Effects of Morphological Characteristics and Biomass on Germination and Growth of Bulbil for Production of Leaf Garlic (잎마늘 생산을 위한 마늘 주아의 무게 및 형태가 발아와 생장에 미치는 영향)

  • The objective of this study was to find out the most optimal bulbil size and planting densities of garlic bulbils to germination for production of leaf garlic. In the first this experiment, the bulbils classified by the size of bulbil to Big (>0.2) and Small (${\leq}0.2$), were planted at $13,680bulbils/m^2$ and $18,240bulbils/m^2$ (Big), $18,240bulbils/m^2$ and $22,800bulbils/m^2$ (Small), respectively. The germination rate, plant height and total yield were measured. In the second experiment, length, width, fresh weight and dry weight of 200 garlic bulbils were measured to analyze the correlation of days to first germination, T50 and germination rate. In the first experiment, the germination rate showed the highest at Big bulbils. The density of $18,240bulbils/m^2$ was the most suitable for cultivation because of the highest germination rate. In the case of yield, it was found that both of the fresh weight and the dry weight of Big bulbil was 2.8 times heavier than Small bulbil. Therefore, Big bulbil with the density of $18,240bulbils/m^2$ was the most suitable for cultivation because of the highest yield. The correlation between each factor of morphological characteristics had a positive correlation between fresh weight, dry weight and width. Also, the germination rate had a positive correlation with fresh weight and width. And, days to first germination and T50 had a negative correlation with fresh weight and width. In conclusion, the optimal planting density is $18,240bulbils/m^2$ and the optimal size of bulbil was 0.2g or more. However, thicker and heavier bulbils could lead to higher and faster germination.

Hypoglycemic Effect of Culture Broth of Bacillus subtilis S10 Producing 1-Deoxynojirimycin (1-Deoxynojirimycin을 생산하는 Bacillus subtilis S10 배양액의 혈당강하 효과)

  • 1-Deoxynojirimycin (DNJ) is a strong $\alpha$-glucosidase inhibitor which inhibits hyperglycemia in animals. To select the Bacillus strains highly producing DNJ, 4,000 strains were isolated from soil and grain samples. By the inhibitory activity against $\alpha$-glucosidase, nine Bacillus strains were selected and then identified by 16S rDNA sequencing. B. subtilis S10 was finally selected as the best strain for the production of DNJ. Various carbon sources and nitrogen sources in culture medium were evaluated for the highest production of DNJ. As the results, the optimized concentration of carbon source and nitrogen source was 1.0% galactose and 1.6% polypeptone and the concentration of DNJ produced was 0.75 g/L. The effect of culture supernatant of B. subtilis S10 on lowering blood glucose level was investigated in streptozotocin (STZ)-induced diabetic mice model. Mice were randomly assigned to control group (saline) and three test groups such as acarbose group, silkworm powder group and B. subtilis S10 group. After eight-week oral feeding, blood glucose levels of the B. subtilis S10 and silkworm powder groups were respectively $209.1{\pm}19.6\;mg/dL$ (59.1%) and $208.6{\pm}39.8\;mg/dL$ (59.0%) lower than $510{\pm}10\;mg/dL$ of the control group. These results indicated that the culture supernatant of B. subtilis S10 was able to reduce the blood glucose level in STZ-induced diabetic mice.

A Study on the Maintenance Plan of Trees in Gwanghalluwon Garden for Representing Traditional Space (전통공간 재현을 위한 광한루원의 수목정비방안 연구)

  • This study aims to draw up measures to planting tree and maintain a landscape in traditional space. Preceding comprehensive theoretical consideration of selected species of trees and tree maintenance. And analysis of present condition of planting in cases of Gwanghalluwon Garden, then draw a maintenance plans of planting through species of trees and landscape of planting recorded in literature. The results were as follows. First, Analysis of selected species of trees and tree maintenance that traditional space. A dispute about the selection species of trees in traditional space has been continued until today. Because unconditional reject of foreign trees are limited. In this context, should be sublated that hasty blind faith of records and dichotomous preparation plans such as removal of foreign trees and implicitly planting of native plants. Secondly, Planted trees in Gwanghalluwon garden was investigated and found to the species of trees used in traditional space such as Pine(Pinus densiflora S. et Z.), Sawleaf Zelkova(Zelkova serrata), Ginkgo(Ginkgo biloba), Crape Myrtle(Lagerstroemia indica L.). But, present planting irrelevant to traditional space, except Gwanghallu pavilion area from the spatiality. Thirdly, A look at the records or literature that maintenance of planting through historical research are limited. Because literature was recorded Salix spp., Crape Myrtle, Bambusoideae(Pseudosasa japonica), lotus only among planted trees in Gwanghalluwon garden. Fourth, Gwanghallu zone have nature of history and sense of place. And It was going to restore the appearance on historical. Consequently maintenance plan of planting of Gwanghallu zone should be maintain the current state. Wanwol pavillion zone can be recognized as the original form because they look similar to the Gwangallu zone's buildings. Therefore, it is necessary to secure the sense of place different from Gwanghallu zone by buffer planting for composition of transition space. Wolmaejip zone and lawn zone was marketplace in outside of castle and large forest. Accordingly, this area should be symbolic restoration of the Yulrim(栗林) and representation of the marketplace in outside of castle through aggregation of facilities and administrative facilities in Gwanghalluwon garden. East lawn of the Wanwol pavilion zone is should be maintained the current state that opened place in terms of using thought linked with the Wanwol pavilion zone. Boundary zone of the Gwanghalluwon garden is difficult to associate in terms of historical research and authenticity. Therefore, application of cultural landscape that appeared in literature is be worth.

