Removal of Organophosphorus Pesticides during Making and Fermentation of Kimchi (배추김치의 담금 및 숙성과정중 유기인계 농약의 제거)

  • The removal of three pesticides which were residued in chinese cabbage was investigated during making process of Kimchi. When chinese cabbage was washed by water, the removal rates of three pesticides were 62.0%, 54.8% and 61.1% for pirimiphos-methyl, chlorpyrifos and prothiofos, respectively. Pesticides remaining in chinese cabbage after washing by water were also removed from 22.4% to 23.8% by salting. During the fermentation of kimchi for 24 days at 4。C, the pH was lowered 4.5 from 5.8 and the residual amount of pesticides was decreased by 51.4% to 69.4% for three Pesticides remaining after washing and salting On the other hand, when Kimchi was fermented under various temperature for 11 days, the residual amount of chlorpyrifos was decreased up to 29.2%, 45.0% and 77.3% of initial concentration at 4, 10 and 20 。C, respectively. The residual amount of chlorpyrifos in Kimchi was decreased up to 16.3% by heating at 100 。C for 6.5 minutes.

Removal Efficiency of Pesticide Residues on Apples by Ultrasonic Cleaner (초음파 세척기를 이용한 사과의 잔류농약 제거 효과)

  • In order to determine the removal efficiency of pesticide residues on apples by ultrasonic cleaner, apples(Fuji var.) were artificially treated by dipping in water solution of fenitrothion EC, chlorpyrifos EC and phenthoate EC. The treated apples were washed by the ultrasonic cleaner with water, 30% ethanol solution or 0.2% detergent solution, respectively. The removal efficiency of fenitrothion, chlorpyrifos and phent-hoate by ultrasonicated washing in water for 0.5min. was 39.2%, 32.0% and 50.4%, respectively, but there was a tendency to decrease the removal efficiency of the pesticide residues as the duration of ultrasonicated washing period is increased. The maximum removal efficiency of the pesticide residues washed in the detergent solution was observed by ultrasonicated washing the samples for 5min. and it was recorded as 33.5% ; fenitrothion, 30.1% ; chlorpyrifos and 48.3% ; phenthoate, respectively. However it's appeared that the longer the ultrasonicated washing period in 30% ethanol solution the more pesticides from the apples were removed and the maximum removal efficiency of the pesticides was recorded as 66.2%(fenitrothion ; washed for 15min.), 41.7%(chlorpyrifos ; washed for 10min.), 74.2%(phenthoate ; washed for 10min.).

Characteristics of resistance to chlorpyrifos in diamondback moth(Plutella xylostella L.) collected from Chinese cabbage alpine farmland at Gangwon-do, Korea (강원도 고랭지 Chlorpyrifos 포장저항성 배추좀나방(Plutella xylostella L.)의 저항성 특성)

  • A field-resistant strain of the diamondback-moth(Plutella xylostella L.), collected from Chinese cabbage alpine farmland at Gangwon-do, Korea, was used for determination of the characteristics of resistance to chlorpyrifos using the activities of esterases and glutathione-S-transferase(GST), protein sequestration and AChE insenstivity. Although the activities of esterases extracted from resistant strain and susceptible strain were not significantly different, isozyme bands shown on the electrophoresis were different. GST activity from field resistant strain was 1.5-fold higher than that of susceptible. No differences were shown between resistant and susceptible ones in protein sequestration. The insensitivities of AChE to chlorpyrifos, however, extracted from susceptible strain was 460-fold higher than those of resistant. These results indicated that the insensitivity of AChE is the major factor for developing the resistance and activities of GST might be a minor factor.

