전통간장으로부터 분리한 Cladosporium sp.과 Sterigmatomyces sp. 미생물의 동정 및 NaCl 농도에 따른 미생물 효소활성 분석 (Identification of Microorganisms, Cladosporium sp. and Sterigmatomyces sp., Proliferated on the Surface of Traditional Soy Sauce, and the Effect of NaCl Concentration on Their Enzymatic Activity)

  • 전통방식으로 숙성되고 있는 한식간장의 표면으로부터 분리된 바위꽃과 메밀꽃이라 불리는 미생물을 18S rDNA ITS1과 ITS4 지역의 염기서열를 분석한 결과, 각각 Cladosporium 종들과 S. halophilus의 염기서열과 높은 상동성을 보였다. 이에 따라 바위꽃 미생물은 Cladosporium sp. NK1로, 메밀꽃 미생물은 S. halophilus NK2로 명명하였다. YPD 액체 배지를 이용한 최적 생장 pH와 온도에 있어서 Cladosporium sp. NK1은 각각 pH 6과 22-$27^{\circ}C$였고, S. halophilus NK2는 각각 pH 5-7과 $22^{\circ}C$였다. Cladosporium sp. NK1는 NaCl를 첨가하지 않은 YPD 액체배지에서 가장 잘 생장하였으나, protease 및 amylase 효소활성에 있어서는 각각 10%와 15% NaCl이 첨가된 YPD 배양액 시료에서 가장 높았다. S. halophilus NK2는 10% NaCl이 첨가된 YPD 액체배지까지 매우 잘 생장하였으며, protease 활성은 10% NaCl이 첨가된 YPD 배양액 시료에서, amylase 활성은 5% NaCl이 첨가된 배양액 시료에서 가장 높았다. Cladosporium sp. NK1과 S. halophilus NK2의 lipase 활성은 보이지 않았다. 간장의 관능평가 결과 Cladosporium sp. NK1 균주는 간장의 맛에서, S. halophilus NK2는 향미에서 유의적 차이를 보였다.

  • 산마늘에서 분리한 Cladosporium sp.는 Allium속(屬) 식물에서 보고된 C. allii나 C. allii-cepae와는 연쇄상의 다격벽(多隔壁) 분생포자(分生胞子)와 무격벽(無隔壁) 분생자경(分生子梗)을 형성하는 점에서 구별된다. 이 균은 기타 다른 기질(基質)에서 보고된 Cladosporium spp.와 비슷한 점이 없다. 따라서 이 균은 신종(新種)으로 판단되며, Cladosporium alliicola H.D. Shin & U. Braun으로 명명(命名)한다.

Cladosporium sp. is the Major Causal Agent in the Microbial Complex Associated with the Skin Sooty Dapple Disease of the Asian Pear in Korea

  • Skin sooty dapple disease, a fungal disease that lowers Asian pear fruit quality, has emerged recently in Korea but has not yet been thoroughly characterized. This disease affects the surface of fruit, leaves, and young shoots of the Asian pear, typically appearing as a dark or pale black dapple on the fruit surface. The disease initiates on the fruit with small circular lesions that become bigger, eventually spreading to form large circular or indefinite lesions. Sparse dark or flourishing white-greyish aerial mycelia and appearance of a dark or pale black dapple on the fruit surface are typical signs of this disease. The disease was severe during cold storage of the Niitaka and Chuhwangbae varieties, but more limited on the Gamcheonbae and Hwangkeumbae varieties. To identify causal pathogens, 123 fungal isolates were obtained from lesions. The fungi that caused typical skin sooty dapple disease symptoms in our bioassay were identified. Based on their morphological characteristics, 74% of the isolates were Cladosporium sp. and 5-7 % of the isolates were Leptosphaerulina sp., Tripospermum sp., or Tilletiopsis sp. None of the isolates caused severe soft rot by injection to a wound plug, but some of the Cladosporium sp. isolates caused mild maceration. Therefore this microbiol complex cannot account for the soft rot also observed in stored fruits. The high frequency of isolation of Cladosporium sp. from disease tissues and bioassay on pear fruit surface suggest that Cladosporium sp. could be a major pathogen in the microbial complex associated with skin sooty dapple disease of the Asian pear in Korea.

