The transformation-associated recombination (TAR) cloning technique allows selective isolation of chromosome regions or genes from complex genome. The procedure requires knowledge of relatively small genomic sequences that reside adjacent to the chromosome region of interest. This method involves homologous recombination during spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences (hooks). To examine whether TAR cloning can be applied to the isolation of gene homologues, we chose the HPRT genes from human and mouse genome. As results, the yield of positive clones for HPRT gene from human and mouse genome when using a TAR vector containing mHPRT hook or hHPRT hook was almost same level. Analysis of the gap regions in mHPRT revealed that they contain abnormalities that could result in instability of the sequences. In conclusion, we were able to use the TAR cloning technology to isolate gene homologue (orthologue) from nonidentical genome. Moreover, the use of the TAR cloning system may accelerate work on closing the remaining gaps in mammalian genome to achieve the goal of annotation of all mammalian genes.

A patient who went through maxillectomy can have soft palate defects including oronasal fistulas and suffer from dysphagia and dysarthria due to velopharyngeal insufficiency. This defect causes the food to enter nasal cavity and creates hypernasal sound which debilitates a quality of life. An obturator can rehabilitate the substantial oral tissue defects. The maxillary obturator separates the nasopharynx from the oropharynx during speech and deglutition by closing of the defect. For edentulous obturator patient, it is difficult to obtain proper retention due to reduced peripheral sealing. Therefore, the contours of the defects must be used to maximize the retention, stability, and support. Hollow type obturator can improve physiologic function by reducing weight than the traditional obturator. This case report describes a patient with hemi-maxillectomy who recovers mastication, speech, deglutition, and appearance with a maxillary obturator using physiological border molding of the velopharyngeal area and double-processing method.

This study evaluated groundwater quality around an uncontrolled landfill in W sity, Korea, which was monitored for about two years (2005-2006). Parameters of concern include redox-sensitive indicators such as pH, DO, EC, ORP, DOC (dissolved organic carbon), NH3, NO3 and SO4. About 10 years have elapsed after closing dumping of municipal wastes in the landfill. Leachates showed widely varying concentrations in COD(136${\sim}$263 mg/L), T-N(121${\sim}$186 mg/L), and NH3-N(14${\sim}$369 mg/L). Groundwater at the immediate downgradient of the landfill showed weakly acidic pH condition but very high levels of EC (3,000-4,000 ${\mu}S/cm$), which indicated that the groundwater was largely affected by the landfill leachate. Cl, a conservative ion, showed over 200 mg/L at the landfill border, but a gradually decreasing level with distance from the landfill, representing dispersion and dilution (natural attenuation) due to mixing with surrounding groundwater and replenished rainwater. Redox potential showed negative value at the landfill border but it increased up to 350 mV at downgradient wells, which indicated conversion of redox condition from reducing oxic ones. Ammonia, was largely enriched at most of the monitoring wells and its level greatly exceeded drinking water standard. In summary, all the parameters evidenced occurrence of natural attenuation with distance and with time but further monitoring is still required.
Biennial serial cephalometric radiographs of 47 children (26 boys, 21 girls) were used to analyze the craniofacial growth changes in Korean children from 8 years to 16 years of age. A craniofacial model was designed for this study. It consisted of 72 anatomical points and 98 derived points. The craniofacial changes of these samples during these ages might be summarized as follows: 1. Mandibular growth to cranial base was more forward than maxillary growth. 2. Mandibular growth during this period was found to be a forward-upward (bite closing) rotation of the mandible. 3. Growth changes in total mandibular length (Ar-Gn) showed a pubertal growth spurt at 12-14 years of age in boys and 10-12 years of age in girls. Synchrony of the growth spurts on total mandibular length (Ar-Gn) and standing body height was found. 4. The pubertal spurts occured in the growth of total cranial base (Ba-Na) at 10-12 years of age in boys and 8-10 years of age in girls. The time of the spurts of the cranial base growth was 2 years ahead of that of the total mandibular growth. 5. Synchrony of growth spurts in anterior facial height (Na-Me), posterior facial height (S-Go) and body height was found. 6. The whole craniofacial changes during this period were plotted by using a X-Y plotter and personal computer. A simple profilogram for an diagnostic tool was obtained.
The purpose of this study was to evaluate the force degradation rate of synthetic elastomeric chains during space closing phase of orthodontic treatment. Two kinds of synthetic elastomeric chains(RMO, 3M) were selected which were commonly used In clinics. All of the samples were extended and tested for 4 weeks under the simulated intraoral condition. The results can be summarized as follows : 1. Time related residual force showed typical logarithm function. Residual force after 4 weeks was $41.2\~64.6\%$ of original force, and difference between two kinds of elastomeric chain existed. 2. Elastic force decreased greatly during first 10 minutes, so $20\~25\%$ of original force disappeared. After that, this decreasing tendency was diminished significantly, average rate of elastic force after 1 week to 4 weeks were $1.5\%$ demonstrating rather constant force. 3. Even though the same brand of elastomeric chiain was used, as extension rate of elastomeric chain increased, force degradation rate increased by decreasing of residual force.

