Identification of csp Homolog in Bradyrhizobium japonicum

  • Low-temperature adaptation and protection for environmental stresses were studied in the gram-negative soil bacterium Bradyrhizobium japonicum 61A101c. B. japonicum was more resistant to alcohol, $H_2O_2$, heat and freezing following a pretreatment at $4^{\circ}C$, resulting in approximately 10 to 1,000 folds increased survival compared to mid-exponential-phase cells grown at an optimal temperature at $28^{\circ}C$. This phenomena relate to the cold shock protein expressed when cells are exposed to a downshift in temperature. To confirm the presence of cold shock protein genes in B. japonicum, a PCR strategy was employed using a degenerate primer set, which successfully amplified a putative csp gene fragment. Sequence analysis of the PCR product(200bp) revealed csp-like sequences that were up to 96% identical to csp gene of S. typhimurium.

  • The Korean Next Generation Reactor(KNGR) is a Pressurized Water Reactor adopting direct vessel injection(DVI) to optimize the performance of emergency core cooling system(ECCS). In a certain accident, however, pressurized thermal shock(PTS) of the vessel due to the sudden contact with the injected cold water is expected. In this paper, an accident of Main Steam Line Break(MSLB) has been numerically investigated with direct vessel injections and an increased volume flow rate in some cold legs. Using FLUENT code, temperature distributions of the fluid in the downcomer and of reactor vessel including the core region have been calculated, together with the distribution of convective heat transfer coefficient(CHTC) at the cladding surface of the reactor vessel. The result shows that some parts of the core region of the reactor vessel have higher temperature gradient expressing higher thermal stress.

  • There are many difficult problems in analyzing the gas flow in puffer type circuit breaker such as complex geometry, moving boundary, shock wave and so on. To predict the interruption performance accurately, these should be considered in the simulation. In this paper, the analysis procedure of the cold gas flow in the circuit breaker is presented. Euler equation is solved by FVFLIC method which is an explicit time difference scheme for an unsteady flow computation. Moving boundaries are treated with a cell elimination-addition technique. The pressure and density in front of piston are calculated from the rate of the cell volume change. The presented method is applied to the real circuit breaker model and the pressure in front of the piston is good agreement with the experimental one.

  • 충돌제트는 미사일 또는 수직/단거리 이착륙기의 배기가스가 지면이나 화염 편향기 등에 부딪힐 때 발생하며 노즐의 형상, 노즐과 평판과의 거리등에 따라 유동양상이 달라진다. 본 연구에서는 초음속 노즐을 통해 과소 팽창된 제트가 평판에 부딪힐 때 형성되는 평판 압력분포 및 버블 발생 등에 대한 시험결과를 선행된 충격파 풍동을 이용한 시험 결과와 비교하고 초음속 유동시험장치 이용시의 유동 특성을 규명하였다. 초음속 유동시험장치와 충격파 풍동의 평판에서 압력분포는 저압 챔버 내의 수분과 온도 때문에 차이가 나타났고 과소 팽창비에 따른 압력분포는 서로 유사한 경향을 보여주었다.

  • There are various characteristics called for in work roll and intermediate rolls for cold rolling mills. Among these characteristics, the two main requirements are to ensure the quality of the rolled products and to reduce roll cost. To achieve these needs, resistance to wear, to thermal shock and to contact fatigue are especially important. This paper describes that new material(named DSR1) for intermediate rolls which greatly increases rolling campaign and improves resistance to wear has been developed. DSR1 was successfully manufactured and has been used in the cold rolling mill. It showed that Trial product was homogenous in hardness distribution and sufficient usable diameter. Also in service test, trial product is much more excellent rolling performance than conventional $5\%Cr$.

  • Earth's bow shock is a transition layer across which properties of plasmas change irreversibly. Although some features of the bow shock are well described by continuities of fluxes of various macroscopic quantities, particle dynamics across the transition layer is very complicated. Observed phase space distributions show multiple ion beams and partially thermalized ions around the transition layer. In some cases, both hot magnetosheath ions and cold solar wind ions simultaneously exist in the magnetosheath. Electrons around the transition layer usually have flat-top distributions with temperature anisotropy. From the observed properties of the phase space distributions we will discuss thermalization processes that occur across the shock transition.

Expression Analysis of the csp-like Genes from Corynebacterium glutamicum Encoding Homologs of the Escherichia coli Major Cold-Shock Protein CspA

  • Three csp-like genes were identified in the Corynebacterium glutamicum genome and designated cspA, cspB, and cspA2. The genes cspA and cspA2 encode proteins, comprising of 67 amino acid residues, respectively. They share 83% identity with each other. Identity of those proteins with Escherichia coli Csp proteins was near 50%. The cspB gene encodes a protein composed of 127 amino acids, which has 40% and 35% sequence identity with CspA and CspA2, respectively, especially at its N-terminal region. Analysis of the gene expression profiles was done using transcriptional cat fusion, which identified not only active expression of the three genes at the physiological growth temperature of $30^{\circ}C$ but also growth phase-dependent expression with the highest activity at late log phase. The promoters of cspA and cspA2 were more active than that of cspB. The expression of the two genes increased by 30% after a temperature downshift to $15^{\circ}C$, and such stimulation was more evident in the late growth phase. In addition, the cspA gene appeared to show DNA-binding activity in vivo, and the activity increased at lower temperatures. Interestingly, the presence of cspA in multicopy hindered the growth of the host C. glutamicum cells at $20^{\circ}C$, but not at $30^{\circ}C$. Altogether, these data suggest that cspA, cspB, and cspA2 perform functions related to cold shock as well as normal cellular physiology. Moreover, CspA and its ortholog CspA2 may perform additional functions as a transcriptional regulator.

  • 제트 편향기 설계를 위한 기초 자료로 활용하기 위해 꼭지각 $60^{\cire}$인 쐐기에 충돌하는 초음속 제트에 대해 연구하였다. 초음속 제트가 쐐기에 충돌할 때 발생되는 주요 특징은 충격파 간섭과 유동의 박리현상이다. 초음속 유동장치를 이용하여 유동가시화, 표면압력분포, 등압력분포를 측정하고 실험결과와의 비교 및 검증을 위해 유동해석을 하였다 주요변수로는 과소팽창비, 노즐출구에서 꼭지점까지 거리, 설계마하수를 고려하였다. 쐐기 충격파가 이격 또는 부착되는 조건을 확인하였으며 유동장에 지배적 영향을 끼치는 인자는 자유제트의 강도, 쐐기 충격파와 배럴 충격파의 간섭에 의해 형성되는 충격파 패턴임을 알 수 있었다.
