It has been reported that hepatic glutathione (GSH) levels are decreased in diabetic patients, and glucagon increases hepatic efflux of GSH into blood. The signaling pathways responsible for mediating the glucagon effects on GSH efflux, however, are unknown. The signaling pathways involved in the regulation of GSH efflux in response to glucagon and insulin were examined in primary cultured rat hepatocytes. The GSH concentrations in the culture medium were markedly increased by the addition of glucagon, although cellular GSH levels are significantly decreased by glucagon. Insulin was also increased the GSH concentrations in the culture medium, but which is reflected in elevations of both cellular GSH and protein. Treatment of cells with 8-bromo-cAMP or dibutyryl-cAMP also resulted in elevation of the GSH concentrations in the culture medium. Pretreatment with H89, a selective inhibitor of protein kinase A, before glucagon addition markedly attenuated the glucagon effect. These results suggest that glucagon changes GSH homeostasis via elevation of GSH efflux, which may be responsible for decrease in hepatic GSH levels observed in diabetic condition. Furthermore, the present study implicates cAMP and protein kinase A in mediating the effect of glucagon on GSH efflux in primary cultured rat hepatocytes.

Somatic cell nuclear transfer (SCNT) is a useful tool for animal cloning, but the efficiency of producing viable offspring by SCNT is very low. To improve this efficiency in the production of cloned pigs, it is critical to understand the interactions between uterine function and cloned embryos during implantation. Lysophosphatidic acid (LPA) is a lipid mediator that plays an important role in the establishment of pregnancy in pigs; however, LPA production in the uterine endometrium of pigs carrying SCNT-cloned conceptuses has not been determined. Therefore, we investigated expression of ENPP2, an LPA-generating enzyme, in the uterine endometrium of gilts with conceptuses derived from SCNT during the implantation period. Uterine endometrial tissue and uterine flushing were obtained from gilts carrying SCNT-derived conceptuses and from gilts carrying conceptuses resulting from natural mating on d 12 of pregnancy. Our results demonstrated no difference in the level of ENPP2 mRNA expression in the uterine endometrium between gilts carrying SCNT-derived conceptuses and gilts carrying naturally-conceived conceptuses, but secretion of ENPP2 protein into the uterine lumen did decrease significantly in pigs with SCNT-derived conceptuses. These results indicate that expression and secretion of ENPP2, which are critical for appropriate LPA production and successful pregnancy, are dysregulated in the uterine endometrium of pigs carrying SCNT-derived conceptuses.
One of the reasons to causing blood coagulation in the tissue of xenografted organs was known to incompatibility of the blood coagulation and anti-coagulation regulatory system between TG pigs and primates. Thus, overexpression of human CD73 (hCD73) in the pig endothelial cells is considered as a method to reduce coagulopathy after pig-to-non-human-primate xenotransplantation. This study was performed to produce and breed transgenic pigs expressing hCD73 for the studies immune rejection responses and could provide a successful application of xenotransplantation. The transgenic cells were constructed an hCD73 expression vector under control porcine Icam2 promoter (pIcam2-hCD73) and established donor cell lines expressing hCD73. The numbers of transferred reconstructed embryos were $127{\pm}18.9$. The pregnancy and delivery rate of surrogates were 8/18 (44%) and 3/18 (16%). The total number of delivered cloned pigs were 10 (2 alive, 7 mummy, and 1 died after birth). Among them, three live hCD73-pigs were successfully delivered by Caesarean section, but one was dead after birth. The two hCD73 TG cloned pigs had normal reproductive ability. They mated with wild type (WT) MGH (Massachusetts General Hospital) female sows and produced totally 16 piglets. Among them, 5 piglets were identified as hCD73 TG pigs. In conclusion, we successfully generated the hCD73 transgenic cloned pigs and produced their litters by natural mating. It can be possible to use a mate for the production of multiple transgenic pigs such as ${\alpha}-1,3-galactosyltransferase$ knock-out /hCD46 for xenotransplantation.
닭의 간은 해독작용, 당의 저장, 혈장 단백질의 합성 등 주요 기능을 하는 것으로 알려져 있다. 본 연구는 간에서 성장 단계별로 발현량에 차이를 보이는 단백질들을 비교해 보았다. 2차원적 전기 영동에 의해 분리된 300개 이상의 단백질들이 확인되었으며 이 중 성장 단계별 특이적인 13개의 단백질은 MALDI-TOF MS에 의하여 분석이 되어졌다. 본 연구를 통하여 밝혀진 단백질들은 생화학적인 연구에 중요한 자료를 제공할 것으로 사료된다.