To set up an objective basis for the evaluation of the stroboscopic findings, video-strobolaryngoscopic images of vocal fold vibration in 5 female and 5 male normal speakers were analyzed using an image analysis computer program called KSIP(Kay Storoboscopy Image Processing, Kay Elemetrics Corp., NJ, USA). Four consecutive vibratory cycles were compared in comfortable, louder, high-pitched /ee/ phonation for every subject. findings mostly replicated earlier studies including glottal chinks which were observed in most female speakers throughout the cycles and clear distinction between female and male speakers in their vibratory patterns as well as intensity and frequency-re-lated differences. However, there were some findings incompatible with those from previous studies which may be attributable to technical problems. This study may provide an objective basis of the stroboscopic findings such as image shape, amplitude, area, and their changes according to frequency and intensity variations. We anticipate that funker study with larger samples ran provide an objective criteria for normal vibratory characteristics of the laryngostro-boscopic findings.

Moyamoya disease is characterized by progressive bilateral narrowing of the internal carotid arteries(ICA) and their main branches, and formation of a fine vascular network (moyamoya vessels) at the base of brain. Mild intellectual or motor impairment, or both can be seen in moyamoya disease patient. Patients having intellectual disability usually have malocclusions associated with oral habits such as tongue thrusting, bruxism, or clenching. A patient with moyamoya disease and cerebral palsy visited Chonbuk National University hospital for dental examination. She showed mild openbite on anterior teeth, and had oral habits such as mouth breathing, tongue thrusting, and drooling. She was treated with myofunctional therapy(MFT) whose purpose was to strengthen orofacial muscles. Lip closing force and bite force were recorded to evaluate the muscle tone. Tongue crib was delivered to restrict tongue thrusting habit, since her compliance to the MFT was decreased after 6 months of treatment. With the MFT and tongue crib, the openbite was fairly improved. When myofunctional therapy has difficulty in obtaining good compliances, the treatment should not be effective enough. Although intellectually disabled patients are not compliant enough, thus a habit breaking device should be needed to treat the openbite caused by oral habits, dentist should make an effort to do MFT, since it could reduce a treatment period, and enhance the stability.

This paper is to study the examples for rubber damage and weaken reliability of timing belt, constant velocity joint boot and weather strip in vehicle. The first example, when the service man replaced the new timing-belt of rubber material, he assembled the belt that was weaken it's contact surface because of material transform. He knew the abnormally tearing failure by friction action between belt and sprocket. The second example, it certified the fact that the grease is leaked out boot protecting of constant velocity joint by split of rubber surface because of durability badness. The third example, the weather stripe took the minutely tearing because of damage produced the material transform by crack of chemistry change. It certified the production phenomenon of a tiny noise by coming with outside air because of overlapped the rubber of weather stripe when driving after closing the door. Therefore, the driver must always manage the rubber system part of vehicle.
Objective: All patients who have the disc displacement with reduction (DDwR) are always not progressive but some of them with significant catching or locking progress to disc displacement without reduction (DDw/oR). In this study, we suggest opening exercise therapy using Locking-free Appliance (LA) to be helpful to prevent progressive derangement. Methods: Retrospectively, a total 37 patients who received exercise treatment using LA was analyzed from January 2010 to December 2011. Patients had been instructed in proper exercise technique to practise for opening and closing of mouth wearing the appliance 30 times a day in locking-free position. Results: None of the patients took a turn for worse to DDw/oR (37 patients). The 16 patients (43.2%, locking-free group) of them could open their mouth without intermittent condylar locking. Measurement of data was analyzed using and Mann-Whitney test. There was no statistically significant difference for age, gender, presence of palpational pain, locking period, VAS quantitative pain score between locking-free group and locking group. Conclusions: In this study, some patients could open their mouth without intermittent locking and did not take a turn for worse to DDw/oR after opening exercise therapy with LA. Furthermore, LA had advantages which were short wearing time, low costs and minimizing patients' uncomfortable compared to conventional treatment method. This therapy can be more appropriate new treatment to prevent progress to DDw/oR.
Statement of problem: The previous unilateral bite force recorder has several limitations for taking long time for measuring and causing discomfort to subjects. Because of these reasons, it could not use widely for epidemiological researches. However, 'Dental Prescale System' which is new equipment for measuring bite force, is more convenient for measuring bite force than previous unilateral recorder. Purpose: The purpose of this study was to compare a new technique(Dental Prescale System) using pressure sensitive foils for recording of maximal jaw closing force with conventional measurement using unilateral bite force recorder Material and method: This studies included 22 healthy dental students in college of dentistry Yonsei university in Rep of Korea. Mean age was 23.9 years. All subjects had continuous dental arches and no significant malocclusion or signs from the teeth and craniomandibular system. The Dental Prescale System (Fuji Film, Tokyo, Japan) consists of a horseshoe-shaped pressure sensitive sheet (50H, R type) and a computerized scanning system.(FPD705) We also used unilateral bite force recorder(Denbotics Co. Seoul, Rep.Korea) for comparing with Dental Prescale. Results and conclusion: The total bite force recorded with Dental Prescale System (1423 N) was systematically higher than that recorded by unilateral bite force recorder. (256 N) However, the maximum bite force values measured in the two ways were significantly correlated (r=0.46, p<0.05). The Dental Prescale bite force calculated for first molar (208 N) was lower than that recorded by unilateral bite force recorder. (256 N) The two values were also very significantly correlated. (r=0.66. p<0.001) There were significantly different in bite force between two measurement methods. The reasons were first, unilateral bite force recorder measured only the bite force of a part of teeth, and Dental Prescale measured the total teeth force. Second, in measurement, a difference in muscle contraction appeared by an extent of mouth-opening. Third, unilateral bite force recorder has the reducing effect of the bite force by protective tape. Fourth, Dental Prescale has limitations during the computer scanning procedure. Therefore, Dental Prescale System is considered to be a very promising alternative to be conventional bite force recording methods.
