Cytotoxic Effects of Strawberry, Korean Raspberry, and Mulberry Extracts on Human Ovarian Cancer A2780 Cells

  • Reactive oxygen species are tumorigenic by their ability to increase cell proliferation, survival, and cellular migration. The purpose of the present study was to compare the antioxidant activity and cytotoxic effects of 3 berry extracts (strawberry, Korean raspberry, and mulberry) in A2780 human ovarian carcinoma cells. Except for raspberry, the ethyl acetate or methylene chloride fractions of berries containing phenolic compounds exerted dose dependent free radical scavenging activities. In the raspberry fractions, the hexane fraction also exhibited potent antioxidant activity. The cytotoxic effects of berries extracts in A2780 human ovarian carcinoma cells were measured using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Surprisingly, co-treatment with n-butanol (BuOH) fractions of berries showed stronger cytotoxic effects compared to the other fractions. These findings suggest that potent anticancer molecules are found in the BuOH fractions of berries that have stronger cytotoxic activity than antioxidants.

  • This paper provides an introduction to life-span simulation and numerical approach to support the performance design processes of reinforced concrete structures. An integrated computational system is proposed for life-span simulation of reinforced concrete. Conservation of moisture, carbon dioxide, oxygen, chloride, calcium and momentum is solved with hydration, carbonation, corrosion, ion dissolution. damage evolution and their thermodynamic/mechanical equilibrium. Coupled analysis of mass transport and damage mechanics associated with steel corrosion is presented for structural performance assessment of reinforced concrete. Multi-scale modeling of micro-pore formation and transport phenomena of moisture and ions are mutually linked for predicting the corrosion of reinforcement and volumetric changes. The interaction of crack propagation with corroded gel migration can also be simulated. Two finite element codes. multi-chemo physical simulation code (DuCOM) and nonlinear dynamic code of structural reinforced concrete (COM3) were combined together to form the integrated simulation system. This computational system was verified by the laboratory scale and large scale experiments of damaged reinforced concrete members under static loads, and has been applied to safety and serviceability assessment of existing structures. Based on the damage details predicted by the nonlinear finite element analytical system, the life-span-cost of RC structures including the original construction costs and the repairing costs for possible damage during the service life can be evaluated for design purpose.

Corrosion behaviors of SS316L, Ti-Gr.2, Alloy 22 and Cu in KURT groundwater solutions for geological deep disposal

  • Deep geological disposal using a multibarrier system is a promising solution for treating high-level radioactive (HLRW) waste. The HLRW canister represents the first barrier for the migration of radionuclides into the biosphere, therefore, the corrosion behavior of canister materials is of significance. In this study, the electrochemical behaviors of SS316L, Ti-Gr.2, Alloy 22, and Cu in naturally aerated KAERI underground research tunnel (KURT) groundwater solutions were examined. The corrosion potential, current, and impedance spectra of the test materials were recorded using electrochemical methods. According to polarization and impedance measurements, Cu exhibits relatively higher corrosion rates and a lower corrosion resistance ability than those exhibited by the other materials in the given groundwater condition. In the anodic dissolution tests, SS316L exposed to the groundwater solution exhibited the most uniform corrosion, as indicated by its surface roughness. This phenomenon could be attributed to the extremely low concentration of chloride ions in KURT groundwater.

Bioactive characteristics of an implant surface coated with a pH buffering agent: an in vitro study

  • Purpose: The purpose of this study was to evaluate the effectiveness of conventional sandblasted, large-grit, acid-etched (SLA) surface coated with a pH buffering solution based on surface wettability, blood protein adhesion, osteoblast affinity, and platelet adhesion and activation. Methods: Titanium discs and implants with conventional SLA surface (SA), SLA surface in an aqueous calcium chloride solution (CA), and SLA surface with a pH buffering agent (SOI) were prepared. The wetting velocity was measured by the number of threads wetted by blood over an interval of time. Serum albumin adsorption was tested using the bicinchoninic acid assay and by measuring fluorescence intensity. Osteoblast activity assays (osteoblast adhesion, proliferation, differentiation, mineralization, and migration) were also performed, and platelet adhesion and activation assays were conducted. Results: In both the wetting velocity test and the serum albumin adsorption assay, the SOI surface displayed a significantly higher wetting velocity than the SA surface (P=0.000 and P=0.000, respectively). In the osteoblast adhesion, proliferation, differentiation, and mineralization tests, the mean values for SOI were all higher than those for SA and CA. On the osteoblast migration, platelet adhesion, and activation tests, SOI also showed significantly higher values than SA (P=0.040, P=0.000, and P=0.000, respectively). Conclusions: SOI exhibited higher hydrophilicity and affinity for proteins, cells, and platelets than SA. Within the limits of this study, it may be concluded that coating an implant with a pH buffering agent can induce the attachment of platelets, proteins, and cells to the implant surface. Further studies should be conducted to directly compare SOI with other conventional surfaces with regard to its safety and effectiveness in clinical settings.

