Middle East Respiratory Syndrome Coronavirus-Encoded Accessory Proteins Impair MDA5-and TBK1-Mediated Activation of NF-κB

  • Middle East respiratory syndrome coronavirus (MERS-CoV) is a newly emerging coronavirus which is zoonotic from bats and camels. Its infection in humans can be fatal especially in patients with preexisting conditions due to smoking and chronic obstructive pulmonary disease (COPD). Among the 25 proteins encoded by MERS-CoV, 5 accessory proteins seem to be involved in viral evasion of the host immune responses. Here we report that ORF4a, ORF4b, and ORF8b proteins, alone or in combination, effectively antagonize nuclear factor kappa B ($NF-{\kappa}B$) activation. Interestingly, the inhibition of $NF-{\kappa}B$ by MERS-CoV accessory proteins was mostly at the level of pattern recognition receptors: melanoma differentiation-associated gene 5 (MDA5). ORF4a and ORF4b additively inhibit MDA5-mediated activation of $NF-{\kappa}B$ while that of retinoic acid-inducible gene 1 (RIG-I) is largely not perturbed. Of note, ORF8b was found to be a novel antagonist of MDA5-mediated $NF-{\kappa}B$ activation. In addition, ORF8b also strongly inhibits Tank-binding kinase 1 (TBK1)-mediated induction of $NF-{\kappa}B$ signaling. Taken together, MERS-CoV accessory proteins are involved in viral escape of $NF-{\kappa}B$-mediated antiviral immune responses.

Pro-tumorigenic roles of TGF-β signaling during the early stages of liver tumorigenesis through upregulation of Snail

  • Many studies have focused on the tumor suppressive role of $TGF-{\beta}$ signaling during the early stages of tumorigenesis by activating the target genes involved in cytostasis and apoptosis. We investigated the effects of $TGF-{\beta}$ inhibition on early tumorigenesis in the liver, by employing diverse inhibitory methods. Strikingly, $TGF-{\beta}$ inhibition consistently suppressed hepatic tumorigenesis that was induced either by activated RAS plus p53 downregulation or by the co-activation of RAS and TAZ signaling; this demonstrates the requirements for canonical $TGF-{\beta}$ signaling in tumorigenesis. Moreover, we found that Snail is the target gene of the $TGF-{\beta}$ signaling pathway that promotes hepatic carcinogenesis. The knockdown of Snail suppressed the early tumorigenesis in the liver, as did the $TGF-{\beta}$ inhibition, while the ectopic expression of Snail restored tumorigenesis that was suppressed by the $TGF-{\beta}$ inhibition. Our findings establish the oncogenic $TGF-{\beta}$-Smad-Snail signaling axis during the early tumorigenesis in the liver.

오이풀 뿌리 분획물의 화장품 소재로서의 약리활성 연구 (The research of pharmacological activation for Sanguisorbae Radix Fractions as cosmetic material)

  • Objectives : Sanguisorbae Radix(SO) is a plant in the family Rosaceae, which grows widely in open fields Korea. It has been used as traditional medicine for thousands of years, as a treatment for anti-inflammatory and it is widely used for throat infection, tonsilitis, conjuctivitis and lymphadentis. In this study, investigated skin antiaging and anti-bacterial by using SO fractions water, acetone and butanol, chloroform. Methods : The effects of anti-microbial on SO fractions and elastase inhibition activity, collagenase inhibition activity were experimented. Results : 1. The ethyl acetate fraction showed the strongest antimicrobial activity against Staphylococcus epidermidis. 2. The elastase inhibition rate and collagenase inhibition rate of the water fraction of SO was the highest other factions. Conclusions : From the above results, it was confirmed the SO has sufficient potentiality applying itself to industry and also SO can be utilized as antimicrobial natural materials and antiaging cosmetics.

