Testicular fat deposition attenuates reproductive performance via decreased follicle-stimulating hormone level and sperm meiosis and testosterone synthesis in mouse

  • Objective: Testicular fat deposition has been reported to affect animal reproduction. However, the underlying mechanism remains poorly understood. The present study explored whether sperm meiosis and testosterone synthesis contribute to mouse testicular fat deposition-induced reproductive performance. Methods: High fat diet (HFD)-induced obesity CD1 mice (DIO) were used as a testicular fat deposition model. The serum hormone test was performed by agent kit. The quality of sperm was assessed using a Sperm Class Analyzer. Testicular tissue morphology was analyzed by histochemical methods. The expression of spermatocyte marker molecules was monitored by an immuno-fluorescence microscope during meiosis. Analysis of the synthesis of testosterone was performed by real-time polymerase chain reaction and reagent kit. Results: It was found that there was a significant increase in body weight among DIO mice, however, the food intake showed no difference compared to control mice fed a normal diet (CTR). The number of offspring in DIO mice decreased, but there was no significant difference from the CTR group. The levels of follicle-stimulating hormone were lower in DIO mice and their luteinizing hormone levels were similar. The results showed a remarkable decrease in sperm density and motility among DIO mice. We also found that fat accumulation affected the meiosis process, mainly reflected in the cross-exchange of homologous chromosomes. In addition, overweight increased fat deposition in the testis and reduced the expression of testosterone synthesis-related enzymes, thereby affecting the synthesis and secretion of testosterone by testicular Leydig cells. Conclusion: Fat accumulation in the testes causes testicular cell dysfunction, which affects testosterone hormone synthesis and ultimately affects sperm formation.

Effects of Estrogen on the Transcriptional Activities of Catecholamine Biosynthesizing Enzymes in the Brain and Adrenal Gland of Ovariectomized Rats (난소 절제 흰쥐의 뇌와 부신에서의 Catecholamine Biosynthesizing Enzyme들의 전사에 미치는 Estrogen의 효과)

  • Dopamine(DA), norepinephrine(NE), and epinephrine(E) belong to a class of neurotransmitters known as catecholamine (CA) which are synthesized and secreted by mammalian brain and adrenal medulla. CA regulate several behavior patterns connected with breeding, and regulate GnRH-gonadotropin hormone axis' vitality between hypothalamus-pituitary gland linking with reproduction freeze. The present study examined effects of sex steroid hormone on the transcriptional activities of CA biosynthesis enzymes, tyrosine hydroxylase(TH), dopamine $\beta$ -hydroxylase(DBH), and phenylethaolamine-N-methyl transferase(PNMT). Mature female rats were ovariectomized(OVX) and implanted with 17 $\beta$-estradiol(E$_2$: 500 $\mu\textrm{g}$/ml) or sesame oil. Forty-eight hours after implantation all the animals were sacrificed. Total RNAs were extracted immediately and were applied to semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR). The expression level of TH was appeared by hypothalamus > SNc> adrenal medulla orders in OVX+Oil group, and by SNc > hypothalamus) adrenal medulla orders in OVX+E$_2$ group. Treatment with E$_2$ significantly increased TH expression in SNc and adrenal medulla but in hypothalamus, the reduced TH expression was observed. The expression level of DBH was appeared by adrenal medulla > SNc > hypothalamus orders in OVX+Oil group and in OVX+E$_2$ group. Administration of E$_2$ significantly reduced DBH expression in SNc, and increased in adrenal medulla. Two cDNA products, large(PNMT1) and small(PMNTs) species of 110bp difference, were amplified in SNc and hypothalamus, but only PNMTs was observed in adenal medulla. The PNMTs expression level was in the order of adrenal medulla > hypothalamus > SNc in both OVX+Oil and OVX+E$_2$ group. The PNMTs expression in SNc and adrenal medulla was significantly increased byE$_2$. The present report demonstrated that estrogen effects on transcriptional activities for CA biosynthethic enzymes were tissue specific in adrenal medulla as well as different region of brain. These results suggest that it might be crucial relationship between the type of estrogen receptor and CA enzyme gene expression.

