Reactive oxygen species potentially cause damage to cellular components including lipids, protein and DNA; this oxidative damage plays a key role in the pathogenesis of neurodegenerative disease, cardiovascular disease, metabolic disease and cancer. On the basis of the oxidative stress hypothesis, a number of studies have been performed to search for an efficient and safe antioxidant. Although in vitro studies have provided promising results, only a limited number of natural and synthetic antioxidants have been developed for clinical application due to their low efficacy and side-effects. Thus, the discovery of new antioxidants with marked efficacy and safety has attracted worldwide attention in recent decades. Since plants are recognized as important sources of natural antioxidants, our research has focused on the discovery of new naturally occurring antioxidants from medicinal plants. The purpose of this review is to open a new prospect in the field of search for natural antioxidants from medicinal plants by summarizing our recent findings. Using in vitro bioassay systems such as 2,2-diphenyl-1-picrylhydrazyl, superoxide radical scavenging tests and lipid peroxidation models, we have tested over than 350 species of medicinal plants for their antioxidant activity and selected several of them for further investigation. During the research on the discovery of effective natural antioxidants from the medicinal plants selected, we have isolated several new and known antioxidant compounds that include stilbene glycosides, phenolic glycosides, flavonoids, oligostilbenes, and coumarins. Our results suggest that the presence of antioxidant compounds in the medicinal plants might be associated with the traditional use to treat inflammation, cardiovascular disease and various chronic diseases.
Oxidative stress or accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. Glutamate is one of the major excitatory neurotransmitter in the central nervous system (CNS). Glutamate contributes to fast synaptic transmission, neuronal plasticity, outgrowth and survival, behavior, learning and memory. In spite of these physiological functions, high concentration of glutamate causes neuronal cell damage, acute insults and chronic neuronal neurodegenerative diseases. Heme oxygenase-1 (HO-1) enzyme plays an important role of cellular antioxidant system against oxidant injury. NNMBS020, the water-insoluble fraction of the 70% EtOH extract of root barks of Dictamnus dasycarpus, showed dominant neuroprotective effects on glutamate-induced neurotoxicity in mouse hippocampal HT22 cells by induced the expression of HO-1 and increased HO activity. In mouse hippocampal HT22 cells, NNMBS020 makes the nuclear accumulation of Nrf2 and stimulates extracellular signal-regulated kinase (ERK) pathway. The ERK MAPK pathway inhibitor significantly reduced NNMBS020-induced HO-1 expression, whereas the JNK and p38 inhibitors did not. In conclusion, the water-insoluble fraction of the 70% EtOH extract of root barks of D. dasycarpus (NNMBS020) significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2 and ERK pathway in mouse hippocampal HT22 cells.

Sensory factors are important determinants of appetite and food choices but little is known about factors affecting taste acuity and preference of Koreans. Any factors causing deficits in sweet taste perception may lead to over consumption of simple sugar, which is related to several chronic diseases. This study was conducted to determine factors affecting sweet taste sensitivity and preference. Subjects were 30 government employees who were serving as school dietitians or in the area of public health while they were studying in the program for the qualification to become nutrition teachers. Sweet taste threshold and the optimally-preferred sweetness of omija jelly were determined by a sensory evaluation and general characteristics, health-related lifestyles, dietary habits and food preferences were determined using a self-administered questionnaire. For the subjects of this study, detection threshold concentration of sucrose solution was $0.184{\pm}0.06%$ and optimally-preferred sucrose concentration of omija jelly was $13.88{\pm}2.28%$ and there was no significant correlation between the sweet taste sensitivity and preference. Subjects who had higher(${\ge}4$ out of 10) physical or psychological stress and who had late getting-up time (after 7am) tended to have lower sweet taste threshold (higher sensitivity) than their counterpart. The sweet taste preference determined by optimally-preferred sucrose concentration of omija jelly tended to be lower in the subjects who eat slowly. Those who answered in the questionnaire to prefer sweet foods did have significantly higher optimally-preferred sucrose concentration of omija jelly. Further research is required to determine whether decreased sensitivity and increased preference for sweetness can increase the actual intake of simple sugar. This study was designed to assess diet and health-related factors of older adults in Korea. Subjects females were 2,660 adults aged 50 and over living in Korea. Males were 847 persons and were 1813 persons. The mean weight and height for males and females were 63.8 $\pm$ 0.3kg / 164.0 $\pm$ 0.2cm and 57.0 $\pm$ 0.2kg /150.6 $\pm$ 0.1cm respectively. BMI (body mass index), body fat, and percent fat were significantly greater in females than in males. The muscle mass and body water were significantly greater in males than in females. Twenty-one percent of total subjects lived alone and 26% with spouse only. Most of the subject's self-reported income was in middle level (65%) or low level (24%). Proportion of subjects who answered 'very poor' or 'poor' on perceived health status was higher in older group. The 50-64 years old group was facing more stress than 65yr and over group. Among male subjects,38.4% were current-smokers and 22.0% were ex-smokers. But only 6.5% of female subjects were current-smokers. Males turned out to have better dietary habits-meal frequency per day, mealtime regularity, regular meal size and balanced eating-than females (p < 0.001). This study revealed that the diet and health-related factors affect nutritional status and chronic diseases of the elderly. For better management and evaluation of health status of the elderly, more effective nutritional assessment tools should be developed.
