The anti-allergy activity of Cinnamomum cassia cortex extract and its fractions were investigated in mice. The extract exhibited potent anaphylactic shock and Arthus reaction. Among the fractions obtained in the successive fractionation with n-hexane, butanol and acetone, the butanol insoluble portion was shown to have the activities.

This study was carried out to investigate the effect of Cinnamomum camphora on melanogenesis. The MeOH extract of Cinnamomum camphora inhibited mushroom tyrosinase activity in dose-dependent manner. Moreover, it significantly suppressed the melanin production in melan-a cells at the concentration of $100{\mu}/m{\ell}$. The MeOH extract was partitioned with ethyl acetate, n-butanol and water. Among them, the BuOH soluble fraction exhibited significant inhibitory effect on mushroom tyrosinase. In addition, the BuOH soluble fraction reduced the melanin production in melan-a cells. But, the BuOH soluble fraction had less inhibition effects on melan-a cell originated tyrosinase. So, it was performed western blotting for melanogenic proteins (tyrosinase, tyrosinase-related protein (TRP-2)) using melan-a cells. The BuOH soluble fraction inhibited the protein expression of tyrosinase at the concentration of $100{\mu}/m{\ell}$. The results suggested that the BuOH soluble fraction of C. camphora might be a potent inhibitor of melanin biosynthesis in melan-a cells.
목적 : 한의학 분야에서 육계(CCE: Cinnamomum cassia)는 혈류 순환이 저하된 상태에 널리 쓰이는 약재이다. 이 연구에서는 CCE와 그 활성 혼합물인 cinnamic acid가 in vitro와 in vivo에서 발휘하는 신생혈관 형성 작용에 대해 알아보고자 한다. 방법 : CCE와 CA의 신생혈관형성 작용을 알아보기 위하여 Human unbilical endothelial cells(HUVECs)와 Bovine arterial endothelial cells(BAECs) 그리고 Matrigel assay를 이용하였다. 결과 : In vitro에서 CCE와 그 활성 혼합물은 HUVEC 증식, 이동, 모세혈관-유사 관 형성을 유도한다. CCE에서의 25-50% 증가(1 또는 10ng/ml 용량에서)는 CA에서 증식과 유사하게 나타났다. HUVEC에서는 증식이 뚜렷하게 나타났으나 BAEC에서는 증식이 뚜렷하게 나타나지 않아 증식 효과는 내피세포의 기원에 따라 다른 것으로 나타났다. BAEC는 CCE와 CA에서 증식은 보이지 않았지만 이동과 모세혈관-유사 관 형성 또는 분화를 보였다. 또한 CCE와 CA는 HUVEC에서 VEGF 발현과 Flk-1/KDR 발현을 각각 2.2배와 2.5배 증가시켰다. 결론 : CCE와 그 활성 혼합물은 in vivo와 in vitro에서 혈관 내피세포의 VEGF 발현을 통하여 신생혈관 형성 작용을 유도하며 이는 앞으로 다른 한약 추출물의 신생혈관형성 촉진 작용 연구에 모델을 제시할 것으로 보인다.
The in vitro antioxidative activities of the 33 plant-derived essential oils and 37 phytochemicals including 3 Mentha arvensis leaf-, 2 Thymus vulgaris leaf- and 2 Syzygium aromaticum flower-derived isolates were determined by the inhibition of linoleic acid autoxidation, the generation of superoxide anion and scavenging of DPPH radical. They were then compared to those of the widely used plant-derived antioxidants (pyrogallol and quercetin) and synthetic antioxidant BHT. At a concentration of 0.01%, potent antioxidative effect was observed in the essential oils from Cinnamomum cassia roots, Mentha arvensis leaves, Ginkgo biloba fruits, and Syzygium aromaticum flowers. Of the phytochemicals used, eugenol and isoeugenol at 0.01% showed potent antioxidative activity, and their activities were comparable to those of pyrogallol, quercetin, and BHT. The Cinnamomum root-, the Mentha leaf-, the Ginkgo fruit-, and the Syzygium flower-derived materials may be a good source for an alternative to the currently used antioxidants.
Twenty five methanolic plant extracts were investigated to determine the anticancer activity against sonic hedgehog (shh)/Gli signaling pathway dependent cancer, PANC-1 human pancreatic cancer cells, through three screening programs. All extracts were inspected their inhibitory properties on sonic hedgehog-conditioned medium (shh-CM) induced alkaline phosphatase (ALP) activity in C3H10T1/2 mouse mesenchymal stem cells to examine whether the plant extracts affect the shh/Gli signaling pathway. Next, plant extracts were screened the ability to suppress the cell proliferation of PANC-1 human pancreatic cancer cells. Finally, active plant extracts from the two screening systems were evaluated for the suppressive effect on Gli-mediated transcriptional activity in PANC-1 cells. Among active plants, Cinnamomum cassia suppressed Gli-mediated transcriptional activity leading to the down-regulated expression of Gli-target genes such as Gli-1 and Patched-1 (Ptch-1). This study provides the consideration for the important role of natural products in drug discovery process as well as the basis for the further analysis of active plant and potential identification of novel bioactive compounds as inhibitors of Gli and therapeutic candidates against shh/Gli signaling pathway dependent cancers.