건물(建物) 환경(環境)의 진균(眞菌) 분포(分布) 조사(調査) (A Mycological Survey in Indoor Environments)

  • 서울에서 1983년 12월 26일부터 1984년 2월 11일 까지 각종 건물실내환경에서 채취한 진균을 배양분리한 결과에서 다음과 같은 결론을 얻을 수 있었다. 총 26종류의 진균이 검출되었다. 병원에서는 14종류의 진균이 검출되었는데, 이중에서 Cladosporium sp.와 Penicillium sp.가 주종을 이루며 Alternaria sp., Chaetomium sp. 그리고 효모등이 비교적 많이 검출되었다. 일반주택에서는 22종류의 진균이 검출되었으며, 여기서는 Penicillium sp.가 상당분을 차지하고, Alternaria sp., Aspergillus sp., Cladosporium sp., Rhizopus sp., Trichoderma sp.가 주종을 이룬다. 아파트에서의 진균종류도 대체로 주택과 같았다. 이 조사에서는 특별히 진균의 정량적인 분석이 이루어지지 않았으므로 확실한 위해도의 여부는 가리기 힘들다. 따라서 이들 진균을 종까지 분류하고 각각에 대한 정량분석을 하여 공중위생적인 위해도와의 관계를 추정하는 조사가 앞으로 요망된다. 이상에서 진균성 식중독을 유발하는 균의 분포가 보편적으로 인정되므로, 분포가 넓은 여러 종의 진균에 대해서는 실내 환경내에서 오염 또는 증식되지 않도록 하는 대책이 필요하다. 또한 많은 사람이 모이는 공중 장소에 대해서는 검출되는 진균의 종류에 대하여서는 적절한 위생적인 규제가 있어야 하겠다.

  • The Investigation of organisms damaged to papers and cellulose materials of Cultural Property in the Ch'ang Dok Palace The investigation of the airborne fungi, the attached fungi to the papers and cellulose materials of Cultural Properties in addition to the insects inhabiting at the Ku Sonwon Jon, Shin Sonwon Jon and Yonwa ch'anggo in the Ch'ang Dok Palace carried out from Jul. 10 to Jul. 21,1984.The results are summarized as follows ;1. Isolation and identification of the airborne fungi from the three storages were Cladosporium sp., Alternaria alternata, As pergillus cervinus, A. flavus, A. nidulance, A. oryzae, A. terreus, A. versicolor, A. wentii, Penicillium adametzii, P.albicans, P.canescens, P. citreo-viride, P. citrinums, P. decumbens, P. frequentans, P. funiculosum, P.herquei, P.implicatum, P.multicolor, P.nigricans, P.nonatum,P.purpurogenum, P.roqueforti, P.viridicatum, Trichodema viride, Geotrichumcandidum, Curvvlaria lunata, Torula hebarum, T.thermophila, Itersoniliasalmonicolor, Drechsclera avenue, Candida sp., Acremonium sp., and Botrytis sp., It was found that thirty five species in thirteen genera was isolated. Among them, the dominant species was Cladosporium sp., and the order was Penicillium, Aspergillus, Alternaria and so on.2. The attached fungi directly isolated from the papers and cellulose materials of Cultural Properties were twenty-nine species in fourteen genera, namely, Acremonium sp., Albertiniella sp., Alternaria alternata, Aspergillus clavatus,A.niger, A.ornatus, A.versicolor, Botrytis sp., Bysochlamys sp., Carpenteles sp.,Chaetomium globosum, Cladosporium sp., Eurotium sp., Mucor sp., Penicilliumcanescens, P.chermesium, P.citrinum, P.frequentans, P.funiculosum, P.herqueiP .implicatum, P.javanicum, P.luteum, P.purpurogenum, P.thomii, P.viridicatum, Torula thermophila, Trichoderma koningi and T.viride. Among them, the mostfungi distributed on the surface of the papers and cellulose materials was Penicillium and the order was Aspergillus, Alternaria, Cladosporium, Trichodermaand so on.3. The insects collected the three storages were ten genera and ten species including 916 specimens. By classifying the insects collected, the most species of the insects was Stenoscelodes hayashii of 857 specimens occupied about 93% of the total insect. And the other insects were collected as Microgamme costipennisAnobium pertinax, Xenomimetes alni, Anthrenus verbasci, Holoparmecus signatus,Thermobia domestica, Halyomorpha brevis, Drosophila coracine and Brattaorientalis. As described above, it could be known that the most airborne fungi was Cladosporium and the order was Penicillium, Aspergillus, Alternaria in the three storages. And the most attached fungi distributed on the surface of papers and cellulose materials was Penicillium and the other fungi were Aspergillus, Alternaria Cladosporium, Trichoderma and so on. Accordingly, from the results, itwas assumed that the major part of airborne fungi were attached to the papers and cellulose materials of Cultural Properties. The paper and cellulose materials of Cultural Properties in Ch'ang Dok Palace were chiefly damaged by S.hayashii in Coleoptera.

