollis Mot.(Coleoptera: Elasteridae) and Analysis of Factors Assiciated with Resistance (청동방아벌레(Selatosomus puncticollis Mot.) 유충에 의한 감자 품종별 피해도 검정과 저항성 관련요인 분석)

  • In a previous report, 50 potato cultivars were screened for infestation levels by the coppery click beetle (Selatosomus puncticollis Motschulsky) in the field. Subsequently, we selected 10 potato cultivars (Anco, Atlantic, Bintje, Dejima, Denali, Jopung, Irish Cobbler, Namsuh, Shepody, and Superior) to evaluate feeding preferences by wireworms, and to analyze some factors associated with resistance. The injury rates and number of holes in potato tubers damaged by larvae of S. puncticollis were checked in the field and laboratory. Additionally, some of their chemical characters (contents of glycoalkaloids, total-nitrogen, Ca, K, Mg, sugars, and starch) were quantified. And finally correlation analysis was conducted to see whether there is a possible relationship between these characteristics and the damage level. The tuber injury rates by S. puncticollis larvae were generally high showing 19% to 73% of damage level. The highest number of tuber hole damaged by S. puncticollis larvae was found on cv. Namsuh, but generally fewer on cvs. Anco, Atlantic, Bintje, Denali and Superior. No activities for $\alpha$-chaconine, $\alpha$-solanine and $\alpha$-tomatine at a concentration of 2,500 ppm were found to S. puncticollis larvae. The contents of glycoalkaloids in tuber were different depending on cultivars. In tubers, cv. Superior contained the highest level of 18.8 mg%, but cv. Irish Cobbler had the lowest level of 6.39 mg%. Concentrations of reducing sugars and total free sugars in tubers of cv. Namsuh were 0.71 % and 2.95%, but 0.26% and 1.77% in those of cv. Dejima, respectively. For the content of potato starch, cvs. Bintje, Dejima and Irish Cobbler showed higher level, but cvs. Jopung and Shepody lower. The highest contents of total nitrogen, Ca, K and Mg in tuber were found on cvs. Anco, Atlantic, Anco and Jopung, respectively. From correlation analyses, injury rate by S. puncticollis larvae was correlated with total nitrogen content (r = -0.71435*) and total sugar content in tuber (r = 0.78018*). Such information will become essential in developing integrated pest management programs and also in breeding new potato cultivars resistant to the wireworms.

The Comparative Studies on the Terrestrial Insect Diversity in Protected Horticulture Complex and Paddy Wetland (시설원예단지와 논습지의 육상곤충 다양성 비교분석)

  • Agricultural ecosystem is recognized as a space for providing a variety of services, in addition to the food production that it originally encompassed, such as water purification, biological habitat, air purification, soil conservation, and landscape development. The construction of greenhouses in agricultural landscapes can cause deterioration of ecosystem services because of the increase of impermeable area and loss of biological habitats. This study aimed to compare insect diversity between different types of constructed greenhouses and paddy ecosystems. The target study area was selected by considering the distribution status of horticultural complexes and was classified as Single Vinyl Greenhouse, Multi Vinyl Greenhouse or Glass Greenhouse and they were compared with four paddies. The study locations were in Gu-Mi, Bu-Yeo, Ginje and Jin-Ju. A total of 2,333 individual insects belonging to 9 orders, 38 families, 76 genus, and 80 species were collected. The composition of orders was Hemiptera (22.37%), Coleoptera (18.42%), Hymenoptera (14.47%), Orthoptera (11.84%), and Diptera (10.53%). The average number of collected species were in the order Paddy (39.38 species) > Single Vinyl Greenhouse (35.50 species) > Multi Vinyl Greenhouse (22.50 species) > Glass Greenhouse (24.00 species). The Diversity Index (H') was Paddy (4.76) > Single Vinyl Greenhouse (4.57) > Multi Vinyl Greenhouse (4.12), and Glass Greenhouse (4.12). The Richness Index (RI) was Paddy (7.72) and Single Vinyl Greenhouse (7.03) > Multi Vinyl Greenhouse (4.99) and Glass Greenhouse (5.32). From our results, it can be seen that the biological diversity features of insects decreased when greenhouses are constructed.However, Single Vinyl Greenhouse is noted to promote insect diversity more than that by Multi Vinyl Greenhouse and Glass Greenhouse. Hence, when constructing greenhouses, it is necessary to consider insect habitat to conserve insect diversity.