Analysis of the Relationship between MHC-DRB1 Gene Polymorphism and Hydatidosis in Kazakh Sheep

  • The objective of this work was to analyze the relationship between ovine major histocompatibility complex (MHC) DRB1 gene polymorphism and genetic resistance to hydatidosis in Kazakh sheep. The Ovar (ovine MHC) class II DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of 702 Kazakh sheep, including 302 sheep with hydatidosis and 400 health controls. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique using five restriction enzymes, i.e., MvaI, HaeIII, SacI, SacII and Hin1I, yielding 14 alleles and 28 genotypes. Comparing the frequency of genotypes in hydatidosis sheep with the control group, it was found that the genotype frequencies of MvaIbc, Hin1Iab, SacIIab, HaeIIIde, HaeIIIdf and HaeIIIdd in control sheep were significantly (p<0.01) higher than in hydatidosis sheep, indicating that a significant correlation existed between these genotypes and resistance to hydatidosis. Genotype frequencies of MvaIbb, SacIIaa, Hin1Ibb and HaeIIIef in sheep with hydatidosis were extremely significantly (p<0.01) higher than in the control group, and the genotype frequency of HaeIIIab was significantly higher (p<0.05), indicating that a marked correlation existed between these genotypes and susceptibility to hydatidosis. By way of analyzing haplotype with these resistant genotypes, the hydatidosis resistant haplotype MvaIbc-SacIIab-Hin1Iab of Kazakh sheep was screened out, and then verified through artificial hydatid infection in sheep. The results indicated that the infection rate of sheep with the resistant haplotype of hydatidosis was significantly lower (p<0.01) than without this resistant haplotype. It showed that the genic haplotype MvaIbc-SacIIab-Hin1Iab of Ovar-DRB1 exon 2 was the resistant haplotype of hydatidosis in Kazakh sheep.

An Exploratory Study on Fitness Consumer: Focusing on Established and Outsider Relations of the Body among Fitness Members of Gangnam and Gangbuk (피트니스 소비자에 대한 탐색적 고찰: 강남과 강북 피트니스 회원 간 몸의 기득권자-아웃사이더 권력관계를 중심으로)

  • This article is a socio-cultural research on the fitness members by employing an original synthesis of the work of Elias and Bourdieu. The purpose of this research is to provide a multidimensional and in-depth analysis of fitness members, by researching the relationship between body groups with differing economic and cultural capital (e.g. Gangnam and Gangbuk, private and public center). Through interviews and participant observation, the established and outsider relations of fitness members in both Gangnam and Gangbuk are examined (Gangnam 12: Gangbuk 12). Participants in social space differentiated by fitness capital and socio-economic positions gather in certain spaces that identify them as members of the same class location: the established body in Gangnam (Gangnam E), the outsider body in Gangnam (Gangnam O), the established body in Gangbuk (Gangbuk E), and the outsider body in Gangbuk (Gangbuk O). The E-O figuration of the body in fitness clubs shows differences in their body tastes and habitus (selecting a fitness centre, body ostentation, social assessment, making muscles, participation in other sports) and civilizing process (fitness manners and etiquette). The fitness centers in Gangnam and Gangbuk were not simply spaces for exercise but symbolic spaces that both recreate and perpetuate socio-cultural hierarchies between members.

