This paper represents our efforts to diagnose nutrient stresses using physiological instruments in cucumber plants. The stresses could be detected by measuring and analyzing the difference of chlorophyll content, photosynthetic efficiency(Fv/Fm), differential temperature(DT), stomatal resistance and light absorbance values between deficient and controlled plants. From the all over experiments, the stresses could be first diagnosed in the 8th day after treatment and the overall diagnosis rate was estimated at more than 50%.
Phaeodactylum tricornutum (Bacillariophyceae) is a marine diatom which has been supplied as a food of bivalves. In this study, growth responses of P. tricornutum to some nitrogen sources and sodium were investigated by measuring cell number and contents of chlorophyll a in culture. In medium with nitrogen and sodium, brisk cell division occurred and maximum growth rate was respectively found in the medium with 150 mg/l of nitrate and 10 mg/l of ammonium and urea. At 10-500 mg/l ammonium and urea and 200-500 mg/l nitrate, specific growth rate decreased slightly. However, no cell division observed in sodium-deficient medium, regardless of presence or absence of nitrogen. This suggests that sodium is required for the nitrogen uptake of P. tricornutum, resulting nitrogen uptake leading to cell division. Also the upper limits of ammonium and nitrate for the growth of P. tricornutum seem to be 10 mg/l and 500 mg/l, respectively.

To clarify the formation process and characteristics of oxygen deficient water mass in Gamak Bay, oxygen deficiency was weekly observed from 17 June to 12 September 2005. Surface water temperature was significantly lower in the outer bay than in the inner bay, whereas the bottom water temperature was higher in the central area of bay than in the outer and inner bay. The vertical stratification of water mass was strongly formed during the period, and thermocline was observed between 3 and 5m deep. The oxygen deficiency in the bottom layer began to appear at early July in the inner bay and gradually spread to the center area of the bay in early August. The mean transparency and light attenuation coefficient($K_d$) in water mass was 4.0m and 0.47, respectively. Average concentrations of nutrient and chlorophyll ${\alpha}$ in the bottom layer were significantly higher than those in surface, and those concentrations were significantly higher in the inner bay than in the outer bay. During the formation of oxygen deficiency in the bottom layer, oxygen penetration depth in the bottom sediment were extremely shallow, and oxygen consumption rate in the bottom sediment were lower than that in the area where oxygen deficient water mass disappeared. Dissolved oxygen concentrations in the bottom layer are negatively correlated with nutrient concentrations, whereas those in the surface layer did not show a significant relationship with nutrient concentrations. Elevated loss of oxygen in the bottom water mass was attributed to the increase of the oxygen consumption rates in sediments and the decomposition of organic matter by microorganism.

Though there is an on-going need for the diversity of ornamental plants for in-door environment, their growth and stress adaptability in comparatively low light intensity condition require further studies for implementation. Here investigates the growth and chlorophyll fluorescence parameters of Farfugium japonicum (L.) Kitam, Liriope muscari (Decne.) L.H.Bailey and Acorus gramineus Aiton under several light intensities which were based on common in-door environment. The growth measurement of the plants included the quantity, length, width and SPAD value of the leaves. Calculated values of Fm/Fo, Fv/Fm, Pi_Abs, ETo/RC and DIo/RC were used as the parameters of the chlorophyll fluorescence under 10, 50, 100 and 200 PPFD (μmol·m-2·s-1) light intensities. In-door plants group was put in a closed room allowing no other light sources for 10 weeks and the control group was put in glass-greenhouse for the same period. The overall in-door growth of L. muscari was not significantly different in all light intensities compared to the control group and even showed the higher SPAD values. Also, an increasing tendency of Pi_Abs value under 10 to 100 PPFD was observed implying that L.muscari could adapt well to in-door environment. Measurement of A. gramineus growth mostly showed the highest values in the control group especially in the number of the leaves. Nevertheless, chlorophyll fluorescence parameters showed no significant value difference between in-door and the control groups and thus, A. gramineus might have possibility of successful adaptation to in-door environment. F. japonicum showed deficient growth in plant height and leaf length compared to the control but, it seemed to be able to sustain ornamental value under in-door light intensities. Furthermore, Pi_Abs and DIo/RC values were increased under in-door light conditions suggesting potential adaptability of F. japonicum.
Immature embryos of Hordeum spontaneum were cultured on B5 and CI medium (Cheng's modified MS) to induce callus formation. CI media containing lmg/$\ell$ and 2mg/$\ell$ 2,4-D were more effective than B5 medium with lmg/$\ell$ 2,4-D for the initiation of callus. Total 883 calli were induced from 1,060 immature embryos plated. Callus induction frequency was 83%. Calli were transferred to differentiation media after one subculture to regenerate plants. Forty six plants were regenerated from 608 calli. Seventeen plants were chlorophyll deficient. There was no significant difference for plant regeneration among genotypes and media effects in calli which had been induced from immature embryos of seven types of trisomies. The overall regeneration frequency was 7.6%.