A Note on the Deveolopment of Tribolium castaneum (Coleoptera: Tenebrionidae) on Brown Rice Oryzae sativa L. (현미에서의 거짓쌀도둑, Tribilium castaneum(딱정발레목:거저리과)의 발육)

  • The developmental period(mean $\pm$ SE) of the red flour beetle, Tribolium castaneum was studied on brown rice at four constant temperature(25, 28, 34, $36^{\circ}C$ $\pm$ $0.5^{\circ}C$, RH 70%)conditions. Based on the results, the lower and the upper developmental threshold temperature for the beetle were also estimated. The duration of the egg stage was estimated to be 7.6 $\pm$ 0.25, 4.8 $\pm$ 0.10, 3.0 $\pm$ 0.03 and 2.5 $\pm$ 0.09 days at 25, 28, 34, $36^{\circ}C$, respectively. That of the larval stage was estimated to be 53.3 $\pm$ 1.49, 33.4 $\pm$ 0.57, 30.6 $\pm$ 0.70, 31.0 $\pm$ 1.18; that of the pupal stage to be 12.1 $\pm$ 0.17, 7.8 $\pm$ 0.09, 5.7 $\pm$ 0.09, 5.0 $\pm$ 0.07 ; that of the overall period which spans from eggs to the adult emergence to be 72.3 $\pm$ 1.67, 46.0 $\pm$ 0.55, 39.4 $\pm$ 0.64, 38.7 $\pm$ 1.15 days at 25, 28, 34, $36^{\circ}C$, respectively. The duration of the egg and the pupal stage were similar to those of the results reared on other diets such as wheat and maize. Fed on brown rice, however, the duration of the larval stage was significantly retarded than on wheat. The mortality of the beetle during ,development was higher on brown rice than on wheat reported, indicating that brown rice is a poor diet compared with wheat. The lower developmental threshold temperature was estimated to be $20.0^{\circ}C$ for all stages of the beetle. The upper one was estimated to be $40.2^{\circ}C$ for the overall stage. The modal value of the moulting times during the larval stage was 7. However, as temperature increased,.the frequency of the beetle that moulted more than 7 times became higher. The sex ratio of the beetle was not affected by temperature.

Effect of Water temperature on the Climbing up of Larvae of Firefly, Luciola lateralis (Coleoptera: Lampyridae) (애반딧불이(Luciola lateralis) 유충의 상륙에 미치는 수온의 영향)

  • This study was conducted to investigate the relationship between the climbing up event of Luciola lateralis larvae and water temperature. In the laboratory condition, the larvae did not come out of water at constant temperature condition of 19.3${\pm}$$0.3^{\circ}C$ and tried to climb up at varying temperature condition of 20${\sim}$$22^{\circ}C$ (mean temperature 20.9${\pm}$$0.9^{\circ}C$) without success. However, they climbed up at constant temperature condition of 20.8${\pm}$$0.6^{\circ}C$. The frequency of the larval climbing up was highest as 63.6% at 20${\sim}$$21^{\circ}C$. The most larvae climbed up at approximately $21^{\circ}C$ of average daily water temperature, In the natural condition, the larvae climbed up at 19.8${\sim}$$21.7^{\circ}C$ and the frequency of the larval climbing up was high as 80.4% at 20${\sim}$$21^{\circ}C$. The larval climbing up was also little observed at 19${\sim}$$20^{\circ}C$ at which no larval climbing up was found in the laboratory experiment. No larvae attempted to climb up when water temperature was below $19^{\circ}C$ and above $22^{\circ}C$. It took 18 days from climbing up of larvae to appearance of adults. In its habitat, the range of water temperature required for larvae to climb up is assumed to be 19.6${\sim}$$21.8^{\circ}C$ and the most suitable water temperature may be 20.4${\pm}$$2.3^{\circ}C$. Therefore, the range of water temperature required for larvae to climb up is assumed to be 19.8${\sim}$$21.7^{\circ}C$, and the suitable water temperature range for larvae may be 20${\sim}$$21^{\circ}C$, and the most suitable temperature is thought to be $21^{\circ}C$.

