Coagulase-negative Staphylococcus sp #39, isolated from raw-milk showed reduced susceptibility to vancomycin. The minimun inhibitory concentration for strain #39 was at 8$\mu\textrm{g}$ of vancomycin per ml. Transmitting electron microscopy displayed that this strain had a 2.5-3.5 times thicker cell wall than a vancomucin sensitive strain of Staphylococcus sp. The strain #39 also had an increased cell volume. These data indicate that the reduced susceptibility may be due to the thickness of the cell wall of the test strain.
A total of 251 clinical isolates (human origin, 43 strains and bovine udder origin 208 strains) of the Staphylococcus that fermented mannitol aerobically were tested for their ability to produce coagulase, DNase, and thermostable nuclease. Of these, 158 isolates coagulated human or bovine plasma, produced DNase, and thermostable, nuclease and were identified as St. aureous, 146 of which produced a 1+ to 3+ clot. The remaining 12 isolated produced a -clot in citrate treated plasma but produced 1+ to 3+ clot in ethylenedi-aminetetraacetic acid (EDTA) treated plasma. It was found that 7 coagulase positive isolates failed to produced thermostable nuclease. In these organisms, we found out of the clot formation is not by coagulase activity but utilization of citrate, because EDTA treated plasma is not coagulated. Among 93 isolates which did not coagulate citrate-or EDTA treated plasma and thermostable nuclease negative, 28 strains produced DNase were identified as St. epidermidis, and other strains were not identification further. It was found that thermostable nuclese production appears to be a consistent property of St. aureus and the test is easy to perform, is rapid became quite distinct within 2 to 4 hour, and is not influenced by as many factors and variations as the coagulase test.
We evaluated the performance of a novel screening test, PBP2a MRSA rapid kit (Dinona Inc., Iksan, Korea), for methicillin-resistant Staphylococcus aureus (MRSA) based on a immunochromatographic assay. The test is able to detect penicillin-binding protein 2a (PBP2a) using the nasal specimens from health care workers. The nasal specimens were obtained from 69 healthcare workers and were incubated in enrichment broth followed eight hours incubatin in BHI with cefoxitin $4{\mu}g/mL$. These broth were tested by PBP2a Rapid Kit. The enrichment broths were also directly tested for tube coagulase using the conventional identification method. 19 of 22 MRSA showed positive results by PBP2a rapid test and direct coagulase test (the sensitivity for detection of MRSA, 86.36%). While, 8 of 47 non-MRSA showed false positive results for the two tests. All of the 8 non-MRSA which showed false positive were co-colonizing isolates with MRCNS and MSSA. In addition, 46 of 49 methicillin-resistant staphylococci (MRS) showed positive results for PBP2a MRSA rapid kit (the sensitivity for detection of MRS, 93.8%), and all of 20 non-MRS showed negative results (specificity, 100%). The combination of PBP2a MRSA rapid kit and direct coagulase test showed the good sensitivity for detection of MRSA from anterior nares but frequently showed false positive results from the co-colonizing carrier with MRCNS and MSSA.

A total of 129 clinical isolates of Staphylococcus species was characterized by the tests of coagulase production, haemagglutination, mannitol fermentation, DNase production and hemolysis. Ninety-nine out of them showed positive reactions to the tests, therefore they were identified as Staphylococcus aureus. The isolates showing positive reaction in haemagglutination test also showed 100% of tube coagulase positive reaction. The haemagglutination test was a reliable method for identifying Staphylococcus aureus in the clinical laboratory. S. aureus produced stronger hemolysis with human blood agar than with sheep blood agar. Antibiotic resistant S. aureus isolates(S-46, S-112, S-126) had 4 to 6 p]asmid DNA elements. The S-112 strain had 6 plasmid DNA elements(1.8, 2.2, 3.7, $26.3{\sim}50$, and 70 Mdaltons), the S-126 had 4 elements(2.6, 4.2, $4.6{\sim}60Md$), and the S-46 had 1 element(${\sim}100Md$). PPSA strain had 4 plasmid DNA elements(2.5, 4.2, $4.6{\sim}60Md$) and S. aureurs(ATCC) strain contained 9.4, 26.3 and ${\sim}50Md$ plasmid DNA elements.

