본 연구는 전통 식재료로 많이 사용되는 10종의 약용식물 및 채소류 추출물과 10종 혼합추출물의 항산화 효과를 살펴보기 위해서 디자인되었다. 세포 생존률은 MTS assay로, 항산화효과는 DPPH free radical 소거능, Riboflavin 유래 Superoxide 억제활성(SQA)에 의한 항산화활성을 측정하였다. 그 결과 세포생존률은 10종 약용식물 및 채소류 추출물과 10종 혼합추출물 모두 0.5mg/mL에서 대조군과 유사한 생존율을 보였다. DPPH free radical 소거능은 Cinnamomum cassia Blume, Eugenia caryophyllata Thunb. Arctium lappa, Total extract가 우수하였고, Riboflavin 유래 Superoxide 억제활성(SQA)은 Cinnamomum cassia Blume, Arctium lappa, Prunus mume Sieb. et Zucc., 우수하였으나, Total extract가 가장 우수한 항산화 효과를 보였다. 전통식재료로 사용되는 약용식물 및 채소 추출물과 혼합추출물의 항산화 효과를 비교한 결과, 단독으로 사용하는 것보다 혼합물로 사용시 항산화능이 증가되었다. 이는 항산화 기능성 소재로 활용이 가능한 것으로 사료되며, 향후 항산화 소재 개발 시 가치가 있을 것으로 사료된다.
약용식물의 열수 추출물이 in vitro에서 alcohol dehydrogenase (ADH)와 aldehyde dehydrogenase (ALDH)의 활성에 미치는 영향을 확인하였다. 약용식물에 20배의 증류수를 넣고 $80^{\circ}C$에서 8시간 추출하여 얻어진 추출액을 시료로 사용하였다. 50종의 약용식물 중에서 마늘과 육계 추출물이 숙취해소 천연소재로서의 활용 가능성이 가장 높게 나타났다. 마늘 추출물은 ADH에 비해 ALDH의 활성을 2배 이상 촉진시킴으로써 acetaldehyde의 분해가 잘 되게 하였다. 육계 추출물은 ALDH의 활성에 비해 ADH의 활성을 획기적으로 저해함으로써 acetaldehyde의 생성을 크게 억제하였다. 육계 추출물은 농도에 비례하여 ADH와 ALDH의 활성을 저해하였으며, $45.33{\mu}g/mL$의 농도에서 ADH의 활성을 52.8% 저해하였고 ALDH의 활성을 11.0% 저해하였다.
Antioxidative and antimicrobial activities of essential oils of cassia (Cinnamomum cassia) and dill (Anethum graveolens L.) were investigated. Essential oils used in this study were added to soybean oil and stored at $65^{\circ}C$ for 9 days to examine their antioxidant activities using peroxide value (POV). The results showed that dill essential oil possessed a higher antioxidant activity than cassia essential oil. Strong antimicrobial activity was observed in cassia essential oil, whereas low activity was observed in dill essential oil against the test microorganisms. Heat stability and cell growth inhibitions were investigated with different concentrations of cassia oil. Results showed that cassia oil had thermal stability in a wide range of $70-160^{\circ}C$. Cassia inhibited cell growth of Bacillus cereus KCTC 1022, Micrococcus luteus A TCC 9341 and Escherichia coli ATCC 25922, but not great on Salmonella typhimurium ATCC 14028 at level of 200 ppm. In conclusion, the results indicate that dill essential oil could be a potential candidate for an antioxidative agent, while cassia essential oil could be suitable for use as an antimicrobial agent in the food industry.
CC-MS analysis on the essential oil (CC-oil) of Cinnamomum cassia stem bark led to the identification of cinnamaldehyde (CNA, 1), 2-hydroxycinnamaldehyde (2-CNA), coumarin (2), and cinnamyl acetate. The major volatile flavor in CC-oil was found to be 2-CNA. Coumarin was first isolated from this plant by photochemical isolation and spectroscopic analysis. CNA and CC-oil showed potent cytotoxicity, which was effectively prevented by N-acetyl-L-cysteine (NAC) treatment. Intraperitoneal administration with CNA considerably decreased malondialdehyde (MDA) formation and glutathione S-transferase activity in rats. These results suggest that CC-oil and CNA can regulate the triggering of hepatic drugmetabolizing enzymes by the formation of a glutathione-conjugate.
This study was undertaken to test for anti-Hepatitis B virus (HBV) activity of the aqueous extracts prepared from Eugenia caryophyllate, Ephedra sinica, Cinnamomum cassia. Aqueous extracts were assayed for the inhibition of HBV replication by measurement of HBV DNA and surface antigen (HBsAg) levels in the extracellular medium of HePG2 2.2.15 cells. All extracts decreased the levels of extracellular HBV virion DNA at concentrations ranging from 128 to 256 $\mu$g/ml and inhibited the production of HBsAg dose-dependently. Our findings suggest that these three hebal medicinal plants may have potential to develop as specific anti-HBV drugs in the future.
