Antimicrobial Activities of Korean Sword Bean (Canavalia gladiata) Extracts (한국산 작두콩 추출물의 항균활성)

  • The antimicrobial activities of three parts of Korean sword bean , such as whole seed, dotyledon and hull were investigated . Yields were higher hull other parts in water and methanol extract. Methanol extract in seed parts was fractionated with different solvents, such as hexane, chloroform, ethyl acetate, butanol and water, yields of water fractino showed 3.3% , 2.5% and 0.3% in whole seed, 채쇼ledon and hull, respectively. In solvent extracts using methanol, hexane, chloroform, and water , methanol extract showed the most effective antimicrobial activities. Antimicrobial activities of ethyl acetate fraction of methanol extractwas higher than those of other fractions.

Comparative Study on the Cytotoxic Activities of Red Ginseng of Korea and China (한국 및 중국 홍삼의 암세포 증식억제 효과 비교연구(III))

  • A study was performed to compare the anticancer effects of Korean and Chinese red ginseng roots. The whole crude extracts or chloroform, methanol and acetone fractions of the crude extracts were added in the culture medium of three cancer cell lines, a mouse leukemia cell line ($P_{388}$), a human colon carcinoma cell line (HT-29) and a human rectal carcinoma cell line (HRT-18), to screen the growth inhibition effects. The results are summarized as follows : 1. Crude extracts of both Korean and Chinese red ginseng roots inhibited the proliferation of all the three cancer cell lines tested in a dose dependent manner. However, the growth inhibition effects of Korean red ginseng extracts were significantly greater than that of Chinese red ginseng. 2. An acetone fraction showed the greatest antiproliferative effects among the 11'hole crude extracts, chloroform, methanol and acetone fractions of the crude extracts. 3. These results suggest that the active antiproliferative components of the crude extracts are present mostly in the acetone fraction.

Studies on the Biological Activity of Astragalus membranaceus Extracts

  • Physiological activities of hot water extract and solvent fractions isolated from Astragalus membranaceus were examined and the antioxidative, fibrinolytic, thrombin inhibitory and a-glucosidase inhibitory activity were measured. The hot water extract of Astragalus membranaceus was fractionated into hexane, chloroform, ethyl acetate, butanol and water fractions, and each of these fractions was individually assayed. The antioxidative activities of ethyl acetate and chloroform fractions were 89.96% and 87.36%, respectively. Using the fibrin plate method, only the hot water extract showed a plasmin activity of 0.41 units/ml. The thrombin inhibitory activity of the ethyl acetate fraction was the highest with a value of 82.73%. The hot water extract displayed a-glucosidase inhibitory activity of 64.91%. In conclusion, the hot water extract and the ethyl acetate fraction can be used as materials for the development of biofunctional foods to prevent cardiovascular diseases.

  • This study was designed to investigate the possible utilization of Zizyphus jujuba leaves as a source of functional ingredients. The physiological activity of different solvent fractions prepared from ethanol extract of Zizyphus jujuba leaves were analyzed. Xanthine oxidase inhibitory effect was very high in all fractions except chloroform fraction. The very high electron donating ability was observed in the ethylacetate fraction and the effect was similar to 0.1% tocopherol. Nitrite scavenging effect of all fractions was more than 40% even at low concentration of 1mg/ml and was increased with increasing concentration. Angiotensin I converting enzyme inhibitory activity was appeared in ethyl acetate and chloroform fractions only at high concentraton.

  • Antibacterial activity test of solvent fractions, sesquiterpenoid lactones, and Compound I and II extracted from Chrysanthemum boreale M. and Chrysanthemum indium L. were performed against four microorganisms. Among the tested substances, antibacterial activities were appeared against B. subtilis and V. parahaemolyticus at the chloroform fraction, sesquiterpenoid lactones and Compound I extracted from C. boreale. But chloroform fraction and sesquiterpenoid lactones extracted from C. indicum were showed weakly than those of C. boreale. Compound II and all fractions extracted from C. indicum were not appeared against the all tested microorganisms.

  • In the present study, in attempt was made to observe the effect of lipid components in pantocrin and antler velvet and sponge layers extracts on the aldolase activity in the rat spinal nerves. The antler chloroform-methanol (2:1) extracts are known to be composed of neutral lipids, cholesterol, cerebrosides, spingomyelin, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and gangliosides. The antler extracts were studied on the aldolase activity in the rat spinal nerves. The aldolase activity was measured by the method of Lehninger. To investigate the components of tile extracts which affect the enzyme activity, the chloroform-methanol (2:1) extracts of antler were fractionated into petroleum ether soluble and insoluble fractions and their effects on the enzyme activity were compared. It was found that the chloroform-methanol (2:1) extracts of the antler sponge and velvet layers as well as those petroleum ether soluble fractions obtained from the chloroform-methanol (2:1) extracts, decrease the aldolase activity by 18-23%. However, pantocrin showed increasing effect on the aldolase activity by 42.6%. The petroleum ether insoluble fraction of the chloroform-methanol (2:1) -extract showed no significant increasing activity (about 13%) on the spinal nerves aldolase. The components of pantocrin ind the petroleum ether insoluble fractions were attempted to analysize by thin layer (silica gel) and gas liquid chromatography.

