This study is intended to understand the styles, colors and color arrangements of costumes expressed on Kisan Kim Jun-geun's genre paintings of Joseon in the late 19th century. The paintings vary according to the status and gender of the person wearing the costume as well as the time period and different situations the person belongs to. Also, this study aims to examine the meaning and value of Kisan's paintings in terms of the history of Korean costumes. The result of this study are as follows: The basic and common attire for people of all status and class is the jeogori(jacket), baji(pants) for men and Banhoijang jeogori, chima(skirt) for women. Men wear Po(coat) that represents their status and age, and they put on various hats according to different weather conditions or work-skill even when in the same status and class. However, women wear the Baeja(vest), Durumagi(coat) and head-dresses when they are cold. Overall, the costume patterns and shapes that appear on Kisan's paintings show the same patterns and structures when compared to different data during the same time period. Thus, they provide useful information to help not only understand the changes of patterns and structures of costumes, but also the situations and emotions of the people of that time period. Moreover, it can be understood that various colors and color arrangements that reflect the situations and emotions of the late 19th century were used. This study that analyses the colors and color arrangements used in Kisan's genre paintings can provide the very basic and systematically arranged data to help understand the unique colors in Korea. These data can also provide important information to understand the use of dyes and pigments during that time. Therefore, Kisan's genre paintings are meaningful and valuable in terms of the history of Korean costumes for these are the data, from which we can review the costumes, colors and color arrangements in the years from 1890 to 1910.
In Korean beef market, one of the major problems is mislabeling or fraudulent distribution of Holstein dairy meat or imported beef as domestic Hanwoo meat. Therefore, there has been a great need for a development of technology to identify beef breeds in meat and meat products. This study was carried out to develop the accurate and reliable method for the identification of beef breed using PCR-RFLP marker of MC1R, MGF and TYRPl genes affecting coat colors in cattle. A single base substitution (G\longrightarrowT transition) at the codon for amino acid position 104 of MC1R gene was identified between Hanwoo and Holstein and Angus breeds. The change at this position creates Msp I restriction site in Holstein and Angus, but not in Hanwoo. When the DNA amplified products (537 bp) was digested with Msp I, Hanwoo meat showed a single band of 537bp, while two fragments of 329bp and 208 bp were observed in Holstein meat and Angus breed, respectively. Thus, breed-specific RFLP marker in the MC1R gene can be used to distinguish between Hanwoo meat and Holstein and Angus meats. In the RFLP genotype of MGF gene, the frequency of r/r type was 75% in Manwoo, whereas the frequency of R/R was 80% in Hereford breed. Holstein and Angus breeds showed 100% for R/r type. Therefore, Hanwoo meat showed significant difference in the MGF genotype frequencies compared with those of Holstein meat and imported beef cattle breeds. However, TYRP1 gene showed the same genotype in all breeds examined. Thus, this TYRP1 gene can not be used as a molecular marker for breed identification. As a consequence, we suggest that RFLP markers of the MC1R and MGF coat color genes could be used as DNA marker for identification of Hanwoo meat from Holstein and imported meats.

We previously observed that Bacillus subtilis spores from sspE mutants presented a lower germination capacity in media containing high salt concentrations (0.9M NaCl). This deficiency was attributed to the absence of SASP-E (gamma-type small-acid-soluble protein), rich in osmocompatible amino acids released by degradation. Herein we observed that, in addition, this mutant spore presented a reduced capacity to use L-alanine as germinant (L-ala pathway), required longer times to germinate in calcium dipicolinate ($Ca^{2+}$-DPA), but germinated well in asparagine, glucose, fructose, and potassium chloride (AGFK pathway). Moreover, mild sonic treatment of mutant spores partially recovered their germination capacity in L-ala. Spore qualities were also altered, since sporulating colonies from the sspE mutant showed a pale brownish color, a higher adherence to agar plates, and lower autofluorescence, properties related to their spore coat content. Furthermore, biochemical analysis showed a reduced partition in hexadecane and a higher content of $Ca^{2+}$-DPA when compared with its isogenic wild-type control. Coat protein preparations showed a different electrophoretic pattern, in particular when detected with antibodies against CotG and CotE. The complementation with a wild-type sspE gene in a plasmid allowed for recovering the wild-type coat phenotype. This is the first report of a direct involvement of SASP-E in the spore coat assembly during the differentiation program of sporulation.
Seed coat anthocyanin can be purified by soaking 3 times in methanol solution supplemented with one percent of HCl. Anthocyanin content was very wide range in collected lines and average anthocyanin content of black seed coat lines was 15.07 permillage, but that of white mottled on brown seed coat lines was 0.31 permillage. In black seed coat lines green seed embryo type has more anthocyanin in amount compare to yellow seed embryo. Anthocyanin accumulation was promoted in late maturing lines compare to early maturing lines. Positive correlations were observed among 100 seed weight, days to flowering, days to growing and anthocyanin content, but negative correlation between days from flowering to maturity and anthocyanin content. Collected black seed coat lines were divided into two maturity groups. Group VI which has longer than group V in days to maturity accumulated more anthocyanin compare to group V. When the seeding date was May 15, highest anthocyanin content was observed.