Antioxidant and Antiproliferative Activities of the Halophyte Angelica japonica Growing in Korean Coastal Area (한국 연안지역에 서식하는 갯강활의 항산화 및 암세포증식 억제 활성)

  • This study evaluated the antioxidizing and antiproliferative effects of Angelica japonica extract and its solvent-partitioned fractions. A dried sample of the halophyte A. japonica was extracted twice using methylene chloride (CH2Cl2) and extracted twice again using methanol (MeOH). The combined crude extracts were then fractionated by solvent polarity into distilled water (water), n-butanol (n-BuOH), 85% aqueous methanol (85% aq.MeOH), and n-hexane fractions. The antioxidant activities of the crude extracts and their solvent-partitioned fractions were assessed according to their DPPH radical and peroxynitrite scavenging abilities, formation of intracellular reactive oxygen species (ROS), DNA oxidation, NO production, and ferric reducing antioxidant power (FRAP). The crude extract showed significant antioxidant activity in the overall antioxidizing bioassay systems. Among solvent-partitioned fractions, good antioxidant activities were observed in n-BuOH and 85% aq.MeOH fractions and significantly correlated with the polyphenol and flavonoid contents of the samples. Furthermore, all samples tested, including the crude extract, not only showed cytotoxic effects against human cancer cells (AGS, HT-29, MCF-7, and HT-1080) but also prevented cell migration in a dose-dependent manner in the wound healing assay using HT 1080. Among the solvent-partitioned fractions, the 85% aq.MeOH fraction most effectively inhibited the invasion of HT-1080 cells. Therefore, these results suggest that A. japonica may be a potential antioxidizing and antiproliferative agent.

Three sesquiterpene lactones suppress lung adenocarcinoma by blocking TMEM16A-mediated Ca2+-activated Cl- channels

  • Transmembrane protein TMEM16A, which encodes calcium-activated chloride channel has been implicated in tumorigenesis. Overexpression of TMEM16A is associated with poor prognosis and low overall survival in multiple cancers including lung adenocarcinoma, making it a promising biomarker and therapeutic target. In this study, three structure-related sesquiterpene lactones (mecheliolide, costunolide and dehydrocostus lactone) were extracted from the traditional Chinese medicine Aucklandiae Radix and identified as novel TMEM16A inhibitors with comparable inhibitory effects. Their effects on the proliferation and migration of lung adenocarcinoma cells were examined. Whole-cell patch clamp experiments showed that these sesquiterpene lactones potently inhibited recombinant TMEM16A currents in a concentration-dependent manner. The half-maximal concentration (IC50) values for three tested sesquiterpene lactones were 29.9 ± 1.1 µM, 19.7 ± 0.4 µM, and 24.5 ± 2.1 µM, while the maximal effect (Emax) values were 100.0% ± 2.8%, 85.8% ± 0.9%, and 88.3% ± 4.6%, respectively. These sesquiterpene lactones also significantly inhibited the endogenous TMEM16A currents and proliferation, and migration of LA795 lung cancer cells. These results demonstrate that mecheliolide, costunolide and dehydrocostus lactone are novel TMEM16A inhibitors and potential candidates for lung adenocarcinoma therapy.