Stroke is one of the leading causes of death in Korea. Because of their sequelae, strokes are categorized as a sudden-onset, constant course chronic illness which needs continuous efforts for rehabilitation. Unfortunately, there are few community based rehabilitation program for post-stroke patients who stay at home. The authors developed a community based selp-help management program for post-stroke patients to enhance their rehabilitation process. The program consists of five sessions and each session contains health education. ROM exercise, ADL training, and stress management like foot reflexology. A professor and two graduate students of nursing college coordinated the program. To test the effects of the program we conducted a 5 week program to the 10 conveniently selected post-stroke patients who were living in Kang-buk district of Seoul. The Questionnaires about ADLs, IADLs, depression and life satisfaction were asked to the all subjects before and after program. The hand grisp power and muscle strength of four limbs were measured at the end of each sessions. The analysis of data revealed that the program was effective to increase the ADLs, IADLs, and muscle strength and to decrease the depression levels of subjects. However, there was no significant difference between pre and post hand grisp power and life satisfaction. Because the program was effective to Improve the physical and psycholocial function of subjects, we suggest continual development and Implementation of community based self-help management programs.
The purpose of this study was to estimate the prevalence of hypertension, and to assess the risk factors associated with hypertension in elderly (over 65 years old) Koreans, using data from the 4th Korean National Health & Nutrition Examination Survey (the 4th KNHANES), 2007~2009. A total of 1,887 participants were analyzed for assessing the risk factors after excluding those who took hypertension medicines or underwent diet therapy (hypertension perceivers). On the other hand, in analyzing prevalence, a total of 3,526 people were analyzed, including hypertension perceivers. Hypertension was more frequently found in female (64.1%) and urban (62.0%) groups compared to male (55.6%) and rural (57.4%) groups. The mean age of the hypertensive group was significantly higher than that of normal group. The mean values of total cholesterol levels were also significantly higher in the hypertensive group, while body weight, waist circumference, fasting blood glucose, hemoglobin, hematocrit, LDL-cholesterol, and HDL-cholesterol were not. Multiple logistic regression showed that smoking was significantly related to the prevalence of hypertension, but alcohol drinking, stress, obesity, hypercholesterolemia, hypertriglyceridemia, diabetes, anemia, and nutrient intakes were not. The results of this study does not support clear relations of hypertension with chronic diseases including obesity, hyperlipidemia, anemia, and diabetes as well as nutrient intakes among a Korean elderly population. A prospective long-term research study is needed to establish the effects of these factors on hypertension.
Urticaria is a common dermatopathy, coming under "eun-jin(隱疹) in Korean Oriental medicine. We researched sixty-two patients of urticaria who had visited the Dept of Dermatology Oriental medical hospital Dongeui universty from January 2002 to April 2004 and had taken the Oriental herb medicine as Sungmagalguntanggamibang (SGT, 升摩葛根湯加味方) for verification of voluntariness of Korean Oriental medical treatment. In our time, urticaria is tending upwards, consequently the air pollution by industrialization, patronizing of convenience food by simplification of dietary life, stress and an abuse of medicine. Especially in case of chronic urticaria we're under going hardships in elimination of it's symptoms, because it's prognosis is inferior. We inspect the effects of SGT from various viewpoints and meet with following results. Results: 1. AS amount of taking the SGT increase, the effect in taking SGT period and the condition of present time become more satisfactory. 2. As continuous Time of urticaria decrease, the effect in taking SGT period become more satisfactory. 3. Etiology of urticaria is classified into the deficiency both of qi-blood(氣血兩虛), the moisture-heat of spleen-stomach(脾胃濕熱), the heat of blood(血熱), The wind-cold(風寒), The wind-heat(風熱), spleen-stomach accumulated heat(腸胃實熱) ect. The SGT is available in this order. 4. The effect in taking SGT period connects the quality of life of after treatment and the quality of life of after treatment is improved comparing with that of before treatment. The above results demonstrate that SGT effectively suppresses recurrent, pruitus and erythematous urticaria.
