Development of Phytosterol Ester-added Cheddar Cheese for Lowering Blood Cholesterol

  • This study was carried out to investigate the effect of phytosterol ester addition on lowering blood cholesterol in cholesterol-reduced Cheddar cheese. For cholesterol removal, separated cream was treated with 10% ${\beta}$-cyclodextrin at 800 rpm, then blended with remaining skim milk and homogenized with 1,000 psi at $70^{\circ}C$. Experimental cheeses were manufactured by five different levels of phytosterol addition. After the cholesterol reduction process by ;${\beta}$-cyclodextrin, the cholesterol removal rate was in the range of 91.0 to 92.1%. Amount of short-chain free fatty acid and free amino acids increased with an increase of phytosterol ester, and those were significantly different from that of control in all ripening periods. All rheological properties also increased with an increase of phytosterol ester during ripening period. In sensory analysis, the scores of rancid, bitterness Cheddar flavor and off-flavor intensities increased significantly, while texture was decreased during ripening in phytosterol ester-added groups. Total blood cholesterol was reduced by 18% when rats were fed Cheddar cheese treated with 8% phytosterol. The present study indicated that phytosterol ester addition resulted in a profound lowering effect of blood with cholesterol-reduced Cheddar cheese.

  • This study was carried out to investigate the effect of phytosterol ester addition on lowering blood cholesterol in cholesterol-reduced Cheddar cheese, which was manufactured by the mixture of cholesterol-reduced cream and skim milk. After the cholesterol reduction process by ${\beta}$-CD treatment, the cholesterol removal rate was in the range of 91.2 to 92.1%. In animal study, 18% of total blood cholesterol was lowered in 8% phytosterol ester-added Cheddar cheese, which was significantly different from that of control. The present study indicated that phytosterol ester addition showed a profound lowering effect of blood with cholesterol-reduced Cheddar cheese.

A Korean Family with Cholesterol Ester Transfer Protein Deficiency

  • A 32-year-old female patient and her sister show high levels of high density lipoprotein (HDL) cholesterol in regular health checkups, since female patient was 11 years old. The patient's serum total cholesterol was 285 mg/dL and HDL cholesterol was 113 mg/dL. Her sister's total cholesterol was 240 mg/dL and the HDL cholesterol measured to be 90 mg/dL. Lipoprotein pattern and cholesteryl ester transfer activity gene analysis were examined in these patients. We found c.1321+1G>A (IVS14+1G/A) hetero mutation in cholesteryl ester transfer protein (CETP) genes. Generally, CETP mediates transfer and exchange of triglycerides and cholesteryl ester between plasma lipoproteins. Also we investigated a key role of HDL-CE and Apo A-1 metabolism. Patients with low levels of CETP have increased serum HDL levels. We hereby report two Korean cases of CETP deficiency in a family. Brief literature review ensues with the cases.

  • The effect of milk in low and high cholesterol diets were investigated on serum cholesterol esterification. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high (1.01% w/w) cholesterol-diets groups. Bothlow and high cholesterol groups were consisted of three groups : control , LM(low heat milk), and HM (high heat milk) groups. After feeding these experimental diets for six weeks, serum cholesterol (free cholesterol and cholestryl ester) concentration and activity of lecithin.: cholesterol acyltransferase (LCAT) were measured, and serum lipoprotein profile was examined using gel column chromatography. According to the result, activity of LCAT was elevated independently by intakes of high cholesterol and milk , which resulted in the increase of daily turnover of serum cholesteryl ester. However, the turnover of HDL-cholesteryl ester increased only by milk. LCAT activity was moderately correlated with levels of total-and HDL-free choelsterol. It is concluded from the present study that milk had the cholesterol-lowering effect which partly appears to be mediated through facilitated reverse cholesterol transport.

