Microcystic Meningiomas: Its Immunohistochemical and Genetic Aspect

  • The authors report three microcystic meningiomas with its characteristic immunohistochemical findings and chromosomal pattern. Three patients with surgically treated microcystic meningioma were studied for its radiological, histopathological findings, and chromosomal analysis was done in the one patient. Tumors were convexity meningioma in the frontal area. The tumors were enhanced homogenously in the two, and enhanced in homogenously with multiple small cysts in the other one on preoperative magenetic resonance image. Pathological examination showed marked nuclear pleomorphism, many small cysts, hyaline thickening in blood vessel wall, and mucinous background, compatable to microcystic type. EMA and vimentin were positive on the immunohistochemical stain. Chromosomal analysis showed tetrasomies of chromosome 5, 13, 17, and 20, and trisomies of chromosome 6, 7, 9, 11, 12, 16, 19, and 21, which are quite different from those of benign meningioma.

Haplotype Phylogeny of a 200kb Region in the Human Chromosome X Terminal Band (q28)

  • The haplotypes of a 200 kb region in the human chromosome X terminal band (q28) were analyzed using the International HapMap Project Phasell data, which had been collected for three analysis panels (YRI, CEU, and CHB+JPT). When multiple linkage disequilibrium blocks were encountered for a panel, the neighboring haplotypes that had crossover rate of 5% or more in the panel were combined to generate 'haploid' configurations. This resulted in 8, 7, and 5 'haploid' configurations for the panels of YRI, CEU, and CHB+JPT, respectively. The multiple sequence alignment of these 'haploids' was used for the calculation of allele-sharing distances and the subsequent principal coordinate analysis. Two 'haploids' in CEU and CHB+JPT were hypothesized as 'parental' in light of the observations that the successive recombinants of these haploids can model two other haploids in CEU and CHB+JPT, and that their configurations were consistent with those in YRI. This study demonstrates the utility of haplotype phylogeny in understanding population evolution.

  • Meiotic metaphase chromosomes of the testis of two storage insects, tobacco moth (Ephestia elutella H.) and cigarette beetle (Lasioderma serricorne F.) were examined to study their karyotypes. The number of haploid chromosomes of the tobacco moth was 31 and the karyotype was characterized by 29 metacentric and 2 submetacentric chromosomes. The metaphase chromosome length of tobacco moth ranged ca. 1.44-2.11${\mu}{\textrm}{m}$ and the average length was 1, 78${\mu}{\textrm}{m}$. The number of haploid chromosomes of the cigarette beetle at metaphase was 10 and the karyotype was characterized by 8 metacentric and 2 submetacentric chromosomes. The range of metaphase chromosome length of the cigarette beetle was ca. 1.79- 2.39${\mu}{\textrm}{m}$ and the average length 2.09${\mu}{\textrm}{m}$.

Benzidine dihydrochloride에 의한 Chinese hamster lung 세포의 적응반응 (Adaptive Response in Chinese Hamster lung Cells by Benzidine Dihydrochloride)

  • We studied adaptive response in CHL cells by benzidine dihydrochloride, a derivative of benzidine, which was a major mutagenic agent in dye industry. Chromosome aberration analysis was used for the identification of adaptive response to this mutagen. Adaptive and reactive doses were confirmed by cell proliferation rate curve. Cell proliferation rate curve was obtained from the mitotic indices of cells treated with various concentrations of benzidine dihydrochloride for 24 hours. Marked adaptive responses to benzidine dihydrochloride in the induction of chromosome aberration were observed in CHL cells by pre-treatment with low concentrations of benzidine dihydrochloride (0.0047 mg/$m\ell$ or 0.0094 mg/$m\ell$) for 24 hours following post-treatment with high concentrations (0.0187, 0.0375, 0.075, 0.15 mg/$m\ell$) for 24 hours. These adaptive responses were found mostly in the type of chromatid breaks and chromatid exchanges. There is no difference in these results between two adaptive doses, 0.0047 mg/$m\ell$ and 0.0094 mg/$m\ell$. The amount of adaptive response, however, was dependent on post-treatment doses.

양식산 무지개송어 (Oncorhynchus mykiss) 2배체 및 유도된 3배체의 염색체 다형현상 (Chromosomal Polymorphism in Diploid and Induced Triploid Rainbow Trout, Oncorhynchus mykiss)

  • 국내에서 양식되고 있는 무지개송어의 유전학적 동정 (genetic stock indentification)의 일환으로 염색체 분석을 실시한 결과 염색체 수 2n=60 및 2n=61의 개체 간 염색체 다형 현상이 발견되었다. Idiogram 분석 결과 이와 같은 염색체 다형 현상은 Robertsonian 전좌에 의해 중부 염색체 (metacentiric chromosome) 1개가 1쌍의 단부 염색체(acrocentric chromosome)로 나누어지기 때문 (Robertosoman fission)이었다. 염색체수 60개 및 61개인 개체의 집단 내 빈도는 각각 $38\%$$62\%$였으며 염색체 수 61개인 개체들에서는 암컷의 비율이 수컷에 비해 높게 나타났다. 염색체 다형 현상을 보인 친어들로부터 3배체를 유도한 결과 유도된 3배체의 염색체 수 분포는 90, 91, 92 및 93개로 나타났으나 염색체 arm의 수는 모두 156개로 2배체와 동일하게 Robersonian 전좌에 기인한 다형 현상을 나타내었다.

