페놀 화합물의 Hexavalent Chromium독성에 대한 세포 보호효과 (Cytoprotective Effect of Phenolic Compounds Against Hexavalent Chromium-Induced Cytotoxicity)

  • Objectives : In order to evaluate the cytotoxicity of hexavalent chromium, the cytoprotective effect of phenolic compounds against hexavalent chromium-induced cytotoxicity, cell viability, cell adhesion ability, lactate dehydrogenase(LDH) activity, and morphological changes of cells were examined. Methods : We measured the cytotoxicity of hexavalent chromium with 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyltetrazolium bromide(MTT), 2,3-bis-[2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium-5-caboxanilide (XTT), LDH and DPPH methods. Results : The cytotoxicity of hexavalent chromium($IC_{50}$, $44.0-51.0{\mu}M$) was high according to the toxic criteria. Cytoprotective effect of phenolic compounds against $IC_{50}$ value of hexavalent chromium in cell morphology increased in a concentration-dependent manner. Conclusions : These results suggest that 3,4,5-trihydroxybenzoic acid may be used as a cytoprotective agent against chromium(IV)-mediated cytotoxicity.

벤조산 유도체의 Chromium Trioxide 독성에 대한 세포 재생효과 (The Cell Regenerative Effect of Benzoic Acid Derivatives Against Chomium Trioxide-Induced Cytotoxicity)

  • In order to evaluate the cytotoxicity of chromium trioxide, and the cell regenerative effect of phenolic acid against chromium trioxide-induced cytotoxicity, cell viability, cell adhesion activity, lactate dehydrogenase (LDH) activity, and morphological changes of cells were performed in these cultures. The toxicity of chromium trioxide (${IC}_{50}$, 44.0 ${\mu}M$) was high according to the toxic criteria. Cell regeneration of benzoic acid derivatives against ${IC}_{50}$ value of chromium trioxide in cell morphology was increased in concentration-dependent manner. These results suggest that benzoic acid derivatives may be used as a cell regenerative agent against chromium-mediated cytotoxicity.

5-Fluorouracil and Interleukin-2 Immunochemotherapy Enhances Immunogenicity of Non-Small Cell Lung Cancer A549 Cells through Upregulation of NKG2D Ligands

  • Background: The aim of this study was to investigate the anti-cancer effects and mechanisms of immunochemotherapy of 5-fluorouracil (5-FU) and interleukin-2 (IL-2) on non-small cell lung cancer (NSCLC) A549 cells. Materials and Methods: In order to detect whether 5-FU+IL-2 could effectively inhibit tumor growth in vivo, we established an A549-bearing nude mouse model. The cytotoxicity of natural killer (NK) cells was evaluated using a standard chromium release assay. To evaluate the relevance of NK cells in 5-FU+IL-2-mediated tumor inhibitory effects, we depleted NK cells in A549-bearing mice by injecting anti-asialo-GM-1 antibodies. Effects of 5-FU+IL-2 on the expression and promoter activity of NKG2D ligands (MICA/MICB) in A549 cells in vitro were also assessed. Results: In A549-bearing nude mice, combination therapy significantly inhibited tumor growth in comparison with monotherapy with 5-FU or IL-2 and enhanced the recognition and lysis of tumor cells by NK cells. Further study of mechanisms showed that NK cells played a vital role in the anticancer immune response of 5-FU+IL-2 immunochemotherapy. In addition, the combination therapy synergistically stimulated the expression and promoter activity of MICA/MICB. Conclusions: 5-FU and IL-2 immunochemotherapy significantly inhibited tumor growth and activated NK cytotoxicity in vivo, and these effects were partly impaired after depleting NK cells in tumor-bearing mice. Combination treatment of 5-FU and IL-2 upregulated the expression and the promoter activity of MICA/MICB in A549 cells, which enhanced the recognition of A549 cells by NK cells. All of the data indicated that immunochemotherapy of 5-FU and IL-2 may provide a new treatment option for patients with lung cancer.

연꽃추출물이 6가 크롬으로 유도된 세포독성에 대한 보호효과에 관한 연구 (Study on the Protective Effect of Nelumbo nucifera GAERTN Extract on Cultured Cerebral Neuroglial Cells Damaged by Hexavalent Chromium)

  • 중금속의 하나인 $Cr0_3$의 세포독성과 이에 대한 연꽃(Nelumbo nucifera GAERTN(NNG))의 수술에서 채취한 연꽃추출물의 방어효과를 배양 대뇌신경교세포(C6 glioma cell)를 대상으로 항산화 측면에서 조사하였다. $Cr0_3$의 세포독성을 조사하기 위하여 배양된 C6 glioma 세포를 $4{\sim}55{\mu}M$ 농도의 $Cr0_3$로 48시간동안 처리하였다. XTT 분석법에 의하여 세포생존율을 조사하였다. 그 결과 $Cr0_3$는 처리 농도에 비례하여 C6 glioma세포의 생존율을 유의하게 감소시켰으며 $Cr0_3$$55{\mu}M$ 농도 처리에서는 $XTT_{50}$ 값이, $10{\mu}M$에서는$XTT_{90}$ 값이 나타났다. 따라서 세포독성판정기준에 의하여 $Cr0_3$는 C6 glioma세포에 고독성인 것으로 나타났다. 한편, $Cr0_3$의 세포독성에 대한 연꽃추출물의 방어효과를 항산화 측면에서 조사하기 위하여 먼저 연꽃추출물의 항산화활성을 SOD-유사활성에 의하여 조사하였다. 그 결과 $130{\sim}150{\mu}g{\cdot}mL^{-1}$의 연꽃추출물 농도에서 SOD-유사활성이 대조군에 비하여 모두 증가하였으며 특히 $150{\mu}g{\cdot}mL^{-1}$의 NNG추출물의 처리에서는 대조군에 비하여 유의하게 증가한 것으로 나타났다(p<0.05). 이는 또한 비교군으로 사용한 $15{\mu}M$, vitamin E의 SOD유사활성과 매우 유사한 활성을 나타냈다. 따라서 연꽃추출물은 SOD 유사활성을 보임으로서 항산화활성을 나타냈다. 한편, $Cr0_3$의 세포독성에 대한 연꽃추출물의 방어효과를 조사하기 위하여 $Cr0_3$$XTT_{50}$ 농도인 $55{\mu}M$에서 배양 C6 glioma 세포를 처리하기 전에 연꽃추출물이 $90-120{\mu}g{\cdot}mL^{-1}$ 농도로 각각 포함된 배양액에서 2시간 동안 배양한 후 이에 대한 세포생존율을 조사하였다. 그 결과 연꽃추출물을 처리한 실험군에서는 $Cr0_3$을 처리한 실험군에 비하여 유의한 세포생존율의 증가를 보였다. 본 실험 결과는 연꽃추출물이 $Cr0_3$의 세포독성을 효과적으로 방어하였음을 알 수 있었다. 이상의 실험 결과로부터 $Cr0_3$는 배양 C6 glioma세포에 고독성을 나타냈으며 항산화활성을 가지고 있는 연꽃추출물은 $Cr0_3$에 의한 세포독성을 효과적으로 방어함으로서 $Cr0_3$의 독성이 산화적 손상과 관련이 있음을 알 수 있었다.
