Effect of Monascus koji on Blood Pressure and Serum Cholesterol Composition of SHR by Chronic Dietary Administration (홍국 장기투여가 SHR의 혈압 및 혈중 콜레스테롤 조성에 미치는 영향)

  • The antihypertensive and cholesterol- lowering effects of Monascus koji (M. koji) prepared with Monascus ruber IFO32318 were examined in spontaneously hypertensive rats (SHR). Control was fed a normal diet and groups I, II or III were fed diets containing 0.03%, 0.1% or 0.3% M. koji for 8 weeks, respectively. After 8 weeks, all animals were fed normal diets in the following 2 weeks. The blood pressure of rats fed M. koji added diets were significantly attenuated as compared with control and the diet containing high concentration of M. koji had a tendency of stronger antihypertensive effect. These differences lasted throughout the experimental period when they were fed experimental diet. For 2 weeks after the 8 weeks of experimental diet all groups were fed the same normal diet and the differences of blood pressure caused by M. koji disappeared. In rats fed the M. koji added diet, the serum total cholesterol (TC), HDL cholesterol (HDL-C), LDL cholesterol (LDL-C) were not differed compared with control, but VLDL cholesterol (VLDL-C) was significantly lowered. M koji also significantly decreased serum risk factors, both TC/HDL-C ratio and non HDL-C/HDL-C ratio. Consequently, it is suggested that M. koji may play an important role to attenuate hypertension and to improve serum lipid Profiles.

Changes of the blood chemistry components in serum of the rat after oral administration of caffeine (Caffeine 투여시 Rat의 혈액내 혈액화학성분의 변화)

  • This study was conducted to identify the effects of caffeine on the change of blood chemistry components in the serum of the rat(Sprague-Dawley, female). The experimental groups were divided into 7 groups according to the time lapsed after a single oral administration of 100mg/kg caffeine(that is control, 2, 4, 8, 24, 48 and 72 hrs lapsed group) to the rats. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid and phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase (AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The results obtained from this study were summarized as follows ; 1. The concentrations of serum glucose were significantly higher($\rho$<0.01) between 4 (143.0 mg/dl) and 8 hrs(138.0mg/dl) in comparison to the control(101.1mg/dl) after a single oral administration of caffeine(100mg/kg). Whereas there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and albumin/globulin(A/G) ratio in comparison to the control. 2. The concentrations of total cholesterol and HDL-cholesterol in serum were significantly higher ($\rho$<0.01) between 4(77.4mg/dl, total cholesterol) and 8 hrs(64.7mg/dl, HDL-cholesterol) in comparis to the control(62.8, 46.7mg/dl) after a single oral administration of caffeine (100 mg/kg). On the other hand, the concentrations of triglyceride in serum were significantly lower($\rho$<0.01) after 8 hrs(38.8mg/dl) in comparison to the control(66.5mg/dl). 3. The activities of AST in serum was significantly higher($\rho$<0.05) from 2 hrs(149 U/L) to 8 hrs(178 U/L) in comparison to the control(112 U/L) after a single oral administration of caffeine (100mg/kg). The activities of ALT in serum were significantly higher($\rho$<0.01) at 4(45.5 U/L), 24(49.3 U/L), 48(46.8 U/L) and 72 hrs(42.3 U/L) in comparison to the control (39.7 U/L) after a single oral administration of caffeine (100mg/kg) On the other hand, there were no significant differences in the activities of ALP in comparison to the control.