어성초 분말 및 즙의 급이가 고콜레스테롤혈증 흰쥐의 간장, 뇌 및 신장의 지질성분에 미치는 영향 (Effects of Houttuynia cordata Thunb Powder and Juice on Lipid Composition of Liver, Brain and Kidney in Dietary Hypercholesterolemic Rats)

  • For the investigation to the effects of H. cordata on prevention of hypercholesteromia, dietary hypercholesteromic rats were fed for 4 weeks with basial diet containing 5, 10, 15, 20% H. cordata powder, 5, 10 and 15% H. cordata juice. Total cholesterol and triglyceride contents of livers were significantly lower in experimental groups than in control group. Phospholipid contents in livers were not significant difference between control and experimental groups. Contents of free cholesterol in livers were ranged from 0.4$\pm$0.2 to 0.7$\pm$0.5mg/kg and cholesteryl ester contents were lower in H. cordata juice added groups than H. cordata powder added groups. Total cholesterol content in brains were not significant difference between cholesterol not added, H. cordata powder added at 15, 20% and its juice added group at 15%. The more powder and juice of H. cordata were added to basial diets, the lower contents of triglyceride and phospholipid were detected in brains. Contents of free cholesterol and cholesteryl ester in brains were not significance in all experimental groups. In kidneys, total cholesterol contents were not significant in control group and H. cordata powder of 5% was added group. Triglyceride, phospholipid and cholesteryl ester contents in kidney were not significant in all groups. Contents of free cholesterol in kidney were equally significant between cholesterol free and 15% H. cordata juice treated group.

한국산 해면의 지질 성분 연구 (Lipids Constituents of the Korean Marine Sponges)

  • 한국 남해안 소흑산도에서 채집한 해면 V.japonica H.로부터 통상적인tetradecanoic 및 hexadecanoic methyl ester 이외에 탄소가 홀수인 pentadecanoic, heptadecanoic methyl ester와 가지달린 12-methyltetradecanoic, 14-methylpentadecanoic, 15-methylhexadecanoic 및 14-methylhexadecanoic methyl ester가 발견되었으며, 주 sterol 성분으로는 cholesterol과 소량의 ergost-25-ene-3,5,6-triol이 검출되었다. 한편, 여수 돌산도에서 채집한 해면 H.sinapium의 주 sterol성분은 5${\alpha}$-cholestan-3${\beta}$-ol이었고 cholesterol은 소량으로 얻어졌다. 지방산으로는 팔미틴산, 팔미토레인산, 올레인산, 스테아린산 및 리놀레인산 에틸에스터 이외에 상당량의 arachidonic산 에틸에스터가 함유되어 있었다. 상기 지역에서 채집한 또 다른 해면에서는 상당량의 tetradecanoic tetradecyl 및 hexadecyl ester와 sterol 성분으로 cholesteryl acetic 및 fatty acid ester가 함유되어 있었다.

백하수오 추출액이 고지혈증 및 Streptozotocin 유발 당뇨성 흰쥐의 혈청 지질성분 및 효소활성에 미치는 영향 (Effects of Cynanchum wilfordii Extract on Serum Lipid Components and Enzyme Activities in Hyperlipidemic and Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was designed to observe the effects of the feeding Cynanchum wilfordii extract on the improvement of the blood glucose, lipid components in the serum of dietary hyperlipidemic and streptozotocin(STZ) -induced diabetic rats(S.D. strain, ♂) fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-Cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose in serum were significantly higher in the cholesterol administration groups((group 2(cholesterol+water), 4(cholesterol+Cynanchum WIlfordii 3.5g% extract)) than those in the control group (group1 , basal diet+water). But the concentrations of total cholesterol. atherosclerotic index, LDL, LDL- cholesterol. free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum were remakably lower in the group 4 than those in the group 2. In the STZ(55mg/kg B.W.)-induced diabetic groups((group 3(STZ, IP.)+water), 5(STZ(IP.)+Cynanchum WIlfordii 3.5g% extract? the serum total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose concentrations actions were rather lower in the group 5 than those in the group 3. In the ratio of HDL -cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Cynanchum wilfordii extract administration groups were higher percentage than III the groups 2 and 3. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase (ALP) and lactate dehydrogenase(LDH) in serum were rather lower in the Cynanchum wllfordii extract administration groups (group 4,5) than in the cholesterol diet group(group 2) and STZ-induced diabetic group (group 3). From the above research, the physiological activity substances in Cynanchum wllfordii were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic and STZ-induced diabetic rats. And particularly, physiological activity substance in Cynanchum wilfordii was more effective therapeutic regimen for the control of metabolic derangements in adult disease.