The anticoagulant properties of Cinnamomum cassia bark-derived materials were evaluated against platelet aggregation induced by arachidonic acid (AA), collagen, platelet activating factor (PAF), or thrombin, and these effects were then compared to those of three commercially available compounds (cinnamic acid, cinnamyl alcohol, and aspirin). The active constituent obtained from C. cassia barks was isolated by silica gel column chromatography and high pressure liquid chromatography (HPLC), and was characterized as trans-cinnamaldehyde by MS, $^1H-NMR$, $^{13}C-NMR$, and IR spectroscopy. With regard to 50% inhibitory concentration ($IC_{50}$) values, cinnamaldehyde was found to effectively inhibit platelet aggregation induced by AA ($IC_{50},\;43.2\;{\mu}M$) and collagen ($IC_{50},\;3.1\;{\mu}M$). By way of comparison, cinnamaldehyde proved to be a significantly more potent platelet inhibitor against platelet aggregation induced by collagen than aspirin. The effect exerted by cinnamaldehyde against platelet aggregation induced by AA was 1.2 times less than that of aspirin. These results indicate that cinnamaldehyde may prove useful as a lead compound for the inhibition of platelet aggregation induced by AA and collagen.
Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.
Cinnamomum migao plants often face different degrees of drought in karst habitats, which can lead to plants' death, especially in the seedling stage. Widespread of arbuscular mycorrhizal (AM) fungi in karst soils have the potential to address this drought, which is a threat to C. migao seedlings. We inoculated C. migao seedlings with spores from Glomus lamellosum and Glomus etunicatum, two AM fungi widely distributed in karst soils, to observe seedling growth response after simulated drought. Our results showed that 40 g of G. lamellosum and G. etunicatum significantly promoted the growth of C. migao seedlings, 120 days after inoculation. Following a 15-day drought treatment, root colonization of the seedlings with G. lamellosum or G. etunicatum had lower the accumulation of malondialdehyde (MDA) and increased the accumulation of enzymes and osmotic substances in the seedlings. The relative water content in different organs (roots, stems, and leaves) of the drought-stressed seedlings was higher in plants with G. lamellosum or G. etunicatum than in plants without AM fungi colonization. Our results showed that inoculation with AM fungi was an effective means to improve the drought resistance of C. migao seedlings.
In this study, we selected two essential oils, Citrus unshiu and Cinnamomum cassia with superior antioxidant effects from the essential oils of 7 wild plants in South Korea and examined their antimicrobial activity against Candida albicans, which causes dermatitis to identify the antimicrobial components in the essential oils. As a result of measuring DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, SC50 value of the Citrus unshiu essential oil was 0.010 mg/mL, while for the Cinnamomum cassia essential oil, SC50 value was 0.09 mg/mL. In addition, when ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was measured, SC50 value of the Citrus unshiu essential oil was 0.09 mg/mL, while for the Cinnamomum cassia essential oil, it was 0.06 mg/mL, exhibiting high antioxidant activity. For the minimum inhibitory concentration (MIC), the essential oil of Cinnamomum cassia was 1.25 mg/mL and that of Citrus unshiu was 5 mg/mL, demonstrating a high antimicrobial activity of the Cinnamomum cassia essential oil. Through the thin layer chromatography (TLC) bioassay, we assessed the antimicrobial activity against C. albicans according to the fraction components of the two essential oils. Also, by using preparative TLC (prep. TLC), we obtained the active fractions, and by performing GC/MS analysis of the components with the same Rf value, we identified the antimicrobial-active components. As a result, the main components having antioxidant and antimicrobial activities were cinnamyl acetate, eucalyptol, linalool, and citral of the Cinnamomum cassia essential oil and linalool from the Citrus unshiu essential oil. Also, based on the analysis of the fractional components that showed antioxidant and antimicrobial activities in both of the two essential oils, it was found that linalool has antioxidant activity, while cinnamyl acetate, eucalyptol, citral, and geranyl acetate have antioxidant and antimicrobial activities.
The inhibitory activities against bacterial ${\alpha}-amylase$of oriental drugs from animals, plant and mineral origin were investigated. In final screening test, it was found that Areca catechu L., Cinnamomum cassia Pres 1. and Ephedra sinica Stapf had stronger inhibitory activities against ${\alpha}-amylase$ than other oriental drugs used in this experiment.