Effects of Fish Oil and Some Seed Oils on Lipid Composition of Serum in Rats (어유 및 종자유의 급이가 흰쥐의 혈청 지질 성분에 미치는 영향)

  • This study was designed to observe the effects of the fish oil and some seed oils on the improvement of the lipid composition in rats. In order to induce the triglyceridemia in the rats of the Sprague-Dawley, 12% coconut oil and 3% each of olive oil, lard, fish oil, perilla oil, corn oil, red pepper seed oil and evening primrose oil were administered to the rats for tweets. Total cholesterol concentrations of serum were lower in the fish oil, perilla oil and corn oil groups and by for higher in the red pepper seed oil and evening primrose oil groups than in the olive oil group(control group). HDL-cholesterol concentrations were a little higher in the red pepper oil and evening primrose oil groups. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, all groups were higher percentage than the control group. Cholesteryl ester concentrations of serum were high in n-6 PUFA rich red pepper seed oil and evening primrose oil group. In the ratio of cholesteryl ester concentrations to total cholesterol, all groups(70.0~74.4%)were higher than the control group(62%). Phospholipid concentrations of serum were low in the fish oil and perilla oil groups and triglyceride concentrations were remarkably lower in the fish oil and evening primrose oil groups than in the control group. LCAT activities of serum were higher in the lard group than in the control group, but lower in the other groups.

  • Influence of magnesium and calcium on the serum cholesterol concentration in rabbit blood serum was studied. The subjects were fed with the basal diet and magnesium, calcium and Korea Ginseng. The results obtained are summarized as follows: The subjects body weight fed a basal diet, magnesium, calcium and ginseng powder added did not increased, but the weight of liver was increased. Total cholesterol level of the subject fed ginseng powder 4mg. added diet group showed the lowest concentration. The subjects fed with a basal diet and magnesium/calcium(1:2) added diet showed the lower than anyother group. In case of free cholesterol and cholesterol ester, the subject fed with ginseng powder tap. magnesium/calcium(1:2) added diet showed the lowest concentration. The subjects of the lowest concentration of total cholesterol showed the lowest concentration of low density lipoprotein(LDL) and high density lipoprotein(HDL)/total cholesterol ratio was the higher level than any other group. In case of triglyceride and phospholipid, the subject fed with ginseng powder showed the triglyceride concentration gradually do creased, but phospholipid level was increased positively. In case of electrolytes, sodium and potassium of the subject fed with magnesium, calcium and ginseng powder 4mg added diet showed 120mEq/l 1 and 6.8mEq/l, respectively.

Effects of Dietary Onion Supplementation on Growth Performance and Cholesterol Level of Blood in Finishing Pigs (비육돈 사료에 양파의 첨가가 성장 및 혈중 콜레스테롤 함량에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary onion supplementation on the growth rate and blood cholesterol level in finishing pigs. Sixty $Duroc\;{\times}\;Yorkshire\;{\times}\;Landrace$ pigs ($69.75{\pm}0.79kg$ average BW) were used in a 50 day growth assay. Dietary treatments included 1) corn-SBM (Con), 2) 1)+dietary onion rice bran 1.0% (ORB1), 3) 1)+dietary onion rice bran 2.0% (ORB2). For overall period, average daily weight gain and average daily feed intake were increased by the addition of dietary onion compared with Con (linear effect; p<0.01). Feed efficiency ratio was successfully improved by supplementation of dietary onion (linear effect; p<0.01). The total cholesterol concentrations in serum decreased as the concentration of onion in the diets was increased (linear effect, p<0.06). Also, LDL-cholesterol concentrations in serum tended to decrease in pigs fed onion than in those fed Con diet group (linear effect, p<0.04; quadratic effect, p<0.09). The HDL-cholesterol and triacylglyceride concentrations in serum were not influenced by the onion supplementation. The results obtained from this feeding trial suggest that the dietary onion supplementation for finishing pigs weighed 70 to 108 kg had improved growth performance and reduced total cholesterol level of serum.

