Identification of Biologically Active Substances from Lilac(Syringa vulgaris L.) (라일락 잎에 함유된 생리활성물질의 동정)

  • Inhibitory substance in the water extracts from lilac(Syringa vulgaris) leaves was determined in terms of the allelopathic chemicals. The water extracts from S. vulgaris leaves inhibited the germination and root growth of Digitaria sanguinalis and L. sativa, indicating that a biological substances are presented in the lilac leaves. The phenolic acids were separated and tentatively identified from S. vulgaris leaves by gas chromatography and there were composed of higher contents of p-coumaric acid, salicylic acid, pyrogallol, and catechol. Polyphenolic compounds such as rutin (5.3%), scopoletin (3.3%), kaempferol (2.9%), and other polyphenolic compotmds were detected from lilac leaves. The mixtures of $10^{-6}M$ of pyrogallol with all the concentrations of catechol had high inhibition of the shoot growth on D. sanguinalis and E. crus-galli regardless of the catechol concentrations.

Inhibition of NF-ĸB, Bcl-2 and COX-2 Gene Expression by an Extract of Eruca sativa Seeds during Rat Mammary Gland Carcinogenesis

  • The effect of Eruca sativa seed extract (SE) on nuclear factor kappa B (NF-${\kappa}B$), cyclooxygenase-2 (COX-2) and B-cell lymphoma-2 (Bcl-2) gene expression levels was investigated in rat mammary gland carcinogenesis induced by 7,12 dimethylbenz(${\alpha}$)anthracene (DMBA). DMBA increased NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and lipid peroxidation (LP), while, decreased glutathione-S-transferase (GST) and superoxide dismutase (SOD) activities and total antioxidant concentration (TAC) compared to the control group. After DMBA administration, SE treatment reduced NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and LP. Hence, SE treatment reduced inflammation and cell proliferation, while increasing apoptosis, GST and SOD activities and TAC. Analysis revealed that SE has high concentrations of total flavonoids, triterpenoids, alkaloids and polyphenolic compounds such as gallic, chlorogenic, caffeic, 3,4-dicaffeoyl quinic, 3,5-dicaffeoyl quinic, tannic, cinnamic acids, catechin and phloridzin. These findings indicate that SE may be considered a promising natural product from cruciferous vegetables against breast cancer, especially given its high antioxidant properties.

Polyphenol Analysis and Peroxynitrite Scavenging Effect of the Extracts from Eight Korean Mountainous Vegetable (한국 산채류 8종 추출물의 폴리페놀 함량분석 및 Peroxynitrite 소거효과)

  • Mountainous vegetables are usually used in the early stage of plant growth. We attempted to identify the quantity of polyphenols (caffoeoylquinic acids (CQs) and flavonoids) and peroxynitrite scavenging effect ($ONOO^-$) of eight Korean mountainous vegetables. The prominent characteristics were as follows: Only the roots of two plants Taraxacum platycarpum and Ixeris dentata contained chicoric acid. Five CQs were identified in the leaves of Cacalia firma whereas only 3-p-coumaroylquinic acid in the petioles of the plant was shown. The quantities of polyphenols such as quinic acid or tartaric acid derivatives in the mountainous vegetables were generally high, though those of flavonoids were very low. The $IC_{50}$ of chlorogenic acid, chicoric acid and kaempferol were 0.31, 0.12 and $0.25\;{\mu}g/ml$, respectively.

Antioxidant activity and phytochemical contents of the extracts from different parts of Moringa oleifera (모링가 부위별 추출물의 항산화 활성 및 파이토케미컬 함량)

  • Moringa (Moringa oleifera), which is native to tropical and subtropical climates, now grows in Korea due to its warming climate. Since it is also imported and used for food and other products in various countries, it is necessary to analyze the properties of moringa being imported into Korea. In this study, the antioxidant activity and total phenol and flavonoid content were high in the leaves and twigs and showed a positive correlation. The mineral content was also high in the leaves and twigs, and potassium ions were the most abundant mineral. In addition, seven phenolic acids (caffeic acid, chlorogenic acid, cinnamic acid, coumaric acid, ferulic acid, gallic acid, and syringic acid) and four flavonoids (kaempferol, myricetin, quercetin, and rutin) were detected in the leaves, twigs, and stems, and the ferulic acid and quercetin content was particularly high. The results of this study can be used as comparative data for domestic moringa growers in the future.

