The Effect of Feeding Juice and Powder of Houttuynia cordata Thunb on Serum Lipids in Rats (어성초 즙과 분말을 첨가한 식이가 흰쥐의 혈청지질개선에 미치는 영향)

  • This study was designed to observe the effects of feeding Houttuynia cordata juice and powder on the improvement of the lipids in the serum of dietary hypercholesterolemic rats. Experimental groups such as 5% cellulose for control group, 5%, 10%, 15%, 20% H. cordata powder for group 2, 3, 4, 5, respectively, and 5%, 10%, 15% H. cordata juice for group 6, 7, 8, respectively, were admi nistered to the male rats of the Sprague Dawley for 4 weeks. Concentrations of total cholesterol in serum were significantly lower in all experimental groups than in the control group, and particularly, the lowest in group 8. The ratio of HDL cholesterol to total cholesterol was the highest in group 5. Atherosclerotic index was lower in group 5 and 8. Concentrations of free cholesterol and cholesterol ester in serum were significantly lower in all experimental groups than in the control group, and then concentrations of free cholesterol and cholesterol ester were the lowest in group 5 and 8, re spectively. Concentrations of triglyceride and phospholipid were the lowest in the group 5. Con centrations of chylomicron and VLDL were significantly lower in all experimental groups than in the control group. And the concentration was remarkably the lowest in group 5. Concentration of glucose in blood was not significantly different, but lower in the group 5 and 8. Concentrations of Ca, P, Mg, Fe were not significantly different in all experimental groups. The GOT and GPT activites were lower in all experimental groups than in the control group, especially the lowest in group 7 of GOT and group 8 of GPT. The LDH activity was the lowest in group 8.

Tu-Chung Leaf Meal Supplementation Reduced an Increase in Lipid Accumulation of Chickens Stimulated by Dietary Cholesterol

  • The effect of tu-chung (Eucommia ulmoides, Oliver) leaf meal on reducing lipid accumulation in chickens fed 1% cholesterol containing diet was studied. Forty male White Leghorn chickens aged 56 days were weighed and divided into four groups of ten chickens, and fed diets with or without 1% dietary cholesterol which were supplemented with 0 and 5% tu-chung. Tu-chung supplementation to the diet without cholesterol increased acetyl-CoA carboxylase (p<0.01) but decreased 3-hydroxy-3-methylglutaryl-CoA reductase activities (p<0.01) with no effect on fatty acid synthetase activities. However, its supplementation to the diet with cholesterol had no effect on these three enzyme activities as compared with the cholesterol containing diet without tu-chung. Tu-chung supplementation to the diet without cholesterol increased hepatic triglyceride (p<0.01), whereas its supplementation to the diet with cholesterol decreased it (p<0.01). Tu-chung supplementation to the diet with cholesterol decreased plasma cholesterol ester, free cholesterol, phospholipids (p<0.05) and triglyceride (p<0.01) as compared with the cholesterol containing diet without tu-chung. Supplementation of tu-chung to the diet without cholesterol decreased plasma free cholesterol (p<0.05). It is concluded that tu-chung leaf meal reduced an increase in lipid accumulation in chickens stimulated by 1% cholesterol feeding.

Effects of High Polyunsaturated Fat Diet on Human Plasma Cholesterol Esterification and Transfer (고 불포화 지방산 식이가 인체 혈청 Cholesterol의 Esterification과 Transfer에 미치는 영향)

  • The reaction of lecithin cholesterol acyltransferase(LCAT) and cholesteryl ester transfer protein (CETP) are important in cholesterol esterification and transfer for the reverse cholesterol transport(RCT) system. The purpose of this study were to assess the effects of fatty acid unsaturation on RCT system. After 12 female human subjects consumed a prescribed high saturated fat diet prior to the period, two groups of six subjects were provided with a high PUFA(corn oil) or a high SFA(butter) as major fats in a 40 en % fat diet. Butter feeding increased plasma total-(34%), esterified-(96%), HDL_3$-(23%), LDL-(20%), and VLDL plus LDL(35%) cholesterol while corn oil feeding decreased esterified-(25%), LDL(15%) cholesterol and TG(27%). There were significant differences of fats on total-(p=0.0001), esterified-(p=0.0001), total HDL-(p=0.005), $HDL_2$-(p=0.01)and LDL-(p=0.0001) cholesterol. LCAT activity did not change during the period but highly correlated to apo A-I in HDL which was increased in the corn oil group. The 2.5 fold increased CETP activity in the butter group during the period might be related to changes in plasma VLDL plus LDL cholesterol level which were increased in the butter group.

