  • Spatial variation of heavy metal accumulation was investigated in Manila clam Ruditapes philippinarum collected from several tidal flats. Sediment metal levels varied highly among the sites, which was attributed primarily to differences in Fe and organic carbon contents and in part to gain size. Significant differences in metal concentrations also were found in the clam tissue among the different sampling sites. However, except for a few metals (Mn, Zn, Pb), which showed some elevation, the variations in the clam tissue were not related to the variations in the sediment. This is likely because most metals in filter-feeding herbivores such as R. philippinarum accumulated as a result of feeding on suspended particles such as phytoplankton and organic detritus in the water column, not in bottom sediment. In addition, tissue weight for a specific shell size varied significantly among the sites, and increased tissue mass indicating a good nutritive condition likely caused a subsequent dilution of body metals leading to reduced weight-specific concentrations of some metals (Cd, Zn, Cu, Co).

  • This study screened the biological activity of an acidified gill extract of the Manila clam Ruditapes philippinarum including antimicrobial, hemolytic, membrane permeabilization, and DNA-binding activity, and purified the antimicrobial material. The acidified gill extract showed potent antimicrobial activity against Bacillus subtilis and Escherichia coli without significant hemolytic activity, but showed no membrane permeabilization or DNA-binding ability. An antimicrobial material was purified from the acidified gill extract using C18 reversed-phase and cation-exchange high-performance liquid chromatography (HPLC). Treatment of the purified material with trypsin completely abolished all of the antibacterial activity against Bacillus subtilis, suggesting that the purified material is a proteinaceous antibiotic. The molecular weight of the purified material was 2571.9 Da, but no primary structural information was obtained due to N-terminal blocking. A future study should confirm the primary structure. Our results suggest that the Manila clam gill contains proteinaceous antibiotics that have a role in first-line defense. This information could be used to better understand the Manila clam innate immune system.

  • Shell waste from the butter clam Saxidomus purpuratus and littleneck clam Ruditapes philippinarum is a large by-product of shellfish aquaculture, and it is desirable to convert it into value-added products for industrial applications. In this study, calcium carbonate (CaC) polymorphs from butter clam (BCSP) and littleneck clam (LCSP) shell powders and commercial CaC were characterized using Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The results revealed that the optimal calcination conditions to eliminate organic substances and improve solubility for both BCSP and LCSP were $800^{\circ}C$ for 8 h in an electrical furnace. Calcination improved the white index of the butter clam (BCCP) and littleneck clam (LCCP) calcined powder compared with shell powders. The calcium content in BCCP (51.1%) was higher than that of LCCP (44.9%) or commercial calcium oxide (CaO, 44.7%). The XRD patterns of BCCP and LCCP were similar to that of CaO. Cubic-like crystals of CaC and irregular crystals of BCCP and LCCP were observed by SEM. The FT-IR and XRD analyses revealed the presence of calcite and aragonite in the BCSP and aragonite in the LCSP, whereas the CaC contained calcite. These results indicate that butter and littleneck clam shells are potential biomass resources for calcium carbonate and calcium oxide.

  • Immmune parameters of the Manila clam Ruditapes philippinarum collected from four tidal flats, Nudong, Gonam, Hwangdo and Bangpo on Anmyeon-do, Korea were optimized and evaluated at the single cell level using flow-cytometry Hemocytes were withdrawn from the sinus of each clam, and total hemocyte counts (THC), phagocytosis rate, hemocyte mortality (HM) and DNA damage of hemocyte were analyzed. The highest hemocyte counts was recorded from the clams collected from Gonam, followed by Hwangdo, Nudong and Bangpo (P<0.001). Phagocytosis rate and hemocyte mortality of Gonam and Nudong clams were significantly higher than those of clams from Hwangdo and Bangpo (P<0.001). DNA damage in the clams from Nudong was higher twice than that of clams from Gonam (P<0.05). We suggest that the flow-cytometry has a high potential for evaluation of immunity of marine bivalves.

  • Growth patterns of the Manila clam, Ruditapes philippinarum in each tidal level were studied in the intertidal zone in Tae-an on the west coast of South Korea from April to August 2009. Environmental factors and the specimens of R. philippinarum were monthly collected at three intertidal locations: low intertidal zone, low-middle intertidal zone and middle intertidal zone. Temperature of sediment in low intertidal zone were lower than those in other intertidal locations. Concentrations of Chl-${\alpha}$ were not significantly different between in each tidal level. Exposure hours in low intertidal zone were shorter than those in other intertidal locations. Density of each year class in low intertidal zone were higher than those in low-middle intertidal zone and middle intertidal zone. Growth factors of R. philippinarum in low intertidal zone were better than those in other intertidal locations. The growth rate of under one year class individuals of low intertidal zone were higher than those in low-middle intertidal zone and middle intertidal zone, before the main recruitment period of R. pilippinarum. Although the growth patterns of R. philippinarum may be affected by various environmental factors, we suggests that the exposure duration is the main affect in the growth patterns of R. philippinarum in intertidal zone.