蔗糖과 葡萄糖이 白鼠肝臟 및 血淸 Cholesterol의 含量에 미치는 影響 (The Effect on the Change of Liver and Serum Cholesterol Contents of Albino Rats by the Feeding of the Suerose and Glucose)

  • 成熟雄性白鼠를 標準食餌群과 10%, 25% 및 40% 葡萄糖食餌群 그리고 10%, 25% 및 40% 蔗糖食餌群으로 나누어 各核當食餌를 2개월간 급식시킨후 肝臟 및 血淸中 cholesterol 含量을 觀察한바 다음과 같은 結論을 얻었다. 1. 肝臟內 總臟器當 cholesterol 含量은 total cholesterol 과 ester 型 cholesterol 은 標準食餌群에 比하여 40% 葡萄糖食餌群은 현저히 증가하였고 free 型 cholesterol 은 標準食餌群에 比하여 各食餌群 모두 증가하였다. 2. 血淸 100ml 中 cholesterol 含量은 total cholesterol 에서는 標準食餌群에 比하여 10% 葡萄糖食餌群 및 蔗糖食餌群은 감소하였으나 40% 蔗糖食餌群은 증가하였다. free型 cholesterol 은 10%, 40% 葡萄糖食餌群 및 10% 蔗糖食餌群에서는 減少하였고 ester型 cholesterol은 10%蔗糖食餌群 및 葡萄糖食餌群에서는 감소하였으나 40%蔗糖食餌群에서는 증가하였다. 이상의 결과로 보아 체내 cholesterol 含量은 食餌中 蛋白質의 量이 一定할때도 食餌性糖質의 量과 種類에 따라서 영향을 받는다는 事實을 알수 있다.

가시발새우 키토산 식이가 식이성 고지혈증 흰쥐의 지질대사에 미치는 영향 (Effects of Dietary Lobster Shrimp Chitosan on Lipid Metabolism in Diet-Induced Hyperlipidemic Rats)

  • This study was designed to investigate effects of lobster shrimp chitosan on serum and liver lipid levels in rats fed high fat and cholesterol diets. Male Sprague-Dawley rats of 4-weeks old, weighing 70$\pm$5g, were fed experimental diets for 3 weeks: basal diet+5% cellulose(group 1), high lipid diet+5% cellulose(group 2, control group), high lipid diet+5% balloon flower root(group 3), high lipid diet+5% cellulose(group 4, high lipid diet+5% lobster shrimp chitosan(group 5). Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid in serum and liver were determined. The results obtained were as followes: Serum total cholesterol concentration were significantly reduced in all supplemented group compared with control. HDL-cholesterol concentrations were higher in groups 1, 4, and 5 and the ratios of HDL-cholesterol to total cholesterol concentration were higher in groups 1, 4, while the atherogenic-indices were lower in groups 1, and 4 than control. Concentrations of LDL, LDL-cholesterol and free cholesterol, cholesteryl ester were significantly lower in all supplemented groups than the control group. Concentrations of phospholipid and triglyceride were lower in groups 3, 4, 5, while activities of GPT and GOT were significantly lower in groups 4, 5 than the other group. Concentrations of calcium, magnesium and phosphorous were high in all treatment groups compared to the control group. Liver contents of total cholesterol, free cholesterol and cholesteryl ester were lower in the groups 1, 3, 4, and 5 than in the control group, especially lower in groups 1, and 5. Liver triglyceride and phospholipid concentrations were significantly lower in the groups 1, and 5 than in the other groups. The results from the data suggested that the experimental diet containing 5% lobster shrimp chitosan was effective to reduce the serum and liver lipids.

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats.