The anticoagulant properties of Cinnamomum cassia bark-derived materials were evaluated against platelet aggregation induced by arachidonic acid (AA), collagen, platelet activating factor (PAF), or thrombin, and these effects were then compared to those of three commercially available compounds (cinnamic acid, cinnamyl alcohol, and aspirin). The active constituent obtained from C. cassia barks was isolated by silica gel column chromatography and high pressure liquid chromatography (HPLC), and was characterized as trans-cinnamaldehyde by MS, $^1H-NMR$, $^{13}C-NMR$, and IR spectroscopy. With regard to 50% inhibitory concentration ($IC_{50}$) values, cinnamaldehyde was found to effectively inhibit platelet aggregation induced by AA ($IC_{50},\;43.2\;{\mu}M$) and collagen ($IC_{50},\;3.1\;{\mu}M$). By way of comparison, cinnamaldehyde proved to be a significantly more potent platelet inhibitor against platelet aggregation induced by collagen than aspirin. The effect exerted by cinnamaldehyde against platelet aggregation induced by AA was 1.2 times less than that of aspirin. These results indicate that cinnamaldehyde may prove useful as a lead compound for the inhibition of platelet aggregation induced by AA and collagen.
The anti-allergy activity of Cinnamomum cassia cortex extract and its fractions were investigated in mice. The extract exhibited potent anaphylactic shock and Arthus reaction. Among the fractions obtained in the successive fractionation with n-hexane, butanol and acetone, the butanol insoluble portion was shown to have the activities.
The essential oil obtained by steam distillation from medicinal plants of Cinnamomum cassia and Prunellae Herba. Analysis of essential oils were performed on GC/MS selective detector. Separations were performed fused silica capillary column. The carrier gas was ultra pure helium with a flow of 1 $m{\ell}/min$ and the splitless injector temperature was set as $280^{\circ}C$. The column temperature program was as follows: initial temperature of $70^{\circ}C$ for 4 min, and increased by $2^{\circ}C/min$ 70 to $100^{\circ}C$ (held 2 min), After that the temperature was varied from 100 to $200^{\circ}C$ at $5^{\circ}C/min$ (held 20 min), increase to $280^{\circ}C$ (held 5 min) at $10^{\circ}C/min$, in a total run time of 73 min. Ten volatile flavor components were identified from C. cassia and ten volatile flavor components were identified from Prunellae Herba. Strong inhibition of growth of Vibrio parahaemolyticus was obtained with all doses of C. cassia tested. Moreover, antimicrobial activity of C. cassia occurred in a dose dependant manner.
본 연구에서 육계 추출물로부터 cinnamyl alcohol을 분리·정제하였다. 연구 결과, H3 분획100 ㎍/mL에서 부정근 형성율이 286.6%였으며, 표준품 cinnamyl alcohol 1 mM (134.2 ㎍/mL)의 형성율이 290%로 유사한 결과를 나타내었다. 따라서 육계 methanol 추출물에 함유되어있는 cinnamyl alcohol이 녹두의 부정근 형성을 유도하는 주된 화합물일 것으로 판단된다. 육계에서 동정한 cinnamyl alcohol은 현재 식품첨가제 및 향료로써 사용되고 있으므로 안전성이 이미 보장되어 있으며, 한약재로써 사용된 육계 잔류물을 활용하여 부정근 형성 유도물질을 추출한다면비용적인 측면에서 또한 긍정적인 요인으로 작용할 것으로 판단된다. 따라서 육계는 유기농 농자재로써 충분히 사용 가능할 뿐만 아니라 신규 천연 식물생장조절제의 개발용 소재로서도 활용 가능할 것으로 사료된다.
목적 : 한의학 분야에서 육계(CCE: Cinnamomum cassia)는 혈류 순환이 저하된 상태에 널리 쓰이는 약재이다. 이 연구에서는 CCE와 그 활성 혼합물인 cinnamic acid가 in vitro와 in vivo에서 발휘하는 신생혈관 형성 작용에 대해 알아보고자 한다. 방법 : CCE와 CA의 신생혈관형성 작용을 알아보기 위하여 Human unbilical endothelial cells(HUVECs)와 Bovine arterial endothelial cells(BAECs) 그리고 Matrigel assay를 이용하였다. 결과 : In vitro에서 CCE와 그 활성 혼합물은 HUVEC 증식, 이동, 모세혈관-유사 관 형성을 유도한다. CCE에서의 25-50% 증가(1 또는 10ng/ml 용량에서)는 CA에서 증식과 유사하게 나타났다. HUVEC에서는 증식이 뚜렷하게 나타났으나 BAEC에서는 증식이 뚜렷하게 나타나지 않아 증식 효과는 내피세포의 기원에 따라 다른 것으로 나타났다. BAEC는 CCE와 CA에서 증식은 보이지 않았지만 이동과 모세혈관-유사 관 형성 또는 분화를 보였다. 또한 CCE와 CA는 HUVEC에서 VEGF 발현과 Flk-1/KDR 발현을 각각 2.2배와 2.5배 증가시켰다. 결론 : CCE와 그 활성 혼합물은 in vivo와 in vitro에서 혈관 내피세포의 VEGF 발현을 통하여 신생혈관 형성 작용을 유도하며 이는 앞으로 다른 한약 추출물의 신생혈관형성 촉진 작용 연구에 모델을 제시할 것으로 보인다.