The xenotransplantation of pig organs and cells can be related with a risk of transmission of infectious diseases to human. Previous findings indicate that the regulatory region of PERV for retroviral transcription, replication and integration into the cellular DNA is located on the 5' Long Terminal Repeat (LTR). The objective of this study is the investigation of methylation and deletion status of the PERV 5' LTR region which can be used for regulating PERV expression. We compared the sequences of genomic DNA and bisulfite-treated genomic DNA from PK-15 cells expressing PERV to observe the methylation status of the 5' LTR. Our results showed that the CpG sites of U3 were methylated and methylation was inconsistent in the R and U5 regions. Also, variable numbers of 18 bp repeats and 21 bp repeats were detected on 5' LTR by sequencing analysis. The consistent U3 methylation might be indicative of host suppression of expression of the retroviruses.

The transfection efficiency of a transgene into pig and goat fetal fibroblast cells (PFF and GFF, respectively) was tested using co-transfection of a human tissue-type plasminogen activator (tPA) transgene and neomycin-resistant ($Neo^r$) gene, followed by G418 selection. To initially test G418 resistance, GFF and PFF were incubated in culture medium containing different concentration of G418 for 2 weeks, and cell survival was monitored over time. Based on the obtained results, the concentrations chosen for G418 selection were 800 ug/ml and 200 ug/ml for GFF and PFF, respectively. For co-transfection experiments, the pBC1/tPA and $Neo^r$ vectors were co-transfected into GFF and PFF ($1{\times}10^6$ cells in each case) using the FuGENE6 transfection reagent, and resistant colonies were obtained following 14 days of G418 selection. We obtained 96 and 93 drug-resistant colonies of GFF and PFF, respectively, only 54 and 39 of which, respectively, continued proliferating after drug selection. PCR-based screening revealed that 23 out of 54 analyzed GFF colonies and 5 out of 39 analyzed PFF colonies contained insertion of the tPA gene. Thus, the experimentally determined transfection efficiencies for tPA gene co-transfection with the $Neo^r$ gene were 42.6% for GFF and 12.8% for PFF. These findings suggest that co-transfection of a transgene with the $Neo^r$ gene can aid in the successful integration of the transgene into fetal fibroblast cells.
We employed human SK-MEL-28 cells as a model system to identify cellular proteins that accompany N-(4-methyl)phenyl-O-(4-methoxy)phenyl-thionocarbamate (MMTC)-induced apoptosis based on a proteomic approach. Cell viability tests revealed that SK-MEL-28 skin cancer cells underwent more cell death than normal HaCaT cells in a dose-dependent manner after treatment with MMTC. Two-dimensional electrophoresis in conjunction with matrixassisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry analysis or computer matching with a protein database further revealed that the MMTC-induced apoptosis is accompanied by increased levels of caspase-1, checkpoint suppressor-1, caspase-4, NF-kB inhibitor, AP-2, c-Jun-N-terminal kinase, melanoma inhibitor, granzyme K, G1/S specific cyclin D3, cystein rich protein, Ras-related protein Rab-37 or Ras-related protein Rab-13, and reduced levels of EMS (oncogene), ATP synthase, tyrosine-phosphatase, Cdc25c, 14-3-3 protein or specific structure of nuclear receptor. The migration suppressing effect of MMTC on SK-MEL-28 cell was tested. MMTC suppressed the metastasis of SK-MEL-8 cells. It was also identified that MMTC had little angiogenic effect because it did not suppress the proliferation of HUVEC cell line. These results suggest that MMTC is a novel chemotherapeutic and metastatic agents against the SK-MEL-28 human melanoma cell line.
