Object: Death rate due to hypertension, atherosclerosis, ischemic heart disease and cerebral infarction induced by Westernized diet and increased average life span is on the rise. Decrease in blood circulation, activation of thrombus generation and intravascular lipid accumulation, cited as the principal causes of the above mentioned diseases in recent studies, result in circulatory disturbance and blood vessel obstruction leading to ischemic cell death of heart, brain and peripheral vessels. Method: We investigated the biochemical changes in microvascular permeability, aggregation of platelet and the intravascular lipid accumulation in induced-diabetic rat using Streptozotocin. We also studied the effects of Woohwangcheongsirn-won after oral administration on blood circulation, platelet function and lipid metabolism. The results are as follows: I. Woohwangcheongsim-won increased blood circulation in microvessels. 2. Woohwangcheongsim-won increased the reduced erythrocyte deformability in diabetes. 3. Woohwangcheongsim-won induced the reduction of contents of 2, 3-DPG, but failed to affect the reduced contents of ATP in erythrocyte in diabetes. 4. Woohwangcheongsim-won reduced the activity of Ca/sup 2+/-ATPase in the membrane of erythrocyte. 5. Woohwangcheongsim-won reduced the platelet aggregation evoked by platelet agglutinin factor. 6. Woohwangcheongsim-won reduced the production of platelet-derived granules. 7. Woohwangcheongsim-won reduced the production of metabolites of arachidonic acid in diabetes, and also reduced the production of increased thromboxane B2. 8. Woohwangcheongsim-won reduced the synthesis of oxidized LDL-cholesterol. In conclusion, Woohwangcheongsim-won enhanced blood circulation in microvesseles, erythrocyte deformability and inhibited the increased platelet aggregation and the synthesis of oxidized LDL-cholesterol in diabetes. 폴리코사놀의 콜레스테롤 감소 효과에 대한 기전은 아직 명확하게 규명되지 못하고 있다. 최근 몇몇 연구들은 지방산 합성 경로를 통한 콜레스테롤 합성의 조절에 sterol regulatory element-binding proteins(SREBP-1c)의 역할을 제시하였다. 그러나 현재까지 SREBP-1c에 의해 조절되는 지방산 합성에서 폴리코사놀의 효과에 대한 연구는 전무하다. 그러므로 본 연구의 목표는 SREBP-1c에 의해 매개되는 지방산 합성이 폴리코사놀의 콜레스테롤 감소 효과와 관계하는지를 조사하는 것이다. 7주령의 C57BL/6 수컷 생쥐를 7개군(군당 7마리)으로 나누고 8주간 다음과 같이 처리하였다; 1) 정상식이군(정상 대조군), 2) 고지방식이군(high-fat diet, HFD, 음성대조군), 3) 고지방식이+에탄올처리군(Pol-0), 4) 고지방식이+1 mg/kg 폴리코사놀처리군(Pol-1), 5) 고지방식이+2 mg/kg 폴리코사놀처리군(Pol-2), 6) 고지방식이+4 mg/kg 폴리코사놀처리군(Pol-4),7) 고지방식이+simvastatin 50 ㎍/kg 처리군(양성 대조군). 폴리코사놀과 simvastatin은 고지방식이를 유지하는 동안 매일 동일 시간에 처리하였으며 체중과 음식섭취량은 8주 동안 매주 측정하였다. 8주 후, 혈중 콜레스테롤 수치를 측정하였으며, 간의 조직학적 분석과 SREBP-1c와 지방산의 발현을 조사하였다. 폴리코사놀은 농도-의존적으로 체중과 음식섭취량을 감소시켰다. 혈중 콜레스테롤 수치는 Pol-1과 Pol-4군에서 유의하게 감소하였으며, SREBP-1c와 FAS의 발현 역시 Pol-4 군에서 유의하게 감소하였다. 이러한 결과들은 폴리코사놀의 콜레스테롤 감소 효과가 SREBP-1c와 FAS의 발현 억제에 기인하여 일어날 수 있음을 시사한다.
