  • Transgenic coho salmon, Oncorhynchus kisutch containing a growth hormone gene construct have been examined for their hormone levels and ability to growth for 90 days in winter season. Food intake of the transgenic coho was approximately 4-fold higher than that of nontransgenic coho salmon of similar size, but feed efficiency of the transgenic coho was 1.1-fold lower than that of size-matched control. Specific growth rates of body weight of the transgenic coho were approximately 1.4-fold (length) or 3-fold(weight) higher than that of nontransgenic coho salmon. GH, total-T$_4$ and total-T$_3$ levels were Increased approximately 2-fold compared to size control salmon. The transgenic animals also displayed head, jaw and opercular abnormalities typical of the effects of this gene construct in coho salmon, indicating that some imbalance in growth processes were induced.

  • For identification of morphometric characteristics in rainbow trout, Oncorhynchus mykiss, coho salmon, O. kisutch, and their allotriploid, twenty one body traits were measured, and the ratios of each trait to fork length, length from postorbital to the origin of caudal fin and head length were calculated. The ratios of fin length to fin height in anal, dorsal and adipose fins showed significant differences among these two species and their allotriploid $(p\leq0.01)$. Allotriploid could be distinguished easily from two parents species in the ratios of length of origin of ventral fin to fork length and anal fin length to anal fin height.

  • This study was conducted to evaluate plant proteins as a replacement for a fishmeal diet in the rearing of coho salmon Oncorhynchus kisutch. Twelve groups of 20 fish averaging 34.0±0.62 g were randomly distributed into 12 rectangular tanks (250 L). Four experimental diets included a control diet containing 60% fishmeal (Control), and three other diets that replaced 20% of fishmeal with soy protein concentrate (SPC), fermented soybean protein concentrate (F-SPC), and enzyme-processed soy protein concentrate (E-SPC). At the end of the feeding trial, fish that were fed Control, SPC and E-SPC diets showed significantly higher weight gain, specific growth rate, feed efficiency, and protein efficiency ratio than those that were fed F-SPC diet. However, there were no significant differences among the fish that were fed Control, SPC, and E-SPC diets. No significant differences were observed in crude protein, crude lipid, and ash of whole body among the fish that were fed all the diets. Therefore, these results indicated that 20% of fishmeal could be replaced by E-SPC or SPC without any adverse effects on the growth performance of coho salmon.

  • Hybird and allotriploid between female rainbow trout (Oncorhynchus mykiss) and male coho salmon (O. kisutch) were produced by artificial fertilization and heat shocks. Hatching and survival rate of allotriploid at 2 month after hatching was $77.6\%$ and $54.5\%$ respectively, and these rates clearly exceeded those of their hybrid. Cell and nuclear sizes of the erythrocyte of hybrid were intermidiate of their parents and those of allotriploid were larger than thier hybird. The somatic chromosome number of viable hybrid was 2n = 60 and that of allotriploid was $90\~93$ with chromosomal polymorphism. Allotriploid karyotpe was constituted by two sets of rainbow trout chromosome and one set of coho salmon chromosome.

  • Mitotic analyses of the coho salmon (Oncorhynchus kitsuch) revealed a mode of 2n=60 whit 46 meta or submetacentric and 14 acrocentric chromosomes (NF=106). Individuals with 2n=61 were also found in both sexes in the same population; the karyotype is composed of 45 meta or submetacentric and 16 acrocentric chromosomes (NF=106). This result may indicate that Robertsonian chromosomal polymorphism occurred in the population of the coho salmon.

  • A feeding study was conducted for 85 days to determine if soybean meal can be substituted for fish meal in a diet of coho salmon (Oncorhynchus kisutch) fingerlings. Solvent extracted soybean meal was used in the experimental diet at $0{\%},\;10{\%},\;30{\%},\;50{\%}$ and $70{\%}$ levels to make up about $40{\%}$ protein in the final diets. The results of feeding trial indicated that soybean meal can be used up to $30{\%}$ (replacing $34.3{\%}$ fish meal) as a major protein source with no adverse effects on the growth of coho salmon(P>0.05) . However, both growth rate and survival were decreased in the groups of fish fed over $50{\%}$ soybean meal diets.

  • Disease including parasite, bacteria and virus cause serious mortality to salmonid fish in the aquaculture. In this study, we investigated the current disease status of the rainbow trout (Oncorhynchus mykiss) and coho salmon (Oncorhynchus kisutch) in Yanayang, Pyeongchang, Jeongseon and Yeongwol of Gangwon province in 2021 and performed molecular characterization of those pathogen. For parasites, Ichthyophthirius multifiliis was observed at 2 farms. For bacteria, we identified Aeromonas sobria from kidney of rainbow trout using phylogenetic analysis of gyrB gene. A. salmonicida were isolated from necrosis site of gill cover and fin in coho salmon and necrotic lesion of fin in rainbow trout. Phylogenetic analysis using vap gene indicated that A. salmonicida isolated in this study were clustered with previously reported A. salmonicida subsp. salmonicida isolates. For virus, JRt-Nagano type of infectious haematopoietic necrosis virus was detected in rainbow trout, but infectious pancreatic necrosis virus and Oncorhynchus masou virus were not detected. These results provide useful information for the prevention of disease spread and transmission when cultivating new species such as Atlantic salmon in Korea.

  • For the purpose of identification of inheritance in allotriploid between rainbow trout (Oncorhynchus mykiss) and coho salmon (O. kisutch), five isozymes, lactate dehydrogenase (LDH), malate dehydrogenase (MDH), isocitrate dehydrogenase (IDH), phosphoglucose isomerase (PGI) and phosphoglucomutase (PGM) from skeletal muscle in two species and their allotriploid were analyzed. All of these loci showed differences between two species and their allotriploid except PGI. Generally, coho salmon was more monomorphic in these isozyme loci than rainbow trout. Their allotriploids showed intermediate patterns between the parental species in those isozyme loci except PGI. As a result of this study, LDH, MDH, IDH and PSM may be used as useful genetic markers in these two species, and they also be of use in studying hybrid and allotriploid in salmonids.