곰솔(Pinus thunbergii)의 침엽에서 분리한 내생균의 분자적 동정 (Molecular Identification of Endophytic Fungi Isolated from Needle Leaves of Pinus thunbergii)

  • 곰솔(Pinus thunbergii Parl.)의 잎에 서식하는 내생균의 다양성을 확인하기 위하여 충남 보령의 해안가에 서식하는 곰솔에서 잎을 채취하여 총 19개의 균주를 분리하였다. 분리된 내생균을 ITS 지역과 LSU 지역의 염기서열을 분석하여 동정한 결과 Anthostomella sepelibilis, Beltrania sp., Cladosporium sp., Coniochaeta velutina, Entonaema pallida, Lophodermium sp., Nigrospora sphaerica 등 총 7개 분류군의 내생균을 확보할 수 있었다. 이들 중 Beltrania sp.에 속하는 종이 가장 높은 빈도로 분리되었으며 내생균의 서식 환경이 곰솔의 잎에 서식하는 내생균의 분포에 영향을 미칠 수 있음을 확인하였다.

The Mycobiota of Air Inside and Outside the Meju Fermentation Room and the Origin of Meju Fungi

  • The fungi on Meju are known to play an important role as degrader of macromolecule of soybeans. In order to elucidate the origin of fungi on traditional Meju, mycobiota of the air both inside and outside traditional Meju fermentation rooms was examined. From 11 samples of air collected from inside and outside of 7 Meju fermentation rooms, 37 genera and 90 species of fungi were identified. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp., Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, Asp. nidulans, Aspergillus sp., Cla. cladosporioides, Eurotium sp., Penicillium sp., Cla. tenuissimum, Asp. niger, Eur. herbariorum, Asp. sydowii, and Eur. repens were collected with high frequency. The concentrations of the genera Aspergillus, Eurotium, and Penicillium were significantly higher in inside air than outside air. From this result and those of previous reports, the origin of fungi present on Meju was inferred. Of the dominant fungal species present on Meju, Lichtheimia ramosa, Mucor circinelloides, Mucor racemosus, and Scopulariopsis brevicaulis are thought to be originated from outside air, because these species are not or are rarely isolated from rice straw and soybean; however, they were detected outside air of fermentation room and are species commonly found in indoor environments. However, Asp. oryzae, Pen. polonicum, Eur. repens, Pen. solitum, and Eur. chevalieri, which are frequently found on Meju, are common in rice straw and could be transferred from rice straw to Meju. The fungi grow and produce abundant spores during Meju fermentation, and after the spores accumulate in the air of fermentation room, they could influence mycobiota of Meju fermentation in the following year. This could explain why concentrations of the genera Aspergillus, Eurotium, and Penicillium are much higher inside than outside of the fermentation rooms.

Four Endophytic Ascomycetes New to Korea: Cladosporium anthropophilum, C. pseudocladosporioides, Daldinia eschscholtzii, and Nigrospora chinensis

  • Ascomycota is the largest phylum of the Fungi, including approximately 6,600 genera. They are often isolated from soils, indoor air, and freshwater environments, but also from plants as pathogens or endophytes. In this study, four species of Ascomycota (two of Cladosporium and one of each Daldinia and Nigrospora) were collected from the leaves of four woody plants (Camellia japonica, Ginkgo biloba, Quercus sp., Vitis vinifera). Their cultural characteristics were investigated on five different media (PDA, V8A, CMA, MEA, CZA) at 3 days after incubation at $25^{\circ}C$ in darkness. BLASTn search and phylogenetic analysis were performed using the internal transcribed spacer (ITS) rDNA sequences, in addition to tef1 gene sequences for Cladosporium species. Based on the cultural, morphological, and phylogenetic data, the isolates were identified as Cladosporium anthropophilum, Cladosporium pseudocladosporioides, Daldinia eschscholtzii, and Nigrospora chinensis. Previously, some members of Cladosporium and Nigrospora have been recorded as endophytes inhabiting the leaves and stems of various plants, whereas Daldinia eschscholtzii is a wood-inhabiting endophyte or wood-decaying fungus. To our knowledge, this is the first report of these four ascomycetes in Korea.

