Efficiency of Gas-Phase Ion Formation in Matrix-Assisted Laser Desorption Ionization with 2,5-Dihydroxybenzoic Acid as Matrix

  • Numbers of matrix- and analyte-derived ions and their sum in matrix-assisted laser desorption ionization (MALDI) of a peptide were measured using 2,5-dihydroxybenzoic acid (DHB) as matrix. As for MALDI with ${\alpha}$-cyano-4-hydroxy cinnamic acid as matrix, the sum was independent of the peptide concentration in the solid sample, or was the same as that of pure DHB. This suggested that the matrix ion was the primary ion and that the peptide ion was generated by matrix-to-peptide proton transfer. Experimental ionization efficiencies of $10^{-5}-10^{-4}$ for peptides and $10^{-8}-10^{-7}$ for matrices are far smaller than $10^{-3}-10^{-1}$ for peptides and $10^{-5}-10^{-3}$ for matrices speculated by Hillenkamp and Karas. Number of gas-phase ions generated by MALDI was unaffected by laser wavelength or pulse energy. This suggests that the main role of photo-absorption in MALDI is not in generating ions via a multi-photon process but in ablating materials in a solid sample to the gas phase.

p-Coumaric Acid Potently Down-regulates Zebrafish Embryo Pigmentation: Comparison of in vivo Assay and Computational Molecular Modeling with Phenylthiourea

  • p-Coumaric acid is an organic compound that is a hydroxyl derivative of cinnamic acid. Due to its multiple biological activities p-coumaric acid has been widely studied in biochemical and cellular systems and is also considered as a useful therapeutic candidate for various neuronal diseases. However, the efficacy of p-coumaric acid on zebrafish developmental regulation has not been fully explored. In this study, therefore, we first investigated the action mechanism of the p-coumaric acid on the zebrafish development in a whole-organism model. p-Coumaric acid treated group significantly inhibited the pigmentation of the developing zebrafish embryos compared with control embryos without any severe side effects. In addition, p-coumaric acid down-regulated more effectively in a lower concentration than the well-known zebrafish's melanogenic inhibitor, phenylthiourea. We also compared the molecular docking property of p-coumaric acid with phenylthiourea on the tyrosinase's kojic acid binding site, which is the key enzyme of zebrafish embryo pigmentation. Interestingly, p-coumaric acid interacted with higher numbers of the amino acid residues and exhibited a tight binding affinity to the enzyme than phenylthiourea. Taken all together, these results strongly suggest that p-coumaric acid inhibits the activity of tyrosinase, consequently down-regulating zebrafish embryo pigmentation, and might play an important role in the reduction of dermal pigmentation. Thus, p-coumaric acid can be an effective and non-toxic ingredient for anti-melanogenesis functional materials.

Characterization of Physicochemical Properties of Ferulic Acid

  • Ferulic acid (3-methoxy, 4-hydroxy cinnamic acid) is a flavoid component possessing antioxidant property. The compound is currently under development as a new drug candidate for the treatment of the dementia. The objective of this preformulation study was to determine the physicochemical properties of ferulic acid. The n-octanol to water partition coefficients of ferulic acid were 0.375 and 0.489 at the pHs of 3 and 10, respectively. Accelerated stability study for ferulic acid indicated that the t 90 value for the drug was estimated to be 459 days at $25^{\circ}C$. Ferulic acid was also found to be unstable under the relative humidity of more than 76%, probably because of the hygroscopic nature of the drug. In order to study compatibility of ferulic acid with typical excipients, potential change in differential scanning calorimetry spectrum was studied in 1: 1 binary mixtures of ferulic acid and typical pharmaceutical excipients (e.g., Aerosil, Avicel, CMC, Eudragit, lactose, PEG, PVP, starch and talc). Avicel, CMC, PVP and starch were found to be incompatible with ferulic acid, indicating the addition of these excipients may complicate the manufacturing of the formulation for the drug. Particle size distribution of ferulic acid powder was in the size range of 10-190 $\mu$m with the mean particle size of 61 $\mu$m. The flowability of ferulic acid was apparently inadequate, indicating the granulation may be necessary for the processing of the drug to solid dosage forms. Two polymorphic forms were obtained by recrystallization from various solvents used in formulation. New polymorphic form of ferulic acid, Form II, was obtained by recrystallization from 1,4-dioxane. The equilibrium solubility for Form I was approximately twice of that for Form II. The dissolution rate of Form II was higher than that of Form I in the early phase (<6 min). Therefore, these physicochemical information has to be taken in the consideration for the formulation of ferulic acid.