Staphyylococci are responsible for over 80 per cents of the suppurative diseases encountered in medical practice. They cause most suppurative infections of the skin but may also invade and produce severe infections in any other parts of the body. In order to know the carrier rate of staphylococci between the parahospital workers and in the hospital workers, the author undertook isolation of S aureus from nasal cavity on 68 cases of freshmen, 31 cases of sophomores, and 37 cases of juniors in Busan National University, and 30 cases of nursing students, 30 cases of nurses, 30 cases of nurses in charge of operating room and 30 cases of doctors in Busan National University Hospital. The tested total cases were 256 cases which were 136 Cases in parahospital workers and which were 120 cases in hospital workers. The biochemical characters of S aureus strains isolated were studied on coagulase test, mannitol test, hemolysis test and sensitivity test to antimicrobial agents. The results obtained were as follows: 1. S aureus were isolated 49 cases(29.7%) from nasal cavity parahospital workers and were isolated 67 cases(55.8%) from nasal cavity on hospital workers. 2. Among 40 strains of S aureus isolated from parahospital worker's nasal cavity coagulase positive were 29 cases(72.1%), and coagulase negative were 11 cases(27.5%). And mannitol positive were 29 cases(72.5%), and mannitol negative were 11 cases(27.5%). 3. Among 67 strains of S aureus isolated from hospital worker's nasal cavity coagulase positive were 59 cases(88.1%), and coagulase negative were 8 cases(11.9%). And mannitol positive were 49 cases(73.1%), and mannitol negative were 18 cases(26.9%). 4. The hemolysis test of each erythocytes on coagulase and mannitol positive S aureus isolated were sensitive to rabbit(40 cases: 81.6%), guinea pig(26 cases: 53.6%), sheep(13 case: 26.5%), and were not sensitive to chicken and human erythrocytes, respectively. 5. The hemolysis test of each erythocytes on 10 strains of coagulase and mannitol negative S aureus isolated were not sensitive to all of, erythrocytes. 6. The sensitivity test to the various chemotherapeutic agents was almost sensitive to the strains isolated from parahospital workers, but was almost resistant to the strains isolated from hospital workers.
Staphylococcus aureus is one of the major pathogens that can cause staphylococcal infection and food poisoning. In this study, we compared conventional culture methods and real-time PCR for detection of S. aureus in artificially inoculated milk, sausage, raw pork, and vegetable salad. The performance of a coagulase test for confirming S. aureus was also compared with a colony PCR test. Bulk food samples (500 g each) were artificially inoculated with S. aureus and divided into 20 samples (25 g or mL each). All samples were added to tryptic soy broth (225 mL/sample) with 10% NaCl and incubated at $37^{\circ}C$ for 24 h. After the enrichment, broth cultures were streaked onto Baird-Parker (BP) agar with egg yolk tellulite, and incubated at $37^{\circ}C$ for 24 h. In addition, 1 mL of broth cultures was collected to perform real-time PCR. Two suspicious colonies from the BP agar were picked up and plated on nutrient agar and incubated at $37^{\circ}C$ for 24 h followed, by a coagulase confirmation test and a colony PCR analysis. There were no statistical differences between culture methods and realtime PCR in food samples with low background microflora, such as milk and sausage. However, a significant statistical difference was found between the culture methods and real-time PCR for raw pork and vegetable salad. Furthermore, the colony PCR test of the presumptive colonies on BP agar for confirming S. aureus is more accurate and efficient than the coagulase test for unprocessed foods.