  • The effect of methanolic extract of cashewnut (Anacardium occidentale) shell extract was seen on conidial germination of Erysiphe pisi and powdery mildew development in pea (Pisum sativum). Maximum conidial germination inhibition of E. pisi on glass slides was observed at 300 ppm. Similar effect on floated pea leaves was observed after 48 h at the same concentration. Conidial germination on intact untreated pea leaves was also assessed on II and IV nodal leaves while IV and II nodal leaves were treated with the extract and vice versa. There was tremendous reduction in conidial germination on all the nodal leaves. The disease intensity of pea powdery mildew was significantly reduced by methanolic extract of cashewnut shells. Maximum reduction was observed with 200 ppm where 39% disease intensity was recorded in comparison to 96.53% in the control. The phenolic acid content of pea leaves following treatments with this extract varied and no defmite pattern was observed. Out of several phenolic compounds, namely, gallic, ferulic, chlorogenic, and cinnamic acids, only gallic acid was found to be present consistently in all the treatments with varied amounts.

Comparative phenolic compound profiles and antioxidative activity of the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) according to cultivation years

  • Background: The study of phenolic compounds profiles and antioxidative activity in ginseng fruit, leaves, and roots with respect to cultivation years, and has been little reported to date. Hence, this study examined the phenolic compounds profiles and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free-radical-scavenging activities in the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) as a function of cultivation year. Methods: Profiling of 23 phenolic compounds in ginseng fruit, leaves, and roots was investigated using ultra-high performance liquid chromatography with the external calibration method. Antioxidative activity of ginseng fruit, leaves, and roots were evaluated using the method of DPPH free-radical-scavenging activity. Results: The total phenol content in ginseng fruit and leaves was higher than in ginseng roots (p < 0.05), and the phenol content in the ginseng samples was significantly correlated to the DPPH free-radical-scavenging activity ($r=0.928^{****}$). In particular, p-coumaric acid ($r=0.847^{****}$) and ferulic acid ($r=0.742^{****}$) greatly affected the DPPH activity. Among the 23 phenolic compounds studied, phenolic acids were more abundant in ginseng fruit, leaves, and roots than the flavonoids and other compounds (p < 0.05). In particular, chlorogenic acid, gentisic acid, p- and m-coumaric acid, and rutin were the major phenolic compounds in 3e6-yr-old ginseng fruit, leaves, and roots. Conclusion: This study provides basic information about the antioxidative activity and phenolic compounds profiles in fruit, leaves, and roots of Korean ginseng with cultivation years. This information is potentially useful to ginseng growers and industries involved in the production of high-quality and nutritional ginseng products.

The Allelopathic Effects of Aqueous Extracts of Hypochaeris radicata L. on forage Crops (서양금혼초(Hypochaeris radicata L.)의 수용추출물이 사료작물에 미치는 allelopathy 효과)

  • This experiment was performed to examine the allelopathy effect of allelochemical substance on the crop plants. According to the experiment of the allelochemical substances in Hypochaeris radicata by HPLC, there are the differences at each part of plants. However, it is ascertained that there are 14 kinds of phenolic compounds ingredients that are $\rho$-hydroxybenzoic acid, chlorogenic acid, catechin, caffeic acid, syringic acid, salicylic acid, $\rho$-coumaric acid, ferulic acid, naringin, hesperidin, myricetin, trans-cinnamic acid, quercetin and naringenin. The chemicals like caffeic acid, ferulic acid, and naringenin are commonly included. The result of the chemical experiment shows that there are the differences at each season and part of plants. The leaves in May and blossoms and roots in July contain lots of phenolic acids. It is very high contents such as salicylic acid 2085.6 ${\mu}g/g$ and quercetin 1522.0 ${\mu}g/g$, especially in roots of plants. The result on the growth of crop plants treated by the aqueous extract of Hypochaeris radicata shows that the value of the control group and the test group are same in some cases. However, because the treat value of test group is towel'than that of control group in all items of the experiment, it is cofirmed that the growth of crop plants was inhibited and that the inhibitory effect was increased as its density of treatment was increased. The result of change in quantity shows that there are the differences at each kind of crop plants, but the inhibitory effect was increased as its concentration of treatment was increase with entire. As results, it is confirmed that H. radicata has the allelopathy effect to the crop plants. Especially the inhibitory effect on growth is high in gramineous crop, italian ryegrass and leguminous crop. purple alfalfa.