Bionomics and a Lana Sampling Method of Coppery Wireworm, Selatosomus puncticollis (Motschulsky), (Coleoptera: Elateridae) in Potato Field (청동방아벌레(Selatosomus puncticollis Motschulsky)의 생태적 특성 및 감자포장내 유충밀도 조사법)

  • The occurrence pattern and bionomics of Selatosomus puncticollis (Motschulsky) were surveyed in three net house (6$\times$70m) in the field and in a laboratory ($20^{\circ}C$, RH$75\%$, L16/DB). Emergence of hibernated adults of S. puncricollis started from early May, reached peak at early June and diminished in late June. Occurrence rate to total number was $42.1\%$ in early June and $31.7\%$ in late May, when most of hibernated adults emerged at these periods. Adults started to oviposit from mid June, and eggs hatched from early July Larvae turned into pupae from mid July and emerged to adults from mid August. Egg-period was 23 days and pupal period was 21 days. It is estimated that preovipositional period and larval period were approximately 10 months and 30 months, respectively. Wireworms were distributed at the soil depth of 10-5 cm, $56.8\%$ in 1997 and $45.8\%$ in 1998. To establish bait techniques to attract wireworms in the soil, six baits: pieces of potato, carrot, and sweet potato, wheat grains, corns, and flour dough wrapped with gauze, were buried at 15 cm of soil depth, and collected after 5 and 10 days. The numbers of wireworms attracted by potato pieces, wheat grains and sweet potato pieces after 10 days were 1.8, 1.6, and 1.4/bait, respectively. Therefore, burying potato pieces at 15 cm of soil depth and collected after 10 days could be recommended as a wireworm baiting technique.

Selection of Indicator Insects for the Evaluation of Agricultural Environment (농업환경 평가를 위한 지표곤충 선발)

  • To select indicator insects, the abundance of which is one of the major criteria for the evaluation of agricultural environment, was designated and monitored in the three sites (Hongcheon A, B, and Yangpyeong) from 2001 to 2003. Characteristics of rice cultivation are usual cultivation with 2 or 3 times sprayed chemical agents per year in Hongcheon A site, duck cultivation with non-sprayed chemical agent, green manure and mowing in Hongcheon B site, and pond snail with non-sprayed chemical agent, green manure and mowing in Yangpyeong site. Dominant floral species are Digitaria ciliaris (Retz.) Koel., Persicaria thunbergii (Siebold & Zuccarini) H. Gross, Echinochioa crus-galli (L.) Beauvois, and Humulus scandens (Lour.) Merr. in Hongcheon A site, Digitaria ciliaris (Retz.) Koel., Persicaria thunbergii (Siebold & Zuccarini) H. Gross, and Echinochloa crus-galli (L.) Beauvois in Hongcheon B site, and Digitaria ciliaris (Retz.) Koel., Setaria viridis (L.) Beauv., and Persicaria langiseta (De Btuyn) Kitagawa in Yangpyeong site. Degree of green naturality (DGN) was higher in Hongcheon B site (2.0) and Yangpyeong site (3.6) than in Hongcheon A site (1.6). Number of species and individual of insects collected in the three sites were abundant on August and September. The species diversity was higher in Hongcheon B and Yangpyeong than in Hongcheon A. And Coleoptera was dominant in the all sites. The dominance index (DI) was the lowest, but diversify index (H'), evenness index (E) and richness index (RI) were the highest in Hongcheon B site. Based on three results, indicator insects for the evaluation of agricultural environment were selected as Dolichus halensis in Hongeheon A site, as Pheropsophus. javanus and Polionemobius mikado in Hongcheon B and Yangpyeong sites.