The Origin of Meju Fungi - Fungal Diversity of Soybean, Rice Straw and Air for Meju Fermentation

  • Meju is a brick of dried fermented soybeans and is the core material for Jang such as Doenjang and Ganjang. Jang is produced by addition of salty water to Meju and is considered the essential sauces of authentic Korean cuisine. Meju is fermented by diverse microorganisms such as bacteria, fungi and yeasts. It is known that fungi play an important role in the Meju fermentation and they degrade macromolecules of the soybeans into small nutrient molecules. In previous study, 26 genera and 0 species were reported as Meju fungi. However, it is not comprehensively examined where the fungi present on the Meju are originated. In order to elucidate the origin of the fungi present on the Meju, the mycobiota of 500 samples soybean kernels, 296 rice straw pieces and air samples of Jang factories was determined in 0, 2 and 7 Jang factories respectively. Forty-one genera covering 86 species were isolated from the soybeans and 33 species were identical with the species from Meju. From sodium hypochlorite untreated soybeans, Eurotium herbariorum, Eurotium repens, Cladosporium tenuissimum, Fusarium fujikuroi, Aspergillus oryzae/flavus and Penicillium steckii were the predominant species. In case of sodium hypochlorite-treated soybeans, Eurotium herbariorum, E. repens and Cladosporium tenuissimum were the predominant species. Of the 4 genera and 86 species isolated from soybeans, 3 genera and 33 species were also found in Meju. Thirty-nine genera and 92 species were isolated from the rice straws and 40 species were identical with the species from Meju. Fusarium asiaticum, Cladosporium cladosporioides, Aspergillus tubingensis, A. oryzae, E. repens and Eurotium chevalieri were frequently isolated from the rice straw obtained from many factories. Twelve genera and 40 species of fungi that were isolated in the rice straw in this study, were also isolated from Meju. Especially, A. oryzae, C. cladosporioides, E. chevalieri, E. repens, F. asiaticum and Penicillium polonicum that are abundant species in Meju, were also isolated frequently from rice straw. C. cladosporioides, F. asiaticum and P. polonicum that are abundant in low temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $5^{\circ}C$ and $25^{\circ}C$, while A. oryzae, E. repens and E. chevalieri that are abundant in high temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $25^{\circ}C$ and $35^{\circ}C$. This suggests that the mycobiota of rice straw have a large influence in mycobiota of Meju. Thirty-nine genera and 92 species were isolated from the air of Jang factories and 34 species were identical with the species from Meju. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp. Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, A. nidulans, Aspergillus sp., C. cladosporioides, Eurotium sp., Penicillium sp., C. tenuissimum, A. niger, E. herbariorum, A. sydowii, and E. repens were collected with high frequency. 콩 종자(種子)에서 분리(分離)한 사상균(絲狀菌), 그 병원성(病原性) 및 종자(種子) 소독(消毒)에 관한 연구(硏究) (Fungi Associated with Soybean Seed, their Pathogenicity and Seed Treatment)

  • 콩 종자(種子) 14개(個) 시료(試料)를 Blotter법(法)으로 조사(調査)한 결과(結果) 부생성(腐生性) 사상균(絲狀菌)은 Alternaria tenuis, Arthrobotrytis sp., Aspergillus spp., Cephalosporium sp., Cladosporium sp., Cylindrocarpon sp., Fusarium equiseti, F. moniliforme, F. semitectum, F. solani, Pennicillium spp. 및 Rhizopus sp. 등이 검출(檢出)되었고, 병원성사상균(病原性絲狀菌)은 Cercospora kikuchii, Colltotrichum truncatum, Diaporthe phaseolorum sojae 및 Fusarium oxysporum 등이 검출(檢出)되었다. C. kikuchii, C. truncatum, D. phaseolorum sojae 및. F. oxysporum에 의해서 자연상태(自然狀態)에서 이병(罹病)된 종자(種子)로부터 유묘(幼苗)의 병징발현(病徵發現)이 어떻게 일어나는가를 조사(調査) 논의(論議)하였다. C. truncatum, D. phaseolorum sojae 및 F. oxysporum에 의해서 병든 종자(種子)는 발아율(發芽率)이 유묘생육(幼苗生育)에 영향을 미쳤다. 병원성(病原性) 사상균(絲狀菌)의 인공접종(入工接種) 결과(結果) 모두 병원성(病原性)이 나타났으나 F. oxysporum은 약하게 나타났다. C. kikuchii, C. truncatum, D. phaseolorum sojae 및 F. oxysporum에 대한 효과적(效果約)인 종자소독제(種子消毒劑)는 Benlate-T, Homai, Tecto 및 Sisthane이었으며, Arasan, Captan, Busan-30 및 Mercron은 C. kikuchii에는 효과(效果)가 낮았으나 기타(其他) 병원균(病原菌)에는 효과(效果)가 있었다. 본(本) 실험(實驗)에 공시(供試)된 약제(藥劑)는 병든 종자(種子)의 발아율(發芽率)을 증진(增進)시키는 효과(效果)가 있었다.
