Changes in Chlorphyll Contents and Volatile Compounds of Angelica keiskei Kimchi during Fermentation (신선초 김치 숙성 중 Chlorophyll 함량 및 휘발성 향기 성분의 변화)

  • Changes in the contents of volatiles and chlorphylls of Angelica keiskei Kimchi during fermentation were investigated. Total chlorphyll contents decreased gradually during the fermentatin period. Chlorphyll a decreased rapidly from 1.38 to 0.5$\mu\textrm{g}$/cm by the 5th day of fermentation with no further changes at 7th day, while chlorophyll b did not show any change by 3rd day and decomposed rapidly thereafter. Major voltile compounds identified in the kimchi were sabinene, $\alpha$-pinene and $\alpha$-terpinolene, which were gradually decreased during fermentation. There were significant differnce in color, sourness, bitterness and overall acceptability in sernsory evauation of Angelica keiskei Kimchi during fermentation at 2$0^{\circ}C$.

Accelerated and restrained effects of gibberellic acid on the growth of Chlorella (Chlorella의 생장에 미치는 gibberellic acid의 촉진 및 억제효과)

  • This investigation was proceeded to define the effects of gibberellic acid on the growth of Chlorella by determining the contents of chlorphyll and changes in various components in Chlorella cells according to the concentration of treated GA. The growth of Chlorella was accelerated with telative low concentrations of GA(10, 40 ppm) and was restrained with relative high concentrations of GA(70, 100, 200 ppm). The synthetic ability of chlorophyll of GA was inhibited generally in proportion to the concentration of treated-GA and the higher the concentration of GA was applied, the longer time was required in the restoration. The contents of RNA, protein and soluble carbohydrate were increased PCA-soluble amino acid and polysacharide were decreased in those cell components between the accelerated and restrained group. Consequently, the effect on accelerated growth in relative low concentrations of GA is considered to be caused by the powerful effet on expansion growth of GA. It is presumed that the effect of restrained growth in relative high concentrations of GA is due to the inhibitory effect on the chlorophyll synthesis.

  • The effects of heavy metals on the growth rate and phosphate metabolism of Chlorella elliposidea cells were investigated. Chlorella cells were cultured in the media treated with Hg(0.3, 0.7, 0.9 ppm), Cd(1, 5, 15ppm), and Zn(1, 5, 50ppm) for 6days. Aliquots cells were taken out at the inoculation and at intervals during the culture, and measured packed cell vlolume and optical density. The inhibitions of heavy metals on the growth rate and chlorophyll contents were traced. Also after 6 days culture, the amounts of inorganic phosphate and organic compounds of various fractions in Chlorella cells were observed. The turbid effects of heavy metals on the growth rate and chlorphyll contents of Chlorella cells were in order of Hg>Cd>Zn. Because heavy metals depressed the biosynthesis of inorganic polyphosphates and nucleic acids and turn over of inorganic phosphates, the amounts of various phosphate compounds were decreased. The inhibitory effect of photosynthesis by heavy metals resulted in lower contents of carbohydrate. Due to the turbidity of biosynthesis of amino acids by heavy metals, contents of protein were reduced in comparison with those of control. It is suggested conciusively that the minimum concentrations affected by heavy metals on the growth rate and phosphate metabolism of Chlorella cells were 0.7 ppm Hg, 15ppm Cd, 50ppm Zn.

Cytotoxicity of the ethanol extracts of mulberry leaves, branches and silkworm feces (상엽, 상지 및 잠분 에탄올 추출물의 품종별 세포독성 효과)

  • The cytotoxicity of the ethanol extracts of varietal samples of mulberry leaves and branches and silkworm feces was measured using CT-26 cells originated from murine metastatic colon cancer, using dye uptake assay in order to find potential anticancer agents. Two ethanol extracts (varietal mulberry leaves and mulberry branches) were prepared from 16 varietal mulberries and used as partial extract materials for the activity assay. Among these, the ethanol extracts from Shinkwangppong leaves showed a little anticancer activity, and those from Sugaeppong, Cheongunppong and Gumsulppong branches showed some anticancer activity as well as cytotoxicity. In contrast, ethanol extracts from freeze-dried, the 3rd day of 5th instar feces showed more potent anticancer activity than that of other mulberry leaves, mulberry branches and other 5th silkworm instar larva feces on the basis of high UV absorbance at 665 nm.