Effect of Enhancement on Functionality of Germinated Adzuki Bean (Angularis angularis var. nipponensis) with High Hydrostatic Pressure (HHP) Treatment (고압처리에 의한 발아팥의 기능성 향상 효과)

  • We investigated the influence of germination and high hydrostatic pressure (HHP) treatment conditions on the conversion of functional compounds and antioxidant activity in adzuki bean. The adzuki bean germinated at $25^{\circ}C$ for three- or six-days, and was later subjected to HHP at 0.1, 50, 100, or 150 MPa for 24 h. The highest polyphenol content (5.36 mg gallic acid equivalents (GAE)/g) and flavonoid content (0.91 mg catechin equivalents (CE)/g) were observed after germination for six days and HHP treatment at 100 MPa for 24 h, respectively. The total phenolic acid contents increased with increasing applied pressure from 88.86 to $208.26{\mu}g/g$ (100MPa, 24h). Phenolic acids are divided into two categories; those that exhibit increased content upon HHP treatment, and those that exhibit decreased content. The increasing phenolic acids were gallic acid, chlorogenic acid, (+)-catechin, ${\rho}-coumaric$ acid, ferulic acid, heperidin, salicylic acid, protocatechuic acid, cinnamic acid, naringenin. The total anthocyanin content decreased with increasing applied pressure from 22.42 mg/100 g to 6.28 mg/100 g (150 MPa, 24 h). The highest ABTS radical scavenging activity (8.02 mg eq AA/g) and DPPH radical scavenging activity (1.22 eq Trolox/g) were observed after germination for six days and HHP treatment at 100MPa for 24h, respectively. These results suggested that the combination of HHP and germination can lead to improved functionality in adzuki bean.

A Comparative Study on Anthocyanin and Polyphenol Contents in Colored Agricultural Products (유색 농산물 중 안토시아닌과 폴리페놀 함량 비교 연구)

  • In this study, 17 kinds of polyphenols and 5 kinds of anthocyanins were analyzed to compare the contents of polyphenols and anthocyanins in 76 colored agricultural products. A total of 17 polyphenols were analyzed simultaneously by 9 phenolic acids (gallic acid, protocatechuic acid, chlorogenic acid, vanillic acid, caffeic acid, syringic acid, p-coumaric acid, t-ferulic acid, t-cinnamic acid) and 8 flavonoids ((+)catechin, syringic aldehyde, rutin, epicatechin gallate, naringin, luteolin, naringenin, kaempferol) and 5 anthocyanins (delphinidin-3-glucoside, delphinidin-3-rutinoside, cyanidin-3-galactoside, cyanidin-3-glucoside, cyanidin-3-arabinoside) were simultaneously analyzed. The total content of 17 polyphenols was determined as seoritae $255.1{\pm}7.5{\mu}g/g$, seomoktae $275.8{\pm}5.3{\mu}g/g$, black rice $78.5{\pm}4.6{\mu}g/g$, black sesame $75.8{\pm}3.2{\mu}g/g$, blueberry $143.3{\pm}5.5{\mu}g/g$, aronia $195.2{\pm}4.9{\mu}g/g$ and blackcurrent $131.6{\pm}3.2{\mu}g/g$, the highest content was found in the order of seomoktae > seoritae > aronia > blueberry > blackcurrant > black rice > black sesame. The total content of 5 anthocyanins was determined as seoritae $82.4{\pm}17.2{\mu}g/g$, seomoktae $95.2{\pm}6.1{\mu}g/g$, black rice $74.1{\pm}9.7{\mu}g/g$, black sesame were not detected, blueberry $110.8{\pm}1.9{\mu}g/g$, aronia $218.9{\pm}6.1{\mu}g/g$ and blackcurrent $209.7{\pm}4.0{\mu}g/g$, the highest content was found in the order of aronia > blackcurrant > blueberry > seomoktae > seoritae > black rice. These results indicated that seomoktae and aronia possessed the high level of functional components and further study will be needed to develop high value-added foods based on the colored agricultural products.

Comparison of Nutritional Components and Physicochemical Properties of Small Colored Potatoes and Small Regular Potatoes (칼라꼬마감자와 일반꼬마감자의 영양성분 및 이화학적 특성)

  • This study examined the nutritional components and physicochemical properties of small colored potatoes and small regular potatoes as a natural health food source. To accomplish this, the general and antioxidative contents of small colored potatoes and small regular potatoes were measured. Total contents of carbohydrates, crude protein, crude lipid, and ash were 88.1%, 4.9%, 0.9%, and 6.4%, respectively. Small colored potatoes contained 76.5 kcal, while their total dietary fiber was 4.0%. Total proteins consisted of 17 different kinds of amino acids. Regarding their mineral contents, K was the most abundant mineral, followed by P, Mg, and Ca. Total phenol contents of the 70% ethanolic extracts of small colored potatoes were $48.2{\pm}1.2$ mg GAE/g. Total flavonoid contents of the 70% ethanolic extracts were $13.1{\pm}0.3$ mg RE/g. Overall, small colored potatoes had higher amounts of nutrients and physicochemical properties than small regular potatoes. The general nutrients and other antioxidant bioactive materials in small colored potatoes were also potential materials for good health food. It is expected that follow up studies of small colored potatoes through developing processed food and evaluation of their functional properties would provide useful information as a source of functional foods.