Differential Sensitivity of Rice Cultivars to HPPD-Inhibiting Herbicides and their Influences on Rice Yield (HPPD 억제 제초제에 대한 벼 생태형간 약해 차이 및 수량 영향)

  • A field study was conducted at Miryang in 2011 to evaluate differential sensitivity of six rice cultivars, three Indica${\times}$Japonica rice (long grain, cv. Hanareumbyeo, Hanareum2, Dasanbyeo) and three Japonica rice (short grain, Nampyung, Ilpumbyeo and Junamjosaeng) cultivars, to benzobicyclon, mesotrione+pretilachlor (MP) and butachlor (control) in transplanting rice and their treatment influences on rice yield. All Indica${\times}$Japonica rice cultivars exhibited susceptible to both benzobicyclon and MP as reflected by higher visual injury and carotenoid biosynthesis inhibition when compared with Japonica rice cutlivars. The percentage of foliar chlorosis of Indica${\times}$Japonica rice was only 0.9~4.7% for benzobicyclon at 13 days after treatment (DAT) and 30.3~64.4% for MP at 5DAT but it increased rapidly to 88.4~91.2% at 12~20DAT. However, most of the Japonica cultivars are tolerant to bezobicyclon and MP. There was no visible leaf chlorosis but carotenoid biosynthesis was slightly inhibited. Based on relative carotenoid content reduction of benzobicyclon and MP to butachlor, the Indica${\times}$Japonica rice showed 4.6~15.6 fold higher compared with the Japonica rice. The heading date of the injured rice plant was delayed by 3 days and panicle number per square meter and ripened grain ratio were reduced as compared with the control treatment of butachlor. Rice yield of the Indica${\times}$Japonica rice cultivars treated with benzobicyclon and MP was reduced by 7~10%, 3~5%. respectively. The result indicates that rice cultivars vary in tolerance to HPPD-Inhibiting herbicides and Indica${\times}$Japonica rices were more susceptible than the Japonica rices to MP and benzobicyclon. Rice yield of the Indica${\times}$Japonica rices was also significantly reduced by the those herbicide treatments.

  • Purpose: This paper proposes a performance evaluation method for state-of-the-art facility inspection/diagnostic equipment through a trend survey of equipment and standardization systems of US, Japan, and Korea. This paper also suggests the priority of developing a performance evaluation method through expert interviews and surveys. Method: In this study, report for the last 5 years of FMS, state-of-the-art equipment of facility maintenance companies/safety diagnosis specialist agencies and papers/research reports/patents of NTIS were analyzed to identify recent trends of facility inspection/diagnostic equipment usages. standardization system of US, Japan, and Korea were analyzed to figure out a suitable form of a performance evaluation method for the domestic situation. And expert interview and survey were conducted to identify the priority of developing a performance evaluation method. Result: The performance evaluation method must be developed by the shape that only evaluates performance, regardless of types of equipment, on inspection item level for creative technology development. The priority of developing the performance evaluation method was identified as crack detection of concrete for durability evaluation and displacement/deformation/fatigue detection of concrete and steel for stability evaluation. Conclusion: The performance evaluation method will be developed firstly for the crack detection of concrete for durability evaluation and displacement/deformation/fatigue detection of concrete/steel for stability evaluation. In order to promote creative technology development, the performance evaluation method should be developed in a form that provides standardized specimens or testbeds and can be applied regardless of types of technologies.