  • The mouse lymphoma thymidine kinase (tk+/-) gene assay (MOLY) using L5178Y tk+/- mouse lymphoma cell line is one of the mammalian forward mutation assays. It is well known that MOLY has many advantages and more sensitive than the other mammalian forward mutation assays such as x-linked hyposanthine phosphoribosyltransferase (hprt) gene assay. The target gene of MOLY is a heterozygous tk+/- gene located in 11 chromosome of L5178Y tk+/- cell, so it is able to detect the wide range of genetic changes like point mutation, deletion, rearrangement, and mitotic recombination within tk gene or deletion of entire chromosome 11. MOLY has relatively short expression time (2-3 days) compared to 1 week of hprt gene assay. MOLY can also induce relatively high mutant frequency so a large number of events can be recorded. The bimodal distribution of colony size which may indicate gene mutation and chromosome breakage potential of chemicals according to mutation scale such as large normal-growing mutants and small slow-growing mutants can be observed in this assay. The statistical analysis of data can be performed using the MUTANT program developed by York Electronic Research in association with Hazelton as recommended by the UKEMS (United Kingdom Environmental Mutagen Society) guidelines. This report reviewed MOLY using the microtiter cloning technique (microwell assay).

국내 토종개의 염색체 조사에 관한 연구 (Study on chromosomes survey of Korea native dogs)

  • The karyotype of the domestic dog is widely accepted as one of the difficult mammalian karyotypes to work. In contrast to many other animals, knowledge about the canine karyotype is quite sparse. The dog has a total of 78 chromosomes; all 76 autosomes are acrocentric in morphology and show only a gradual decrease in length. But appear to be quite small and difficult to identify unambiguously. To purchased standardization of chromosome in Korea native dog, there were analyzed by conventional trypsin/Giemsa staining (GTG-banding techniques), and were compared with 4, 6, 8, 11, 13, 17 chromosome. There were no variations in karyotypes which were analyzed by conventional GTG-banding techniques, but differences were observed in G-banding patterns with Sapsaree, Jindo, Gyeongju DongGyeong dogs, Welshi-Corgi. It is not clear that these disagreements in G-banding patterns between strains of dog were caused by chromosome polymorphism or a difference in interpretation. Comparative analysis of the distribution patterns of conserved segments defined by dog paints in the genomes of the Korea native dogs demonstrates that their differences in the karyotypes of these three species could have resulted from acrocentric banding patterns.

A Study on Effect of Meiosis Background Concepts on the High School Students' Understanding of Meiosis

  • The objectives of this study were to investigate what kind of background concepts is necessary to help that high school students easily understand meiosis and to find out how these background concepts affect students' understanding of meiosis. To achieve these objectives, first this study surveyed meiosis background concepts that high school teachers think. Based on 8 background concepts - nuclear phases, chromosome, mitosis, reproduction, gamete, gene, mother/daughter cell - of previous survey, the questionnaire was made for the 10th(724) and 11th(862) grade students and then was analyzed for the effect of meiosis background concepts on the high school students' understanding of meiosis. Results of the analysis revealed that the influential background concepts are as follow; cell cycle, chromosome in the advanced level, mother/daughter cell, mitosis, chromosome, nuclear phases in the intermediate level, mother/daughter cell, nuclear phases, gene in the low level. And the achievement according to item types was differed not by meiosis achievement, but by each background concepts.

Characterization of Quantitative Trait Loci (QTL) for Growth using Genome Scanning in Korean Native Pig

  • Molecular genetic markers were genotyped used to detect chromosomal regions which contain economically important traits such as growth traits in pigs. Three generation resource population was constructed from a cross between the Korean native boars and Landrace sows. A total of 193 F2 animals from intercross of F1 were produced. Phenotypic data on 7 traits, birth weight, body weight at 3, 5, 12, 30 weeks of age, live empty weight were collected for F2 animals. Animals including grandparents (F0), parents (F1), offspring (F2) were genotyped for 194 microsatellite markers covering from chromosome 1 to 18. Quantitative trait locus analyses were performed using interval mapping by regression under line-cross model. To characterize presence of imprinting, genetic full model in which dominance, additive and imprinting effect were included was fitted in this analysis. Significance thresholds were determined by permutation test. Using imprinting full model, four QTL with expression of imprinted effect were detected at 5% chromosome-wide significance level for growth traits on chromosome 1, 5, 7, 13, 14, and 16.