Indigo [鴉靑] was used as a symbolic color in 15th century diplomacy. This study reinforces the historical research for characteristic and shapes of indigo colored royal clothing gift in King Seongjong's reign with a focus on po [袍] and ieom [耳掩]. Clothing made of fur that was combined with satin damask, silk gossamer [綃] were frequent gift, and a sable coat was a symbol of high-ranking clothes. Another clothing's materials were satin damask, sheer fabric gauze [羅], cotton, tabby with silk and ramie [紵絲]. The indigo color of King Seongjong's reign was recommended not for luxury. Historical research factors for indigo clothing gift were extracted from noblemen's relics. Danryeong and jigryeong are shaped of narrow sleeves and trapezoid gusset pleated in and out. Sable coat can consist of satin damask outshell, sable lining, and double oblique collar. Heohyung are presumed types of short sleeved fur vests. Ieom can be reconstructed with an indigo fabric outshell and sufficient fur. Dapho has gusset pleated wide and narrow. As additional gifts, the shapes of yoseon-cheopri and cheopri were analyzed. We selected a commercial fabric similar to relics and an indigo powder dyeing method to remedy a lack of traditional material. The reconstructed data were presented as flat drawings and samples.
Here I describe a multiplex allele specific PCR-based approach for the rapid detection between Hanwoo and Holstein meat associated with Melanocortin 1 receptor (MC1R) gene. Specific and universal oligonucleotide primers were used in combination to detect the presence of a single nucleotide polymorphism within the bovine MC1R DNA sequence. The presence of the bovine MC1R gene is indicated by the production of a single control PCR product, whilst positive samples generate an alternative smaller specific product over the same region. The mutations in MC1R104 codon revealed depending on the presence or absence of an indicative fragment amplified from the wild-type allele of this codon. As little as 0.39 ng and 1.56 ng of genomic DNA of Hanwoo and Holstein could be detected by MAS-PCR assay, respectively. This technique, which is widely used in human genetic screening, provides a reliable and sensitive result that has not been documented for the identification of bovine coat color. The MAS-PCR assay approach was proven to be useful in complementing routine beef DNA analysis for differentiation of these MC1R variants and it would facilitate the screening of deceiving sales of Holstein meat in the butcher shop.
The melanocortin 1 receptor (MC1R) plays an important role in regulation of melanin pigment synthesis within mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat color variations within several mammalian species including cattle. To develope a rapid and accurate method for the identification of Hanwoo, we performed a modified PCR-RFLP analysis of MC1R gene using single nucleotide polymorphism (SNP) within MC1R as a target. A size of 538 bp (537 bp for Hanwoo) was amplified by PCR, digested with Hpa II, and electrophoresed on a 1.5% agarose gel. A PCR product from Hanwoo showed a single band of 537 bp, whereas two fragments of 328 bp and 210 bp were detected in both Holstein and Black angus. The current result suggests that the PCR-RFLP using our primers and enzyme digestion system would be very accurate, easy and reproducible method to discriminate between Hanwoo and Holstein/Black angus meat.

As a result, Hanwoo samples showed FAM-positive signal only, whereas other samples showed VIC-positive. This result suggests that the TaqMan$^{(R)}$ MGB probe based real-time PCR technique would be very accurate, easy and reproducible method to discriminate between Hanwoo meat and Holstein/Black angus meat.

Korean native cattles (Han woo) are divided yellow, tiger(blinder), and black by coat color. Chikso has tiger coat color, which cultured about 1,700 heads within Korea. However there were a few researches reported about preservation and production. This study was conducted to investigated that the effect of follicle stimulating hormone (FSH) dosage(240 mg and 280 mg) on the embryo production, pregnancy and calf birth weights. There were no difference total embryo and transferable embryo between FSH 240 mg or 280 mg treatment, but pregnancy rate was increased 46% in embryos derived from 280 mg group than 240 mg group's 29.6%. The birth weight of Chikso calves was similar between artificial insemination(25.6 kg) and embryo transfer(24.8 kg) groups.