미강 추출 상업용 유통 감마오리자놀의 콜레스테롤 자동산화에 의한 C-7 산화 콜레스테롤 유도체 생성 저해 효과가 수용성 모델 시스템을 이용하여 검토되었다. C-7 콜레스테롤 산화 유도체 (C-7 oxidized cholesterol derivatives: C-7 OCDs) 생성을 위해 콜레스테롤과 감마오리자놀이 분산된 수용성 모델시스템은 구리이온을 촉매로 pH 5.5와 $80^{\circ}C$의 가혹 조건에서 20시간 동안 반응되었다. 산화 유도 기간에 따른 C-7콜레스테롤 산화 유도체 (7-ketocholesterol, 7{\alpha}$-hydroxy-cholesterol과 7b-hydroxycholesterol)의 생성 정도와 감마오리자놀 및 콜레스테롤 변화 추이 정도가 핵산과 에틸아세테이트를 이용한 용매 추출법과 고속 액체크로마토그래프 (high-performance liquid chromatography) 테크닉을 이용 정량적으로 분석되었다. 분석 결과 콜레스테롤 산화 유도 기간에 따른 7-ketocholesterol 생성비율은 7-hydroxycholesterol 이성체 (${\alpha}$-형:${\beta}$-형) 대비 약 2:1의 비율로 생성되었으며, 7-hyoxycholesterol 이성체에 있어서는 ${\alpha}$-형 대비 ${\beta}$-형의 생성 정도가 약 1:2의 비율로 나타났고, 총 C-7 산화콜레스테롤의 생성은 상대적인 고농도(300 ppm) 감마오리자놀 처리 모델 시스템에서 효과적으로 저해되었다.

Cholesterol gallstones are a digestive disease of high prevalence that has many risk factors; for this reason, research has focused mainly on how to prevent it rather than how to treat it. Many molecules of the hepatic, bile and intestinal systems are involved in the pathophysiology of this disease, making it very difficult to find a therapeutic target. The pharmacological treatment is limited, so when gallstones generate symptoms, medical treatment indicates gallbladder removal. Ursodeoxycholic acid is used to dissolve cholesterol stones, and ezetimibe and statins are other drugs with possible applications in the treatment of this disease. Given the small number of drugs that have been developed for treating this disease, the research of natural products becomes of paramount importance. Resources such as black radish, glucosinolates, fenugreek, capsaicin, curcumin, garlic, and onion, have all shown significant effects in the prevention and treatment of cholesterol gallstones. In this review, we made a synthesis of the scientific reports that deal with these natural products and that can serve as antecedents for finding a way to treat the most common disease of the gallbladder.
Essential hypertension is considered to be a multifactorial disease that is influenced not only by environmental factors but also by genetic factors. Genes involved in lipoprotein synthesis, modification and metabolism are candidates for essential hypertension. The purpose of this study was to estimate gene frequencies of paraoxonase/arylesterase (PON1) gene in Korean population and investigate the relationship between genotypes of this gene and essential hypertension or cardiovascular risk factors. In order to estimate the genotype frequencies, Alw I RFLP of PON1 gene was used as genetic marker. There were no significant differences in allele and genotype frequencies between normotensives and essential hypertensives, respectively. However, Alw I RELP of PON1 gene were significantly associated with plasma HDL-cholesterol level in Korean population (one-way ANOVA test, p=0.008). Therefore, our result suggest that this RFLP of PON1 gene may be protective marker on cardiovascular disease in Korean population.
Smith-Lemli-Opitz syndrome (SLO) is a rare, autosomal recessive disease caused by an inborn error in cholesterol synthesis. Patients with this disease suffer from multiple malformations due to reduced activity of 7-dehydrocholesterol reductase (DHCR7), which increases 7-dehydrocholesterol (7DHC) and 8-dehydrocholesterol (8DHC) concentrations and decreases cholesterol concentration in body fluids and tissue. Here, we describe Korean siblings with SLO who were diagnosed recently, and performed a review of literature about Korean cases with SLO to date. Microcephaly and syndactyly of the second and third toes are the most common physical finding in SLOS patients. Other malformations including growth failure, cleft palate or bifid uvula, various heart malformation, genital ambiguity in males are also accompanied. Not all patients showed low levels of serum cholesterol, so DHCR7 mutation analysis can be helpful to confirmative diagnosis. Two mutations on p.R352 locus (p.R352W and p.R352Q) are commonly identified in Korean SLO patients. Although rare in Korea, SLO should be considered in the differential diagnosis of growth failure with intellectual disability, especially in patients with multiple congenital anomalies.
