차가버섯 추출물이 소화기계 암세포의 증식 및 Caspase-3 활성에 미치는 영향 (Effect of Inonotus Obliques Extracts on Proliferation and Caspase-3 Activity in Human Castro-Intestinal Cancer Cell Lines)

  • We studied the effects of hot water extract of Inonotus obliquos mushroom on the proliferation and apoptosis of the human colon adenocarcinoma, HT-29 and the human stomach adenocarcinoma, SNU-484 cell. Cells were maintained with Dulbecco's modified Eagle medium/Ham's F-12 nutrient mixture supplemented with 10% fetal bovine serum at 37$^{\circ}C$ in a humidified $CO_2$. For the cell proliferation experiments, cells were seeded in 35 mm dishes, and were treated with the various concentrations of the extract for the different time course. Apoptosis was measured by caspase-3 activity. When we incubated HT-29 cells for 24, 48, 72, and 96 hours after treatments, the cell proliferation was more suppressed with more treatment time. In case of the human stomach cancer cell, SNU484, the extract significantly decreased the cell number. Thus, the treatment of 1.5 mg/$m\ell$ extract decreased almost half of the cell number. Caspase-3 activity in HT-29 was increased by the treatment of mushroom extracts. In SNU484, caspase-3 activity tended to increase in proportion to the amounts of the extracts and the treatment of Inonotus obliquos affected the activity a lot. Therefore, Inonotus obliquos is suggested for the prevention of gastro-intestinal cancer and strongly recommended for the treatment of stomach cancer. (Korean J Nutrition 36(1) : 18~23, 2003)

Production of Cellulase and Xylanase by Aspergillus niger KKS

  • A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.

Fate of Parthenogenetic Mouse Embryos Aggregated with ES Cells

  • The present study examined the developmental ability of embryonic stem (ES) cells aggregated with mouse parthenogenetic embryos. Oocytes obtained from superovulated female mouse (BCF1) were treated with 7% ethanol and 5 $\mu\textrm{g}$/$m\ell$ cytochalasin B (CB) for producing pathenotes and in vitro fertilized with fresh sperm for producing normal embryos. The reporter vector (pNeoEGFP) were inserted into ES cells (129S4/svJae) by electroporation. At the 8-cell stage, in vitro fertilized embryos and pathenotes, which the zona pellucida was removed, were co-cultured with 5~10 ES cells for 4 hr. After in vitro fertilized embryos and parthenotes aggregated with ES cells were incubated to blastocyst stage, and these blastocysts transferred into the uterus of pseudopregnant recipients. The fertilized embryos aggregated with ES cells were successfully developed to offspring, but the parthenotes aggregated with ES cells failed to develop offsprings. However, genomic DNA of ES cells was detected in the pathenogenetic fetus by polymerase chain reactions at 15 day post gestation. In this study, results indicated that parthenotes aggregated with ES cells showed possible development to fetus. In the future, this method may help to produce transgenic chimera from parthenotes aggregated with ES cells.

Optimal Culture Conditions for the Production of a Novel Extracellular Alkaline Lipase from Yarrowia lipolytica NRRL Y-2178

  • Lipases are industrially useful versatile enzymes that catalyze numerous different reactions. Among lipases functioning under extreme conditions, alkaline lipase is useful in detergent industry. Lipase from yeast strain Yarrowia lipolytica NRRL Y-2178 was most active under alkaline condition, and initial medium pH for most lipase production was also alkaline [Lee et al., 2007, J Microbiol Biotechnol, 17(6)]. High lipase production was achieved using Y. lipolytica NRRL Y-2178. Optimal incubation time for lipase production at $25^{\circ}C$ was 72 h. Optimal temperature, when incubated for 72 h, was $27.5^{\circ}C$. Lipase production but not cell growth was very sensitive to concentrations of glucose and glycerol as efficient carbon sources, showing optimal concentrations of 1.0 and 1.5% (w/v), respectively. Lipase production was highly stimulated by $Ca^{2+},\;K^+,\;and\;Na^+$, but was inhibited by $Co^{2+},\;Cu^{2+},\;Mn^{2+},\;Na^+,\;and\;Fe^{2+}$. Maximum lipase production at 0.1 mM $Ca^{2+}$ for 72 h incubation at $27.5^{\circ}C$ was 649 units/mL.

Single-Step Purification of Proteins of Interest from Proteolytically Cleaved Recombinant Maltose-binding Protein (MBP) Fusion Proteins by Selective Immunoprecipitation of MBP

  • The maltose binding protein (MBP) fusion protein system is a versatile tool to express and isolate recombinant proteins in E. coli. In this system, MBP fusion proteins are efficiently isolated from whole cell lysate using amylose conjugated agarose beads and then eluted by competition with free maltose. Since MBP is a rather large molecule (∼42 kDa), for further experiments, the MBP part is usually proteolytically cleaved from the fusion protein and subsequently removed by ion-exchange chromatography or rebinding to amylose columns after washing out excess and MBP-bound maltose. In the present study, we have developed an improved method for the removal of cleaved MBP, which is advantageous over conventional methods. In this method, factor Xa cleaved MBP fusion proteins were incubated with Sepharose beads conjugated with MBP specific monoclonal antibodies and then precipitated buy centrifugation, resulting in highly purified proteins in the supernatant.