  • Gonad maturation of Manila clam, Ruditapes philippinarum was induced in this study using a recirculation system over 8 weeks in early spring. Clams used in the experiment were collected in $15^{th}$ April 2010 from the west coast of Korea, as the surface water temperature remained $11^{\circ}C$. To induce gametogenesis and subsequent maturation seawater temperature was elevated $1^{\circ}C$ per day over 10 days to reach $20^{\circ}C$. For the experiment, clams were raised in 120 L quadrangle tank maintained with re-circulated seawater system over 57 days. Water quality parameters including the water temperature, salinity dissolved oxygen, ammonium ion and nitrate levels in the tanks were monitored daily. Mixture of concentrated microalgae including Tetraselmis, Isochrysis, Pavlova and Thalassiosira weissflogii was supplied to clams twice a day, and quantity of the daily ration was adjusted as 3% of clam body dry weight. Histology was applied to examine gonad maturation. Daily monitoring of the water quality parameters indicated that the recirculation system supplied suitable environment to Manila clam; the nitrogenous components stayed below toxic levels (< 0.2 mg/L). At the beginning of the study, clams were mostly in early developing stage. As the seawater temperature reached $20^{\circ}C$, 10 days after the experiment, 20% of clams reached late development at 12 days. First ripe clams were observed at 42 days and 40% of clams were in ripe and ready for spawning at the end of study, 57 days after the experiment. In this study, gametogenesis of Manila clam was successfully induced by elevating water temperature and supplying commercially produced microalgae in a recirculation tank system.

  • Oxygen consumption by the Manila clam, Ruditapes philippinarum (Bivalvia: Veneridae) maintained in a culture tank for 7-10 weeks was measured with an automatic intermittent-flow-respirometer (AIFR). There was evidence for an endogenous tidal rhythm of 12.4 h in oxygen consumption, at irregular intervals of between about 7 and 18 hours during experiments of between 22 h 5 min and 70 h 32 min (duration) recording for fasting animals in constant darkness at constant temperatures in the absence of tidal rhythms.

완도 영흥지선 연안의 천연 바지락 자원에 대한 연구 (Study on natural short-necked clam, Ruditapes philippinarum, stocks in Yeongheung coast of Wando Island, Korea)

  • In order to understand the effect of introduction of oyster rack culture on natural short-necked clam, Ruditapes philippinarum, stock, we investigated the status of natural clam stock prior to introduction of oyster rack culture at Yeongheung Coast of Wando Island. The growth equation of the clam was estimated as: $L_t=61.46{\cdot}(1-e^{-0.172(t+0.155)})$ from ring radius composition of shell. Instantaneous coefficient of total mortality and natural mortality were calculated as: 2.4087/year and 0.478/year, respectively. The age at first capture was estimated to be 2.55 year. The total biomass was estimated to 3.23 ton in the bed (0.8 ha). Applied by these parameters, the annual recruit biomass and the current yield per recruit (Y/R) was corresponded to $114.7individuals/m^2$ and $92.0g/m^2$, respectively. Maximum sustainable yield (MSY) was estimated 0.77 ton/year which was close to annual catches, 0.74 ton/year, in the area.

Temporal Variation in the Reproductive Effort and Tissue Biochemical Composition in Manila Clam, Ruditapes philippinarum from a Sand Flat on the East Coast of Jeju Island Korea

  • In the present study, we investigated temporal variation in the reproductive effort and biochemical contents in adult Manila clam Ruditapes philippinarum at Shi-Heung-Ri beach on the east coast of Jeju Island. Gonad-somatic index (GSI), a ratio of the egg mass to the total biomass determined using ELISA increased dramatically from late May to June (7.1% to 19.5%). In mid-July, GSI dropped to 15.1%, indicating spawning on a small scale during this period. GSI also declined dramatically from late August (15.5%) to September (4.3%), suggesting that massive spawning occurred during this period. A positive correlation was observed between clam size and potential fecundity, while potential fecundity ranged between 1.19-8.40 million eggs/clam. Total proteins and lipids in the tissue increased from late spring to mid-summer, coinciding with an increase in GSI. Protein, lipid and carbohydrate contents in the tissue were lowest in January, indicating that clams were suffering from poor nutritional circumstances, which may be associated with poor food supply from the environment. Monthly increase and/or decrease in the body weight were closely related to gonad maturation and subsequent spawning, which was also linked to temporal changes in the contents of protein, lipid and carbohydrates in the tissue.