이상 저온조건이 딸기의 생육, 수량 및 생리활성 성분에 미치는 영향 (Influence of Abnormally Low Temperatures on Growth, Yield, and Biologically Active Compounds of Strawberry)

  • The present study aimed to investigate the effects of low temperature on the growth, yield, quality, and biologically active compounds of strawberry and obtain basic information for developing a technology for stable growth of strawberry in greenhouses. Growth of strawberry, including leaf number, area, and length, plant height, and dry weight was better at the optimum growth temperature of $20^{\circ}C$ than at a lower temperature of $15^{\circ}C$. At the low temperature of $15^{\circ}C$, the cultivar 'Maehyang' was more tolerant and displayed better growth rate than 'Seolhyang'. At $15^{\circ}C$, the fruit production per week and fruit weight was lower than that at $20^{\circ}C$. In contrast, fruit length and diameter were not significantly different between the two growth temperatures. Growth temperature also did not affect the fruit color index, Hunter L, a, b value, or fruit firmness. However, the sugar content of strawberries grown at $15^{\circ}C$ was higher by 0.8 and 1.5 Brix for 'Seolhyang' and 'Maehyang', respectively, than of those grown at $20^{\circ}C$. There was no difference in the content of fisetin, a biologically active compound, for 'Seolhyang' at both growth temperatures, however, the fisetin content of 'Maehyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$. Cinchonine and ellagic acid content of 'Seolhyang' was higher at $20^{\circ}C$ than at $15^{\circ}C$, whereas that of 'Maehyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$. Quercetin content showed no significant differences with respect to growth temperature, however, it tended to increase at $20^{\circ}C$. The cinnamic acid content of 'Seolhyang' was higher at $15^{\circ}C$ than at $20^{\circ}C$, whereas that of 'Maehyang' increased at $20^{\circ}C$. Collectively, the biologically active compounds of strawberry were affected by growth temperature. Moreover, the content of these compounds tended to increase at $20^{\circ}C$, the optimum growth temperature, rather than at the sub-optimal growth temperature of $15^{\circ}C$.

서양금혼초(Hypochaeris radicata L.)의 수용추출물이 사료작물에 미치는 allelopathy 효과 (The Allelopathic Effects of Aqueous Extracts of Hypochaeris radicata L. on forage Crops)

  • 서양금혼초(Hypochaeris radicata Linne)에 함유되어있는 화학물질이 사료작물의 미치는 allelopathy효과를 구명하기 위하여 본 실험을 실시하였다 서양금혼초에 함유되어 있는 allelochemicals 물질을 HPLC에 의해 성분 분석한 결과 계절적, 부위별로 차이가 있었으나 $\rho$-hydroxybenzoic acid, chlorogenic acid, catechin, caffeic acid, syringic acid, salicylic acid, $\rho$-coumaric acid, ferulic acid, naringin, hesperidin, myricetin, krans-cinnamic acid, quercetin, naringenin 등 14 종류의 phenolic compounds 성분을 확인하였고 caffeic acid, ferulic acid, naringenin 3종류는 공통으로 함유하고 있는 것으로 분석되었다. 5월에는 일, 7월에는 꽃과 뿌리에서 많은 양의 phenolic acids가 함유되어있고 특히 뿌리에 sali-cylic acid $2085.6{\mu}g/g$,와 quercetin $1522.0[\mu}g/g$,로 함량이 높았다. 서양금혼초 수용성 추출액으로 처리한 사료작물의 생장을 조사한 결과 품종에 따라 처리구와 대조구의 값이 같은 경우도 있으나 모든 조사 항목에서 대조구 보다 처리 값이 낮아 사료작물의 생장이 억제 된 것이 확인되었고 처리농도가 높을수록 강한 억제효과를 나타내었다. 수량의 변화를 조사한 결과 사료작물 품종에 따라 차이가 있으나 전체적으로 억제효과를 나타내었으며 처리농도가 높을수록 강한 억제효과를 나타내었다. 이러한 결과 서양금혼초는 사료작물에 대한 allelopathy 효과가 있음을 밝혀냈다. 특히 화본과 사료작물의 경우 italian ryegrass, 콩과 사료작물의 경우는 purple alfalfa가 생장억제효과가 높게 나타났다.

Chemical Composition of Cactus Pear Seed Oil: phenolics identification and antioxidant activity

  • Objectives: The chemical composition of cactus pear seed oil (Opuntia ficus-indica [L.] Mill.) was analyzed in terms of its fatty acid composition, tocopherol content, phenolic identification, and the oil's phenolic-rich fraction antioxidant power was determined. Methods: Fatty acid profiling was performed by gas chromatography coupled to an FI detector. Tocopherols and phenolic compounds were analyzed by LC-FLD/UV, and the oil's phenolic-rich fraction antioxidant power was determined by phosphomolybdenum, DPPH assay and β-carotene bleaching test. Results: Fatty acid composition was marked by a high unsaturation level (83.22 ± 0.34%). The predominant fatty acid was linoleic acid (66.79 ± 0.78%), followed by oleic acid (15.16 ± 0.42%) and palmitic acid (12.70 ± 0.03%). The main tocopherol was γ-tocopherol (172.59 ± 7.59 mg/kg. In addition, Tyrosol, vanillic acid, vanillin, ferulic acid, pinoresinol, and cinnamic acid were identified as phenolic compounds in the analyzed seed oil. Moreover, the oil's phenolics-rich fraction showed a significant total antioxidant activity, scavenged DPPH up to 97.85%, and effectively protected β-carotene against bleaching (97.56%). Conclusion: The results support the potential use of cactus pear seed oil as a functional food.