Pre-Harvest Residual Characteristics of Boscalid and Pyraclostrobin in Paprika at Different Seasons and Plant Parts (파프리카 재배 중 살균제 boscalid와 pyraclostrobin의 사용시기에 따른 작물 부위별 생산단계 잔류특성)

  • Recent outbreak of new diseases and pests which were introduced from abroad, seriously hampered both quality and safety of paprika fruits. This study has been carried out to aid an establishment of guideline for safe use of pesticides and reduction of their residues on paprika. Systemic fungicides boscalid and pyraclostrobin of either mixed (a.i.; 13.6+6.8%) or single (a.i.; 47 and 18.8%, respectively) water dispersible granule formulation(WG) products were sprayed with recommended or double dosage on paprika grown in green house at March and June. To draw pre-harvest residue limit, residues of each fungicide were analyzed from fruits collected eight times from 18 to 1 day pre-harvest. The biological half-lives of both boscalid and pyraclostrobin in mixed formulation in March and June were slightly shorter than those of single formulation which ranged from 14.4 to 20.1 days. Residue levels of both fungicides of single formulation in fruits in June were about one lower compared to those in March. However, application of double dosage frequently exceeded MRLs from fruits grown both seasons. These results showed that residue levels on fruits persisted longer period of time, more than two weeks, and so the case applied in winter season. The dissipation of fungicides on leaves and fruits was compared. The distribution of both fungicides in leaves was 20-200 times higher than that of fruits and persisted up to 18 days of pre-harvest period at the concentration of 10-40 ${\mu}g\;g^{-1}$. This study indicated that the mixed formulation product exhibited low residues in fruits, but high and long enough to pathogen growth in leaves.

  • Greenhouse experiment was conducted to evaluate tolerance of six rice cultivars, three Indica${\times}$Japonica rice (long grain, cv. Dasanbyeo, Segejinmi and Hanareumbyeo) and three Japonica rice (short grain, Nampyung, Ilpumbyeo and Junamjosaeng) cultivars, to mesotrione+pretilachlor (MP) and bensulfuron-methyl+mesotrione+pretilachlor+pyriftalid (BMPP) in transplanting rice. Two herbicides were applied at 90 g and 180 g a.i. $ha^{-1}$ to three to four leaf stage rice at 5 and 15 days after transplanting, respectively. Related study was also conducted to compare $GR_{50}$ for Indica${\times}$Japonica and Japonica rice cultivars at different MP rates. Response to two herbicides varied with respect to rice cultivars and herbicide rates. All Indica${\times}$Japonica rice cultivars exhibited susceptible to both MP and BMPP as reflected by increased visual injury, shorter plant height and higher plant dry matter reduction when compared with nontreated rice cutlivars. The degree of foliar chlorosis by MP at 90 g a.i. $ha^{-1}$ was 5~6 at 7 days after treatment(DAT) but it was decreased to 3~6 at 14DAT. The degree of leaf chlorosis treated with 180 g a.i. $ha^{-1}$ MP was 6~7 at 7DAT and it was also decreased to 3~8 at 14 DAT. The plant hight of Indica${\times}$Japonica rices was inhibited by 18~43% at application of 90 g a.i. $ha^{-1}$ MP and 30~50% at 180 g a.i. $ha^{-1}$ MP. The dry matter reduction was greater than that of plant height inhibition, showing 46~73% at 90 g a.i. $ha^{-1}$ MP and 65~82% at 180 g a.i. $ha^{-1}$ MP. Similar leaf chlorosis injury and growth inhibition of rice cultivars was observed in the BMPP treatment. The injury and growth inhibition by MP and BMPP increased with increase in herbicide rate from 90 g a.i. $ha^{-1}$ to 180 g a.i. $ha^{-1}$. However, most of the Japonica cultivars are tolerant to MP and BMPP at both rates. There was no visible leaf chlorosis but plant height and dry matter production were slightly reduced at 180 g a.i. $ha^{-1}$. Based on $GR_{50}$ value determined in reduction of shoot dry weight for MP, the Indica${\times}$Japonica rice showed 12.9 fold lower compared with the Japonica rice. The result indicates that rice cultivars vary in tolerance to herbicides of MP and BMPP and Indica${\times}$Japonica rices were more susceptible than the Japonica rices to the MP and BMPP.