Effects of Fish Oil and Some Seed Oils on Lipid Composition of Serum in Rats (어유 및 종자유의 급이가 흰쥐의 혈청 지질 성분에 미치는 영향)

  • This study was designed to observe the effects of the fish oil and some seed oils on the improvement of the lipid composition in rats. In order to induce the triglyceridemia in the rats of the Sprague-Dawley, 12% coconut oil and 3% each of olive oil, lard, fish oil, perilla oil, corn oil, red pepper seed oil and evening primrose oil were administered to the rats for tweets. Total cholesterol concentrations of serum were lower in the fish oil, perilla oil and corn oil groups and by for higher in the red pepper seed oil and evening primrose oil groups than in the olive oil group(control group). HDL-cholesterol concentrations were a little higher in the red pepper oil and evening primrose oil groups. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, all groups were higher percentage than the control group. Cholesteryl ester concentrations of serum were high in n-6 PUFA rich red pepper seed oil and evening primrose oil group. In the ratio of cholesteryl ester concentrations to total cholesterol, all groups(70.0~74.4%)were higher than the control group(62%). Phospholipid concentrations of serum were low in the fish oil and perilla oil groups and triglyceride concentrations were remarkably lower in the fish oil and evening primrose oil groups than in the control group. LCAT activities of serum were higher in the lard group than in the control group, but lower in the other groups.

  • This experiment was designed to observe some influence on the serum components of growing rats maintained with basal diet supplemented with sausage. Forty male Albino rats were used as the experimental animals and the subjects were divided into four diet groups (control group; feeding of basal diet containing 10.8% and 10.4% of protein and lipid respectively, A group; feeding of sausage containing 12.5% and 27.6% of protein and lipid respectively supplemented with basal diet every day, B group; feeding of sausage supplemented with basal diet once a three-day and C group; feeding of sausage supplemented with basal diet once a five-day) and each group was fed on the corresponding diet for forty-days. The contents of protein, lipid and cholesterol in their serum were determined. The result obtained are summarized as follows: 1) The final body weight and liver weight of the A group were increased more than other groups. 2) The protein contents in the serum of the B group was increased more than other groups. 3) The total lipid contents in the serum of the A group and B group were decreased compared with control group. 4) The total cholesterol, ester form cholesterol and free form cholesterol in the serum of the A group were decreased, but the total cholesterol and ester form cholesterol in that of the C group were increased compared with control group.

Changes in Fatty Acid and Cholesterol Composition of Koran Styled Beef Broths (Gom-Guk) during Cooking (쇠고기 곰국의 조리중 지방산 및 Cholesterol 조성변화)

  • The content of total lipid, cholesterol and fatty acid in Korean styled various beef broths was examined before and during toiling to determine the influence of cooking time. The values obtained from the broths of brisket, ox-tail and small-intestine were determined by using G. C. The highest total lipid value among small-intestine, brisket and ok-tail was in the small-intestine. The content of total lipid in the boiled meat decreased during cooking with the exception of the brisket. Total lipid content found in the broth of brisket meat increased until 6hr and thell decreased at cooking time of 8hr and 10 hr. The highest total lipid contents in the broths of brisket, ox-tail and small-intestine were found when the cooking time were 6 hr, 10 hr and 4 hr, respectively. The main fatty acids found in the broths of brisket, ok-tail and small-intestine were $C_{14}$, $C_{16}$, $C_{18}$, $C_{18:1}$, and $C_{18:1}$. The content of $C_{18:1}$ and $C_{18:2}$ from the brisket meat increased until 4hr's cooking, especially $C_{18:2}$ increased significantly until 6 hr cooking. The highest ratios of unsaturated to saturated fatty acid (UNS/S) in the meat of brisket and ox-tail were found in the 2hr and 4hr cooking, respectively. The ratios of UNS/S in the broths of both of brisket and small-intestine, and ok-tail were highest in the 4 hr and 2 hr cooking, respectively. The content of total cholesterol (TC) was the highest in the small-intestine and the highest percentage of ester cholesterol was 5.4 in the brisket. The content of TC in small-intestine, brisket and ox-tail was decreased significantly after 2 hr cooking. Precentage of the ester in the brisket was the highest when the cooking time was 8 hr. Only trace amount of total cholesterol was found in the broth.