Antioxidative Activity and Cytotoxicity on Human KB cell of Extracts from Rhododendron mucronulatum Turcz. Flower (진달래꽃 추출물의 항산화 효과 및 인체 KB cell에 대한 세포독성)

  • This study was carried out to investigate the antioxidative activity and cytotoxicity on human KB cell of extract from Rhododendron mucronulatum flower, and also to determine the contents of total polyphenol and flavonoid. The methanol extract of Rhododendron mucronulatum flower was fractionated with various solvents such as hexane, chloroform, ethyl acetate, butanol and water. The DPPH radical scavenging activities of ethyl acetate fraction had stronger activity than other fractions. So, antioxidative substances of ethyl acetate fraction were crude purified by silica gel column chromatography. The DPPH radical scavenging activities of crude purified fraction 2 and 3 were more than 90% at 40 ppm. In the presence of $100{\mu}g/mL$, growth inhibition on human KB cell by WST-1 assay showed 81.2% in chloroform fraction and 74.6% in hexane fraction. The contents of total polyphenol and flavonoid of ethyl acetate traction were 32.70% and 20.30%, respectively, The antioxidative activity showed correlation with total polyphenol and flavonoid contents.

Effects of subfractions of Coptidis Rhizoma extract on the nitric oxide production in LPS-stimulated BV2 microglial cells (황련 추출물의 분획화 및 BV2 microglial cells에서 LPS에 의해 유도되는 nitric oxide 생성억제효과 검정)

  • Objectives : Uncontrolled activation of microglia may directly toxic to neurons by releasing various substances such as inflammatory cytokines, nitric oxide(NO), prostaglandin E2 and superoxide. In this study, the effects of the several subfractions isolated from Coptidis Rhizoma extract were investigated on NO production in LPS-stimulated BV2 microglial cells, Methods : Coptidis Rhizoma extract prepared with 80% methanol, and then fractionated with ethylacetate, chloroform, n-butanol and water. BV2 cells were pretreated four subfractions of Coptidis Rhizoma with various concentrations, and then stimulated with LPS. Cytotoxicity of each fraction was measured by MTT assay. NO production was determined in culture surpernatants by Griess reagent. Results : Ethylacetate, chloroform and butanol fractions of Coptidis Rhizoma extract significantly decreased LPS-induced NO production in BV2 cells as a dose-dependent manner without cytotoxicity. Ethylacetate fraction of Coptidis Rhizoma extract was most effective on inhibition of NO production in LPS-stimulated BV2 cells compared with other fractions. Conclusion : This data indicates that Ethylacetate fraction of Coptidis Rhizoma extract shows strong antiinflammatory effects through inhibition of LPS-induced microglial activation.

Inhibition of lipopolysaccharide-stimulated inflammatory mediator production in RAW264.7 macrophages by Foeniculum vulgare fruit extract (LPS로 자극한 RAW264.7 대식세포주에서 회향 추출물에 의한 염증성 매개물의 생성 억제)

  • 이물질 침입에 대한 인식의 결과 NO, PGE$_2$, TNF-, IL-6와 같은 여러 신호전달물질의 분비가 개시되며 이들을 억제하는 물질을 항염증제라고 볼 수 있다. 본 연구에서는 회향(Foeniculum vulgare Mill.) 열매 추출물이 mouse macrophages RAW264.7 세포에서 lipopolysaccharide(LPS)로 유도한 NO(iNOS 산물), PGE$_2$(COX-2 산물) 및 cytokines (TNF-$\alpha$, IL-6) 생성 억제에 미치는 영향을 살펴보았다. 회향 열매의 methanol 추출물 및 분획물(chloroform, butanol, and aqueous fractions)은 4~100$\mu$g/mL 농도에서 LPS가 활성화된 대식 세포에서 NO 생성을 억제하였으며 독성을 나타내지 않았다. LPS가 유도한 PGE$_2$ 생성은 butanol 분획(100 $\mu$g/mL)에 의해서만 유의적으로 감소하였다(P<0.05). 회향 열매 추출물 및 분획물은 TNF-$\alpha$의 생성을 유의적으로 감소시켰으며 IL-6의 생성은 methanol extract(4~100 $\mu$g/mL), chloroform fraction(4 $\mu$g/mL), butanol fraction(4 and 100$\mu$g/mL) 및 aqueous fraction(4~100 $\mu$g/mL)에 의해 감소되었다(P<0.05). 이는 회향 열매 추출물은 염증 상태에서 유용할 것이며 COX-2와 iNOS를 억제하는 butanol 분획은 새로운 항염증제 개발에 사용될 수 있음을 시사하여 주었다.

Antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis from Jeju Island, Korea

  • In this study, we assessed the antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis, from Jeju Island, Korea. Specifically, the antioxidant activity of fractions isolated from 80% methanol extract, and digests produced from five proteases and carbohydrases, were investigated. Almost all the fractions and the 80% methanol extract exhibited higher effects on 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging. The ethyl acetate fraction showed the highest superoxide anion scavenging activity, while both n-hexane and chloroform fractions exhibited higher $H_2O_2$ scavenging activity. Among the enzymatic digests from H. porphyrae and O. unicellularis, all the digests exhibited remarkable DPPH scavenging activities. In nitric oxide inhibition, all the digests recorded significantly higher effects than those of the commercial antioxidants (p < 0.05). Flavozyme and Neutrase digests from H. porphyrae, and Termamyl and Alcalase digests from O. unicellularis, showed significant effects in metal chelating. Lipid peroxidation was significantly inhibited in the ethyl acetate fraction, in the Celluclast and Protamex digests from H. porphyrae, and in the chloroform fraction from O. unicellularis. These findings suggest that the two tidal pool microalgae tested in this study are rich in potential antioxidative compounds, the specific properties of which can be considered for use in the food and pharmaceutical industries.