잔디 Large patch의 생물학적 방제를 위한 길항 미생물의 선발과 항균력 검정 (Antimicrobial test of Antagonistic Microbes for Biological Control of Large patch of Zoysiagrass)

  • A large patch disease caused by Rhizoctonia solani AG2-2(IV) is a serious problem in turfgrass sites including golf courses and sports fields in Korea. The objectives of this study were to isolate some antagonistic microorganisms and to explain some involving mechanisms. Initially single colonies which were formed from the filtrates of various soil samples were obtained from LB culture and then co-cultured with R.solani AG2-2(IV) on PDA plate to explore some antagonistic microbes against for large patch fungus, Rhizoctonia solani AG2-2(IV). Out of total 82 antagonistic isolates which commonly had inhibition effect on Rhizoctonia solani AG2-2(IV) mycelial growth, one candidate (YPIN22) showed the most antifungal effect, which was confirmed by the longest distance from the edge of bacterial colony to the mycelial edge of the Rhizoctonia solani AG2-2(IV) in the dual culture. A succeeding investigation was to test any potential effect of the isolate on growth inhibition of 5 other turfgrass pathogens including R. solani solani AG2-2(IIIB), P. ultimum, C. caudatum, C. lunata, and F.oxysporum. Preliminary result indicated that the new isolate YPIN22 was also found to have antagonistic potential on the growth inhibition of those turfgrass pathogenic fungi, which was explained by inhibition zones ranging from 8 to 22mm. A further explanation of some characteristics of the isolate YPIN22 will be discussed in detail.

Growth Inhibitory Effects of Chlorine Dioxide on Bacteria

  • Chlorine dioxide ($ClO_2$) gas is a neutral chlorine compound. $ClO_2$ gas was proven to effectively decontaminate different environments, such as hospital rooms, ambulances, biosafety level 3 laboratories, and cafeterias. In this study, to evaluate the effects of $ClO_2$ gas, bacteria of clinical importance were applied. Staphylococci, Streptococci and Bacillus strains were applied and Klebsiella, and others e.g., Escherichia coli, Shigella, Salmonella, Serratia were also done for the inhibitory analysis. Bacteria plates were applied with a hygiene stick, namely, "FarmeTok (Medistick/Puristic)" to produce $ClO_2$. $ClO_2$-releasing hygiene stick showed the very strong inhibition of bacterial growth but had different inhibitions to the bacteria above 96.7% except for MRSA of 90% inhibition. It is difficult to explain why the MRSA were not inhibited less than others at this point. It can be only suggested that more releasing $ClO_2$ should be essential to kill or inhibit the MRSA. B. subtilis, S. agalactiae, S. pyogenes, E. coli O157:H7, S. typhi (S. enterica serotype typhi) and S. marcesence were inhibited over 99%. This study will provide fundamental data to research growth inhibition by $ClO_2$ gas with bacteria of clinical importance value.

Lactobacillus rhamnosus와 Lactobacillus paracasei 복합 배양의 피부 미백 및 주름 개선 활성 증진 (Enhancement of Skin Whitening and Anti-wrinkle Activities of the Co-culture of Lactobacillus rhamnosus and Lactobacillus paracasei)

  • 본 연구는 여성의 손으로부터 분리한 두 유산균의 co-culture에 의한 향장활성에 대한 것이다. Lactobacillus rhamnosus (L. rhamnosus)와 Lactobacillus paracasei (L. paracasei)를 함께 배양하는 것은 최초이며, co-culture를 이용한 유산균은 tyrosinase 저해율이 L. paracasei와 L. rhamnosus에 비해 20.68%로 가장 높은 수치를 보였다. 또한, co-culture 유산균은 melanin 생성 저해율에서도 63.5%의 가장 높은 활성을 보였으며, 주름개선활성에 영향을 미치는 MMP-1의 생성량이 3726.3 pg/mL로 확인되었다. 반면 단일 유산균 배양시 얻어지는 MMP-1 생성량은 각각 L. rhamnosus가 13613.5 pg/mL, L. paracasei는 13012.0 pg/mL이 확인되었다. Collagen 생성량을 확인한 결과 co-culture 유산균이 380.7 ng/mL로 323.4 ng/mL의 양이 확인된 L. paracasei와 304.1 ng/mL의 양이 확인된 L. rhamnosus보다 많은 양이 생성됨을 확인하였다. 상기의 결과는 유산균의 co-culture가 향장활성 증진을 시킬 수 있다는 것을 나타내고, 이는 향장 활성에 영향을 미치는 항산화 활성을 측정한 결과 단일 유산균 배양을 통한 결과보다 co-culture 유산균의 항산화 활성이 더 높은 것으로 확인할 수 있었다.