Embryogenesis in the Octopus minor (낙지(Octopus minor)의 배 발생)

  • The small octopus(Octopus minor), a species in the cephalopod class spawns the fewer eggs compared with its relatives such as Octopus vulgaris and Octopus ocellatus and exerts special efforts in nursing eggs hiding in the deep cave beneath the mud flat. Correspondingly, its embryogenic process had been hardly observed. In this study, we induced the small octopus to spawn its eggs in the artificial spawning tube and to nurse the developing embryos under the periodical observation. The early embryogenic change in the fertilized eggs was appearance of blastodisc on the animal pole at 4 days after spawning. A mass of rudiments of several organs developed in the middle of the yolk sac at 28-th day and it moved gradually toward the animal pole. At 38 days after spawning, two eyes appeared on the head and the trunk with heart developed on the apical part of the yolk sac. At 45-th day of embryonic development, the trunk and head occupied a half of the egg capsule and consequently the yolk sac was reduced to a half of its original size. Two eyes became prominent at the bottom of the trunk with several spots at day 60. Embryonic development was completed at about 80 days after spawning and the arms stuffed the lower half of egg capsule. The young fry squeezed out of the egg capsules with their long and slender arms first and mantles later. Soon after hatching, it swam actively and squirted ink.

Integrating Concept Mapping and the Learning Cycle to Teach Genetics and Reproduction to High School Students (고등학생들의 생물학습에서 개념도와 순환학습을 통합한 수업의 효과)

  • Although many studies have investigated the effectiveness of concept mapping and the learning cycle, in Korea none have explored the effectiveness of concept mapping and the learning cycle combined. This study explored the effectiveness of concept mapping, the learning cycle, and a combination of concept mapping/learning cycle(CL) in high school biology class. Students' science achievement, the science related attitudes and scientific inquiry ability was measured. The results indicated that concept mapping, the learning cycle, and CL treatment were significantly different from the traditional one in science achievement(p< .05). However, the three treatments were not significantly different from each other. No significant difference exists among different learnings in high and average-ability students. But, concept mapping was the most effective in low-ability students. For the students' scientific inquiry ability, CL and learning cycle were more effective than concept mapping and traditional learning. No significant difference exists among different learnings in high-ability students. CL and learning cycle were more effective than concept mapping and traditional learning in average and low-ability students. For the students' science related attitudes, concept mapping, the learning cycle, and CL were more effective than the traditional learning. But, there was no significant difference among these three groups.

Expression Analysis of Lily Type Lectin Isotypes in the Rock Bream, Oplegnathus fasciatus: in the Tissue, Developmental Stage and Viral Infection

  • Lectins belong to the pattern-recognition receptors (PRRs) class and play important roles in the recognition and elimination of pathogens via the innate immune system. Recently, it was reported that lily-type lectin-1 is involved when a pathogen attacks in the early immune response of fish. However, this study is limited to information that the lectin is involved in the innate immune response against viral infection. In the present study, the lily-type lectin-2 and -3 of Oplegnathus fasciatus (OfLTL-2 and 3) have been presented to be included B-lectin domain and two D-mannose binding sites in the amino acid sequence that an important feature for the fundamental structure. To investigate the functional properties of OfLTLs, the tissue distribution in the healthy rock bream and temporal expression during early developmental stage analysis are performed using quantitative real-time PCR. OfLTL-2 and 3 are predominantly expressed in the liver and skin, but rarely expressed in other organ. Also, the transcripts of OfLTLs are not expressed during the early developmental stage but its transcripts are increased after immune-related organs which are fully formed. In the challenge experiment with RBIV (rock bream iridovirus), the expression of OfLTLs was increased much more strongly in the late response than the early, unlike previously known. These results suggest that OfLTLs are specifically expressed in the immune-related tissues when those organs are fully formed and it can be inferred that the more intensively involved in the second half to the virus infection.