Hand Exposure of Operator to Chlorpyrifos during Mixing/loading and Risk Assessment (유기인계 살충제 Chlorpyrifos 유제 살포액 조제 시 농작업자의 손 노출량 측정 및 위해성 평가)

  • Exposure and risk assessment during mixing/loading of chlorpyrifos emulsifiable concentrate (EC, 20%) were carried out. Limit of detection and limit of quantitation were 0.02 and 0.1 ng, respectively. Calibration curve linearity and reproducibility were excellent. Recovery of chlorpyrifos from gloves was 72.3-103.4%. During mixing/loading procedure, average hand exposure amount of chlorpyrifos was 3.9 mg which is corresponding to 0.004% of total active ingredient in the prepared spray mixture. In calculation of MOS (Margin of Safety) for risk assessment, male Korean average body weight and AOEL (Acceptable Operator Exposure Level) were used. Nine events of mixing/loading procedure were assumed per day. And 75 percentile of 30 repetition (4.6 mg) was used as for the worst case. MOS was more than 1 for total repetition, indicating mixing/loading work was of least risk. However, MOS of individual repetition was examined, two cases were less than 1 suggesting careful work habit is essential in mixing/loading procedure.

Studies for the Processing Factors of Pesticides during the Milling of Wheat Grain (밀의 제분에 따른 밀가루 중 농약 가공계수 산출 연구)

  • Processing factors of pesticides in milling process of wheat grain, which are consumed and imported at large quantity were examined to establish reasonable MRL of the processed food. Azinphos-methyl, chlorpyrifos, chlorpyrifos-methyl, fenitrothion, malathion, and trichlorfon were selected for the study according to annual usage and the previous detection record in wheat grain. Dipping process for pesticide application was performed in laboratory, while milling process was conducted under pilot plant system. Processing factors were calculated by analyzing residual pesticides on wheat grain and processing products as wheat flour, bran and red dog. Processing factors were 0.05 for azinphos-methyl, 0.06 for chlorpyrifos, 0.05 for chlorpyrifos-methyl, 0.07 for fenitrothion, 0.07 for malathion, 0.06 for trichlorfon, respectively. Recovery test was also performed to establish extraction efficiency of analytical procedure. The recovery value ranged from 93.2% to 98.6% with standard deviation of 0.1-0.9%.

Exposure Assessment of Korean Farmers While Applying Chlorpyrifos, and Chlorothalonil on Pear and Red Pepper (노지고추 및 배 재배 농업인의 방제작업 중 Chlorpyrifos, Chlorothalonil 노출에 관한 연구)

  • Objectives: The objectives of the present study were to establish the exposure monitoring method of chlorpyrifos and chlorothalonil application to pear and field red pepper by vehicle-mounted sprayer, hand-held sprayer and to assess the risk. Methods and Results: Chlorpyrifos met all of requirements of sampling and analysis method(U.S. EPA), but chlorothalonil met only dermal patch method. Potential dermal and inhalation exposure was evaluated in 42 farmers. Compared with the hand-held sprayer application, vehicle mounted sprayer demonstrated producing relatively lower dermal exposure levels with statistical significance. In dermal exposure during hand-held application, there is no significant difference between pear and red pepper. Conclusions: This results show that application method is the main factor of dermal exposure. There was no statistically significant difference between each group of respiratory exposure level. The margin of safety (MOS) was calculated to assess the risk of pesticide exposure. All Chlorpyrifos MOS values are lower than 0.2, which mean working conditions are unsafe. In order to protect farmers, big efforts to control exposure are needed.

Removal Efficiency of Residual Pesticides During Processing of Perilla Jangachi preparation (깻잎장아찌 제조과정 중의 잔류농약 제거 효과 연구)

  • This study was performed to study the removal efficiency of residual organophosphorus pesticides with process for making Perilla Jangachi. Two organophosphorus pesticides(chlorpyrifos-methyl and fenitrothion) were artificially attached to Perilla leaves. Then Perilla leaves were washed with detergent solution for 1minute and rinsed 2 times each for 1 minutes. After washing with neutral detergent solution, Perilla Jangachi was made with 2 steps of optimal condition. As a pretreatment, when soaked with 2% salt concentration solution for 42hours, the removal rate of residual pesticides was 81.75% of chlorpyrifos-methyl and 76.82% of fenitrothion. When Perilla leaves were steamed for 72 seconds after soaking, it became 88.94% and 82.19%, respectively. Finally, after making optimal Perilla Jangachi with 27% onion contents, removal rate was 89.12% of chlorpyrifos-methyl and 82.76% of fenitrothion. Consequently, it appeared that the process for making Perilla Jangachi effectively removed the residual pesticides of Perilla leaves.