The purpose of this study was to examine the effects of the short term education and reinforcement program to health promotion in women in mid-life. This study has been done between February and April, 2004 and the subjects of the study were 26 women in mid-life. The treatment intervention was applied during total 8 weeks as 5 days for the short term education and 7 weeks for reinforcement with 1 time per a week. The short term education included health education for menopause, effect of exercise, healthy diet, management of stress and management of chronic illness and stretching exercise and recreation. The collected data were processed using the SPSS Win(12.0) program and analyzed using Wilcoxon Signed Ranks Test. The result of this study are as follows : 1. The short term education and reinforcement program significantly effected on the total cholesterol, HDL-cholesterol, triglyceride. 2. The short term education and reinforcement program significantly effected on the anxiety, but not depression. 3. The short term education and reinforcement program non significantly effected on the sleep disturbance and self-efficacy of exercise. 4. The short term education and reinforcement program significantly effected on the practice of healthy life. In conclusion, the short term education and reinforcement program effected on the physical health index, anxiety and practice of healthy life.
Oxidative stress is associated with many kinds of chronic diseases. Antioxidants such as polyphenols are compounds that protect cells against the damaging effects of reactive oxygen species. Grape seeds are considered good resources of polyphenols, and grape seed extracts have a very strong antioxidant effect. In the present study, we established a simple gradient reverse-phase high performance liquid chromatography method to determine polyphenol content from three different grape seed resources. An ODS (2), $150\;{\times}\;3.2\;mm$ column has been employed, and six polyphenols have been determined: gallic acid, protochatechuic acid, (+)-catechin, (-)-epicatechin, procyanidin B2, and epicatechin gallate. Catechin and epicatechin were the main polyphenol compounds in all three extracts. The amount of procyanidin B2 was higher in Extract 1 (from a company of China), while Extract 2 (extracted in our lab) and Extract 3 (from a company of USA) contained higher proportions of epicatechin gallate. For the total polyphenol content, Extract 1 was much higher than that of Extract 2 and 3. The results suggest that the dietary dose of grape seed extracts from different resources should be adjusted according to polyphenol content.
Hydrogen assisted cracking (HAC) is one of the most complicated problem in welding. Huge amount of studies have been done for decades. Based on them, various standards have been established to avoid HAC. But it is still a chronic problem in industrial field. It is well known that the main causes of the hydrogen crack are residual stress, crack susceptible micro structures and a certain critical level of hydrogen concentration. Even though the exact generating mechanism is unclear till today, it has been reported that the hydrogen level in the weld metal should be managed less than a certain amount to prevent it. Matsuda studied that the residual hydrogen level in the weld metal can be varied even if the initial hydrogen content is same. It is also insisted in this report that the residual hydrogen concentration is in stronger correlation with hydrogen crack than the initial hydrogen content. But, in practical point of view, the residual hydrogen is still hard to consider because measuring hydrogen level is time and cost consuming process. In this regard, numerical analysis is the only solution for considering the residual hydrogen content. Meanwhile, Takahashi showed the possibility of predicting the residual hydrogen by a rigorous FE analysis. But, few commercial software suitable for solving the weld metal hydrogen has been reported yet. In this study, two dimensional thermal - hydrogen coupled analysis was developed by using the commercial FE software MARC. Since the governing equation of the hydrogen diffusion is similar to the heat transfer, it is shown that the heat transfer FE analysis in association with hydrogen diffusion property can be used for hydrogen diffusion analysis. A series of simulation was performed to verify the accuracy of the model. For BOP (Bead-On-Plate) and the multi-pass butt welding simulations, remaining hydrogen contents in the weld metal is well matched with measurements which are referred from Kim and Masamitsu.