The object of this study was to measure the biocidal effect of povidone on staphylococcus found in tracheal incision site, change following the disinfection frequency and duration, and tolerance to the antibiotics. The data was analysed by percentage and t-test using SAS program. The subjects of this study are 35 tracheostomy patients in and Intensive Care Unite of the hospital located in Daegu city and analysing term was from January 16 to February 26, 2001. The results of this study were as follows. The biocidal effect of povidone on Staphylococcus was strong regardless of time and concentration. Staphylococcus aureus was found on third day and found to be highest concentration on 6th day after disinfection of once/a day. Coagulase negative Staphylococcus was not found from 1st to 3rd day and highest on 4th day after disinfection of once/a day. As to bacteria colonization following the disinfection frequency, twice per day of disinfection was more effective on Staphylococcus aureus than once a day. In tolerance test of Staphylococcus aureus and Coagulase negative Staphylococcus, 72.7% of Staphylococcus aureus showed tolerance in Methicillin, 63.6% in Imipenem, and 37.5% of Coagulase negative Staphylococcus showed tolerance in Methicillin, 12.5% in Imipenem. Both of them do not have any tolerance in Vancomycin. The results of this study can be used as the basis for protection against hospital mediated infection through thorough disinfection. With above results, I suggest the following. First, we should research relation between antiseptics and fungi, virus more deeply. Secondly, all medical personnel should try to protect against the hospital medicated infection. Thirdly, there is a need of training professional disinfection personnel for preventing hospital mediated infection and the progress of nursing science.
Wildlife is a bio-indicator of environmental pollution by antimicrobial resistant bacteria or genes, however, there is no information on antimicrobial resistance in wildlife-origin bacteria. This study aimed to investigate the normal microbiota of staphylococci and their antimicrobial resistance in wildlife that did not take any antimicrobials. After sampling and bacterial isolation/identification, antimicrobial resistance profiles were examined by broth microdilution test, Kirby-Bauer disc diffusion test and mecA genetargeted PCR. Of 90 isolates from wildlife, 83 were coagulase-negative staphylococci while only 7 were coagulase-positive staphylococci. Methicillin-resistance was found in 63 (70%) isolates and 35 of 90 (38.9%) isolates were multidrug-resistant staphylococci. When considering that all of the animals did not take any medication or contacted any medical device before the sampling, the results indicate significantly high prevalence of antimicrobial resistance in wild environments. Further study would be necessary to investigate the transmission route of antimicrobial resistance.
Milk samples were collected from a total of 418 quarters of 214 Holstein cows. Of these, samples which were positive on California Mastitis Test (CMT) and above 200,000 cells/ml by somatic cell counts were subjected to bacteriological examination and antimicrobial susceptibility test. Major pathogens responsible for mastitis included coagulase-negative staphylococci (34.3%), coagulase-positive staphylococci (21.5%), gran-negative rod (coliforms and noncoliforms: 12.6%) and Streptococcus spp. (8.4%). These strains were tested with 13 antimicrobial agents by the Kirby and Bauer standardized disc diffusion method. The isolated pathogens were mostly susceptible to amoxicillin and cephalothin, but were resistant to erythromycin.
Although, in human medicine, strains of methicillin-resistant staphylococi have become the most important causative agents of nosocomial infections, studies on the small animals are very. limited. The aim of this study was to determine mecA gene and susceptibility to antibiotics of staphylococci strains isolated from clinically ill or healthy dogs and cats, during the period August 2002-July 2003. A total of 136 staphylococci (87 coagulase-positive and 49 coagulase-negative) were investigated for antibiotic resistance, using disk diffusion and minimum inhibitory concentration (MIC) test. The mecA gene was detected using the polymerase chain reaction. The isolates belonged to the species S. aureus (53 isolates), S. intermedius (34 isolates), S. epidermidis (26 isolates) and other coagulase-negative staphylococci (CNS, 23 isolates). Of the 136 isolates, 43 (31.6%) were mecA-positive and the frequency of the ,presence of mecA gene varied among the different species. All S. aureus strains were mecA-negative and were found to be susceptible, with an oxacillin MIC $\leq$1 $\mu\textrm{g}$/ml. Five (13.6%) isolates of 36 that exhibited oxacillin resistance on the MIC testing were found to be mecA-negative, suggesting not all mecA-positive strains may be an oxacillin resistant. However, the mecA presence of the strains was correlated with high oxacillin resistance: 71.4% (10 isolates of 14; P < 0.001) for mecA-positive S. intermedius and 72.4% (21 isolates of 29; P < 0.001) for mecA-positive CNS isolates. About 69% (94 isolates of 136) showed resistance to at least one drug, and 22.8% (31 isolates) were resistant to four or more different drug classes. Resistance (36 isolates, 71.7%) to penicillin G was a common finidng. This study suggest that the mecA-positive staphylococci are prevalent in small animals, and selection of antibiotics to treat infections caused by mecA-positive staphylococci may be very limited because of multi-drug resistance.