  • Lactic acid bacteria have been considered as the most beneficial probiotic organisms contributing to inhibition of harmful and putrefactive intestinal bacteria. Among them, Bifidobacterium spp. has been considered as one of the most beneficial probiotic organism that can improve the health of humans, since it is one of the major bacteria flora in human intestine. However, the harmful enzyme-inhibitory activity of lactic acid bacteria of Kimchi, which is a representative Korean fermented food has not been evaluated. (omitted)

Helicobacter pylori 의 생육억제에 대한 유산균, 난황항체 및 목이버섯의 상승효과 (Synergistic Inhibition of IgY, Auricularia auricula, and Lactic Acid Bacteria from Kimchi and Tarak on Helicobacter pylori)

  • 소화성 질환의 중요 인자인 Helicobacter pylori를 저해하는 IgY, 목이버섯, 김치와 타락 유산균을 이용하여 생육 저해 상승 효과를 분석하고자 하였다. 동결 건조된 유산균 배양 상징액을 다양한 효소 처리 결과, 지질 분해 효소를 제외하고는 활성을 나타냈다. GC 분석을 통해 유산균 동결건조액의 지방산 조성은 undecanoic acid($C_{11:0}$), palmitic acid($C_{16:0}$), steraic acid($C_{18:0}$), oleic acid($C_{18:1}$)가 L. mensenteroides LABKW5와 S. thermophilus LAB KW15에서 모두 확인되었으며, eicosadienoic acid($C_{20:2}$)는 LAB KW5에서만 나타났다. 또한 유산균 동결건조액은 다른 식중독균에서도 spot assay의 결과, 그람 음성균 중에서 특히 E. coli O157:H7, E. coli, C. sakazakii 등에서 생육 저해능이 확인되었다. 목이버섯 추출물은 열수 추출과 ethanol를 이용해 분리하였으며, HPLC를 이용하여 목이버섯 추출 다당체를 분석한 결과, glucurono-xylomannan 혹은 glucomannan이 ${\beta}$-glucan과 함께 존재하는 혼합물일 것으로 되었다. 또한, 면역란은 1차 접종 후 11일째부터 주마다 2회씩 회수하여 IgY를 분리, 정제하였다. 실험을 통해 동결 건조된 유산균 배양 상징액, IgY, 목이버섯 추출액을 혼합 배양하여 배양시간에 따른 생육 저해력의 변화를 확인한 결과, 유산균에 의해 H. pylori의 저해 효과가 있었으며, IgY와 목이버섯의 혼합 시 추가 억제 효과가 있는 것으로 나타났다. 그러므로 동결 건조된 유산균 배양 상징액, IgY, 목이버섯 추출액의 복합처리는 H. pylori를 제어하는데 효과적인 것으로 보인다.

Degradation of Rice Straw by Rumen Fungi and Cellulolytic Bacteria through Mono-, Co- or Sequential- Cultures

  • Two strains of rumen fungi (Piromyces rhizinflata B157, Orpinomyces joyonii SG4) and three strains of rumen cellulolytic bacteria (Ruminococcus albus B199, Ruminococcus flavefaciens FD1 and Fibrobacter succinogenes S85) were used as mono-cultures or combinationally arranged as co- and sequential-cultures to assess the relative contributions and interactions between rumen fungi and cellulolytic bacteria on rice straw degradation. The rates of dry matter degradation of co-cultures were similar to those of corresponding bacterial mono-cultures. Compared to corresponding sequential-cultures, the degradation of rice straw was reduced in all co-cultures (P<0.01). Regardless of the microbial species, the cellulolytic bacteria seemed to inhibit the degradation of rice straw by rumen fungi. The high efficiency of fungal cellulolysis seems to affect bacterial degradation rates.