Simultaneous Determination of Plasticizers in Food Simulants Using GC/MS

  • Migration levels of plasticizers, di-n-butyl phthalate (DBP), benzyl-butyl phthalate (BBP), di-n-octyl phthalate(DNOP), di-iso-decyl phthalate (DIDP) and di-iso-nonyl phthalate (DINP), di-(2-ethylhexyl) adipate (DEHA), from 46 poly(vinyl chloride) (PVC) wrap films and 54 PVC gaskets into food simulants were determined using gas chromatography/mass spectrometry (GC/MS). The method was validated with limit of detection (LOD) of $0.01{\sim}0.02\;{\mu}g/mL$ for DBP, BBP, DNOP and DEHA, and $2\;{\mu}g/mL$ for DIDP and DINP. The linearity were found to be > 0.99 for all the compounds in concentration range of $0.1{\sim}81.4\;{\mu}g/mL$, and overall recoveries were ranged from 90.4 ~ 99.6%. DBP, BBP, DNOP, DEHA, DIDP and DINP were not detected in food simulants, except 1 wrap sample from which 0.28 and $0.99\;{\mu}g/mL$ of DEHA were detected respectively when tested with 20% ethanol and n-heptane as food simulants. These values were below the regulatory limitation in European Union (EU).

A Study on the Chloride Migration Properties of High Durable Marine Concrete Using the Expansion Production Admixture (팽창재를 혼입한 고내구성 해양콘크리트의 염화물 확산특성에 관한 연구)

  • Recently, high strength, flowability, and durability of concrete were required according to increase of large scale and high rise structure. However, cracks occurred easily on the high performance concrete. In this reason, using expansion agent for reducing shrinkage cracks were increased, but it did not consider on durability of high performance concrete. Accordingly, this study1 investigated the resistance of shrinkage and damage form salt by mixing CSA expansion agent on the blast-furnace slag cement and mixed cement for the low heat of hydration by three components. The cases that 8% of expansion agent was mixed and the proportion was OPC were expanded till 43.7 times compared with control concrete. For the resistance to the damage of salt, it was improved when mixing ratio was incresed and the maximum size of coarse aggregate growed bigger. In this study, the resistance to the damage of salt of the cases that 8% of expansion agent was mixed was improved about 16% compared with control concrete.

Electrophoretic Mobilities of the Potato Spindle Tuber Viroid RNA Molecules in the Urea-Gradient Gels (감자 걀쪽바이로드(PSTV) RNA 분자의 요소농도기울기겔에서 전기영동적 이동성에 관하여)

  • Low molecular weight plant ribonucleic acids including viroid-RNA molecules which are soluble in 2M lithium chloride were electrophoresed in the 0M to 8M urea-gradient polyacrylamide gel. Although the linear viroid-RNA molecules migrated at a similarrate across the urea-gradient gel under the denaturing temperature, the circular viroid-RNA molecules moved more rapidly at low urea-gradient region than at high urea-gradient region. Consequently, the migration of the circular viroid-RNA molecules showed a sudden shift across the band of linear forms in the midrange of the urea-gradient gels. Electrophoretic mobilities of the circular viroid-RNA molecules seemed to depend mainly on the concentration of urea in the denaturing urea-gradient gels.

Inhibition of MMP-2 and -9 by Crude Extracts and Their Solvent-partitioned Fractions from the Halophyte Atriplex gmelinii (가는갯능쟁이(Atriplex gmelinii) 추출물과 용매분획물의 MMP-2와 MMP-9 활성 저해효과)

  • In this study, the inhibitory effect of Atriplex gmelinii C. A. Mey. against the activity of MMP-2 and MMP-9 secreted from phorbol-12-myristate-13-acetate (PMA)-stimulated HT-1080 cells was evaluated by gelatin zymography and enzyme-linked immunosorbent assay (ELISA), reverse transcription polymerase-chain reaction (RT-PCR), and Western blot assay. Specimens of the halophyte A. gmelinii were extracted twice for 24 hr with methylene chloride ($CH_2Cl_2$), and then twice with methanol (MeOH), in turn. Each extract significantly inhibited the enzymatic activities in gelatin zymography and MMP ELISA kit, and expression of MMP-2 and 9 in mRNA and protein levels. Two crude extracts were combined and then the combined crude extracts were fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water ($H_2O$) fractions, according to solvent polarity. Among solvent-partitioned fractions, the 85% aq.MeOH fraction showed the strongest inhibitory effect against MMP-2 and -9 in gelatin zymography and MMP ELISA kit. In RT-PCR, all solvent-partitioned fractions significantly suppressed mRNA expression of MMP-2 and -9. On the other hand, in Western blot assay, all solvent-partitioned fractions except $H_2O$ significantly reduced expression levels of protein. HT 1080 cell migration was most significantly inhibited by the n-BuOH fraction followed by the 85% aq.MeOH and $H_2O$ fractions. These results suggest that A. gmelinii could be used as a potential source to inhibit tumor cell metastasis.