마늘 숙성 중 생성되는 S-allylmercaptocysteine의 콜레스테롤 생합성 억제 효과에 대하여 Hepatocytes를 이용하여 조사하였다. HepG2 cells을 Dulbecco's modified Eagle's medium (DMEM)에 배양하여 S-allylmercaptocysteine의 농도를 20, 40, 60, 80 및 100 mL 씩 각각 첨가하여 cell viability를 살펴본 결과 20~40 ${\mu}g/mL$에서는 높았으며, 60 ${\mu}g/mL$ 농도에서 약 50%가 유지되었다. S-allylmercaptocysteine을 5, 10, 15 및 20 ${\mu}g/mL$ 농도로 [$^{14}C$]-acetatecholesterol에서 처리하였을 경우 15 ${\mu}g/mL$ 농도에서 cholesterol 생합성이 79%로 억제되었다. Fatty acid synthase의 활성은 0.95 nmol에서 19%의 억제효과를 나타내었으나, Glucose 6-phosphate dehydrogenase (G6PDH)의 활성에는 거의 영향을 미치지 않았다. S-allylmercaptocysteine의 3-hydroxy-3-methylglutaryl CoA (HMG-CoA) reductase의 활성은 용량 의존형으로 감소하였다. 이상의 결과로 보아 마늘 숙성 과정에서 생성되는 주요 성분인 S-allylmercaptocysteine은 간 세포에서 cholesterol의 생합성을 억제하는데 기여하는 것으로 나타났다.
When 19-hydroxycholesterol acetate was added into CSD-10 in Nutrient Broth or in a mineral salts medium consisting of KH$_{2}$PO$_{4}$(0.1%), $K_{2}$HPO$_{4}$(0.1%), NH$_{4}$NO$_{3}$(0.1%), MgSO$_{4}$(0.02%), CaCl$_{2}$(0.002%), and FeCl$_{3}$(0.005%), a substantial amount of 19-hydroxyandrost-4-ene-3,17-dione was accumulated prior to accumulation of estrone. From this result and all of previous works, a tentative degradation pathway of 19-hydroxycholesterol acetate to estrone by CSD-10 was derived. 19-hydroxyandrost-4-ene-3,17-dione seems to be an attractive intermediate for the synthesis of 19-norsteroids.
중합반응을 할 수 있는 vinyl 기와 콜레스테롤 기를 갖는 계면활성제들을 합성하고, 이 계면활성제들을 물 속에서 sonication하여 vesicle 용액을 형성한 다음, 이 용액을 건조하여 분자들이 규칙적으로 배열된 분자 다층 구조를 갖는 막을 합성하였다. 이 막들을 UV를 이용하여 중합한 다음 분자량을 측정하여 중합성을 살펴보았으며, X-ray diffractometer로 분석하여 다층 구조를 어느 정도 잘하고 있는지극 살펴 보았다.
Tolerance of Mycobacterium sp. against organic solvents has been induced for the cholesterol side chain degradation by adding chemicals associated with synthesis of fatty acids or alcohols. Biotin of 300 .mu.g/1 and 0.5% aqueous ethanol solution were optima for the enhancement of ethanol tolerance of the microorganism. The induction of ethanol tolerance by biotin was found to be due to increase of degree of unsaturation of the fatty acids in membranous phospholipid of the cell, especially due to increase of oleic acid content. However when 0.5% of ethanol was added for the ethanol tolerance induction, there was an ambiguous correlation between ethanol tolerance and degree of unsaturation of the fatty acids, in spite of the fact that the induction increased the content of unsaturated fatty acids. Addition of 0.5% of ethanol induced several ethanol shock proteins having molecular weight similar to that of heat shock proteins.