Synthesis and Optimization of Cholesterol-Based Diquaternary Ammonium Gemini Surfactant (Chol-GS) as a New Gene Delivery Vector

  • Amongst a number of potential nonviral vectors, cationic liposomes have been actively researched, with both gemini surfactants and bola amphiphiles reported as being in possession of good structures in terms of cell viability and in vitro transfection. In this study, a cholesterol-based diquaternary ammonium gemini surfactant (Chol-GS) was synthesized and assessed as a novel nonviral gene vector. Chol-GS was synthesized from cholesterol by way of four reaction steps. The optimal efficiency was found to be at a weight ratio of 1:4 of lipid:DOPE (1,2-dioleoyl-L-${\alpha}$- glycero-3-phosphatidylethanolamine), and at a ratio of between 10:1~15:1 of liposome:DNA. The transfection efficiency was compared with commercial liposomes and with Lipofectamine, 1,2-dimyristyloxypropyl-3-dimethylhydroxyethylammonium bromide (DMRIE-C), and N-[1-(2,3-dioleoyloxy)propyl]-N,N,N-trimethylammonium chloride (DOTAP). The results indicate that the efficiency of Chol-GS is greater than that of all the tested commercial liposomes in COS7 and Huh7 cells, and higher than DOTAP and Lipofectamine in A549 cells. Confirmation of these findings was observed through the use of green fluorescent protein expression. Chol-GS exhibited a moderate level of cytotoxicity, at optimum concentrations for efficient transfection, indicating cell viability. Hence, the newly synthesized Chol-GS liposome has the potential of being an excellent nonviral vector for gene delivery.

Effects of Dietary L-Carnitine Supplementation on Lipid Metabolism in Rats fed with Different Types Protein (단백질의 종류가 다른 식이에 L-카르니틴을 첨가했을 때 흰쥐의 지질대사에 미치는 영향)

  • This study is to investigate the effect of dietary L-carnitine supplementation on lipid metabolism in rats fed with isolated soy protein and casein for their source of protein. Four experimental groups were organized and each group had eight Sprague-Dawley male rats with the initial weight of around 180g. The four groups were CO (casein only supplemented group); CC (casein and 3% L-carnitine supplemented group); ISO (isolated soy protein only supplemented group); ISC (isolated soy protein and 3% L-carnitine supplemented group). All groups were supplemented with the experimental diet for four weeks and carnitine comprised 3% of. their diet. The results were as follows; 1. There was no significant difference in food intake among the groups. 2. Final weight gain was significantly lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (P<0.05). The groups with supplemented casein and carnitine showed the effect of weight reduction (p<0.05). 3. Food efficiency ratio was lower in the groups supplemented with isolated soy protein than in the groups supplemented with casein (p<0.01). The groups supplemented with casein and carnitine showed low food efficiency ratio. 4. The serum total lipid was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein (p<0.05). 5. Serum total cholesterol was higher in the groups supplemented with casein than in the groups supplemented with isolated soy protein. 6. There was no significant difference in triglyceride, HDL-cholesterol, and LDL-cholesterol in serum among the groups. 7. Out of the groups supplemented with isolated soy protein the total cholesterol level in liver was low in the groups to which carnitine was supplemented (p< 0.05). However, there was no significant difference of liver total lipid and triglyceride among the groups. 8. There was no difference in TBARS levels and GSH-Px activities in liver among the groups.

Age-related Circulating Inflammatory Markers and Cardiovascular Disease Risk Factors in Korean Women (한국 성인 여성에서 연령에 따른 혈중 염증 표지자와 심혈관계 질환 위험 요인에 대한 연구)