Purpose: This study was conducted to establish whether an ethanol extract of Lycium barbarum leaves (LLE) and an ethanol extract of Lycium barbarum leaves from which chlorophyll has been removed, denoted as LLE(Ch-), have a protective effect against hepatic fat accumulation. Methods: The inhibitory effects of LLE and LLE(Ch-) on liver fat accumulation were examined in C57BL/6 mice with non-alcoholic fatty liver disease (NAFLD) induced by an methionine and choline deficient diet and in HepG2 cells with palmitic acid-induced fat accumulation. Results: The plasma triglyceride, aspartate aminotransferase, and alanine aminotransferase levels were lower in the LLE(Ch-) group, whereas the plasma ALT activity decreased significantly in the LLE group. In both the LLE and the LLE(Ch-) groups, the triglyceride and cholesterol contents in the hepatic tissue were significantly reduced. A greater inhibitory effect on tissue fat accumulation was observed in the LLE(Ch-) group than in the LLE group. In HepG2 cells, LLE and LLE(Ch-) were non-toxic up to a concentration of 1,000 ㎍/mL. Compared to the control group, intracellular fat accumulation in the LLE and LLE(Ch-) groups were significantly reduced at concentrations of 200 ㎍/mL and 500 ㎍/mL, respectively. The expression of phosphorylated adenosine monophosphate-activated protein kinase and phosphorylated acetyl-CoA carboxylase in both LLE groups increased at the concentrations of 100 ㎍/mL and 500 ㎍/mL. The fatty acid synthase expression was suppressed in a concentration-dependent manner at 10 ㎍/mL. Conclusion: The examined two ethanol extracts of LLE inhibit hepatic fat accumulation in NAFLD. This effect was more pronounced in the LLE(Ch-) group. Therefore, these 2 extracts have an anti-steatosis effect and can be used for NAFLD treatment.
Objective of this research was to investigate the effect of nitrogen concentrations in the fertilizer solution on growth and development of nutrient deficiency in leaf perilla (Perilla frutesens). The nutrient concentrations in above-ground plant tissue, petiole sap and soil solution of root media were also determined. Nitrogen deficiency resulted in dwarfed growth, small leaves, and bright yellow color of older leaves. The leaves of deficient plants became uniform yellowing in color and finally necrosis occurred on the deficient leaves. Elevation of N concentrations in the fertigation solution from 0 to 20 mM increased the crop growth in leaf length and width as well as fresh and dry weights of above ground plant tissue. That also resulted in the increase of chlorophyll contents. However, light toxicity symptoms such as abnormal leaf surface appeared on crops grown in 20 mM N fertilization. The plant growth was commercially acceptable in the treatments of 10 and 15 mM N. The plants with acceptable growth had 0.9 to 1.25% in N contents of above-ground plant tissue, 800 to $3,300mg{\cdot}kg^{-1}$ in the $NO_3-N$ concentrations of petiole sap, and 28.7 to $47.3mg{\cdot}kg^{-1}$ in the $NO_3-N$ concentrations of soil solution (1:2 extract) at 75 days after transplanting.
Leaf disks from cultivar 'Kennebec' and one selection line (ND 860-2) were cultured on Murashige-Skoog medium with various combinations of indole acetic acid (IAA) and zeatin riboside. Shoots, roots and callus were induced at various combinations of plant growth regulator levels. The medium containing $3.5mg{\cdot}L^{-1}$ IAA and $4.0mg{\cdot}L^{-1}$ zeatin riboside produced the most plantlets. Rooted regenerants were grown in the greenhouse. The growth of regenerated plants obtained from the MS medium supplemented with $7.0mg{\cdot}L^{-1}$ IAA and $3.0mg{\cdot}L^{-1}$ zeatin riboside was significantly greater than those grown from nodal expalnts. In ND 860-2, a leaf chimera with chlorophyll deficient (light yellow) sectors was found in plants regenerated fiom leaf disks (grown on MS medium supplemented with $3.5mg{\cdot}L^{-1}$ IAA and $3.0mg{\cdot}L^{-1}$ zeatin riboside) but not in plants grown from nodal explants. The phenotypic variability was also observed in the tuber number, size and weight.
Masan bay is one of the polluted enclosed bays, which has red tides problem and the formation of oxygen deficient water in the bottom layer. Most important factors that cause eutrophication and red tide is nutrient materials containing nitrogen and phosphorus which stem from terrestrial sources and nutrients released from sediment. Therefore, to improve of water quality, reduction of these nutrient loads should be indispensible. At this study, the three-dimensional numerical hydrodynamic and eutrophication model, which were developed by Institute for Resources and Environment of Japan, were applied to analyze the processes affecting the phytoplankton production and also to evaluate the effect of water quality improvement plans on phytoplankton production. In field sorvey, the range of concentrations of chlorophyll-a at surface area was found to be 29.17 - 212.5mg/m3, which were exceeding eutrophication criteria. The constant currents defined by integrating the simulated tidal currents over 1 tidal cycle showed the counterclockwise eddies in the southern part of Budo. The general directions of constant currents were found to be southward at surface and northward at bottom over all the bay. The eutrophication model was calibrated with the data surveyed in the field area in June, 1993. The calculated results are in fairly good agreement with values within relative error of 30%. The pollutant load from the sources such as the input from terrestrial release from the sediment was reduced by the rate of 50, 70, 90, 98% to effect of phytoplankton production. Phytoplankton production was reduced to of the 90% reduction of the input loads from terrestrial sources and 8% in 90% reduction of the load from sediment.