Genetic Divergence and Phylogenetic Relationships among the Korean Fireflies, Hotaria papariensis, Luciola lateratis, and Pyrocoelia rufa(Coleoptera: Lampyridae), using Mitochondrial DNA Sequences (미토콘드리아 DNA의 염기서열을 이용한 파파리반딧불이, 애반딧불이 및 늦반딧불이 (딱정벌레목: 반딧불이과)의 유전적 분화 및 계통적 관련)

  • Genetic divergence and phylogenetic relationships among the major Korean fireflies (Hotaria papariensis, Luciola lateralis, and Pyrocoelia rufa) were studied. A portion of mitochondrial COI (403 bp) and 165 rRNA (490~504 bp) genes were sequenced, and the GenBank-registered, homologous 165 rRNA sequences of Japanese fireflies were compared (27 species of Lampyridae, one of Lycidae, and one of Rhgophthalmidae). Greatest DNA and/or amino acid sequence divergence was found when P rufa, belonging to Lampyrinae was compared with H. papariensis and L. lateralis, both belong-ing to Luciolinae, confirming the current taxonomic status of the species. In the PAUP and PHYLIP analyses with 165 rRNA data, grouping of the two geographic samples of H. papariensis with H. tsushimana validate the use of generic name, Hotaria. Nevertheless, lack of sister-group relationship of the two geographic samples of H. papariensis renders further investigation on this group . Although the Korean and Japanese L. lateralis formed a strong monophyletic group, a substantial genetic differentiation was detected between them (2.9% of 165 rRNA gene sequence divergence). Finally, the geographic samples of Korean p. rufa strongly formed a group with Japanese p. rufa, warranting the use of generic name, Pyrocoelia, but the genetic distance observed between the Cheju-Island individual and all others requires further investigation on this subject. Summarized, this study supports the current taxonomic status of the Korean fireflies in that each respectively formed a strong monophyletic group with its own species or genus.

Escape of Pine Wood Nematode, Bursaphelenchus xylophilus, through Feeding and Oviposition Behavior of Monochamus alternatus and M. saltuarius (Coleoptera: Cerambycidae) Adults (솔수염하늘소와 북방수염하늘소의 섭식과 산란행동을 통한 소나무재선충의 이탈)

  • This study was performed to investigate the escape of pine wood nematode (PWN), Bursaphelenchus xylophilus, from two vector species (Monochamus alternatus and M. saltuarius) through oviposition and feeding behavior. First, we checked number of PWNs escaped from M. alternatus emerged from three different cases of pine logs. In case A, healthy pine trees were cut into logs and left in pine forest infected with PWN. In case B, healthy pine trees were cut into logs, left in large screen cage, and let them oviposited by M. alternatus emerged from pine trees infested with PWN. In case C, pine trees which were harboring M. alternatus were cut into logs, and PWN was inoculated artificially. The M. alternatus adults emerged from the above three cases of pine logs were checked in the next year to know how many PWN they were harboring in their bodies. The percentages of M. alternatus harboring PWN (18.3 and 15.6%, respectively) and number of nematodes per vector ($5,713.1{\pm}9,248.3$ and $2,034.1{\pm}4,746.8$ PWNs, respectively) in case A and B logs are similar to each other. However, the percentage and the number in case C (38.3% and $20,083.1{\pm}32,188.3$ PWNs) were higher than those of case A and B. Among 52 M. alternatus adults harboring PWN from all the three cases, 20 adults (38.5%) were harboring more than 5,000 PWNs per beetle. And these 20 adults were harboring 97.9% of the total PWNs in 52 adults. Second, we checked the daily escape of PWNs from M. alternatus and M. saltuarius collected at pine forest infested with PWN. The PWN escaped from their vector body for $34.9{\pm}12.4$ days for M. alternatus, and for $23.9{\pm}16.2$ days for M. saltuarius, reaching at peak escape during the 2nd week of emergence of the two vector species. A 44.5 and 47.2% to the total PWNs escaped from vector body within 2 weeks of vector emergence for M. alternatus and M. saltuarius, respectively. The number of PWNs escaped from each vector was $3,570.6{\pm}5,189.2$ and $1,556.2{\pm}1,710.3$ for M. alternatus and M. saltuarius, respectively.