  • The purpose of this study was to investigate the age-related changes of cardiovascular disease risk factors and inflammatory markers in non-obese Korean women. Subjects were 112 women over 20 years old with body mass index (BMI) less than $30 kg/m^2$ and were divided into 3 groups (< 40 years, $40{\sim}59$ years, ${\ge}60$ years). Mean weight and BMI in the oldest group were significantly higher than those in the other 2 younger groups (p < 0.05). Mean total cholesterol, triglyceride, LDL-cholesterol and apolipoprotein B/apolipoprotein A1 ratio (BAR) in the oldest group were significantly higher than those in the youngest group (p < 0.05), and mean HDL-cholesterol of the oldest group was significantly lower than that of the youngest group (p < 0.05). The older-aged group showed significantly higher mean values of atherogenic index (AI) and LDL/HDL ratio (p < 0.05) than the respective younger-aged group, and AI was significantly correlated with age, nitric oxide and thiobarbituric acid reactive substances (p < 0.01). In addition, mean vascular cell adhesion molecule-l (VCAM-1) tended to be higher in the older-aged group than the younger group. Tumor necrosis factor-${\alpha}$, a proinflammatory maker, was significantly positively correlated with serum homocysteine, a cardiovascular disease risk factor (p < 0.01). In addition, a significantly positive correlation was observed between C-reactive protein and BAR (p < 0.01). Overall results suggested that the aging might affect the increase of cardiovascular disease risk factors including the serum lipid profiles, weight and BMI, and age-related increases of weight and BMI might play a role in changes in certain biomarkers of inflammation.   • This study was attempted to investigate the effect of Composite Preparation (Sam Whang Sasim-Tang: SWST, Whang Ryun Haedok-Tang: WRHT) on the activities of GOT and GPT, the content of total lipids, triglyceride, total cholesterol. Phospholipid and ${\beta}-lipoprotein$ in the serum, and the change ratio of body and liver weight in the experimentally induced hyperlipemic rats, making use of the extract 150, 200, 300 and 500 mg/kg p.o. Significant test was performed by comparision with the values of corresponding experimentally hyperlipemic rats. The activities of S-GPT was significantly decreased in all dose of SWST and WRHT respectively. The activities of S-GPT was significantly decreased in dose of SWST 500 mg/kg and in all dose of WRHT respectively. The content of total lipids, triglyceride, total cholesterol and phospholipids were significantly decreased in all dose of two the extract. The rate of decrease on total lipids and triglyceride were remarkable in dose of SWST 300, 500 mg/kg and WRHT 200, 500 mg/kg, and then the content of total cholesterol was more remarkable in dose SWST 300, 500 mg/kg, and WRHT 150, 300 mg/kg, and the content of phospholipids was more remarkable in dose of the extract 150, 200 and 300 mg/kg. The content of ${\beta}-lipoprotein$ was significantly decreased in dose of WRHT 150, 200 and in all dose of SWST. Increase ratio of the body and liver weight were significantly decreased in dose of two the extract 300, 500 mg/kg respectively.

The Effect of Weight Control on Obese Women (비만 여성의 체중조절 실시를 통한 효과 측정)

  • This study was conducted to investigate the effect of weight control by use of commercial formula diet and nutrition education on 36 obese women over a period of 6 months. During the study it motivated ways for women to control their weight and improve their health, thus analyzing variables that influence weight control. To help weight control, for the first 3 weeks, along with 1 regular mea, 2-3 liquid formula diets of 135㎉ per pack a day were provided within the 1,200-800㎉ range. For 6 months since then, all 3 meals were taken regularly and it was recommended that energy which is 500㎉ less than energy requirement be taken. To evaluate the effect of weight change by the weight control program, anthropometric measurements, biochemicl parameter, and food behavior are measured. The mean body mass index(BMI) was 30.1$\pm$3.8(25.0-43.6) and it was in the upper 5 percentile of the mean BMI of Korean women. The percent of body fat was significantly reduced from 29.1$\pm$2.4 to 26.4$\pm$2.3, showing that the weight control program induced not only a change in weight but also a change in body composition. The body circumference such as waist and hip circumference and skinfold thickness measurements of biceps, triceps, subscapular and surprailiac were reduced significantly, but the mean waist-to-hip ratio didn't change. Singnificantly improved food behavior score measured by the questionnaire indicated that the weight-contorl program induced the change of the food behavior, as well. Weight loss also induced biochemical parameters of the subject, thus total cholesterol, LDL-Cholesterol, and triglycerol which were in the high range of the normal values were reduced to 20.2%(p<0.01), 22.2%(p<0.01), 25.8%(p<0.01), repectively. HDL-cholesterol didn't change significantly, but the ratio of HDL to total cholesterol increased(p<0.01).