본 연구는 tiletamine/zolazepam 대조군과 대조군에 benzodiazepine의 길항제인 flumazenil을 투여한 실험군의 마취 효과와 혈액생화학, Vital sign, 마취회복시간에 대하여 비교하였다. 건강한 6마리의 개들(평균 체중 $5.2{\pm}2.2$)이 실험에 이용 되었다. Tiletamine/zolazepam 을 투여하였고 20분 뒤에 benzodiazepine의 길항제인 flumazenil 0.1 mg을 투여하였다. 마취효과 (Sedation, analgesia, muscle relaxation, posture and auditory response score), Vital sign (심박수, 호흡수, 체온), 혈액 생화학 (GLU, TP, ALT, AST) 검사들이 두 그룹에서 tiletamine/zolazepam 투여전, 투여후 1, 5, 20, 30, 60 그리고 90분에 실시되었다. 또한 마취회복시간 (head up, sternal recumbency, standing and walking times)은 부동화 상태에서 각 행동양식 이 보이는 시간까지 측정되었다. Analgesia score는 투여 후 20분과 30분에, posture score는 투여 후 30분에 그리고 auditory response score는 투여 후 1분과 20분에 TZF그룹이 TZ그룹보다 유의하게 낮아 benzodiazepine에 대한 flumazenil의 길항효과를 나타냈다. 평균 심박수는 두 그룹 모두 투여 후 1분부터 급상승하여 유의하게 정상 심박수 보다 높았다. 평균 호흡수는 TZF그룹이 TZ그룹보다 유의하지 않았으나 높은 호흡수를 보였다. 결론적으로 개에서 flumazenil의 투여는 tiletamine/zolazepam에 대한 길항작용을 나타내었다. 마취로 부터의 회복에 있어서, flumazenil은 sternal recumbency, standing 및 walking times를 단축시켰다.
이 연구의 목적은 개에 있어서 medetomidine에 대한 yohimbine의 길항효과를 알아 보는데 있다. 총 6 마리의 잡종견 (수: 4 마리, 암: 2 마리)을 본 연구에 2주일의 휴약기간을 두고 반복실험을 하였다. 개들의 평균 연령은 4.3세 (2-6세) 이였고, 평균체중은 3.8 kg (2.7-4.6 kg) 이였다. 그룹 M은 대조군으로서 medetomidine을 $40\;{\mu}g/kg$ 용량으로 근육 내 투여하고 15분 후에 saline을 0.2 ml/kg 용량을 정맥 내로 투여하였다. 그룹 MY는 실험 군으로서 medetomidine을 $40\;{\mu}g/kg$ 용량으로 근육 내 투여하고 15분 후에 yohimbine을 0.11 ml/kg 용량을 정맥 내로 투여하였다. 마취시간에 대한 평가로 induction time, sternal time, standing time과 walking time을 측정하였다. Vital signs으로서 심박동수, 호흡수와 직장체온을 측정하였으며, 혈액화학 치로서 glucose, total protein, ALT와 AST를 측정하였다. 실험결과, medetomidine 투여 후 서맥과 호흡저하를 보였으나, yohimbine 투여 후에는 심박수의 증가와 호흡수의 증가를 나타냄으로써 yohimbine이 medetomine으로 유발된 호흡저하와 서맥을 길항하는 효과를 나타내었다. Medetomine 투약 후 yohimbine을 투여한 군에서는 대조군에 비하여 흉와시간, 기립시간과 보행시간을 현저히 단축시키는 영향을 나타내었다. 결론적으로 개에서 yohimbine은 medetomidine에 대한 양호한 길항작용을 나타내었다.
Somatic cell nuclear transfer (SCNT) for miniature pig has been developed for xenotransplantation and many other biomedical experiments. However, the efficiency of SCNT is still very low due to many factors. To optimize the surrogate mother condition for improvement of cloned miniature pigs efficiency, we investigated the effect of the status of surrogate mother on pregnancy, farrowed rate in SCNT pigs. After SCNT with mesenchymal stem cells as donor cells, the SCNT embryos were surgically transferred into the oviduct of surrogated pigs. To compare the effects of status of surrogate pigs on pregnancy, surrogate pigs were prepared by artificial abortion at day 20~29 (Group 1), 30~39 (Group 2), and 40~45 (Group 3) of gestation. After SCNT embryos transfer in three different status of surrogate pigs, Group 2 (56.3%) and 3 (55.6%) had significantly ($p$ <0.05) higher the pregnancy rate than group 1 (0%) at day 30 of gestation. The status of ovulation in surrogate pig also was investigated. Post-ovulation status (54.8%) had higher proportion than pre-ovulation status (38.7%) and ovulation status (6.5%). We obtained 19 cloned miniature piglets from seven surrogate gilts and five piglets are living healthy but fourteen piglets died soon after birth or stillbirth. The weights of piglets greatly differ from 254 to 1,296 g. Microsatellite analysis showed that cloned piglets were genetically different from the surrogate mother and cloned piglets were genetically equal to the donor cell. In conclusion, the present result indicates that artificially abortion method can improve the efficiency of pregnancy after SCNT in pigs. This study will provide available method for the further study and application in the field of xenotransplantation.