Bionomical Characteristics of Luciola lateralis (Coleoptera: Lampyridae) in Mass Breeding (애반딧불이(딱정벌레목: 반딧불이과)의 생육 특성)

  • This study was performed to investigate the bionomical characteristics of Luciola lateralis in Korea. Imago's longevity of female was $17.5{\pm}0.7$ and male was $20.6{\pm}1.0$ days at $23^{\circ}C$, R.H. 80%, and significant difference was not occurred in breeding density. Pre-oviposition period was a pair $2.8{\pm}1.5$, two pairs $2.5{\pm}1.5$, three pairs $1.7{\pm}0.7$, four pairs $1.3{\pm}0.5$ days and pre-oviposition period was shorten in higher breeding density. The mean of laying eggs was $248.9{\pm}80.7$, was not related breeding density. Daily oviposition rate was 1st day 20.8%, 2nd 17.3%, 3rd 14.3%, 4th 12.7%, 5th 7.0%, the rate was highest in 1st day and gradually reduced. Egg period was $21.8{\pm}0.7$ days and hatchability was $95.5{\pm}4.8%$. Larval instar was identified the 9th instar, the evidence was able to its skin and marking pattern. Larval period of each instar was 1st $1.0{\pm}0.1$, 2nd $3.3{\pm}0.2$, 3rd $10.2{\pm}1.0$, 4th $6.8{\pm}0.5$, 5th $6.9{\pm}2.0$, 6th $15.4{\pm}4.1$, 7th $18.3{\pm}5.9$, 8th $25.8{\pm}8.7$, 9th $31.2{\pm}13.2$ days. Matured 9th larva was made of cocoon during $2.8{\pm}0.8$ days and pre-pupal, pupal period was $4.6{\pm}0.9$, $5.6{\pm}0.7$ days. Imago was made hard hind wing during $3.8{\pm}0.4$ days in cocoon and come out.

Comparative Analysis of Nutrients and Hazardous Substances in Zophobas atratus Larvae (탈지 전, 후 아메리카왕거저리(Zophobas atratus) 유충의 영양성분 및 유해물질 비교분석)

  • The nutritional composition and optimal eating stage of the super mealworm, Zophobas atratus (Coleoptera : Tenebrionidae), were investigated to explore its use as a food ingredient. It was determined that 10th instar larvae were most suitable for eating in terms of nutritional value as well as economic aspects. To improve the quality of powder production, the nutritional value of 10th instar larvae before and after degreasing was analyzed. After drying the larvae powder, crude protein was the most abundant nutrient both before (52.3%) and after (60.6%) degreasing while crude fat measured 36.3% and 21.7% before and after degreasing, respectively. In terms of essential amino acids, leucine levels were highest and 1.3 times greater after degreasing (4.5%) than before (3.5%). Oleic acid, the highest unsaturated fatty acid in larvae, was 31.7% after degreasing which was 1.1 times higher than before (33.2%). Among various major minerals, potassium was most abundant and 1.4 times higher after degreasing (1267.0 mg/100 g) than before (879.3 mg/100 g). Harmful substances were 1.3 to 2.0 times lower in the degreased larvae, although mercury or pathogenic bacteria were not detected in either group. We therefore conclude that degreased Z. atratus larvae are more suitable for eating than before degreasing.