Estimation of Optimum Period for Spring Cultivation of 'Chunkwang' Chinese Cabbage Based on Growing Degree Days in Korea (생육도일(GDDs)에 따른 '춘광' 봄배추의 적정 재배 작기 예측)

  • Knowledge of the optimum cultivation period for Chinese cabbage would help growers especially in spring in Korea. Growth and yield of Chinese cabbage in a temperature gradient chamber was evaluated for the growing periods of 64 days from three set of transplanting dates including March 6, March 20, and April 3 in 2017. Air temperature in the chamber was elevated step-by-step, by $2^{\circ}C$ above the ambient temperature. This increment was divided into three phases; i.e. low (ambient+$2^{\circ}C$, A), medium (ambient+$4^{\circ}C$, B), and high temperature (ambient+$6^{\circ}C$, C). The fresh weight of Chinese cabbage was greater under B and C conditions in the first period and A in the second period, which indicated that GDDs affected the fresh weight considerably. However, leaf growth (number, area, length, and width) did not differ by GDDs. Bolting appeared under A condition in the first period, which was caused by low temperature in the early growth stage. Soft rot was developed under C condition in the second period and all temperature conditions in the third period, which resulted from high temperature in the late stage. Fresh weight increased when GDDs ranged from 587 to 729. However, it decreased when GDDs > 729. The maximum expected yield (16.3 MT/10a) was attained for the growing period of 64 days from transplanting date during which GDDs reached 601. The GDDs for optimum cultivation ranged from 478-724 under which the yield was about 95% (15.5 MT/10a) of maximum fresh weight. Such an optimum condition for GDDs was validated at five main cultivation regions including Jindo, Haenam, Naju, Seosan, and Pyeongtaek in Korea. In these regions, GDDs ranged from 619-719. This suggested that the optimum GDDs for Chinese cabbage cultivation would range from 478-724, which would give the useful information to expect the cultivation periods for ensuring maximum yield.

The Effect of 131I Therapy by Taking in Laxatives (방사성요오드 치료 시 완하제 투여 효과)

  • Purpose: Our goals were to evaluate the effect of high dose radioiodine treatment for thyroid cancer by taking in laxatives. Materials and Methods: Twenty patients(M:F=13:7, age $46.3{\pm}8.1\;yrs$) who underwent high dose radioiodine treatment were seperated into Group 1 taking $^{131}I$ 5,500 MBq and Group 2 with the use of laxatives after taking $^{131}I$ 5,500 MBq. The whole body was scanned 16 hours and 40 hours after taking radioactive iodines by using gamma camera, the ROIs were drawn on the gastro-intestinal tract and thigh for calculation of reduction ratio. At particular time during hospitalization, the radioactivity remaining in the body was measured in 1 meter from patient by using survey meter (RadEye-G10, Thermo Fisher Scientific, USA). Schematic presentation of an Origin 8.5.1 software was used for spatial dose rate. Statistical comparison between groups were done using independent samples t-test. P value less than 0.05 was regarded as statistically significant. Results: The reduction ratio in gastro-intestinal 16 hours and 40 hours after taking laxatives is $42.1{\pm}6.3%$ in Group 1 and $72.1{\pm}6.4%$ in Group 2. The spatial dose rate measured when discharging from hospital was $23.8{\pm}6.7{\mu}Sv/h$ in Group 1 and $8.2{\pm}2.4{\mu}Sv/h$ in Group 2. The radioactivity remaining in the body is much decreased at the patient with laxatives(P<0.05). Conclusion: The use in combination with laxatives is helpful for decreasing radioactivity remaining in the body. The radioactive contamination could be decreased at marginal individuals from patients.

Seedling Establishment, Overwintering Ability and Dry Matter Production of Chinese Milk Vetch (Astragalus sinicus L.) in Natural Reseeding Practices (자운영 지속재배시 입모, 월동 및 생육 특성)

  • Seedling establishment, overwintering ability and dry matter production of Chinese milk vetch (Astragalus sinicus L. CMV) in natural reseeding practice were compared with the annual seedling in 2006~2007 and 2007~2008. In natural reseeding, CMV seeds were distributed up to 15 cm soil depth in autumn after rice harvest and the seeds generally emerged from soil at less than 5 cm depth while they all emerged from soil surface in the annual seeding. It took 7-8 d longer in seedling establishment of the natural reseeding practice than the annual seeding. Generally, the natural reseeding practice showed higher seedling stand than the annual seeding because of high seedling survival rate. The high seedling survival rate in natural reseeded plant was attributed to the longer root length than that of the annual-seeded plant. In annual seeding, most of the seeds emerged from the soil surface and the root can not easily penetrate the soil while, in natural reseeding technology, the seeds incorporated into the soil during land preparation emerged from the soil at 0 to 5 cm depths, thereby having longer root length. The long root length contributed to greater ability to survive even under low temperature and low soil moisture conditions during winter. The dry matter production in the natural reseeding practice was also higher than the annual seeding when the temperature is low and soil moisture is not sufficient. This result indicates that natural reseeding technology is more stable and beneficial in seedling establishment and dry matter production as compared with the annual seeding especially under unfavorable environmental condition for CMV growth. This practice could be encouraged in CMV-rice cropping system in the southern parts of Korea.