A Study on Improvement through Functionality Evaluation on Botanic Gardens and Arboreta in Korea (식물원·수목원 기능성 평가를 통한 개선방안 연구)

  • This study evaluated 23 botanic gardens and arboretums under two main categories including professional group and operational group. As for professional group, the main difference between the functions of botanic gardens and arboretums was analyzed. In the case of operational group compared professional group respectively, on categories including importance of function and implementation, seeking to provide fundamental data of botanic gardens and arboretums by enhancing the quality of visitors and examining functional differences. The result of a difference in arboretums' function between professional and operational groups, the study analyzed the importance of botanic gardens and arboretums in terms of detailed categories such as research, education, collection, display, retention, and service for visitors. As a result, professional group showed retention 4.32>education 4.11>research 4.09>service for visitors 4.05, and operational group showed retention 3.70>service 3.52>education 3.41>research 3.37, indicating that both groups had retention as the most selected answer. Looking closely at the functions of the currently implemented detailed categories of botanic gardens and arboretums, such as research, education, collection, display, retention, and service for visitors, the study showed that 91.3% selected rare species and individual multiplication for research function, 78.3% chose education of natural ecosystem for children for education, 73.9% answered space for experiencing nature for display, and 73.9% also replied managon, diof rare species, reproduction, and retention of native plants' species for retention category, and service for visitors showed 95.7% for implementation. The study could not clearly point out the influence between predictors due to absence of an established comprehensive evaluation model. Also, it analyzed and reviewed comparison of functions of professional group and operational group, but in the future, it should conduct study on a systemic and objective classification of botanic gardens and arboretums to clearly examine the difference in views between groups, which also requires, at the same time, study on management policy of botanic gardens and arboretums or suggesting guidelines that suit types of forms, and also study on facilities and program development that can be applied to each class.

The Development of 10 kW Class Tidal Power Generator System - Focusing on Field Experiments with Pipelines (10 kW급 조력발전장치 개발 - 관수로 현장실험을 중심으로)

  • Along with the growing interest in renewable energy development, Korea's west coast is one of the favorable regions for tidal power. Tidal power using tidal barrages that work like hydroelectric dams is a representative method of tidal power through long-term operation, but the promotion of tidal power projects is being delayed or stopped due to impacts on ecological changes, reproduction, water column processes and hydrology. In order to reduce the high construction cost and environmental cost problems caused by tidal power using tidal barrages, in this study, field experiments were conducted to develop and verify the performance of tidal power generation devices applicable to sea areas where dykes are already installed. As a result of conducting five cases of experiments using two water tanks, pipe lines, open channels, and water turbine and generator, the possibility of developing a power generation system capable of generating more than 10 kW output and more than 60% efficiency were confirmed. The results of this study can be used for small-scale tidal power by utilizing the existing dykes of the west coast.

Quantitative Expression Analysis of Functional Genes in Four Dog Breeds (개의 네 품종에서 기능 유전자들에 대한 정량적 발현 분석)

  • One of the domesticated species; the dog has been selectively bred for various aims by human. The dog has many breeds, which are artificially selected for specific behaviors and morphologies. Dogs contribute their life to human as working dogs for guide, rescue, detection or etc. Working dogs requires good personality, such as gentleness, robustness and patience for performing their special duty. Many studies have concentrated on finding genetic marker for selecting the high-quality working dog. In this study, we confirmed quantitative expression patterns of eight genes (ABAT; 4-Aminobutyrate Aminotransferase, PLCB1; Phospholipase C, Beta 1, SLC10A4; Solute Carrier Family 10, Member 4, WNT1; Wingless-Type MMTV Integration Site Family, Member 1, BARX2; BarH-Like Homeobox 2, NEUROD6; Neuronal Differentiation 6, SEPT9; Septin 9 and TBR1; T-Box, Brain, 1) among brains tissues from four dog breeds (Beagle, Sapsaree, Shepherd and Jindo), because these genes were expressed and have functions in brain mostly. Specially, BARX2, SEPT9, SLC10A4, TBR1 and WNT1 genes were highly expressed in Beagle and Jindo, and Sapsaree and German Shepherd were vice versa. The biological significance of total genes was estimated by database for annotation, visualization and integrated discovery (DAVID) to determine a different gene ontology (GO) class. In these analyses, we suppose to these eight genes could provide influential information for brain development, and intelligence of organisms. Taken together, these results could provide clues to discover biomarker related to functional traits in brain, and beneficial for selecting superior working dogs.