Quantitative Analysis of Eleven Bioactive Constituents of a Traditional Herbal Medicine, Yeonggyechulgam-tang using, Liquid Chromatography-Electrospray Ionization Tandem Mass Spectrometry

  • This study proposes a sensitive and selective liquid chromatography-electrospray ionization tandem mass spectrometry method of efficiently assessing the quality of a traditional herbal medicine called Yeonggyechulgam-tang (YGCGT). The following compounds 1 - 11, namely, liquiritin apioside (1), liquiritin (2), liquiritigene (3), coumarin (4), cinnamic acid (5), cinnamaldehyde (6), glycyrrhizin (7), atractylenolide III (8), atractylenolide II (9), atractylenolide I (10), and pachymic acid (11) were separated on a UPLC BEH $C_{18}$ column ($2.1{\times}100mm$, $1.7{\mu}m$) at a column temperature of $45^{\circ}C$ eluted with a gradient condition of 0.1% (v/v) formic acid in distilled water and acetonitrile. The correlation coefficient of the calibration curve of the eleven constituents was ${\geq}0.9936$. The limits of detection and quantification of the compounds 1 - 11 were 0.06 - 4.73 ng/mL and 0.17-14.20 ng/mL, respectively. Using this analytical method, the compound 11 in lyophilized YGCGT decoction extract was not detected, while the compounds 1 - 10 were detected 0.13-166.43 mg/g.

Simultaneous Determination of Albiflorin, Cinnamaldehyde, Cinnamic Acid, Daidzin, Glycyrrhizin, Liquiritin, Paeoniflorin and Puerarin in Galgeun-tang by HPLC-PDA

  • Objectives: We investigated to develop and validate HPLC-PDA methods for simultaneous determination of eight constituents in Galgeun-tang (GGT). Methods: Reverse-phase chromatography using a Gemini C18 column operating at $40^{\circ}C$, and photodiode array (PDA) detection at 230 nm, 254 nm, and 280 nm, were used for quantification of the eight marker components of GGT. The mobile phase using a gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. Results: Calibration curves were acquired with $r^2$ > 0.9999, and the relative standard deviation (RSD) values (%) for intra- and inter-day precision were less than 3.0%. The recovery rate of each component was in the range of 87.33-101.38%, with an RSD less than 7.0%. The contents of the eight components in GGT were 1.98-12.17 mg/g. Conclusions: The established method will be applied for the quantification of marker components in GGT.

Cloning and mRNA Expression Analysis of the Gene Encoding Phenylalanine Ammonia-Lyase of the Ectomycorrhizal Fungus Tricholoma matsutake

  • The ectomycorrhizal fungus Tricholoma matsutake grows symbiotically with Pinus densiflora. Phenylalanine ammonia-lyase (E.C. 4.3.1.24) catalyzes the conversion of L-phenylalanine to trans-cinnamic acid. The role of fungal phenylalanine ammonia-lyase, however, has not been clear until now. In this study, the gene encoding phenylalanine ammonia-lyase (PAL), which was isolated from T. matsutake, was cloned and characterized. The PAL gene (tmpal) consists of 2,160 nucleotides, coding for a polypeptide containing 719 amino acid residues. The deduced amino acid sequence of tmpal from T. matsutake shows high identity (70%) with that from Laccaria bicolor. Comparative analysis of the PAL genes among T. matsutake and other species of the class Agaricomycetes showed that both active sites and binding sites were significantly conserved among these genes. The transcriptional analysis of the PAL gene revealed a differential gene expression pattern depending on the developmental stages (mycelium, primordium, stipe, pileus, and gills) of T. matsutake. These results suggest that the PAL gene in T. matsutake plays an important role in multiple physiological functions.

Whitening Activity of Phenolic Compounds from Rhizome of Phragmites communis

  • Activity guided phytochemical examination of the rhizome of Phragmites communis has led to the isolation of two phenolic acids and two lignans. Structures of these compounds were elucidated as methyl gallate (1), ${\rho}$ -hydroxy cinnamic acid (2), (+) -lyoniresinol (3), (+)-lyoniresinol-9'-O-${\beta}$-D-glucopyranoside (4) by comparisons with previously reported spectral data. To investigate the melanogenesis inhibitory effects of these compounds, the melanin level and tyrosinase activity were examined in B16F10 melanoma cell. Each compound inhibited both tyrosinase activity and melanin synthesis compared with positive control, kojic acid and arbutin. These results suggest that the phenolic compounds from Phragmites communis might be developed as a potent skin whitenting cosmeceuticals.