Effects of ammonia gas on soybean plant (대두(大豆)에 대한 Ammonia 가스의 영향)

  • This experiment was conducted to find out the effects on soybean plant exposed with various concentration and different fumigation periods of ammonia gas ($0.2mg/{\ell}$). The yield of soybean, nitrogen and, chlorophyll contents in leaves, and percentage of destroyed leaf area were investigated. The results were summarized as: 1. The soybean yield losses and percentage of destroyed leaf area were positively correlated with concentration of inflicting ammonia gas respectively. The yield losses was higher at noon exposure time than at night exposure time. 2. The soybean yield was negatively correlated with the percentage of destroyed leaf area. 3. The highest percentage of destroyed leaf area had at afternoon exposure time (14:00-15:00 o'clock) by $0.2mg/{\ell}$ ammonia gas fumigation, and the lowest one had at midnight exposure time (22:00-23:00 o'clock). 4. According to the increasing concentration of ammonia gas, the total and water soluble nitrogen contents in soybean leaves were increased, but the contents of chlorphyll b on decreasing rate. 5. The nitrogen contents in plant were higher in the afternoon exposure than in the morning, but the contents of chlorophyll were higher at night time exposure than at day time. 6. The highest decreasing of absorption spectra of chlorohpyll in chloroplast had at 10:00-11:00 o'clock on fumigation time and the lowest one had at 22:00-23:00 o'clock.

  • We have investigated the effects of abiotic and biotic stresses on leaf senescence using transgenic tobacco plants, in which cellular contents of polyamines were increased by introducing the genes of polyamine and ethylene biosynthesis in sense or antisense orientation. These transgenic plants showed accumulations of polyamines at higher levels than were found in wild-type. Stress-induced senescence was attenuated in transgenic plants cpmpared with wild-type plants, in terms of total chlorphyll loss and phenotypic changes after oxidative stress of hydrogen peroxide($H_2O_2$), high salinity, acid stress (pH3.0), ABA and fungal pathogen(phytophothora parasitica pv.Nicotianae). Transcripts for antioxidant enzyme, glutathionine-S-transferase and catalase, were also more abundant in transgenic plants than wild-type plants. These result suggested that higher expression of those genes caused a broad-spectrum resistance to abiotic stress/biotic stress. These phenomena indicate that polyamines may play an important role in contributing to the antioxidant defense function in plants. Our findings suggest that facilitate the improvement of stress tolerance of crop plants.

THE RELATION BETWEEN QUALITY AND CONTENT OF ZINC AND MAGNESIUM IN DRIED LAVER, PORPHYRA TENERA KJELLMAN (건해태(김)의 아연 및 마그네숨 함양과 품질과의 관계)

  • The growth of sea weeds is greatly affected by the environmental conditions of ambient water. Especially nutrient salts in sea water function as a major factor to the growth of lavers, so that the content of inorganic substances in lavers could he a criterion for quality evaluation of lavers. In this experiment, the relation between the quality and the content of Zinc and Magnesium which are not only physiologically active and closely related with pigments such as chlorophll and phycobilins but also important in quantify is discussed if such a measurement to be an index for quality evaluation. Sixteen samples of layers were collected from three different culture farms, 7 from Jangrim-Busan, 6 from Wando-Jeon Nam, 3 from Hadong-Gyeong Nam, and classified into 3 quality grades to each farm and to whole samples by organoleptic test. Zinc and Magnesium were analyzed by atomic absorption spectrophotometry. For pigment analysis, chlorphyll was extracted with 85 percent acetone, filtered and the absorbance was measured at 660 nm and the residue was further extracted with phosphoric buffer solution to determine the optical density for phycobilins at 560 nm. The result showed that the total content of Zinc and Magnesium varied between the farms, but kept consistent with quality grades from the same farm. The Zinc content was correlated proportion- ally with the content of phycobilins, and Magnesium content in total ash could roughly represent the content of chlorophll except some examples in which the Magnesium content of chlorophyll-ext ractresiduewasexceptionallyhigher. In conclusion the contents of these metals provide an applicable index for quality judgment of lavers.

Eutrophication of Shellfish Farms in Deukryang and Gamagyang Bays (득량만과 가막양 패류양식장의 부영양화)

  • Some environmental parameters on the shell-fish farms in Deukryang and Gamagyang Bays during summer in 1981 were determined to find an eutrophication level for the conservation of the farm. Chlorophyll-a content in the seawater in the Deukryang Bay in September was $1.0{\sim}5.0{\mu}g/l$ with an average of $2.5{\mu}g/l$ In the superficial bottom muds, contents of COD were 5-10 mg/g, ignition loss $5-9\%$, phaeophytin pigment $2{\sim}5{\mu}g/g$, and sulfide 0.1-0.3 mg/g dry mud in both bays. High contents of both organic matters and sulfide were found in the innermost area of the Deukryang Bay and in the north western part of the Gamagyang Bay. All quantities including chlorphyll-a in the water are little less than or similar to those of Hansan-Geoje Bay, one of the most productive shellfish farms in the southern coastal waters in Korea . Eutrophication on both water and bottom mud was under way like other shellfish farms but pollution indices on the bottom mud calculated from the data of CODs and phaeophytin pigments shelved 6-11, which is much lower than those of Jinhae Bay and of the Hansan-Geoje Bay. This means that the bottom muds are in an early stage of eutrophication, unlike the Jinhae and Hansan-Geoje Bays though the water, similar to the other bays, shows a middle stage of eutrophication.