Helicobacter pylori에 의한 세포독성 및 Interleukin-8 생성에 미치는 생약혼합물의 영향 (Inhibitory Effect of Medicinal Plant Extract on Cell Toxicity and Interleukin-8 Production Induced by Helicobacter pylori)

  • The effects of Helicobacter pylori and medicinal plants extract (Leweifang) on the viability and interleukin(IL)-8 production of gastric epithelial cell were investigated. Cells viability was significantly decreased when they incubated with H. pylori or H. pylori toxin. Co-incubation with Leweifang increased H. pylori or H. pylori toxin-inhibited cell growth in a concentration-dependent manner. The production of IL-8 was greatly increased in H. pylori-infected KATO III gastric epithelial cells in a concentration- and time-dependent manner. The increased production of IL-8 was significantly inhibited by Leweifang $(1,000{\sim}5,000{\mu}g/ml)$. These results indicate that Leweifang has protective effect on H. pylori-inhibited cell growth and H. pylori-induced gastric mucosal cell inflammation by suppressing the production of inflammatory cytokine (IL-8) from gastric epithelial cells.

Oxidative modification of ferritin induced by hydrogen peroxide

  • Excess free iron generates oxidative stress that may contribute to the pathogenesis of various causes of neurodegenerative diseases. In this study, we assessed the modification of ferritin induced by $H_2O_2$. When ferritin was incubated with $H_2O_2$, the degradation of ferritin L-chain increased with the $H_2O_2$ concentration whereas ferritin H-chain was remained. Free radical scavengers, azide, thiourea, and N-acetyl-$_L$-cysteine suppressed the $H_2O_2$-mediated ferritin modification. The iron specific chelator, deferoxamine, effectively prevented $H_2O_2$-mediated ferritin degradation in modified ferritin. The release of iron ions from ferritin was increased in $H_2O_2$ concentration-dependent manner. The present results suggest that free radicals may play a role in the modification and iron releasing of ferritin by $H_2O_2$. It is assumed that oxidative damage of ferritin by $H_2O_2$ may induce the increase of iron content in cells and subsequently lead to the deleterious condition.

FRACTIONS, RUMINAL DISAPPEARANCE AND DIGESTION RATE OF DEER FEED NUTRIENTS ESTIMATED USING IN SITU BAG TECHNIQUE IN THE ARTIFICIAL RUMEN

  • A study was conducted to estimate nutritive value of forage sources used in deer diets. Bags containing feedstuffs were incubated four times for periods up to 72 hours in two chemostats filled with ruminal fluid from deer or cattle. Parameters estimated were water-soluble and $65{\mu}m$ filterable plus insoluble digestible fractions(No. = 4) and extent of disappearance(No. = 8) of feed neutral detergent fiber(NDF). Among tested feeds, the ranking of values of these parameters were soybean hulls > alfalfa pellets > corn cobs or rice straw > cottonseed hulls or rice hulls > sawdust, indicating that soybean hulls and alfalfa pellets were more fermentable than other agricultural residues. It is possible to utilize variability among tested feeds of fraction, disappearance rate and (or) extent of DM and NDF when these feeds are used as roughage sources in deer diets.

토끼 수정란 체외 배양액의 개발에 관한 연구 (Study on Development of In Vitro Culture Medium for Rabbit Embryos)

  • This experiment was carried out to improve in vitro development of rabbit one-cell embryos to the blastocyst stage. One-cell rabbit embryos were collected at 19\ulcorner20hr after superovulation induction and incubated at 39\ulcorner in 5% CO2 for 72hr. In order to find optimum conditions in medium that affects the rabbit embryo's development in vitro, RDH medium which mixed with RPMI1640, DMEM and Ham's F10 was compared with the previously reported mediums (Ham's F10 and RD) for embryo development and cell numbers. Three additives (BSA, taurine and glucose) were tested for the development of rabbit one-cell embryos in vitro. When the embryos were cultured in RDH medium, their development was markedly promoted as compared with Ham's F-10 or RD alone. Glucose exhibited no significant effects on embryo development and cell numbers. BSA a, pp.ared to promote transition from morula to blastocyst stage and taurine increased cell numbers of cultured embryos markedly regardless of medium. BSA and taurine together in RDH medium showed the additive effects on embryos development and cell number.

Helper-Independent Live Recombinant Adenovirus Vector Expressing the Hemagglutinin-Esterase Membrane Glycoprotein

  • The hemagglutinin-esterase glycoprotein (HE) gene of bovine coronavirus, coupled with a simian virus 40 early promoter and polyadenylation signal, was inserted into a human adenovirus transfer vector. The transfer vector was used to co-transfect 293 cells along with adenovirus genomic DNA. The hemagglutinin-esterase transcription unit was rescued into the adenovirus genome by homologous in vivo DNA recombination between the vector plasmid DNA and the adenovirus genomic DNA, and a recombinant adenovirus was isolated by several rounds of plaque assays. Thus the recombinant adenovirus carries the hemagglutinin-esterase gene in the early transcription region 3 (E3) of the adenovirus genome in the parallel orientation to the E3 transcription. The recombinant adenovirus synthesized the HE polypeptide in HeLa cells as demonstrated by immunoprecipitation with anti-coronavirus rabbit antisera. The recombinant HE polypeptide could be labelled by $[^3H]$glucosamine, demonstrating that the recombinant HE was glycosylated. Cells expressing the HE polypeptide exhibited hemadsorption activity when incubated with mouse erythrocytes. The HE was transported to the plasma membrane as shown by the cell surface immunofluorescence, indicating that the recombinant HE polypeptide retained its biological activities. Potential for the use of infectious recombinant adenovirus as a live virus-vectored vaccine candidate for bovine coronavirus disease is discussed.