Development of Flow Injection Analysis System for Amperometric Determination of Cholesterol Using Immobilized Enzyme Columns (고정화 효소컬럼을 이용한 콜스테롤 측정용 Flow Injection Analysis 시스템의 개발)

  • A flow injection analysis(FIA) system was developed for the determination of cholesterol using immobilized cholesterol oxidase and cholesterol ester hydrolase. The enzymes were immobilized on controlled pore galas(CPG) by the glutaraldehyde method. The glass colunms packed with immobilized enzymes were found to contain 3-5 I.U. for each enzyme. A hydrogen peroxide sensitive electrode was contructed and applied to the FIA system. The operational conditions for FIA response were investigated and optimized with variation of sampling volume, flow rate and composition of carrier solution. The FIA response were linear upto 60 and 400mg/m1 for free cholesterol and cholesterol ester, respectively. All samples were analyzed with a good precision (<2.5% CV) and accuracy. 23 samples were mea sured succesively within about an hour. Intermittent assays of more than 500 times caused 50% decrease in response sensitivity.

Effect of Meristotheca papulosa on Lipid Concentration of Serum and Liver in Rats Fed High Fat Diet (해조류 갈래곰보가 고지방 식이를 급여한 흰쥐의 혈액 및 간장 지질 농도에 미치는 영향)

  • This study was carried out to investigate the effect of Meristotheca papulosa on lipid concentration of serum and liver in rats fed high fat diet for 4 weeks. Experimental groups ere divided into normal group (5% corn oil), high fat diet group (5% corn oil+20% lard), high fat diet and 5% or 10% Meristotheca papulosa treated group. The weights of kidney, heart and tests were not significantly different among the group, but weight of liver was significantly decreased by supplement of Meristotheca papulosa (10%). The contents of serum total lipid, trigly-ceride, total cholesterol, free cholesterol, cholesterol ester and atherogenic index were decreased, while HDL-cholesterol was increased by Meristotheca papulosa supplementation compared to high fat diet group. Meristotheca papulosa supplement has obvious effects on lowering hepatic total lipid, triglyceride and total cholesterol. Our findings suggest that Meristotheca papulosa supplement can improve lipid metabolism in high fat fed rats.

Effect of Platycodon Grandiflorum Aqueous Extract on Lipid Levels in Rats

  • This study investigated the effects of Platycodon grandiflorum aqueous extract on lipid concentration of serum and liver in rats fed high cholesterol diet Male Sprague-Dawley rats were assigned to three groups (control, low dose of extract, high dose of extract) for four weeks. The serum total cholesterol concentration was significantly lower in the low and high doses of extract groups than in the control group. There was a significant decrease in the free cholesterol, cholesterol ester, LDL-cholesterol, and triglycerides concentrations in serum, and the total cholesterol and the triglycerides contentin liver in the low and high doses of extract groups compared to the control group. When the serum phospholipid concentration was compared among the groups, it was significantly lower in high dose of extract group than in control and low dose of extract group. It can be postulated that Platycodon grandiflorum aqueous extract may possess substantial hypolipidemic properties in rats.

  • Two new metabolites have been characterized from the methylene chloride extracts of Bezoar Bovis, a well known oriental drug : 8-octadecenoic methyl ester and cholesta-3,5-diene. Their structures were deduced from $^1$H-NMR and GC-mass spectra.