Synthesis of Hapten for Indirect Competitive Immunoassay for Measuring 3,5,6-trichloro-2-pyridinol

  • BACKGROUND: In this study, we have attempted to identify a urinary biomarker to assess chlorpyrifos exposure in farmers. The major metabolite and the excretion pathway of chlorpyrifos is 3,5,6-trichloro-2-pyridinol (TCP) in urine. Herein, we describe an adequate synthetic method for TCP hapten for measuring urinary TCP of farmers. METHODS AND RESULTS: First, TCP was prepared by spacer attachment through hydrolysis of thiophosphate ester from chlorpyrifos. After reaction with benzyl bromide, the TCP was transformed into 2,3,5-trichloro-6-benzyloxypyridine. Next, the chlorine in the 2 nd position of the pyridyl ring was substituted into 3-mercaptopropanoic acid spacer arm. Finally, the phenyl group attached to the 6 th position in pyridyl ring was removed for producing the targeted product, 3-(3,5-Dichloro-6-hydroxy-2-pyridyl) thiopropanoic acid. CONCLUSION: Henceforth, this TCP hapten would be used in developing immunoassay studies for the detection and quantitation of urinary TCP of farmers.

Insecticide susceptibilities of rose field-collected populations of western flower thrips, Frankliniella occidentalis in Korea (지역별 장미재배지에서 채집된 꽃노랑총채벌레의 살충제 감수성)

  • The activities against Frankliniella occidentalis adults collected at rose greenhouse of 40 commerical insecticides were investigated. Among them 9 insecticides showed over 80% mortality to the adults; chlorfenapyr, chlorpyrifos- methyl, emamectin benzoate, fenthion, fipronil, phenthoate, spinosad, chlorpyrifos+diflubenzuron and furathiocarb+diflubenzuron. Susceptibility of field populations collected from greenhouse rose at 5 locations in 2001 was evaluated against the 9 insecticides selected. There was considerable difference in susceptibility depending on the treated insecticides and the regions from which the populations were collected. Goyang and Gimhae populations showed lower $LC_{50}$(ppm) values then Imsil, Jincheon and Gaangjin populations to all the tested insecticides. The adults of all field populations showed lower $LC_{50}$ values less than 1 ppm to emamectin benzoate, fipronil and spinosad. From these results, we could recommend 5 insecticides, such as emamectin benzoate, fipronil, spinosad, chlorpyrifos+diflubenzuron and furathiocarb+diflubenzuron as effective ones to control the Frankliniella occidentalis field populations.

Studies on the Insecticide Resistance of the German Cockroach (Blattella germanica L.) II. Resistant Developments and Cross resistance (바퀴(Blattella germanica L.)의 살충제 저항성에 관한 연구. 2. 저항성 발현 및 교차저항성)

  • The German cockroach (Blattella germanical L.) populations were successively selected with chlorpyrifos and permethrin for xis generations. The resulting resistant strains, $R_{chlorpyrifos}$(Rc) and $R_{permethrin}$(Rp) were observed resistant development and the cross-resistance. The result indicated that the values of $LC_{50}$ were increased 3.32 and 2.10 times from the susceptible strain, respectively. In the Rc strains, the values of the $LC_{50}$ of fenvalerate, propoxur, permethrin were to 3.89, 2.97, and 1.31 times higher than that of the suseptible, respectively. In the Rp strains, the values of the $LC_{50}$ of fenvalerate, chlorpyrifos, and propoxur showed 5.23, 1.42, and 1.80 times cross-resistance.
